Eight hundred and fifty sheep sera from 18 herds, located in 15 municipalities of the State of São Paulo, were examined by complement fixation test, indirect enzyme immunoassay and gel diffusion test for antibodies to Brucella ovis. The combination of the serological test results and the clinical and epidemiological data suggest that none of the examined animals was infected. The complement fixation and the gel diffusion tests showed good specificity, but the indirect enzyme immunoassay showed false positive results in nine of the sera.

The objective of the study was establishing the anatomical-structural differences between the skin of non-wool sheep of the Morada Nova breed and wool-on sheep of the Polwarth or Ideal breeds and their relations to the physical-mechanical characteristics of their leather after tanning. Ten animals of both breeds were used, with five animals of approximately one and four years of age of each breed. The animals were slaughtered, and immediately after skinning, samples were taken viewing the microtomy of the skin in the dorsal, lateral, ventral, hind and palette regions, parallel and perpendicular to the head-tail axle, and perpendicular to the grain of the skin. The skins were depilated, chromium-tanned and re-tanned. From the leather, three samples were taken from the two directions considered from the regions studied for physical-mechanical analysis, for tension and tear resistance and distension on the lastometer. The average of the results of the physical-mechanical analysis were compared by Tukey test at the levels of 1 and 5% probability. The breed, the age, the region and the position exerted a positive effect on the values of the resistance of the leather to tension for the Morada Nova four year old sheep, reaching a minimum of 200 kg/cm . The region and the position exerted a positive effect on the values of tear-resistence of the leather for both breeds studied, reaching a minimum of 40 kg/cm. The resistance of the leather on distension was superior to 8 mm, for all the animals studied, not influenced by breed, age, region or position. The skin of the Ideal sheep presented a thermostatic skin layer greater than the reticular layer with great folicular and glandular density, contrary to the skin of the Morada Nova sheep where the thermostatic and reticular layers are of approximately the same thickness, with less folicular and glandular density.

The influence of Toxoplasma gondii on semen variables and sperm morphology of sheep was evaluated in eight reproductive males distributed into three experimental groups: GI, three sheep inoculated with 2.0 × 105 of P strain oocytes; GII, three sheep infected with 1.0 × 106 of RH strain tachyzoites and; GIII two control sheep. Clinical (rectal temperature, cardiac and respiratory frequencies), parasite and serology exams (IIF) were realized. Sperm variables (volume, motility, vigor and concentration) and semen morphology for each sheep were also evaluated. Thus, semen and blood collections were assessed on post-inoculation days (PIDs)-1,3,5,7,11,14 and weekly thereafter up to PID 70. Clinical alterations were observed (hypothermia and anorexia) in infected sheep from groups GI and GII. Parasitic outbreaks were detected in five sheep. All the infected sheep produced antibodies against T. gondii from PID 5 onwards, reaching a peak of 4096 and 8192 for group GI and GII sheep, respectively. Differences (P < 0.05) were observed regarding the ejaculate volume between the inoculated groups (oocytes and tachyzoites) and control. Even though experimental toxoplasmic infection resulted in clinical symptomology in the inoculated sheep, the minimal alterations in sperm pathologies could not be directly attributed to T. gondii. © 2008 Elsevier B.V. The ovine brucellosis caused by Brucella ovis has tropism for reproductive tissues but until now the mechanism of bacterial persistence is not understood. Cytokine expression profiles were studied for 8 months in rams after being experimentally infected with the rough virulent strain of B. ovis (R- B. ovis) to study the pathogenesis of B. ovis and immune mechanism possibly associated to bacteria tropism and persistence. The messenger RNA (mRNA) expression levels of interleukin-1α (IL-1α), IL-1β, IL-6, IL-10, IL-12, interferon-γ (INF-γ) and tumour necrosis factor-α (TNF-α) cytokines were quantified by real-time quantitative RT-PCR (qRT-PCR) in reproductive tissues (epididymus, testicles, ampolae, vesicular glands and bulbourethral glands), and non-reproductive (liver, spleen and kidneys) tissues at 30, 60, 120 and 240 days post infection (dpi). During the acute phase of infection at 30. dpi, the host immune response was most notable demonstrating an up-regulation of several cytokines in reproductive tissues, including the epididymus (IL-6, IL-1β and IL-1α), testicles (INF-γ and IL-12), bulbourethral glands (IL-6 and TNF-α) and ampolae (INF-γ, IL-10, IL-1β and IL-1α). During the development of infection, cytokine gene expression levels decreased, providing evidence of immunosuppression and evidence of immune evasion that favoured persistence of chronic R- B. ovis infection. During the chronic phase of R- B. ovis infection (120 and 240. dpi), cytokine production was down-regulated in the epididymus (IL-1β and IL-1α), testicles (INF-γ and IL-12), and ampolae (INF-γ, IL-10, IL-1β and IL-1α), with the exception of the bulbourethral glands (IL-6 and TNF-α) and epididymus (IL-6); in these tissues, R- B. ovis infection resulted in up-regulation of the pro-inflammatory cytokine IL-6. Herein, we report cytokine expression profiles in tissues of rams experimentally infected with the rough strain of B. ovis, which are associated with bacterial persistence and macrophage activation. © 2012 Elsevier B.V.

B. ovis pathogenicity was evaluated in experimentally inoculated and naturally infected rams. Ten animals were submitted to simultaneous conjunctival and intrapreputial inoculation with 2x109 CFU/ mL of B. ovis REO 198. After that, animals underwent physical examination and blood samples were collected for serology every week. Positive serology results started to be observed in the 3rd week, with fluctuations in titers. Clinical changes began in the 5th week after inoculation and were associated with positive serology in the acute phase of the disease. Presence of B. ovis in semen and urine culture was intermittent. Three non-inoculated animals showed natural infection. B. ovis was shed twice in semen of one serology-negative animal. The study underscored the pathogenic characteristics of B. ovis REO 198 in Santa Inês rams, as well as the importance of animals as potential sources of infection.

Brucellosis remains an economic problem in animals and public health. Worldwide ovine brucellosis caused by Brucella ovis is considered a major cause of infertility in sheep. The factors responsible for persistence of the agent are not known, as well as the mechanisms involved in immune defense and possibly the persistence of the agent. Brucella spp. induces moderate inflammatory response. The nature of the intracellular agent stimulates immune response of the type 1 helper T lymphocytes. Studies of the pathogenesis of ovine brucellosis are scarce. Recent developments have shown that the inflammatory response induced by moderate brucelas represent probably the result of an attempt to escape the immune response and suppression of host immune response. Were reviewed by the mechanisms described by brucelas and Brucella ovis for penetration into the host, escape of the immune response and the immune response generated by the infection.
