Aluminium is omnipresent in everyday life and increased exposure is resulting in a burgeoning body burden of this non-essential metal. Personal care products are potential contributors to the body burden of aluminium and recent evidence has linked breast cancer with aluminium-based antiperspirants. We have used graphite furnace atomic absorption spectrometry (GFAAS) to measure the aluminium content in breast biopsies obtained following mastectomies. The aluminium content of breast tissue and breast tissue fat were in the range 4-437 nmol/g dry wt. and 3-192 nmol/g oil, respectively. The aluminium content of breast tissue in the outer regions (axilla and lateral) was significantly higher (P = 0.033) than the inner regions (middle and medial) of the breast. Whether differences in the regional distribution of aluminium in the breast are related to the known higher incidence of tumours in the outer upper quadrant of the breast remains to be ascertained. Background. The anaerobic spirochaete Brachyspira pilosicoli causes enteric disease in avian, porcine and human hosts, amongst others. To date, the only available genome sequence of B. pilosicoli is that of strain 95/1000, a porcine isolate. In the first intra-species genome comparison within the Brachyspira genus, we report the whole genome sequence of B. pilosicoli B2904, an avian isolate, the incomplete genome sequence of B. pilosicoli WesB, a human isolate, and the comparisons with B. pilosicoli 95/1000. We also draw on incomplete genome sequences from three other Brachyspira species. Finally we report the first application of the high-throughput Biolog phenotype screening tool on the B. pilosicoli strains for detailed comparisons between genotype and phenotype. Results. Feature and sequence genome comparisons revealed a high degree of similarity between the three B. pilosicoli strains, although the genomes of B2904 and WesB were larger than that of 95/1000 (~2,765, 2.890 and 2.596 Mb, respectively). Genome rearrangements were observed which correlated largely with the positions of mobile genetic elements. Through comparison of the B2904 and WesB genomes with the 95/1000 genome, features that we propose are non-essential due to their absence from 95/1000 include a peptidase, glycine reductase complex components and transposases. Novel bacteriophages were detected in the newly-sequenced genomes, which appeared to have involvement in intra- and inter-species horizontal gene transfer. Phenotypic differences predicted from genome analysis, such as the lack of genes for glucuronate catabolism in 95/1000, were confirmed by phenotyping. Conclusions. The availability of multiple B. pilosicoli genome sequences has allowed us to demonstrate the substantial genomic variation that exists between these strains, and provides an insight into genetic events that are shaping the species. In addition, phenotype screening allowed determination of how genotypic differences translated to phenotype. Further application of such comparisons will improve understanding of the metabolic capabilities of Brachyspira species.

The Salmonella enterica serovar Typhi CT18 (S. Typhi) chromosome harbours seven distinct prophage-like elements, some of which may encode functional bacteriophages. In silico analyses were used to investigate these regions in S. Typhi CT18, and ultimately compare these integrated bacteriophages against 40 other Salmonella isolates using DNA microarray technology. S. Typhi CT18 contains prophages that show similarity to the lambda, Mu, P2 and P4 bacteriophage families. When compared to other S. Typhi isolates, these elements were generally conserved, supporting a clonal origin of this serovar. However, distinct variation was detected within a broad range of Salmonella serovars; many of the prophage regions are predicted to be specific to S. Typhi. Some of the P2 family prophage analysed have the potential to carry non-essential "cargo" genes within the hyper-variable tail region, an observation that suggests that these bacteriophage may confer a level of specialisation on their host. Lysogenic bacteriophages therefore play a crucial role in the generation of genetic diversity within S. enterica. (C) 2004 Elsevier Ltd. All rights reserved.

Selenoproteins are characterized by the incorporation of at least one amino acid selenocysteine (Sec-U) encoded by in-frame UGA stop codons. These proteins, as well as the components of the Sec synthesis pathway, are present in members of the bacteria, archaea and eukaryote domains. Although not a ubiquitous pathway in all organisms, it was also identified in several protozoa, including the Kinetoplastida. Genetic evidence has indicated that the pathway is non-essential to the survival of Trypanosoma growing in non-stressed conditions. By analyzing the effects of RNA interference of the Trypanosoma brucei selenophosphate synthetase SPS2, we found a requirement under sub-optimal growth conditions. The present work shows that SPS2 is involved in oxidative stress protection of the parasite and its absence severely hampers the parasite survival in the presence of an oxidizing environment that results in an apoptotic-like phenotype and cell death. This work studied the immiscible blend of elastomeric poly(methyl methacrylate) (PMMA) with poly(ethylene terephthalate) (PET) bottle grade with and without the use of compatibilizer agent, poly(methyl methacrylate-co-glycidyl methacrylate - co-ethyl acrylate) (MGE). The characterizations of torque rheometry, melt flow index measurement (MFI), measuring the density and the degree of cristallinity by pycnometry, tensile testing, method of work essential fracture (EWF), scanning electron microscopy (SEM) and transmission electron microscopy (TEM) were performed in pure polymer and blends PMMA/PET. The rheological results showed evidence of signs of chemical reaction between the epoxy group MGE with the end groups of the PET chains and also to the elastomeric phase of PMMA. The increase in the concentration of PET reduced torque and adding MGE increased the torque of the blend of PMMA/PET. The results of the MFI also show that elastomeric PMMA showed lower flow and thus higher viscosity than PET. In the results of picnometry observed that increasing the percentage of PET resulted in an increase in density and degree crystallinity of the blends PMMA/PET. The tensile test showed that increasing the percentage of PET resulted in an increase in ultimate strength and elastic modulus and decrease in elongation at break. However, in the phase inversion, where the blend showed evidence of a co-continuous morphology and also, with 30% PET dispersed phase and compatibilized with 5% MGE, there were significant results elongation at break compared to elastomeric PMMA. The applicability of the method of essential work of fracture was shown to be possible for most formulations. And it was observed that with increasing elastomeric PMMA in the formulations of the blends there was an improvement in specific amounts of essential work of fracture (We) and a decrease in the values of specific non-essential work of fracture (βWp)

A reciclagem agrícola do lodo de esgoto tem provocado o acúmulo de metais pesados no solo e na água, podendo atingir níveis tóxicos e causar danos às plantas cultivadas, aos animais e ao homem, por meio da cadeia trófica. Neste intuito foi desenvolvido o presente experimento, em condições de campo, entre 2000 e 2002, onde foram avaliados os efeitos da aplicação de lodo de esgoto por dois anos, sobre a extração de metais de transição (essenciais e não) pelo extrator DTPA em um Latossolo Vermelho distrófico (LVd) de textura média. As concentrações dos elementos metálicos: Mn, Fe, Cd, Ni, Co, Pb e Cr não foram detectados pelo método da absorção atômica na solução obtida com o extrator DTPA. A aplicação de lodo de esgoto causou inicialmente pequena elevação no pH do solo, posteriormente a diminuição do mesmo, e manteve-se próximo ao original. Foi possível concluir que, com a aplicação consecutiva do lodo, os teores extraíveis de Fe e Mn nas amostras de solos aumentaram gradativamente nos dois anos agrícolas, com as doses do lodo de esgoto aplicado, época de amostragens, e foram superiores ao tratamento testemunha. O extrator apresentou capacidade restrita para avaliação da fitodisponibilidade dos metais pesados decorrentes das baixas concentrações nas amostras de solo.

Foram utilizadas poedeiras comerciais com 27 semanas de idade, distribuídas em delineamento inteiramente ao acaso, em esquema fatorial 3 x 3, com três repetições de seis aves por tratamento. Os fatores consistiram de três métodos de estimativa da composição de aminoácidos em ingredientes (tabelas brasileiras, equações de predição e fator para correção de aminoácido em função do teor de proteína do ingrediente) e três recomendações de aminoácidos, sendo duas de aminoácidos digestíveis e uma de aminoácidos totais. Os métodos de estimativa da composição de aminoácidos nos ingredientes afetaram apenas a conversão alimentar e a espessura de casca, que apresentaram os melhores resultados com a utilização das tabelas brasileiras. Embora as recomendações de aminoácidos tenham determinado diferenças em todos os parâmetros de desempenho, não afetaram a qualidade dos ovos. O desempenho das aves foi prejudicado pelos níveis de aminoácidos digestíveis, entretanto, ambas as recomendações promoveram desempenho semelhante e inferior ao de aminoácidos totais. O pior desempenho das aves alimentadas com as rações formuladas com aminoácidos digestíveis pode ser atribuído à deficiência em nitrogênio para a síntese de aminoácidos não-essenciais, visto que o nível protéico foi reduzido (12,5% PB), ou ainda à deficiência nos aminoácidos arginina, histidina, isoleucina, leucina e valina, cujos requerimentos mínimos não foram considerados na formulação das rações.

The identification of genes essential for survival is important for the understanding of the minimal requirements for cellular life and for drug design. As experimental studies with the purpose of building a catalog of essential genes for a given organism are time-consuming and laborious, a computational approach which could predict gene essentiality with high accuracy would be of great value. We present here a novel computational approach, called NTPGE (Network Topology-based Prediction of Gene Essentiality), that relies on the network topology features of a gene to estimate its essentiality. The first step of NTPGE is to construct the integrated molecular network for a given organism comprising protein physical, metabolic and transcriptional regulation interactions. The second step consists in training a decision-tree-based machine-learning algorithm on known essential and non-essential genes of the organism of interest, considering as learning attributes the network topology information for each of these genes. Finally, the decision-tree classifier generated is applied to the set of genes of this organism to estimate essentiality for each gene. We applied the NTPGE approach for discovering the essential genes in Escherichia coli and then assessed its performance. (C) 2007 Elsevier B.V. All rights reserved.

Cottonseed meals (CSM) with different protein levels (35, 39 and 40%) were evaluated to determine their chemical composition and measure apparent digestibility coefficients (ADC) of dry matter, protein, amino acids, energy, lipid and minerals for Nile tilapia. Apparent digestibility coefficients were determined using a reference purified diet containing 0.1% of chromic oxide indicator, and test diets contained 60% of reference diet and 40% of test ingredients. Results showed that apparent digestibility coefficients of cottonseed meals were different (p < 0.05). The apparent digestibility coefficients were: dry matter, 53.45-54.32%; crude protein, 71.56-74.81%; energy, 53.80-58.60%; lipid, 85.37-89.11%; phosphorus, 33.75-34.50%; calcium, 41.21-43.75%; iron, 62.02-79.85%; zinc, 0-67.41%; copper, 13.37-14.27%; essential amino acids, 71.39-79.17%; and non-essential amino acids, 77.08-82.58%. The ADC of protein did not reflect the majority of amino acids ADC showing variation in individual digestibility of amino acids and among cottonseed meals. Cottonseed meal can be used as protein succedaneum in Nile tilapia diets.

A presença de elementos não essenciais nas águas superficiais do rio Amazonas é uma preocupação global, o objetivo da pesquisa foi estudar a distribuição dos elementos As, Al, Mn, e Pb na água do estuário do rio Amazonas. As amostras foram coletadas em três regiões distintas: Canal Norte (AP), Canal Sul (PA) e rio Pará (PA) em três profundidades, com um total de 84 amostras. A espectrometria de emissão atômica com plasma indutivamente acoplado (ICPAES) foi utilizada para avaliar os teores de Al, Mn e Pb e a espectrofotometria de absorção atômica com geração de hidretos (HGAAS) foi usada para a análise do As. O As variou de <0,35 a 50,60 µg/L, o Al de <1,88 a 3347,70 µg/L, o Mn de 0,13 a 403,39 µg/L e Pb de <0,18 a 57,78 µg/L. Em média As (canal Sul), Al (todas as regiões), Mn (canal Norte e Sul) e Pb (canal Norte e Sul) se encontraram em valores acima do permitido pela legislação brasileira. Os elementos podem ter origem antrópica originária de atividade de mineração e industrial e origem natural proveniente de decomposição de rochas ígneas da bacia de drenagem do rio Amazonas que disponibilizam o elemento na forma ligada ao material particulado em suspensão.
