A fourth type of RTX determinant was identified in Actinobacillus pleuropneumoniae and was designated apxIVA. When expressed in Escherichia coli, recombinant ApxIVA showed a weak haemolytic activity and co-haemolytic synergy with the sphingomyelinase (beta-toxin) of Staphylococcus aureus. These activities required the presence of an additional gene, ORF1, that is located immediately upstream of apxIVA. The apxIVA gene product could not be detected in A. pleuropneumoniae cultures grown under various conditions in vitro; however, pigs experimentally infected with A. pleuropneumoniae serotypes 1, 5 and 7 started to produce antibodies that reacted with recombinant ApxIVA 14 d post-infection, indicating that apxIVA is expressed in vivo. In addition, sera from pigs naturally and experimentally infected with any of the serotypes all reacted with recombinant ApxIVA. The apxIVA gene from the serotype 1 A. pleuropneumoniae type strain Shope 4074T encodes a protein with a predicted molecular mass of 202 kDa which has typical features of RTX proteins including hydrophobic domains in the N-terminal half and 24 glycine-rich nonapeptides in the C-terminal half that bind Ca2+. The glycine-rich nonapeptides are arranged in a modular structure and there is some variability in the number of modules in the ApxIVA proteins of different serotypes of A. pleuropneumoniae. The deduced amino acid sequences of the ApxIVA proteins have significant similarity with the Neisseria meningitidis iron-regulated RTX proteins FrpA and FrpC, and to a much lesser extent with other RTX proteins. The apxIVA gene could be detected in all A. pleuropneumoniae serotypes and seems to be species-specific. Although the precise role of this new RTX determinant in pathogenesis of porcine pleuropneumonia needs to be determined, apxIVA is the first in vivo induced toxin gene that has been described in A. pleuropneumoniae.

This paper proposes the EvoBANE system. EvoBANE automatically generates Bayesian networks for solving special-purpose problems. EvoBANE evolves a population of individuals that codify Bayesian networks until it finds near optimal individual that solves a given classification problem. EvoBANE has the flexibility to modify the constraints that condition the solution search space, self-adapting to the specifications of the problem to be solved. The system extends the GGEAS architecture. GGEAS is a general-purpose grammar-guided evolutionary automatic system, whose modular structure favors its application to the automatic construction of intelligent systems. EvoBANE has been applied to two classification benchmark datasets belonging to different application domains, and statistically compared with a genetic algorithm performing the same tasks. Results show that the proposed system performed better, as it manages different complexity constraints in order to find the simplest solution that best solves every problem.

We propose a new methodology to evaluate the balance between segregation and integration in functional brain networks by using singular value decomposition techniques. By means of magnetoencephalography, we obtain the brain activity of a control group of 19 individuals during a memory task. Next, we project the node-to-node correlations into a complex network that is analyzed from the perspective of its modular structure encoded in the contribution matrix. In this way, we are able to study the role that nodes play I/O its community and to identify connector and local hubs. At the mesoscale level, the analysis of the contribution matrix allows us to measure the degree of overlapping between communities and quantify how far the functional networks are from the configuration that better balances the integrated and segregated activity

Poly(ADP)-ribose polymerase (PADPRP) has been purified to apparent homogeneity from suspension cultures of the maize (Zea mays) callus line. The purified enzyme is a single polypeptide of approximately 115 kD, which appears to dimerize through an S-S linkage. The catalytic properties of the maize enzyme are very similar to those of its animal counterpart. The amino acid sequences of three tryptic peptides were obtained by microsequencing. Antibodies raised against peptides from maize PADPRP cross-reacted specifically with the maize enzyme but not with the enzyme from human cells, and vice versa. We have also characterized a 3.45-kb expressed-sequence-tag clone that contains a full-length cDNA for maize PADPRP. An open reading frame of 2943 bp within this clone encodes a protein of 980 amino acids. The deduced amino acid sequence of the maize PADPRP shows 40% to 42% identity and about 50% similarity to the known vertebrate PADPRP sequences. All important features of the modular structure of the PADPRP molecule, such as two zinc fingers, a putative nuclear localization signal, the automodification domain, and the NAD+-binding domain, are conserved in the maize enzyme. Northern-blot analysis indicated that the cDNA probe hybridizes to a message of about 4 kb.

A long-term goal in the field of restriction-modification enzymes has been to generate restriction endonucleases with novel sequence specificities by mutating or engineering existing enzymes. This will avoid the increasingly arduous task of extensive screening of bacteria and other microorganisms for new enzymes. Here, we report the deliberate creation of novel site-specific endonucleases by linking two different zinc finger proteins to the cleavage domain of Fok I endonuclease. Both fusion proteins are active and under optimal conditions cleave DNA in a sequence-specific manner. Thus, the modular structure of Fok I endonuclease and the zinc finger motifs makes it possible to create "artificial" nucleases that will cut DNA near a predetermined site. This opens the way to generate many new enzymes with tailor-made sequence specificities desirable for various applications.

Beyond the inherent technical challenges, current research into the three dimensional surface correspondence problem is hampered by a lack of uniform terminology, an abundance of application specific algorithms, and the absence of a consistent model for comparing existing approaches and developing new ones. This paper addresses these challenges by presenting a framework for analysing, comparing, developing, and implementing surface correspondence algorithms. The framework uses five distinct stages to establish correspondence between surfaces. It is general, encompassing a wide variety of existing techniques, and flexible, facilitating the synthesis of new correspondence algorithms. This paper presents a review of existing surface correspondence algorithms, and shows how they fit into the correspondence framework. It also shows how the framework can be used to analyse and compare existing algorithms and develop new algorithms using the framework's modular structure. Six algorithms, four existing and two new, are implemented using the framework. Each implemented algorithm is used to match a number of surface pairs. Results demonstrate that the correspondence framework implementations are faithful implementations of existing algorithms, and that powerful new surface correspondence algorithms can be created. The topic of bioenergy, biofuels and bioproducts remains at the top of the current political and research agenda. Identification of the optimum processing routes for biomass, in terms of efficiency, cost, environment and socio-economics is vital as concern grows over the remaining fossil fuel resources, climate change and energy security. It is known that the only renewable way of producing conventional hydrocarbon fuels and organic chemicals is from biomass, but the problem remains of identifying the best product mix and the most efficient way of processing biomass to products. The aim is to move Europe towards a biobased economy and it is widely accepted that biorefineries are key to this development. A methodology was required for the generation and evaluation of biorefinery process chains for converting biomass into one or more valuable products that properly considers performance, cost, environment, socio-economics and other factors that influence the commercial viability of a process. In this thesis a methodology to achieve this objective is described. The completed methodology includes process chain generation, process modelling and subsequent analysis and comparison of results in order to evaluate alternative process routes. A modular structure was chosen to allow greater flexibility and allowing the user to generate a large number of different biorefinery configurations The significance of the approach is that the methodology is defined and is thus rigorous and consistent and may be readily re-examined if circumstances change. There was the requirement for consistency in structure and use, particularly for multiple analyses. It was important that analyses could be quickly and easily carried out to consider, for example, different scales, configurations and product portfolios and so that previous outcomes could be readily reconsidered. The result of the completed methodology is the identification of the most promising biorefinery chains from those considered as part of the European Biosynergy Project.

A main unsolved problem in the RNA World scenario for the origin of life is how a template-dependent RNA polymerase ribozyme emerged from short RNA oligomers obtained by random polymerization on mineral surfaces. A number of computational studies have shown that the structural repertoire yielded by that process is dominated by topologically simple structures, notably hairpin-like ones. A fraction of these could display RNA ligase activity and catalyze the assembly of larger, eventually functional RNA molecules retaining their previous modular structure: molecular complexity increases but template replication is absent. This allows us to build up a stepwise model of ligation- based, modular evolution that could pave the way to the emergence of a ribozyme with RNA replicase activity, step at which information-driven Darwinian evolution would be triggered. Im vorliegenden Beitrag wird ein Modell für die Beschreibung und Berechnung der Geometrie manuell bedienter Lagersysteme entwickelt und validiert, welches die Grundlage für eine rechnergestützte Planungssystematik bildet. Der modulare Aufbau des Modells ermöglicht durch entsprechende Erweiterungen die Berechnung des Lagerplatzbedarfs unter Berücksichtigung von Lagerstrategien. Auch die Betrachtung von Speziallösungen wie der Verwendung unterschiedlich hoher Lagerplätze ist möglich. Abschließend wird das vorgestellte Modell auf seine Anwendbarkeit zur systematischen Erzeugung verschiedener Layouts als Lösungsalternativen untersucht.

[ES]En este artículo se presenta una experiencia de innovación que se ha articulado, en el seno de un diseño curricular a través de módulos, al objeto de mejorar los procesos de enseñanza-aprendizaje que se están desarrollando en la Facultad de Fª y Ciencias de la Educación de la UPV/EHU. El marco de innovación aprovecha la oportunidad surgida con la configuración de los nuevos planes de estudio para su convergencia en el Espacio Europeo de Educación Superior. El contexto más amplio en el que se enmarca esta experiencia, es el de los cambios organizativos, curriculares y culturales, que está suponiendo la realización de un diseño modular de las nuevas titulaciones de grado (Pedagogía y Educación Social), trabajando en equipos docentes coordinados y contando con el apoyo institucional de la UPV/EHU. La innovación concreta que se presenta, describe minuciosamente uno de los siete módulos que componen la nueva titulación del Grado en Educación Social, y subraya la labor desplegada por el equipo docente que imparte las cinco materias que conforman el módulo en las dos lenguas oficiales: euskara y castellano, al planificar, desarrollar y evaluar la actividad interdisciplinar que les compete de manera coordinada. En este trabajo se reflexiona respecto a las posibilidades que la estructura modular ofrece para acometer un cambio real en la cultura de la práctica docente, y a cómo gestionar factores clave que garanticen el tránsito a un modelo docente activo, participativo y colaborativo. // This paper presents an innovative approach to improving teacher quality that is being developed at the Faculty of Philosophy and Educational Sciences of the UPV / EHU, taking the opportunity arose with the configuration of the new curricula for their convergence in the European Higher Education Area. It is framed in the broader context of organizational change, curriculum and cultural, that is assuming the implementation of a modular design of the new degree (Education and Social Education), working in teams coordinated and with the institutional support of the UPV. The concrete innovation is presented, describes one of the seven modules of the new degree of Social Education, and underlines the work done by the faculty team who teach the five subjects that make up the module in the two official languages: Basque and Spanish, because they must plan, develop and evaluate the interdisciplinary activity in a coordinated manner. We reflect in this paper on the potential that the modular structure offers to undertake real change in the culture of teaching practice, and how to manage key factors to ensure the transition to a teaching model active, participatory and collaborative.

Two seven-residue helical segments, Val-Ala-Leu-Aib-Val-Ala-Leu, were linked synthetically with an epsilon-aminocaproic acid (Acp) linker with the intention of making a stable antiparallel helix-helix motif. The crystal structure of the linked peptide Boc-Val-Ala-Leu-Aib-Val-Ala-Leu-Acp-Val-Ala-Leu-Aib-Val-Ala-Leu-OMe (1) shows the two helices displaced laterally from each other by the linker, but the linker has not folded the molecule into a close-packed antiparallel conformation. Two strong intermolecular NH...O = C hydrogen bonds are formed between the top of the lower helix of one molecule and the bottom of the upper helix in a laterally adjacent molecule to give the appearance of an extended single helix. The composite peptide with Boc and OMe end groups, C76H137N15O18.H2O, crystallize in space group P2(1) with a = 8.802 (1) angstrom, b = 20.409 (4) angstrom, c = 26.315 (3) angstrom, and beta = 90.72 (1)degrees; overall agreement R = 7.86% for 5030 observed reflections (\F(o)\ > 3-sigma(F)); resolution = 0.93 angstrom. Limited evidence for a more compact conformation in solution consistent with an antiparallel helix arrangement is obtained by comparison of the HPLC retention times and CD spectra of peptide 1 with well-characterized continuous helices of similar length and sequence.

Elucidating the intricate relationship between brain structure and function, both in healthy and pathological conditions, is a key challenge for modern neuroscience. Recent progress in neuroimaging has helped advance our understanding of this important issue, with diffusion images providing information about structural connectivity (SC) and functional magnetic resonance imaging shedding light on resting state functional connectivity (rsFC). Here, we adopt a systems approach, relying on modular hierarchical clustering, to study together SC and rsFC datasets gathered independently from healthy human subjects. Our novel approach allows us to find a common skeleton shared by structure and function from which a new, optimal, brain partition can be extracted. We describe the emerging common structure-function modules (SFMs) in detail and compare them with commonly employed anatomical or functional parcellations. Our results underline the strong correspondence between brain structure and resting-state dynamics as well as the emerging coherent organization of the human brain.

Large loads result in expensive foundations which are a substantial proportion of the capital cost of flap-type Wave Energy Converters (WECs). Devices such as Oyster 800, currently deployed at the European Marine Energy Centre (EMEC), comprise a single flap for the full width of the machine. Splitting a flap-type device into smaller vertical flap modules, to make a ‘modular-flap’, might reduce the total foundation loads, whilst still providing acceptable performance in terms of energy conversion.
This paper investigates the foundation loads of an undamped modular-flap device, comparing them to those for a rigid flap of an equivalent width. Physical modelling in a wave tank is used, with loads recorded using a six degree of freedom (DoF) load cell. Both fatigue and extreme loading analysis was conducted. The rotations of the flaps were also recorded, using a motion-tracking system.
