Michael Reactions of the Anions generated by the Metal-Ammonia Reduction of Benzoic Acids.

A novel method for the construction of carboncarbon bonds is described in which anions obtained by the metal-ammonia reduction of benzoic acid and its derivatives undergo ready Michael reaction with methyl crotonate to give the addition products.

Meta-analysis identifies 13 new loci associated with waist-hip ratio and reveals sexual dimorphism in the genetic basis of fat distribution

Waist-hip ratio (WHR) is a measure of body fat distribution and a predictor of metabolic consequences independent of overall adiposity. WHR is heritable, but few genetic variants influencing this trait have been identified. We conducted a meta-analysis of 32 genome-wide association studies for WHR adjusted for body mass index (comprising up to 77,167 participants), following up 16 loci in an additional 29 studies (comprising up to 113,636 subjects). We identified 13 new loci in or near RSPO3, VEGFA, TBX15-WARS2, NFE2L3, GRB14, DNM3-PIGC, ITPR2-SSPN, LY86, HOXC13, ADAMTS9, ZNRF3-KREMEN1, NISCH-STAB1 and CPEB4 (P = 1.9 × 10−9 to P = 1.8 × 10−40) and the known signal at LYPLAL1. Seven of these loci exhibited marked sexual dimorphism, all with a stronger effect on WHR in women than men (P for sex difference = 1.9 × 10−3 to P = 1.2 × 10−13). These findings provide evidence for multiple loci that modulate body fat distribution independent of overall adiposity and reveal strong gene-by-sex interactions.

This study presents a systematical analysis of biochemist Michael Behe's thinking. Behe is a prominent defender of the Intelligent Design Movement which has gaines influence particularly in the United States, but also in elsewhere. At the core of his thinking is the idea of intelligent design, according to which the order of the cosmos and of living things is the handiwork of a non-human intelligence. This "design argument" had previously been popular in the tradition of natural theology. Behe attempts to base his argument on the findings of 20th century biology, however. It has been revealed by biochemistry that cells, formerly thought to be simple, in fact contain complex structures, for instance the bacterial flagellum, which are reminiscent of the machines built by humans. According to Behe these can be believably explained only by referring to intelligent design, not by invoking darwinian natural laws. My analysis aims to understand Behe's thought on intelligent design, to bring forward its connections to intellectual history and worldviews, and to study whether Behe has formulated his argument so as to avoid common criticisms directed against design arguments. I use a large amount literature and refer to diverse writers participating in the intelligent design debate. The results of the analysis are as follows. Behe manages to avoid a large amount of classical criticisms against the design argument, and new criticisms have to be developed to meet his argument. Genome-wide association study of prostate cancer-specific survival

BACKGROUND: Unnecessary intervention and overtreatment of indolent disease are common challenges in clinical management of prostate cancer. Improved tools to distinguish lethal from indolent disease are critical. METHODS: We performed a genome-wide survival analysis of cause-specific death in 24,023 prostate cancer patients (3,513 disease-specific deaths) from the PRACTICAL and BPC3 consortia. Top findings were assessed for replication in a Norwegian cohort (CONOR). RESULTS: We observed no significant association between genetic variants and prostate cancer survival. CONCLUSIONS: Common genetic variants with large impact on prostate cancer survival were not observed in this study. IMPACT: Future studies should be designed for identification of rare variants with large effect sizes or common variants with small effect sizes.

A new strategy for the total synthesis of (±)-seychellene which involves (i) a regiospecific construction of a bicyclo(2.2.2)octene moiety having a methyl group at the bridgehead and (ii) a vinyl radical induced intramolecular Michael addition is described.

Tandem Michael reaction. Synthesis of bridged diketones

Addition of NaOMe, NaOEt, or NaOPr(i) to dispironaphthalenone 1 resulted in the formation of diketones 4a-c and 5a-c. The structure assigned to 4a was confirmed by conversion to the known hemiacetal 3. Similar addition of carbon nucleophiles like diethyl malonate, dimethyl malonate, methyl cyanoacetate, and ethyl cyanoacetate afforded diketones 4d-g. Formation of these compounds has been rationalized.

Potassamide induced in situ alkylation of 5,6-dihydroisoquinoline with allyl bromide and Michael acceptors

Potassamide induced in situ alkylation of 4-cyano-3-methoxy-1-methyl-5, 6-dihydroisoquinoline (1a) with allyl bromide gives the 5-allyl- and 5,9-diallyl-5,6-dihydroisoquinolines (1c and 1d), isoquinoline derivative 2 and 4-allyl-1, 2, 3, 4-tetrahydroisoquinolin-3(2H)-one (3a). However, potassamide induced in situ alkylation of In with buten-2-one, mesityl oxide and acrylonitrile results in the formation of only 5-alkylated 5,6-dihydroisoquinoline derivatives 1e-g along with fully aromatised compound 2.
