The present Thesis studies three alternative solvent groups as sustainable replacement of traditional organic solvents. Some aspects of fluorinated solvents, supercritical fluids and ionic liquids, have been analysed with a critical approach and their effective “greenness” has been evaluated from the points of view of the synthesis, the properties and the applications. In particular, the attention has been put on the environmental and human health issues, evaluating the eco-toxicity, the toxicity and the persistence, to underline that applicability and sustainability are subjects with equal importance. The “green” features of fluorous solvents and supercritical fluids are almost well-established; in particular supercritical carbon dioxide (scCO2) is probably the “greenest” solvent among the alternative solvent systems developed in the last years, enabling to combine numerous advantages both from the point of view of industrial/technological applications and eco-compatibility. In the Thesis the analysis of these two classes of alternative solvents has been mainly focused on their applicability, rather than the evaluation of their environmental impact. Specifically they have been evaluated as alternative media for non-aqueous biocatalysis. For this purpose, the hydrophobic ion pairing (HIP), which allows solubilising enzymes in apolar solvents by an ion pairing between the protein and a surfactant, has been investigated as effective enzymatic derivatisation technique to improve the catalytic activity under homogeneous conditions in non conventional media. The results showed that the complex enzyme-surfactant was much more active both in fluorous solvents and in supercritical carbon dioxide than the native form of the enzyme. Ionic liquids, especially imidazolium salts, have been proposed some years ago as “fully green” alternative solvents; however this epithet does not take into account several “brown” aspects such as their synthesis from petro-chemical starting materials, their considerable eco-toxicity, toxicity and resistance to biodegradation, and the difficulty of clearly outline applications in which ionic liquids are really more advantageous than traditional solvents. For all of these reasons in this Thesis a critical analysis of ionic liquids has been focused on three main topics: i) alternative synthesis by introducing structural moieties which could reduce the toxicity of the most known liquid salts, and by using starting materials from renewable resources; ii) on the evaluation of their environmental impact through eco-toxicological tests (Daphnia magna and Vibrio fischeri acute toxicity tests, and algal growth inhibition), toxicity tests (MTT test, AChE inhibition and LDH release tests) and fate and rate of aerobic biodegradation in soil and water; iii) and on the demonstration of their effectiveness as reaction media in organo-catalysis and as extractive solvents in the recovery of vegetable oil from terrestrial and aquatic biomass. The results about eco-toxicity tests with Daphnia magna, Vibrio fischeri and algae, and toxicity assay using cultured cell lines, clearly indicate that the difference in toxicity between alkyl and oxygenated cations relies in differences of polarity, according to the general trend of decreasing toxicity by decreasing the lipophilicity. Independently by the biological approach in fact, all the results are in agreement, showing a lower toxicity for compounds with oxygenated lateral chains than for those having purely alkyl lateral chains. These findings indicate that an appropriate choice of cation and anion structures is important not only to design the IL with improved and suitable chemico-physical properties but also to obtain safer and eco-friendly ILs. Moreover there is a clear indication that the composition of the abiotic environment has to be taken into account when the toxicity of ILs in various biological test systems is analysed, because, for example, the data reported in the Thesis indicate a significant influence of salinity variations on algal toxicity. Aerobic biodegradation of four imidazolium ionic liquids, two alkylated and two oxygenated, in soil was evaluated for the first time. Alkyl ionic liquids were shown to be biodegradable over the 6 months test period, and in contrast no significant mineralisation was observed with oxygenated derivatives. A different result was observed in the aerobic biodegradation of alkylated and oxygenated pyridinium ionic liquids in water because all the ionic liquids were almost completely degraded after 10 days, independently by the number of oxygen in the lateral chain of the cation. The synthesis of new ionic liquids by using renewable feedstock as starting materials, has been developed through the synthesis of furan-based ion pairs from furfural. The new ammonium salts were synthesised in very good yields, good purity of the products and wide versatility, combining low melting points with high decomposition temperatures and reduced viscosities. Regarding the possible applications as surfactants and biocides, furan-based salts could be a valuable alternative to benzyltributylammonium salts and benzalkonium chloride that are produced from non-renewable resources. A new procedure for the allylation of ketones and aldehydes with tetraallyltin in ionic liquids was developed. The reaction afforded high yields both in sulfonate-containing ILs and in ILs without sulfonate upon addition of a small amount of sulfonic acid. The checked reaction resulted in peculiar chemoselectivity favouring aliphatic substrates towards aromatic ketones and good stereoselectivity in the allylation of levoglucosenone. Finally ILs-based systems could be easily and successfully recycled, making the described procedure environmentally benign. The potential role of switchable polarity solvents as a green technology for the extraction of vegetable oil from terrestrial and aquatic biomass has been investigated. The extraction efficiency of terrestrial biomass rich in triacylglycerols, as soy bean flakes and sunflower seeds, was comparable to those of traditional organic solvents, being the yield of vegetable oils recovery very similar. Switchable polarity solvents as been also exploited for the first time in the extraction of hydrocarbons from the microalga Botryococcus braunii, demonstrating the efficiency of the process for the extraction of both dried microalgal biomass and directly of the aqueous growth medium. The switchable polarity solvents exhibited better extraction efficiency than conventional solvents, both with dried and liquid samples. This is an important issue considering that the harvest and the dewatering of algal biomass have a large impact on overall costs and energy balance.

The scope of my research project is to produce and characterize new crystalline forms of organic compounds, focusing the attention on co-crystals and then transferring these notions on APIs to produce co-crystals of potential interest in the pharmaceutical field. In the first part of this work co-crystallization experiments were performed using as building blocks the family of aliphatic dicarboxylic acids HOOC-(CH2)n-COOH, with n= 2-8. This class of compounds has always been an object of study because it is characterized by an interesting phenomenon of alternation of melting points: the acids with an even number of carbon atoms show a melting point higher than those with an odd one. The acids were co-crystallized with four dipyridyl molecules (formed by two pyridine rings with a different number of bridging carbon atoms) through the formation of intermolecular interactions N•••(H)O. The bases used were: 4,4’-bipyridine (BPY), 1,2-bis(4-pyridyl)ethane (BPA), 1,2-(di-4-pyridyl)ethylene (BPE) and 1,2-bis(4-pyridyl)propane (BPP). The co-crystals obtained by solution synthesis were characterized by different solid-state techniques to determine the structure and to see how the melting points in co-crystals change. In the second part of this study we tried to obtain new crystal forms of compounds of pharmaceutical interest. The APIs studied are: O-desmethylvenlafaxine, Lidocaine, Nalidixic Acid and Sulfadiazine. Each API was subjected to Polymorph Screening and Salt/Co-crystal Screening experiments to identify new crystal forms characterized by different properties. In a typical Salt/Co-crystal Screening the sample was made to react with a co-former (solid or liquid) through different methods: crystallization by solution, grinding, kneading and solid-gas reactions. The new crystal forms obtained were characterized by different solid state techniques (X-ray single crystal diffraction, X-ray powder diffraction, Differential Scanning Calorimetry, Thermogravimetric Analysis, Evolved gas analysis, FT-IR – ATR, Solid State N.M.R).

Hämocyanine sind große, kupferhaltige Sauerstoff-Transportproteine, die bei zahlreichen Schnecken extrazellulär in der Hämolymphe vorkommen. Das Keyhole Limpet-Hämocyanin (KLH) der Schlüssellochschnecke Megathura crenulata dient aufgrund seiner immunstimu-latorischen Eigenschaften seit vielen Jahren als Modellprotein in der Immunologie. In der Klinik wird es als Hapten- und Vakzincarrier sowie als Medikament gegen oberflächliche Harnblasenkarnzinome eingesetzt. Die Quartärstruktur des KLH besteht aus einem Hohl-zylinder mit einer Molekülmasse von 8 MDa und einem Durchmesser von 35 nm. Dieses sogenannte Didekamer setzt sich aus 20 Untereinheiten mit jeweils 400 kDa zusammen. Jede Untereinheit lässt sich weiter in acht funktionelle Einheiten a bis h (engl. Functional Units = FU) mit ~ 50 kDa unterteilen. Die FUs a bis f bilden die Wandregion des Moleküls, während der Kragen aus den FUs g und h geformt wird. Die Struktur der Wandregion sowie der FU-g konnte bisher bereits durch Röntgenstrukturanalysen aufgeklärt werden. Bezüglich der Struktur der FU-h, die sich durch eine spezielle C-terminale Verlängerung von ~ 100 Amino-säuren auszeichnet, sind allerdings noch keine Informationen verfügbar. Um die Architektur des Kragens zu verstehen, wurden im Rahmen dieser Arbeit zunächst Strategien entwickelt, diese spezielle FU in großer Menge und Reinheit zu isolieren. Anschließend konnten Bedingungen gefunden werden, die zur Ausbildung 0,2 mm großer, hexagonaler Kristalle führten. Diese ergaben am Synchrotron eine Auflösung von 4 Å. Durch Auswertung der Röntgenstrukturdaten konnte für die C-terminale Zusatzdomäne der FU-h eine Cupredoxin-ähnliche Typ I-Kupferfaltung ermittelt werden. Der Nachweis eines zusätzlichen Kupfer-atoms innerhalb dieser Domäne bedarf allerdings einer höheren Auflösung der Kristall-struktur. Hämocyanine lassen sich aufgrund ihrer evolutionären Verwandtschaft zu Phenol-oxidasen mit Hilfe verschiedener in vitro-Aktivatoren zur Catecholoxidase und teilweise auch zur Tyrosinase aktivieren. Beim KLH konnte in dieser Arbeit eine eindeutige Diphenolase- und sogar eine schwache Monophenolase-Aktivität der FUs-a und -f nach SDS-Aktivierung nachgewiesen werden. Zudem konnte eine geringfügige intrinsische Diphenolase-Aktivität dieser FUs belegt werden. Die enzymatischen Reaktionen waren sowohl von der gewählten Puffersubstanz, als auch der Anwesenheit bivalenter Kationen abhängig. Tris wirkt vermutlich als allosterischer Effektor und steigerte den Substrat-Umsatz, während Mg2+-Ionen zu einer starken Inhibition der katalytischen Aktivität führten. Die Klärung einer möglichen physiologischen Funktion der Phenoloxidase-Aktivität des KLH sowie potenziellen in vivo-Aktivatoren steht noch aus. Studien zur thermischen Stabilität des KLH resultierten in einer irreversiblen Denaturierung des Proteins. Die Schmelzpunkte deuteten auf eine hohe Tempe-raturstabilität des KLH, vor allem in Anwesenheit bivalenter Kationen. Eine Hämocyanin-typische Abhängigkeit der Hitzeresistenz vom Oligomerisierungsgrad ließ sich nicht feststellen, da sowohl bei der FU-h als auch den KLH-Didekameren eine vergleichbar hohe thermische Stabilität, bei einer nach wie vor vorhandenen Oxygenierung beobachtet wurde.

Monoanionische, stickstoffhaltige Chelatliganden sind sehr populär und werden oft als Baueinheiten zur Synthese von Metallkomplexen eingesetzt. Im Gegensatz zu den bekannten Amidinat- und β-Diketiminatliganden wurde den Triazeniden bisher weniger Aufmerksamkeit geschenkt. Dies liegt hauptsächlich daran, dass wenige, sterisch anspruchsvolle Vertreter dieser Gruppe bekannt waren. Mit der Synthese neuer biphenyl- und terphenylsubstituierter Triazenide sind diese stärker in den Fokus der Forschung gerückt. Durch die Sperrigkeit der Liganden werden unerwünschte Ligandenumverteilungsreaktionen unterdrückt und der Weg zu neuen Metallkomplexen mit bemerkenswerten Eigenschaften geebnet. Diese Arbeit beschäftigt sich mit Triazenidokomplexen der Alkalimetalle, des Magnesiums, des Zinks und des Aluminiums. Zusätzlich zu den monofunktionellen wurden bifunktionelle Triazene, welche aus einer Biphenylgruppe aufgebaut sind und zwei terphenylsubstituierte Triazengruppen tragen, synthetisiert und deren Funktion als Komplexliganden untersucht. Durch das hohe Volumen der Liganden konnten Metallkomplexe hergestellt werden, die bemerkenswerte strukturelle Eigenschaften aufweisen. So bilden beispielsweise alle dargestellten Verbindungen Kristalle, die aus monomeren Baueinheiten aufgebaut sind. Eine homoleptische Magnesiumverbindung mit zwei Triazenidliganden weist eine ungewöhnliche, planare Koordinationssphäre auf, eine Triazenidodiphenylaluminiumverbindung zeigt eine interessante Ausrichtung der Phenylgruppen, die auf π-Stapeleffekte zurückgeführt wird und die Alkalimetalltriazenide weisen sekundäre Metall-π-Aren-Wechselwirkungen auf, womit eine bessere koordinative Absättigung des Koordinationszentrums erreicht wird. Zusätzlich zur strukturellen Charakterisierung mittels Röntgendiffraktometrie wurden, wo es möglich war 1H und 13C NMR-Messungen durchgeführt, Schmelzpunkte bestimmt sowie IR- und UV-VIS-Spektren aufgenommen. Durch die hohe Dichte an aromatischen Systemen innerhalb der Verbindungen, ist es bei einer dreikernigen Zinktriazenidverbindung gelungen mittels 1H NMR-Messungen die starken Einflüsse des Ringstroms nachzuweisen. Außerdem wurden DFT-Berechnungen durchgeführt, um herauszufinden, wie sehr die räumliche Ausrichtung der Bi- bzw. Terphenylsubstituenten gegenüber der N3-Baugruppe der Triazenide die Verteilung der negativen Ladung innerhalb eines monoanionischen Triazenids und somit dessen Koordinationseigenschaften beeinflusst. Diese Effekte konnten durch experimentelle Ergebnisse unterstützt werden.

All of the metals have definite melting points. When a metal is heated above the melting point, it exists as a liquid. Now if the melt is allowed to cool, it will solidify when a temperature corresponding to the melting point is reached. However, if one metal is added to another metal, both of which are mutually soluble in the liquid state, a certain effect can be noted. The melt will not solid­ify when the melting point of the pure metal is reached, but will freeze at a lower temperature.

Los adhesivos se conocen y han sido utilizados en multitud de aplicaciones a lo lago de la historia. En la actualidad, la tecnología de la adhesión como método de unión de materiales estructurales está en pleno crecimiento. Los avances científicos han permitido comprender mejor los fenómenos de adhesión, así como, mejorar y desarrollar nuevas formulaciones poliméricas que incrementan el rango de aplicaciones de los adhesivos. Por otro lado, el desarrollo de nuevos materiales y la necesidad de aligerar peso, especialmente en el sector transporte, hace que las uniones adhesivas se introduzcan en aplicaciones hasta ahora reservadas a otros sistemas de unión como la soldadura o las uniones mecánicas, ofreciendo rendimientos similares y, en ocasiones, superiores a los aportados por estas. Las uniones adhesivas ofrecen numerosas ventajas frente a otros sistemas de unión. En la industria aeronáutica y en automoción, las uniones adhesivas logran una reducción en el número de componentes (tales como los tornillos, remaches, abrazaderas) consiguiendo como consecuencia diseños más ligeros y una disminución de los costes de manipulación y almacenamiento, así como una aceleración de los procesos de ensamblaje, y como consecuencia, un aumento de los procesos de producción. En el sector de la construcción y en la fabricación de equipos industriales, se busca la capacidad para soportar la expansión y contracción térmica. Por lo tanto, se usan las uniones adhesivas para evitar producir la distorsión del sustrato al no ser necesario el calentamiento ni la deformación de las piezas cuando se someten a un calentamiento elevado y muy localizado, como en el caso de la soldadura, o cuando se someten a esfuerzos mecánicos localizados, en el caso de montajes remachados. En la industria naval, se están desarrollando técnicas de reparación basadas en la unión adhesiva para distribuir de forma más uniforme y homogénea las tensiones con el objetivo de mejorar el comportamiento frente a fatiga y evitar los problemas asociados a las técnicas de reparación habituales de corte y soldadura. Las uniones adhesivas al no requerir importantes aportes de calor como la soldadura, no producen modificaciones microestructurales indeseables como sucede en la zona fundida o en la zona afectada térmicamente de las uniones soldadas, ni deteriora los recubrimientos protectores de metales de bajo punto de fusión o de naturaleza orgánica. Sin embargo, las uniones adhesivas presentan una desventaja que dificulta su aplicación, se trata de su durabilidad a largo plazo. La primera causa de rotura de los materiales es la rotura por fatiga. Este proceso de fallo es la causa del 85% de las roturas de los materiales estructurales en servicio. La rotura por fatiga se produce cuando se somete al material a la acción de cargas que varían cíclicamente o a vibraciones durante un tiempo prolongado. Las uniones y estructuras sometidas a fatiga pueden fallar a niveles de carga por debajo del límite de resistencia estática del material. La rotura por fatiga en las uniones adhesivas no se produce por un proceso de iniciación y propagación de grieta de forma estable, el proceso de fatiga va debilitando poco a poco la unión hasta que llega un momento que provoca una rotura de forma rápida. Underhill explica este mecanismo como un proceso de daño irreversible de los enlaces más débiles en determinados puntos de la unión. Cuando se ha producido el deterioro de estas zonas más débiles, su área se va incrementando hasta que llega un momento en que la zona dañada es tan amplia que se produce el fallo completo de la unión. En ensayos de crecimiento de grieta realizados sobre probetas preagrietadas en viga con doble voladizo (DCB), Dessureault identifica los procesos de iniciación y crecimiento de grietas en muestras unidas con adhesivo epoxi como una acumulación de microfisuras en la zona próxima al fondo de grieta que, luego, van coalesciendo para configurar la grieta principal. Lo que supone, igualmente, un proceso de daño del adhesivo en la zona de mayor concentración de tensiones que, posteriormente, conduce al fallo de la unión. La presente tesis surge con el propósito de aumentar los conocimientos existentes sobre el comportamiento a fatiga de las uniones adhesivas y especialmente las realizadas con dos tipos de adhesivos estructurales aplicados en aceros con diferentes acabados superficiales. El estudio incluye la obtención de las curvas de tensión frente al número de ciclos hasta el fallo del componente, curvas SN o curvas de Wöhler, que permitirán realizar una estimación de la resistencia a la fatiga de un determinado material o estructura. Los ensayos de fatiga realizados mediante ciclos predeterminados de carga sinusoidales, de amplitud y frecuencia constantes, han permitido caracterizar el comportamiento a la fatiga por el número de ciclos hasta la rotura, siendo el límite de fatiga el valor al que tiende la tensión cuando el número de ciclos es muy grande. En algunos materiales, la fatiga no tiende a un valor límite sino que decrece de forma constante a medida que aumenta el número de ciclos. Para estas situaciones, se ha definido la resistencia a la fatiga (o límite de resistencia) por la tensión en que se produce la rotura para un número de ciclos predeterminado. Todos estos aspectos permitirán un mejor diseño de las uniones y las condiciones de trabajo de los adhesivos con el fin de lograr que la resistencia a fatiga de la unión sea mucho más duradera y el comportamiento total de la unión sea mucho mejor, contribuyendo al crecimiento de la utilización de las uniones adhesivas respecto a otras técnicas. ABSTRACT Adhesives are well-known and have been used in many applications throughout history. At present, adhesion bonding technology of structural materials is experiencing an important growth. Scientific advances have enabled a better understanding of the phenomena of adhesion, as well as to improve and develop new polymeric formulations that increase the range of applications. On the other hand, the development of new materials and the need to save weight, especially in the transport sector, have promote the use of adhesive bonding in many applications previously reserved for other joining technologies such as welded or mechanical joints, presenting similar or even higher performances. Adhesive bonding offers many advantages over other joining methods. For example, in the aeronautic industry and in the automation sector, adhesive bonding allows a reduction in the number of components (such as bolts, rivets, clamps) and as consequence, resulting in lighter designs and a decrease in handling and storage costs, as well as faster assembly processes and an improvement in the production processes. In the construction sector and in the industrial equipment manufacturing, the ability to withstand thermal expansion and contraction is required. Therefore, adhesion bonding technology is used to avoid any distortion of the substrate since this technology does not require heating nor the deformation of the pieces when these are exposed to very high and localized heating, as in welding, or when are subjected to localized mechanical stresses in the case of riveted joints. In the naval industry, repair techniques based in the adhesive bonding are being developed in order to distribute stresses more uniform and homogeneously in order to improve the performance against fatigue and to avoid the problems associated with standard repair techniques as cutting and welding. Adhesive bonding does not require the use of high temperatures and as consequence they do not produce undesirable microstructural changes, as it can be observed in molten zones or in heat-affected zones in the case of welding, neither is there damage of the protective coating of metals with low melting points or polymeric films. However, adhesive bonding presents a disadvantage that limits its application, the low longterm durability. The most common cause of fractures of materials is fatigue fracture. This failure process is the cause of 85% of the fracture of structural materials in service. Fatigue failure occurs when the materials are subjected to the action of cyclic loads or vibrations for a long period of time. The joints and structures subjected to fatigue can fail at stress values below the static strength of the material. Fatigue failure do not occurs by a static and homogeneous process of initiation and propagation of crack. The fatigue process gradually weakens the bond until the moment in which the fracture occurs very rapidly. Underhill explains this mechanism as a process of irreversible damage of the weakest links at certain points of the bonding. When the deterioration in these weaker zones occurs, their area increase until the damage zone is so extensive that the full failure of the joint occurs. During the crack growth tests performed on precracked double-cantilever beam specimen, (DCB), Dessureault identified the processes of crack initiation and growth in samples bonded with epoxy adhesive as a process of accumulation of microcracks on the zone near the crack bottom, then, they coalesced to configure the main crack. This is a damage process of the adhesive in the zone of high stress concentration that leads to failure of the bond. This thesis aims to further the understanding of the fatigue behavior of the adhesive bonding, primarily those based on two different types of structural adhesives used on carbon-steel with different surface treatments. This memory includes the analysis of the SN or Wöhler curves (stress vs. number of cycles curves up to the failure), allowing to carry out an estimation of the fatigue strength of a specific material or structure. The fatigue tests carried out by means of predetermined cycles of sinusoidal loads, with a constant amplitude and frequency, allow the characterisation of the fatigue behaviour. For some materials, there is a maximum stress amplitude below which the material never fails for any number of cycles, known as fatigue limit. In the other hand, for other materials, the fatigue does not tend toward a limit value but decreases constantly as the number of cycles increases. For these situations, the fatigue strength is defined by the stress at which the fracture occurs for a predetermined number of cycles. All these aspects will enable a better joint design and service conditions of adhesives in order to get more durable joints from the fatigue failure point of view and in this way contribute to increase the use of adhesive bonding over other joint techniques.

A transition as a function of increasing temperature from harmonic to anharmonic dynamics has been observed in globular proteins by using spectroscopic, scattering, and computer simulation techniques. We present here results of a dynamic neutron scattering analysis of the solvent dependence of the picosecond-time scale dynamic transition behavior of solutions of a simple single-subunit enzyme, xylanase. The protein is examined in powder form, in D2O, and in four two-component perdeuterated single-phase cryosolvents in which it is active and stable. The scattering profiles of the mixed solvent systems in the absence of protein are also determined. The general features of the dynamic transition behavior of the protein solutions follow those of the solvents. The dynamic transition in all of the mixed cryosolvent–protein systems is much more gradual than in pure D2O, consistent with a distribution of energy barriers. The differences between the dynamic behaviors of the various cryosolvent protein solutions themselves are remarkably small. The results are consistent with a picture in which the picosecond-time scale atomic dynamics respond strongly to melting of pure water solvent but are relatively invariant in cryosolvents of differing compositions and melting points.

Single-phase Ba(Cd1/3Ta2/3)O-3 powder was produced using conventional solid state reaction methods. Ba(Cd1/3Ta2/3)O-3 ceramics with 2 wt % ZnO as sintering additive sintered at 1550 degreesC exhibited a dielectric constant of similar to32 and loss tangent of 5x10(-5) at 2 GHz. X-ray diffraction and thermogravimetric measurements were used to characterize the structural and thermodynamic properties of the material. Ab initio electronic structure calculations were used to give insight into the unusual properties of Ba(Cd1/3Ta2/3)O-3, as well as a similar and more widely used material Ba(Zn1/3Ta2/3)O-3. While both compounds have a hexagonal Bravais lattice, the P321 space group of Ba(Cd1/3Ta2/3)O-3 is reduced from P (3) under bar m1 of Ba(Zn1/3Ta2/3)O-3 as a result of a distortion of oxygen away from the symmetric position between the Ta and Cd ions. Both of the compounds have a conduction band minimum and valence band maximum composed of mostly weakly itinerant Ta 5d and Zn 3d/Cd 4d levels, respectively. The covalent nature of the directional d-electron bonding in these high-Z oxides plays an important role in producing a more rigid lattice with higher melting points and enhanced phonon energies, and is suggested to play an important role in producing materials with a high dielectric constant and low microwave loss. A homologous series of ultra-violet stabilisers containing 2-hydroxybenzophenone (HBP) moiety as a uv absorbing chromophore with varying alkyl chain lengths and sizes were prepared by known chemical synthesis. The strong absorbance of the HBP chromophore was utilized to evaluate the concentration of these stabilisers in low density polyethylene films and concentration of these stabilisers in low density polyethylene films and in relevant solvents by ultra-violet/visible spectroscopy. Intrinsic diffusion coefficients, equilibrium solubilities, volatilities from LDPE films and volatility of pure stabilisers were studied over a temperature range of 5-100oC. The effects of structure, molecular weight and temperature on the above parameters were investigated and the results were analysed on the basis of theoretical models published in the literature. It has been found that an increase in alkyl chain lengths does not change the diffusion coefficients to a significant level, while attachment of polar or branched alkyl groups change their value considerably. An Arrhenius type of relationship for the temperature dependence of diffusion coefficients seems to be valid only for a narrow temperature range, and therefore extrapolation of data from one temperature to another leads to a considerable error. The evidence showed that increase in additive solubility in the polymer is favoured by lower heat of fusions and melting points of additives. This implies the validity of simple regular solution theory to provide an adequate basis for understanding the solubility of additives in polymers The volubility of stabilisers from low density polyethylene films showed that of an additive from a polymer can be expressed in terms of a first-order kinetic equation. In addition the rate of loss of stabilisers was discussed in relation to its diffusion, solubility and volatility and found that all these factors may contribute to the additive loss, although one may be a rate determining factor. Stabiliser migration from LDPE into various solvents and food simulants was studied at temperatures 5, 23, 40 and 70oC; from the plots of rate of migration versus square root time, characteristic diffusion coefficients were obtained by using the solution of Fick's diffusion equations. It was shown that the rate of migration depends primarily on partition coefficients between solvent and the polymer of the additive and also on the swelling action of the contracting media. Characteristic diffusion coefficients were found to approach to intrinsic values in non swelling solvents, whereas in the case of highly swollen polymer samples, the former may be orders of magnitude greater than the latter.

The aim of this research was to investigate the molecular interactions occurring in the formulation of non-ionic surfactant based vesicles composed monopalmitoyl glycerol (MPG), cholesterol (Chol) and dicetyl phosphate (DCP). In the formulation of these vesicles, the thermodynamic attributes and surfactant interactions based on molecular dynamics, Langmuir monolayer studies, differential scanning calorimetry (DSC), hot stage microscopy and thermogravimetric analysis (TGA) were investigated. Initially the melting points of the components individually, and combined at a 5:4:1 MPG:Chol:DCP weight ratio, were investigated; the results show that lower (90 C) than previously reported (120-140 C) temperatures could be adopted to produce molten surfactants for the production of niosomes. This was advantageous for surfactant stability; whilst TGA studies show that the individual components were stable to above 200 C, the 5:4:1 MPG:Chol:DCP mixture show ∼2% surfactant degradation at 140 C, compared to 0.01% was measured at 90 C. Niosomes formed at this lower temperature offered comparable characteristics to vesicles prepared using higher temperatures commonly reported in literature. In the formation of niosome vesicles, cholesterol also played a key role. Langmuir monolayer studies demonstrated that intercalation of cholesterol in the monolayer did not occur in the MPG:Chol:DCP (5:4:1 weight ratio) mixture. This suggests cholesterol may support bilayer assembly, with molecular simulation studies also demonstrating that vesicles cannot be built without the addition of cholesterol, with higher concentrations of cholesterol (5:4:1 vs 5:2:1, MPG:Chol:DCP) decreasing the time required for niosome assembly. © 2013 Elsevier B.V.

This thesis describes the preparation of polymersomes from poly(ethylene glycol)-block-polycarbonate (PEG-PC) copolymers functionalized with pendant coumarin groups. Coumarin groups undergo photo-reversible dimerization when irradiated with specific ultraviolet wavelengths, so they can be used to prepare polymers with photo-responsive properties. In this case, the pendant coumarin groups enable stabilization of the polymersome membrane through photo-crosslinking of the hydrophobic block. Initially, several novel cinnamoyl and coumarin functionalized cyclic carbonate monomers were synthesized using ester, ether, or amide linkages. While the homopolymerization of these functionalized monomers proved challenging due to their high melting points, both cinnamoyl and coumarin functionalized monomers were successfully copolymerized with trimethylene carbonate (TMC) at 100 ℃ using a catalyst-free melt polymerization process where the TMC doubled as a solvent for the higher melting point monomer. Using this system, polycarbonate copolymers with up to 33% incorporation of the functionalized monomers were prepared. In addition, an investigation of some anomalous polymerization results identified previously unreported triethylamine-based catalysts for the melt polymerization of carbonate monomers. These studies also demonstrated that the catalyst-free polymerization of TMC occurs faster and at lower temperatures than previously reported. Subsequently, the photo-crosslinking of cinnamoyl and coumarin functionalized polycarbonates was compared and coumarin was identified as the more effective crosslinking agent when using 300-400 nm UV. An investigation of the photo-reversibility of the coumarin dimerization revealed no discernible change in the properties of crosslinked networks, but rapid photo-reversion in dilute solutions. The photo-crosslinking and photo-reversion kinetics of the coumarin functionalized polycarbonates were determined to be second-order in both cases. Finally, the self-assembly of PEG-PC diblock copolymers functionalized with coumarin was examined and both reverse solvent evaporation and solvent displacement were found to induce self-assembly, with hydrophilic mass fractions (f-factors) of 12-28% resulting in the formation of solid microparticles and nanoparticles and f-factors of 33-43% resulting in the formation of polymersomes. The stabilization of these polymersome membranes through photo-initiator-free photo-crosslinking was demonstrated with the crosslinking allowing polymersomes to withstand centrifugation at 12,000 x g. In addition, the encapsulation of calcein, as a model small molecule drug, in the stabilized polymersomes was successfully demonstrated using confocal microscopy.

Lanthanum phosphate is one among the lanthanide family of “Rare Earths” following the periodic table of elements. Known under the generic name, Monazite, the rare earth phosphates have melting points above 1900 °C, high thermal phase stability, low thermal conductivity and thermal expansion coefficient similar to some of the high temperature oxides like alumina and zirconia.

The main objectives of this work are the measurement of terpenes solubility in water at different temperatures, and the formulation of Deep Eutectic Solvents based on choline chloride and polycarboxylic acids, that can be used as hydrotropes of aqueous solutions in terpenes, replacing conventional organic solvents. In this work a new experimental methodology was implemented, using dialysis membranes, for the measurement of terpenes solubility in water. Concerning the deep eutectic diagrams formulation, the determination of the melting points of the eutectic mixtures was performed using a visual method. The method used for determining solubilities was previously validated using a well-studied model compound, toluene. The experimental results of terpenes solubilities in water resulted in a very satisfactory coefficients of variation, always below 6%. The experimental solubility data were analysed and the temperature dependence is also studied in a thermodynamic perspective. The compound with the largest solubility dependence with the temperature is geraniol, while thymol presents the smallest. The phase diagrams of DES formulated were quite satisfactory, presenting always eutectic points below to 373.15 K. For some compositions, the systems composed by choline chloride and lactic, or malonic, or myristic acid were liquid at room temperature. In the case of monocarboxylic acids, eutectic is formed at 60% mol of the acid, to dicarboxylic acid is formed at 50% mol of the acid and for tricarboxylic acid these point is formed at 30% mol of the acid. In the future, it will be important to study the effect of DES as hydrotropes in aqueous solutions of terpenes. Furthermore, it would be interesting to study more terpenes in order to assess the effect of the size of the alkyl chain and the structures of the compounds.
