A genetic linkage map of mungbean (Vigna radiata, 2n = 2x = 22) consisting of 255 RFLP loci was developed using a recombinant inbred population of 80 individuals. The population was derived from an intersubspecific cross between the cultivated mungbean variety 'Berken' and a wild mungbean genotype 'ACC 41' (V radiata subsp. sublobata). The total length of the map, which comprised 13 linkage groups, spanned 737.9 cM with an average distance between markers of 3.0 cM and a maximum distance between linked markers of 15.4 cM. The mungbean map was compared to a previously published map of lablab (Lablab purpureus, 2n = 2x = 24) using a common set of 65 RFLP probes. In contrast to some other comparative mapping studies among members of the Fabaceae, where a high level of chromosomal rearrangement has been observed, marker order between mungbean and lablab was found to be highly conserved. However, the two genomes have apparently accumulated a large number of duplications/deletions after they diverged.

Um dos principais objetivos da ciência é perceber a natureza, i.e., descobrir e explicar o funcionamento do mundo que nos rodeia. Para tal, os cientistas precisam de coligir dados e monitorar o meio ambiente. Em particular, considerando que cerca de 70% da Terra é coberta por água, a coleta de parâmetros de caracterização da água de grandes superfícies é uma prioridade. A monitorização das condições da água é feita principalmente através de bóias. No entanto, as bóias disponíveis no mercado não satisfazem as necessidades existentes. Esta é uma das principais razões que levaram o Laboratório de Sistemas Autónomos (LSA) do Instituto Superior de Engenharia do Porto a lançarem um projeto para o desenvolvimento de uma bóia reconfigurável e com dois modos de funcionamento: monitorização ambiental e baliza ativa de regata. O segundo modo é destinado a regatas de veleiros autónomos. O projeto começou há um ano com um projeto do European Project Project [1] (EPS), realizado por quatro estudantes internacionais, destinado à construção da estrutura da bóia e à selecção dos componentes mais adequados para o sistema de medição e controlo. A arquitetura que foi definida para este sistema é do tipo mestre-escravo e é composta por uma unidade de controlo mestre para a telemetria e configuração e uma unidade de controlo escrava para a medição e armazenamento de dados. O desenvolvimento do projeto continuou com dois estudantes belgas que trabalharam na comunicação e no armazenamento de dados. Este projeto, que prossegue com o desenvolvimento da medição e do armazenamento de dados do lado da unidade de controlo escrava, tem os seguintes objetivos: (i ) implementar o protocolo de comunicação na unidade de controlo escrava; (ii ) coligir e armazenar os dados dos sensores no cartão SD em tempo real; (iii ) fornecer dados em tempo útil; e (iv) recuperar dados do cartão SD em tempo diferido. As contribuições anteriores foram estudadas e foi feito um levantamento dos projetos congéneres existentes. O desenvolvimento do projeto atual começou com o protocolo de comunicação. Este protocolo, que foi projetado pelos alunos anteriores, foi um bom ponto de partida. No entanto, o protocolo foi atualizado e melhorado com novas funcionalidades. Esta última componente foi um trabalho conjunto com Laurens Allart, que esteve a trabalhar no subsistema de telemetria e de configuração durante este semestre. O protocolo foi implementado do lado da unidade de controlo escrava através de uma estrutura de múltiplas actividades paralelas (multithreaded). Esta estrutura recebe as mensagens da unidade mestre, executa as ações solicitadas e envia de volta o resultado. A bóia é um dispositivo reconfigurável multimodo que pode ser expandido com novos modos de operação no futuro. Infelizmente, sofre de algumas limitações: suporta uma carga máxima de 40 kg e tem uma área de implantação limitada pela distância máxima à estacão base.

The relationship between estimated and real motor competences was analyzed for several tasks. Participants were 303 children (160 boys and 143 girls), which had between 6 and 10 years of age (M=8.63, SD=1.16). None of the children presented developmental difficulties or learning disabilities, and all attended age-appropriate classes. Children were divided into three groups according to their age: group 1 (N= 102; age range: 6.48-8.01 years); group 2 (N= 101; age range: 8.02-9.22 years); and group 3 (N=100; age range: 9.24-10.93 years). Children were asked to predict their maximum distance for a locomotor, a manipulative, and a balance task, prior to performing those tasks. Children’s estimations were compared with their real performance to determine their accuracy. Children had, in general, a tendency to overestimate their performance (standing long jump: 56.11%, kicking: 63.37%, throwing: 73.60%, and Walking Backwards (WB) on a balance beam: 45.21%), and older children tended to be more accurate, except for the manipulative tasks. Furthermore, the relationship between estimation and real performance in children with different levels of motor coordination (Köperkoordinationstest für Kinder, KTK) was analyzed. The 75 children with the highest score comprised the Highest Motor Coordination (HMC) group, and the 78 children with the lowest score were placed in the Lowest Motor Coordination (LMC) group. There was a tendency for LMC and HMC children to overestimate their skills at all tasks, except for the HMC group at the WB task. Children with the HMC level tended to be more accurate when predicting their motor performance; however, differences in absolute percent error were only significant for the throwing and WB tasks. In conclusion, children display a tendency to overestimate their performance independently of their motor coordination level and task. This fact may be determinant to the development of their motor competences, since they are more likely to engage and persist in motor tasks, but it might also increase the occurrence of unintended injuries.

In order to evaluate the influence of continental crustal rocks on trace element budgets of serpentinized peridotites incorporated into the continental crust, we have analyzed the chemical composition of whole rock samples and minerals of the Geisspfad ultramafic complex (Swiss-Italian Alps). This complex represents a relict oceanic succession composed of serpentinites, ophicarbonates and metabasic rocks, emplaced into crustal gneisses during Alpine collision. Following peak metamorphic amphibolite facies conditions, fluid flow modified some of the trace element contents of ophicarbonates and deformed serpentinites close to the contact with country rocks. The fluid originated from the surrounding continental crustal rocks as documented by the increase of Pb in the serpentinites, and by the strongly negative all) values (-112 parts per thousand) of some ultramafic rocks close to the contact with surrounding gneisses. Little or no modification of the fluid mobile elements Li, B or U was observed in the serpentinite. In-situ analysis of light elements of serpentinite minerals indicate redistribution of light elements coupled to changes of mineral modes towards the outer 100-150 m of the massif. In the centre of the massif, Li is preferentially concentrated in olivine, while Be and B are hosted by tremolite. In contrast, at the outer rim of the massif, Li and Be are preferentially incorporated into diopside, and B into antigorite. This redistribution of light elements among the different minerals is visible in the serpentinite, at a maximum distance of -100-150 m from the ophicarbonate-metabasite contact. Our results show that interaction of ultramafic rocks and crust-derived fluids can be easily detected by studies of Pb and partial derivative D in whole rocks. We argue that small ultramafic bodies potentially record an emplacement-related trace element signature, and that crustal light element values in ultramafic rocks are not necessarily derived from a subducting slab. The objective of this work was to evaluate the spatial distribution of thrips in different crops, and the correlation between meterological parameters and the flight movements of this pest, using immunomarking. The experiment was conducted in cultivated areas, with tomato (Solanum lycopersicum), potato (Solanum tuberosum), and onion (Allium cepa); and non-cultivated areas, with weedy plants. The areas with tomato (100 days), potato (20 days), and weeds were sprayed with casein, albumin, and soy milk, respectively, to mark adult thrips; however, the areas with onion (50 days) and tomato (10 days) were not sprayed. Thrips were captured with georeferenced blue sticky traps, transferred into tubes, and identified by treatment area with the Elisa test. The dependence between the samples and the capture distance was determined using geostatistics. Meteorlogical parameters were correlated with thrips density in each area. The three protein types used for immunomarking were detected in different proportions in the thrips. There was a correlation between casein-marked thrips and wind speed. The thrips flew a maximum distance of 3.5 km and dispersed from the older (tomato) to the younger crops (potato). The immunomarking method is efficient to mark large quantities of thrips.

We have compared the phylogenetic diversity of methicillin-resistant Staphylococcus aureus (MRSA) strains from Switzerland and their phylogenetic relationships with European epidemic clones, using multiprimer random amplification polymorphic DNA (RAPD). Strains included 24 European epidemic clones (59 strains), 66 sporadic strains isolated in Switzerland in 1996-1997, and 15 reference strains of five other Staphylococcus species. Similarity and clustering analysis with the Jaccard's coefficient showed that the maximum genetic distance between MRSA strains was 0.43, whereas the minimum genetic distance between the six Staphylococcus species was 0.97, indicating that the method permits phylogenetic hierarchization. The 24 MRSA clones reported to be epidemic in European countries during the 1990s were distributed into seven different genetic clusters with a maximum distance of 0.29 among them. This clustering pattern was confirmed by the analysis of a subset of MRSA strains by multilocus enzyme electrophoresis at 12 loci. Most of the sporadic Swiss strains were distributed into these seven different genetic clusters, together with the epidemic MRSA clones. This suggests that there is no phylogenetic cluster specific to epidemic clones of MRSA.

The spatial limits of the active site in the benzylic hydroxylase enzyme of the fungus Mortierella isabellina were investigated. Several molecular probes were used in incubation experiments to determine the acceptability of each compound by this enzyme. The yields of benzylic alcohols provided information on the acceptability of the particular compound into the active site, and the enantiomeric excess values provided information on the "fit" of acceptable substrates. Measurements of the molecular models were made using Cambridge Scientific Computing Inc. CSC Chem 3D Plus modeling program. i The dimensional limits of the aromatic binding pocket of the benzylic hydroxylase were tested using suitably substituted ethyl benzenes. Both the depth (para substituted substrates) and width (ortho and meta substituted substrates) of this region were investigated, with results demonstrating absolute spatial limits in both directions in the plane of the aromatic ring of 7.3 Angstroms for the depth and 7.1 Angstroms for the width. A minimum requirement for the height of this region has also been established at 6.2 Angstroms. The region containing the active oxygen species was also investigated, using a series of alkylphenylmethanes and fused ring systems in indan, 1,2,3,4-tetrahydronaphthalene and benzocycloheptene substrates. A maximum distance of 6.9 Angstroms (including the 1.5 Angstroms from the phenyl substituent to the active center of the heme prosthetic group of the enzyme) has been established extending directly in ii front of the aromatic binding pocket. The other dimensions in this region of the benzylic hydroxylase active site will require further investigation to establish maximum allowable values. An explanation of the stereochemical distributions in the obtained products has also been put forth that correlates well with the experimental observations.

Research into transmissible spongiform encephalopathy (TSE) diseases has become a high priority worldwide in recent years yet remarkably little is known about the behaviour of TSE infectivity in the environment. The resilience and stability of prion proteins could lead to soils becoming a potential reservoir of TSE infectivity as a result of contamination from activities such as infected carcass burial or the dispersion of effluents from slaughter houses, or by contamination of pastures by infected animals, (e.g. scrapie in sheep). Knowledge of the fate of prion proteins in soils, and associated physico-chemical conditions which favour migration, can be used to help prevent re-infection of animals through grazing, to protect watercourses and develop good management practices. In two consecutive experiments of 9 and 6 months, the migration of recombinant ovine PrP (recPrP) in soil columns was followed under contrasting levels of microbial activity (normal versus reduced), under varying regimes of soil water content and redox potential, and in two different soil types (loamy sand and clay loam). At each analysis time (1, 3, 6 or 9 months), in both soil types, full-length recPrP was detected in the original contaminated layer, indicating the resilience and stability of recPrP under varied soil conditions, even in the presence of active soil microbial populations. Evidence of protein migration was found in every soil column at the earliest analysis time (1 or 3 months), but was restricted to a maximum distance of 1 cm, indicative of limited initial mobility in soils followed by strong adsorption over the following days to weeks. The survival of recPrP in the soil over a period of at least 9 months was demonstrated. In this study, recPrP was used as an indicator for potential TSE infectivity, although infectivity tests should be carried out before conclusions can be drawn regarding the infection risk posed by prions in soil. However, it has been demonstrated that soil is likely to act as a significant barrier to the dispersion of contaminated material at storage or burial sites. (c) 2007 Elsevier Ltd. Knowledge of tropical raptor habitat use is limited and yet a thorough understanding is vital when trying to conserve endangered species. We used a well studied, reintroduced population of the vulnerable Mauritius Kestrel Falco punctatus to investigate habitat preferences in a modified landscape. We constructed a high resolution digital habitat map and radiotracked 13 juvenile Kestrels to quantify habitat preferences. We distinguished seven habitat types in our study area and tracked Kestrels from 71 to 130 days old during which they dispersed from their natal territory and settled within a home-range after reaching independence. Mean home-range size was 0.95 km(2) characterized by a bimodal pattern of intensity around the natal site and post-independence home-range. Compositional analysis showed that home-ranges were located non-randomly with respect to habitat but there was no evidence to suggest differential use of habitats within home-ranges. Native and semi-invaded forest and grassland were consistently preferred, whereas agriculture was used significantly less than other habitats. No difference was found between the available length of edge dividing native forest and grassland within a home-range when compared to that available within a 2.35-km buffer around their nest-site, based on the maximum distance a juvenile was found to disperse. Repeating the analysis in three dimensions gave very similar results. Our results suggest that Mauritius Kestrels are not obligate forest dwellers as was once thought but can also exploit open habitats such as grassland. Kestrels may be using isolated mature trees within grassland as vantage points for hunting in the same way as they use the natural stratified forest structure. We suggest that the avoidance of agriculture is partly due to a lack of such vantage points. The conservation importance of forest degradation and agricultural encroachment is highlighted and comparisons with the habitat preferences of other tropical falcons are discussed.

A escolha e o uso adequado de pontas de pulverização são essenciais para a correta aplicação de produtos fitossanitários, sendo, portanto, indispensável o conhecimento de suas características. Este trabalho teve o objetivo de caracterizar o perfil de distribuição e o diâmetro de gotas, oferecendo dados para otimizar o espaçamento entre bicos na barra de pulverização. Foram avaliados os perfis de distribuição da ponta de jato plano Teejet XR 110015 VS, a 0,50 m da altura da mesa de deposição, nas pressões de 200 e 300 kPa, e o diâmetro das gotas pelo método de difração de raios laser. As distâncias máximas foram de 0,85 m, calculadas para um coeficiente de variação (C.V.) aceitável para as pressões de 200 e 300 kPa , com os respectivos valores de 9,52 e 9,58%. A distância ótima foi de aproximadamente 0,70 m, para C.V. em torno de 5%. Comparando as pressões, houve diferença significativa para DV0,1 e DV0,5, não havendo diferença para o DV0,9. Embora o aumento da pressão tenha provocado diminuição do tamanho das gotas, não houve diferença significativa de uniformidade entre as duas pressões de trabalho avaliadas. Concluiu-se que o espaçamento máximo entre bicos na barra não deverá ser maior que 0,85 m e que o DV0,5 diminui com o aumento da pressão de 200 para 300 kPa, porém sem alteração significativa da uniformidade de diâmetro de gota.

A minimalist representation of protein structures using a Go- like potential for interactions is implemented to investigate the mechanisms of the domain swapping of p13suc1, a protein that exists in two native conformations: a monomer and a domain- swapped dimer formed by the exchange of a beta- strand. Inspired by experimental studies which showed a similarity of the transition states for folding of the monomer and the dimer, in this study we justify this similarity in molecular descriptions. When intermediates are populated in the simulations, formation of a domain- swapped dimer initiates from the ensemble of unfolded monomers, given by the fact that the dimer formation occurs at the folding/ unfolding temperature of the monomer ( T-f). It is also shown that transitions, leading to a dimer, involve the presence of two intermediates, one of them has a dimeric form and the other is monomeric; the latter is much more populated than the former. However, at temperatures lower than T-f, the population of intermediates decreases. It is argued that the two folded forms may coexist in absence of intermediates at a temperature much lower than T-f. Computational simulations enable us to find a mechanism, `` lock- and- dock'', for domain swapping of p13suc1. To explore the route toward dimer formation, the folding of unstructured monomers must be retarded by first locking one of the free ends of each chain. Then, the other free termini could follow and dock at particular regions, where most intrachain contacts are formed, and thus de. ne the transition states of the dimer. The simulations also showed that a decrease in the maximum distance between monomers increased their stability, which is explained based on confinement arguments. Although the simulations are based on models extracted from the native structure of the monomer and the dimer of p13suc1, the mechanism of the domain- swapping process could be general, not only for p13suc1.
