The parasitic protists in the genus Tritrichomonas cause significant disease in domestic cattle and cats. To assess the genetic diversity of feline and bovine isolates of Tritrichomonas foetus (Riedmuller, 1928) Wenrich and Emmerson, 1933, we used 10 different genetic regions, namely the protein coding genes of cysteine proteases 1,2 and 4-9 (CP1, 2, 4-9) involved in the pathogenesis of the disease caused by the parasite. The cytosolic malate dehydrogenase 1 (MDH1) and internal transcribed spacer region 2 of the rDNA unit (ITS2) were included as additional markers. The gene sequences were compared with those of Tritrichomonas suis (Davaine. 1875) Morgan and Hawkins, 1948 and Tritrichomonas mobilensis Culberson et al., 1986. The study revealed 100% identity for all 10 genes among all feline isolates (=T. foetus cat genotype), 100% identity among all bovine isolates (=T. foetus cattle genotype) and a genetic distinctness of 1% between the cat and cattle genotypes of T. foetus. The cattle genotype of T. foetus was 100% identical to T. suis at nine loci (CP1, 2,4-8, ITS2, MDH1). At CP9, three out of four T. suis isolates were identical to the T. foetus cattle genotype, while the T. suis isolate SUI-H3B sequence contained a single unique nucleotide substitution. Tritrichomonas mobilensis was 0.4% and 0.7% distinct from the cat and cattle genotypes of T. foetus, respectively. The genetic differences resulted in amino acid changes in the CP genes, most pronouncedly in CP2, potentially providing a platform for elucidation of genotype-specific host-pathogen interactions of T. foetus. On the basis of this data we judge T. suis and T. foetus to be subjective synonyms. For the first time, on objective nomenclatural grounds, the authority of T. suis is given to Davaine, 1875, rather than the commonly cited Gruby and Delafond, 1843. To maintain prevailing usage of T. foetus, we are suppressing the senior synomym T. suis Davaine, 1875 according to Article 23.9, because it has never been used as a valid name after 1899 and T. foetus is widely discussed as the cause of bovine trichomonosis. Thus bovine, feline and porcine isolates should all be given the name T. foetus. This promotes the stability of T. foetus for the veterinary and economically significant venereal parasite causing bovine trichomonosis. (C) 2012 Australian Society for Parasitology Inc. Shoot blight symptom was found on persimmon (Diospyros kaki) in southern Western Australia in December 2010. The pathogen was isolated and identified as Diaporthe neotheicola on the basis of morphology, sequence analysis of the internal transcribed spacer (ITS) and the translation elongation factor 1-α (TEF). A pathogenicity test was conducted and Koch's postulates were fulfilled by re-isolation of the fungus from diseased tissues. This is the first report of D. neotheicola causing shoot blight on persimmon in Australia and worldwide. © 2012 Australasian Plant Pathology Society Inc.

Three genera, Cochliobolus, Bipolaris and Curvularia form a complex that contains many plant pathogens, mostly on grasses (Poaceae) with a worldwide distribution. The taxonomy of this complex is confusing as frequent nomenclatural changes and refinements have occurred. There is no clear morphological boundary between the asexual genera Bipolaris and Curvularia, and some species show intermediate morphology. We investigated this complex based on a set of ex-type cultures and collections from northern Thailand. Combined gene analysis of rDNA ITS (internal transcribed spacer), GPDH (glyceraldehyde 3-phosphate dehydrogenase), LSU (large subunit) and EF1-α (translation elongation factor 1-α) shows that this generic complex divides into two groups. Bipolaris and Cochliobolus species clustered in Group 1 along with their type species, whereas Curvularia species (including species named as Bipolaris, Cochliobolus and Curvularia) clustered in Group 2, with its generic type. The nomenclatural conflict in this complex is resolved giving priority to the more commonly used established generic names Bipolaris and Curvularia. Modern descriptions of the genera Bipolaris and Curvularia are provided and species resolved in this study are transferred to one of these genera based on their phylogeny. © 2012 Mushroom Research Foundation.

The occurrence of pathogenic and endophytic species of Phyllosticta on cultivated Citrus in Australia was investigated by DNA sequence analysis of specimens held in plant pathology herbaria and culture collections. Sequences of the internal transcribed spacer region (ITS1, 5.8S, ITS2), and partial translation elongation factor 1-alpha (TEF) gene of 41 Phyllosticta-like isolates from Citrus were compared to those sequences from the type specimens of Phyllosticta recorded from around the world. Phylogenetic analysis resolved all the sequences of Australian accessions into two major clades. One clade corresponded to P. citricarpa, which causes citrus black spot disease. The other clade contained P. capitalensis, which is a known endophyte of Citrus and many other plant species. All included herbarium accessions previously designated as Guignardia mangiferae are now designated P. capitalensis. No Australian isolates were identified as the newly described pathogens of citrus P. citriasiana or P. citrichinaensis, or the endophytes Guignarida mangiferae, P. brazilianiae, or P. citribraziliensis. © 2013 Australasian Plant Pathology Society Inc.

Six new species of Diaporthe, D. beilharziae on Indigofera australis, D. fraxini-angustifoliae on Fraxinus angustifolia subsp. oxycarpa, D. litchicola on Litchi chinensis, D. nothofagi on Nothofagus cunninghamii, D. pascoei on Persea americana and D. salicicola on Salix purpurea from Australia are described and illustrated based on morphological characteristics and molecular analyses. Three of the new species no longer produced sporulating structures in culture and two of these were morphologically described from voucher specimens. Phylogenetic relationships of the new species with other Diaporthe species are revealed by DNA sequence analyses based on the internal transcribed spacer (ITS) region, and partial regions of the β-tubulin (BT) and translation elongation factor 1-alpha (TEF). © 2013 Mushroom Research Foundation.

Several species of Phyllosticta (syn. Guignardia) have been described from orchids worldwide. A new species, Phyllosticta speewahensis, is proposed for a specimen isolated from leaf spots on a hybrid Vanda orchid in northern Queensland, Australia. Phylogenetic analysis of the nrDNA internal transcribed spacer region (ITS) and partial translation elongation factor 1-alpha (TEF1) gene sequences showed that P. speewahensis is most closely related to P. hostae. The likelihood that orchids harbour further cryptic species of endophytic and pathogenic Phyllosticta species is discussed.

Systematic relationships between the rusts on Goodeniaceae and Stylidiaceae were examined using phylogenetic analyses with two loci (internal transcribed spacer, large subunit region) from ribosomal DNA. Fresh specimens and herbarium material of four rust species (Puccinia dampierae, P. gilgiana, P. saccardoi and Uromyces scaevolae) from the Goodeniaceae and one (P. stylidii) from the Stylidiaceae were examined. A further species (P. lagenophorae) that is reported from hosts in Goodeniaceae and Asteraceae was included in our analysis. Our phylogenetic analysis recovered the rusts on Goodeniaceae and Stylidiaceae in clades sister to P. lagenophorae on Asteraceae. This supported a taxonomy in which P. lagenophorae is restricted to Asteraceae. Descriptions or taxonomic notes are provided for all of the known rusts on Goodeniaceae and Stylidiaceae.

Three new species of Tilletia are described from species of Eriachne (Poaceae) in the arid tropics of north-western Australia. In Western Australia, T. mactaggartii sp. nov. infects E. burkittii, and T. geeringii sp. nov. infects E. festucacea. Tilletia marjaniae sp. nov. infects E. pulchella subsp. dominii in Western Australia and the Northern Territory. These species are the first records of Tilletia on Eriachne. Phylogenetic relationships of these species were inferred from internal transcribed spacer of ribosomal RNA region and large subunit ribosomal RNA gene sequences.

The black rot disease of Vitis species and other host genera of Vitacease is caused by Phyllosticta ampelicida and allied taxa which is considered to be a species complex. In this paper, we introduce four new species of Phyllosticta, including two from the P. ampelicida complex, based on a polyphasic characterization including disease symptoms and host association, morphology, and molecular phylogeny. The phylogenetic analysis was conducted based on the ribosomal internal transcribed spacer (ITS) region and a combined multi-locus alignment of the ITS, actin (ACT), partial translation elongation factor 1-alpha (TEF-1), and glyceraldehydes 3-phosphate dehydrogenase (GPDH) gene regions. Our study confirms the phylogenetic distinctions of the four new species, as well as their phenotypic differences with known species in the genus.

Diaporthe (syn. Phomopsis) species are well-known saprobes, endophytes or pathogens on a range of plants. Several species have wide host ranges and multiple species may sometimes colonise the same host species. This study describes eight novel Diaporthe species isolated from live and/or dead tissue from the broad acre crops lupin, maize, mungbean, soybean and sunflower, and associated weed species in Queensland and New South Wales, as well as the environmental weed bitou bush (Chrysanthemoides monilifera subsp. rotundata) in eastern Australia. The new taxa are differentiated on the basis of morphology and DNA sequence analyses based on the nuclear ribosomal internal transcribed spacer region, and part of the translation elongation factor-1α and ß-tubulin genes. The possible agricultural significance of live weeds and crop residues ('green bridges') as well as dead weeds and crop residues ('brown bridges') in aiding survival of the newly described Diaporthe species is discussed.

Rhizoctonia spp. are ubiquitous soil inhabiting fungi that enter into pathogenic or symbiotic associations with plants. In general Rhizoctonia spp. are regarded as plant pathogenic fungi and many cause root rot and other plant diseases which results in considerable economic losses both in agriculture and forestry. Many Rhizoctonia strains enter into symbiotic mycorrhizal associations with orchids and some hypovirulent strains are promising biocontrol candidates in preventing host plant infection by pathogenic Rhizoctonia strains. This work focuses on uni- and binucleate Rhizoctonia (respectively UNR and BNR) strains belonging to the teleomorphic genus Ceratobasidium, but multinucleate Rhizoctonia (MNR) belonging to teleomorphic genus Thanatephorus and ectomycorrhizal fungal species, such as Suillus bovinus, were also included in DNA probe development work. Strain specific probes were developed to target rDNA ITS (internal transcribed spacer) sequences (ITS1, 5.8S and ITS2) and applied in Southern dot blot and liquid hybridization assays. Liquid hybridization was more sensitive and the size of the hybridized PCR products could be detected simultaneously, but the advantage in Southern hybridization was that sample DNA could be used without additional PCR amplification. The impacts of four Finnish BNR Ceratorhiza sp. strains 251, 266, 268 and 269 were investigated on Scot pine (Pinus sylvestris) seedling growth, and the infection biology and infection levels were microscopically examined following tryphan blue staining of infected roots. All BNR strains enhanced early seedling growth and affected the root architecture, while the infection levels remained low. The fungal infection was restricted to the outer cortical regions of long roots and typical monilioid cells detected with strain 268. The interactions of pathogenic UNR Ceratobasidium bicorne strain 1983-111/1N, and endophytic BNR Ceratorhiza sp. strain 268 were studied in single or dual inoculated Scots pine roots. The fungal infection levels and host defence-gene activity of nine transcripts [phenylalanine ammonia lyase (pal1), silbene synthase (STS), chalcone synthase (CHS), short-root specific peroxidase (Psyp1), antimicrobial peptide gene (Sp-AMP), rapidly elicited defence-related gene (PsACRE), germin-like protein (PsGER1), CuZn- superoxide dismutase (SOD), and dehydrin-like protein (dhy-like)] were measured from differentially treated and un-treated control roots by quantitative real time PCR (qRT-PCR). The infection level of pathogenic UNR was restricted in BNR- pre-inoculated Scots pine roots, while UNR was more competitive in simultaneous dual infection. The STS transcript was highly up-regulated in all treated roots, while CHS, pal1, and Psyp1 transcripts were more moderately activated. No significant activity of Sp-AMP, PsACRE, PsGER1, SOD, or dhy-like transcripts were detected compared to control roots. The integrated experiments presented, provide tools to assist in the future detection of these fungi in the environment and to understand the host infection biology and defence, and relationships between these interacting fungi in roots and soils. This study further confirms the complexity of the Rhizoctonia group both phylogenetically and in their infection biology and plant host specificity. The knowledge obtained could be applied in integrated forestry nursery management programmes.

Endophytic fungi isolated from Catharanthus roseus were screened for the production of vincristine and vinblastine. Twenty-two endophytic fungi isolated from various tissues of C. roseus were characterized taxonomically by sequence analysis of the internal transcribed spacer (ITS) region of rDNA and grouped into 10 genera: Alternaria, Aspergillus, Chaetomium, Colletotrichum, Dothideomycetes, Eutypella, Eutypa, Flavodon, Fusarium and Talaromyces. The antiproliferative activity of these fungi was assayed in HeLa cells using the MTT assay. The fungal isolates Eutypella sp-CrP14, obtained from stem tissues, and Talaromyces radicus-CrP20, obtained from leaf tissues, showed the strongest antiproliferative activity, with IC50 values of 13.5 mu g/ml and 20 mu g/ml, respectively. All 22 endophytic fungi were screened for the presence of the gene encoding tryptophan decarboxylase (TDC), the key enzyme in the terpenoid indole alkaloid biosynthetic pathway, though this gene could only be amplified from T. radicus-CrP20 (NCBI GenBank accession number KC920846). The production of vincristine and vinblastine by T. radicus-CrP20 was confirmed and optimized in nine different liquid media. Good yields of vincristine (670 mu g/l) in modified M2 medium and of vinblastine (70 mu g/l) in potato dextrose broth medium were obtained. The cytotoxic activity of partially purified fungal vincristine was evaluated in different human cancer cell lines, with HeLa cells showing maximum susceptibility. The apoptosis-inducing activity of vincristine derived from this fungus was established through cell cycle analysis, loss of mitochondrial membrane potential and DNA fragmentation patterns.

The identification of sea bass (Centropristis) larvae to species is difficult because of similar morphological characters, spawning times, and overlapping species ranges. Black sea bass (Centropristis striata) is an important fishery species and is currently considered to be overfished south of Cape Hatteras, North Carolina. We describe methods for identifying three species of sea bass larvae using polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) assays based on species-specific amplification of rDNA internal transcribed spacer regions. The assays were tested against DNA of ten other co-occurring reef fish species to ensure the assay's specificity. Centropristis larvae were collected on three cruises during cross-shelf transects and were used to validate the assays. Seventy-six Centropristis larva were assayed and 69 (91%) were identified successfully. DNA was not amplified from 5% of the larvae and identification was inconclusive for 3% of the larvae. Those assays can be used to identify sea bass eggs and larvae and will help to assess spawning locations, spawning times, and larval dispersal.

Sporothrix schenckii é um fungo dimórfico e agente etiológico da esporotricose, uma micose profunda que apresenta diferentes manifestações clínicas. As diversas manifestações clínicas desta e de outras doenças infecciosas podem ser relacionadas ao status imune do hospedeiro, a fatores de virulência do patógeno ou a diferentes genótipos. Dados anteriores do nosso grupo demonstram que diferenças na expressão de adesinas do S. schenckii para fibronectina estão diretamente relacionadas à virulência de diferentes cepas. Neste trabalho visamos avaliar caracteres morfológicos bioquímicos e genotípicos de doze isolados de geofílicos, zoofílicos e antropofílicos de S. schenckii, de diferentes origens geográficas, que apresentam diferentes graus de virulência. Foi analisada a morfologia das formas de micélio e levedura de cada isolado. Foi observado que a fase de micélio dos isolados estudados apresentaram morfologia típica com hifas finas e septadas, com conídios obovóides ou ovóides alongados. As leveduras apresentaram pleomorfismo típico da espécie, com células variando do formato ovóide ao alongado. Verificamos ainda a expressão de adesinas para fibronectina e laminina, do antígeno gp70 e, o padrão de bandas antigênicas reconhecidas por anticorpos IgG presentes em soro de pacientes com esporotricose ou de camundongos infectados. Para isso, foram extraídas proteínas de superfície da forma de levedura de cada isolada, sendo os extratos ensaiados por Western blot. Nestes ensaios observamos que os isolados mais virulentos de S. schenckii expressavam mais adesinas para fibronectina e laminina. A presença da gp70 foi detectada em dez dos doze isolados, sendo que apenas os isolados zoofílicos não expressam esta glicoproteína. O padrão antigênico foi variável entre os isolados, não havendo clara relação com a origem e/ou distribuição geográfica. Os dados fenotípicos foram confrontados com dados genotípicos. Para isso, sequenciamos os loci da calmodulina (CAL) e do Internal Transcribed Spacer 1/2 (ITS 1/2) a fim de averiguar se haviam diferenças genotípicas entre os isolados estudados. As análises do sequenciamento do loci CAL e ITS, contudo, apontam a divisão dos isolados em duas espécies filogenéticas, S. schenckii e S. brasiliensis não correlacionada com a distribuição geográfica dos mesmos. Nosso estudo reforça a hipótese de haver uma correlação entre virulência e expressão de adesinas, porém, sem qualquer relação entre a distribuição geográfica dos isolados zoofilicos, antropofílicos ou geofílicos, bem como dos genótipos encontrados.

羊草(Leymus chinensis(Trin.)Tzvel.或Aneurolepidium chinense Trin.),隶属禾本科(Gramineae),大麦族(Hordeae),小麦亚族(Triticieae),赖草属(Leymus),兼具重要的生态价值和经济价值。面向国家重大需求,本论文选择羊草作为主要研究对象,开展了两方面的研究工作,即我国羊草种质资源的遗传多样性评价和赖草属物种的系统发育分析。 羊草是一种多年生根茎型禾草,根据叶色可以划分为黄绿型和灰绿型两种生态型。其分布范围横穿亚欧草原的东部,包括朝鲜和蒙古的西部,以及西伯利亚的西北部,集中分布于我国的东北部。羊草是我国典型草原植物群落中的优势种。它可以在多种土壤和气候条件下正常生长,如松嫩平原、内蒙古草原和黄土高原。从另一个角度讲,不同类型的生境也造就了羊草丰富的遗传多样性。 自上世纪90年代中期开始,我们陆续从我国东北部6个省市收集了293份羊草种质,包括205份灰绿型羊草和88份黄绿型羊草。经过连续3年的观测记录,通过37个重要农艺性状,对293份羊草的遗传多样性进行了评估。依据10个质量性状和27个数量性状,统计了羊草不同性状和不同地域羊草的Shannon遗传多样性指数,同时还使用了主成分分析和通径分析做了相关统计。结果显示:(1)与黄绿型羊草相比,灰绿型羊草具有更高的遗传变异(P<0.05)。结合这两个趋异型羊草的分布范围,可以得出两种类型羊草之间存在稳定的遗传差异;(2)通径分析显示,羊草营养生长性状和遗传多样性两者的组合效应可以解释羊草生殖特性中20.6%的遗传变异;(3)在124-128ºE这一区域,羊草的遗传多样性指数最高(H=2.252),表明这一地区具有最丰富的羊草种质资源。 赖草属(Leymus Hochst.)是一个异源多倍体属,约有34个物种,该属未知基因组的起源一直争论不休。其倍性范围从四倍体(2n=4x=28)、八倍体(2n=8x=56)一直到十二倍体(2n=12x=84)。新麦草属(Psathyrostachys Nevski)只含有Ns一个基因组,约有9个物种。这两个属都是多年生牧草,具有抗旱、抗病和耐盐碱等生物学特性。 应用核糖体ITS(Internal Transcribed Spacer)序列和叶绿体trnL-F序列,我们对13个赖草属物种、小麦族18个属(40份二倍体材料)、以及Elymus californicus和Bromus catharticus,共计57份材料进行了系统发生分析。ITS序列分析表明,赖草属分别与新麦草属和小麦族中一个未知属在进化上具有紧密的关系。ITS谱系树表明赖草属内部存在大量的分化,以及赖草属物种具有多次起源的特征。trnL-F序列分析表明,赖草属物种的母本,部分来自Ns基因组,部分来自Xm基因组,这可能与赖草属物种的地理分布有关,分布于亚欧大陆的物种其母本是新麦草属,而分布于北美的大部分物种其母本是Xm基因组。trnL-F序列分析还表明,E. californicus和赖草属未知的基因组具有紧密的关系。以上研究结果表明:(1)从分子层面证明,赖草属的未知基因组并非来自薄冰草属,或是一个修正的新麦草基因组,其基因组组成应是NsNsXmXm;(2)赖草属物种的母本,部分来自Ns基因组,部分来自Xm基因组,这可能与赖草属物种的地理分布有关;(3)E. californicus的母本是Xm基因组,父本是Ns基因组,该物种应从披碱草属转移至赖草属。
