Haemolymph, heads, salivary glands, crops, midguts, hindguts, and Malpighian tubules from Rhodnius prolixus and Triatoma infestans were extracted in phosphate or Tris buffer saline with calcium, and tested for agglutination and lytic activities by microtitration against both vertebrateerythrocytes and cultured epimatigote forms of Trypanosoma rangeli. Haemagglutination activity against rabbit erythrocytes was found in the crop, midgut and hindgut extracts of T. infestans but only in the haemolymph of R. prolixus. Higher titres of parasite agglutinins were found in R. prolixus haemolymph than T. infestans, whilst the converse occurred for the tissue extracts. In addition, the extracts of T. infestans salivary glands, but not those of R. prolixus, showed a trypanolytic activity that was heat-inactivated and was not abolished by pre-incubation with any of the sugars or glycoproteins tested. T. infestans, which is refractory to infection by T. rangeli, thus appears to contain a much wider distribution of agglutinating and trypanolytic factors in its tissues than the more susceptible species, R. prolixus

The digestive tract of insects is an important natural, physical, and chemical defense barrier against pathogen invasion. Certain. lepidopteran caterpillars are serious pests of agricultural crops and their biology has received much attention, but little is known about the larval noctuid gut. The morphological analysis of the digestive tract in Anticarsia gemmatalis under scanning electron microscopy (SEM) is a good model for studies about its defense mechanism. The material was fixed (2,5% glutaraldehyde solution; 0.1 M-phosphate buffer, pH 7.3), post-fixed (1% osmium tetroxide in the same buffer), dried at critical point, gold coated and analyzed in a SEM 515-Philips. A. gemmatalis digestive tract consists of a straight duct of varying length and diameter, subdivided in three main regions: the foregut formed by the oral cavity, pharynx, esophagus, and crop; the midgut that is the largest portion of the digestive tract without noticeable morphological differentiation along its length; and the hindgut that is morphologically differentiated in pylorus, ileum, colon, and rectum. Although the general morphology of the A. gemmatalis digestive tract is quite similar to the other Lepidoptera species, the anatomical array of the crop muscular layers is quite different comparing with the description for other larval insect.

The digestive tube of 2nd and 3rd instar larvae, pupae and newly emerged adults of Dermatobia hominis (Linnaeus, 1781) was studied anatomically. The specimens were dissected in buffer saline under a stereomicroscope, and the digestive tubes were placed on slides and fixed in 10% buffered formalin. Each tube was measured using a micrometric eye piece, and drawings were made with camera lucida. The results showed that the midgut, the hindgut and the Malpighian tubules with their ducts grow gradually during the larval development. The oesophagus and the salivary glands with their ducts grow only during the moult from the 2nd to the 3rd instar. In the pupal period, salivary glands grow gradually but disappeared after the 20th day. After metamorphosis the digestive tube regressed. This is expected since adult D. hominis lives about nine days without feeding. This fly, similar to other calyptratae muscoid flies shows no vestige of a crop during all post-embrionic development, and the adult has no salivary glands.

O equino é um herbívoro não ruminante capaz de suprir grande parte ou a totalidade da sua demanda nutricional pela ingestão de gramíneas. Apresenta a região ceco-cólica bastante desenvolvida, sendo este o principal sítio de fermentação. Este processo também ocorre na região aglandular do estômago, porém a produção de ácidos graxos voláteis é inferior, quando comparado ao intestino grosso. O conhecimento do sítio de aproveitamento de cada ingrediente é de suma importância para a combinação dos mesmos, favorecendo o ótimo aproveitamento de cada um deles e evitando excessos que podem ser prejudiciais ao metabolismo do equino. O estudo atual do fracionamento de carboidratos, por mais que não seja adaptado a fisiologia e metabolismo dos equinos, nos fornece informações sobre as diferentes frações que o compõem e com isso pode-se inferir sobre a produção de energia por cada fração gerada. A manutenção de equinos em pastejo ou sob o fornecimento de feno ou alimentos volumosos é essencial para a manutenção da atividade e saúde do seu trato digestório. O aporte mínimo de 12% de FDN garante tal situação. Atualmente, com o objetivo de aumentar a disponibilidade de energia, ingredientes como a polpa cítrica, polpa de beterraba e casca de soja, vem sendo utilizados nas formulações. Estes ingredientes de fermentação mais rápida e fácil, são uma alternativa segura para elevar a concentração energética da dieta de equinos, por proporcionar a diminuição do aporte de amido na dieta de equinos. Sabe-se entretanto, que com a elevação na qualidade do volumoso, maiores concentrações de carboidratos solúveis e de rápida fermentação são encontrados disponíveis ao longo do trato digestório dos equinos levando a alguns distúrbios metabólicos como a laminite e a sensibilidade a insulina. Com o correto manejo dos animais em pastejo e conhecimento do ciclo da planta é possível minimizar tais problemas. Objetivou-se revisão da ação fisiológica e metabólica das diferentes frações fibrosas, em cada compartimento do trato digestório, bem como destacar os ingredientes volumosos com suas diversas frações, os quais possibilitam sugerir manejo alimentar adequado para os equinos.

Mosquito larvae are believed to be capable of digesting chitin, an insoluble polysaccharide of N-acetylglucosamine, for their nutritional benefit. Studies based on physiological and biochemical assays were conducted in order to detect the presence of chitinase activities in the gut of the detritus-feeding Aedes aegypti larvae. Larvae placed for 24 h in suspensions of chitin azure were able to digest the ingested chitin. Semi-denaturing PAGE using glycol chitin and two fluorogenic substrate analogues showed the presence of two distinct chitinase activities: an endochitinase that catalyzed the hydrolysis of chitin and an endochitinase that cleaved the short substrates [4MU(GlcNAc)(3)] and [4MU(GlcNAc)(2)] that hydrolyzed the chitobioside [4MU(GlcNAc)(2)]. The endochitinase had an extremely broad pH-activity against glycol chitin and chitin azure, pH ranging from 4.0 to 10.0. When the substrate [4MU(GlcNAc)(3)] was used, two activities were observed at pH ranges 4.0-6.0 and 8.0-10.0. Chitinase activity against [4MU(GlcNAc)(3)] was detected throughout the gut with the highest specific activity in the hindgut. The pH of the gut contents was determined by observing color changes in gut after feeding the larvae with color indicator dyes. It was observed a correlation between the pH observed in the gut of feeding larvae (pH 10-6.0) and the optimum pH for gut chitinase activities. In this work, we report that gut chitinases may be involved in the digestion of chitin-containing structures and also in the partial degradation of the chitinous peritrophic matrix in the hindgut. We report the identification of a potential pheromone for Gnathotrichus materiarius (Fitch) (Col., Scolytidae). The population sex ratio is close to 1:1, and males initiate attacks on host trees. Headspace and hindgut samples from single males showed the presence of the putative pheromone 6-methyl-5-hepten-2-ol, sulcatol. Unmated males released sulcatol for at least 12 days, and ceased producing the pheromone after 20 days. The peak sulcatol release occurred after 2 days. Males cease production of sulcatol 24 h after being paired with females. Single females were unable to initiate galleries, and no sulcatol was detected from their headspace and hindgut samples. The chiral ratio of the pheromone, observed from headspace samples, was 31% (S)-(+)- and 69% (R)-(-)-sulcatol.

The Malpighian tubules of workers of the fire ant Solenopsis saevissima (Myrmicinae) were analyzed by scanning and transmission electron microscopy in order to determine their functional organization and association with the hindgut epithelium. The ants showed six Malpighian tubules with three segments morphologically and structurally different. The proximal segment was long and its cells showed abundant smooth endoplasmic reticulum and lipid droplets, which suggest their role in lipid secretion. The mid segment was long and undulated and it was composed by the cells that showed the typical features of ion transporting epithelia. The distal segment, short and flattened, adheres to the rectum wall. The cells of this segment showed the basal lamina fused to that of the rectum, it is probable that this part of the tubule may play a role in ion and water uptake from the feces. Symbiotische Mikroorganismen aus dem Termitendarm Es ist eine bekannte Tatsache, dass die Hauptaufgabe der Darmflora der niederen Termite im Abbau von Holz liegt. Im Laufe der Millionen Jahre alten Entwicklung der Termiten hat sich in ihrem Darm ein Ökosystem aufgebaut, das aus Protozoen, Archaeen, Bakterien und Hefen besteht. Ziel der vorliegenden Arbeit war die teilweise Erforschung der symbiotischen Zusammenhänge in diesem ökologischen System. Es wurden dabei zwei Gebiete genauer untersucht: Cellulolytische Bakterien im Darm von niederen Termiten Die bisher vorherrschende Meinung sah in den Protozoen die Hauptkomponenten des Celluloseabbaus in Termiten. In dieser Arbeit gelang es 164 cellulolytische Bakterienstämme aus sieben niederen Termitenarten zu isolieren und zu identifizieren. Diese Vielzahl cellulolytischer Bakterienarten könnte ein Indiz dafür sein, dass Bakterien beim Holzabbau von Termiten eine effizientere Rolle spielen als angenommen. Oberflächenbakterien von Mixotricha paradoxa, einem Flagellaten aus dem Darm der niederen Termite Mastotermes darwiniensisMixotricha paradoxa ist ein Beispiel der seltenen Form einer Bewegungssymbiose zwischen Protozoen und Bakterien. Der Flagellat wird von Spirochäten, die auf seiner Oberfläche befestigt sind, vorangetrieben. Zusätzlich leben noch stäbchenförmige Bakterien auf der Hülle. Drei Spirochätenarten und das stäbchenförmige Bakterium konnten identifiziert und lokalisiert werden. Es wird angenommen, dass alle drei Spirochätenarten Mitglieder der Bewegungssymbiose sind.

Termiten beherbergen in ihrem Darm eine einzigartige Flora aus Bakterien, Archaeen, Flagellaten und Hefen. Diese symbiontische mikrobielle Gemeinschaft ist am Abbau von komplexen organischen Verbindungen beteiligt und ermöglicht es den Termiten schwer abbaubares Material wie Holz als Nahrungsquelle zu nutzen. Spirochaeten, eine Gruppe beweglicher Bakterien die sich durch ihre besondere Morphologie und Art der Fortbewegung von allen anderen Mikroorganismen abgrenzen lassen, gehören zu den häufigsten Bakterien im Termitendarm. Ziel der Arbeit war die Isolierung und Charakterisierung bislang unbekannter Spirochaeten aus Termitendärmen. Aus drei niederen Termitenarten konnten sechs spirochaetale Stämme gewonnen und identifiziert werden. Die Isolate ließen sich anhand der 16S rRNA Gensequenzen den Gattungen Treponema und Spirochaeta zuordnen. Im Gegensatz zu allen bislang charakterisierten Spirochaeten zeigte der Stamm SPN1 aus der Termite Neotermes castaneus eine kokkoide Zellform und war unbeweglich. Der Organismus wurde daher als neue Art, Spirochaeta coccoides sp. nov., beschrieben. Bei allen gewonnenen Isolaten handelt es sich um strikt anaerobe Organismen die verschiedene Mono-, Di- und Oligosaccharide fermentieren. Als wesentliche Stoffwechselprodukte konnten Acetat und Ethanol (sowie Formiat bei einem Stamm) identifiziert werden. Weiterhin konnten bei den untersuchten Stämmen eine Reihe von enzymatischen Aktivitäten nachgewiesen werden, die für den Abbau von Lignocellulose im Termitendarm von Bedeutung sind. Die Untersuchungen deuten darauf hin, dass die Spirochaeten eine wichtige Rolle bei der Fermentation von Abbauprodukten der Lignocellulose im Termitendarm spielen.

Beta1-integrins (beta1) represent cell surface receptors which mediate cell-matrix and cell-cell interactions. Fässler and Meyer described chimeric mice containing transgenic cells that express the LacZ gene instead of the beta1 gene. They observed beta1-negative cells in all germ layers at embryonic day E 8.5. Later in development, using a glucose phosphate isomerase assay of homogenized tissue samples, high levels of transgenic cells were found in skeletal muscle and gut, low levels in lung, heart, and kidney and none in the liver and spleen (Fässler and Meyer 1995). In order to study which cell types require beta1 during development of the primitive gut including its derivatives, chimeric fetuses containing 15 to 25% transgenic cells were obtained at days E 14.5 and E 15.5. They were LacZ (beta-galactosidase) stained "en bloc" and cross-sectioned head to tail. In esophagus, trachea, lung, stomach, hindgut, and the future urinary bladder, we observed various mesoderm-derived beta1-negative cells (e.g. fibroblasts, chondrocytes, endothelial cells, and smooth muscle cells) but no beta1-negative epithelial cells. Since the epithelia of lung, esophagus, trachea, stomach, hindgut, and urinary bladder are derived from the endodermal gut tube, we hypothesize that beta1 is essential for the development and/or survival of the epithelia of the fore- and hindgut and its derivatives.
