The acetic acid and phenyl-p-benzoquinone are easy and fast screening models to access the activity of novel candidates as analgesic drugs and their mechanisms. These models induce a characteristic and quantifiable overt pain-like behavior described as writhing response or abdominal contortions. The knowledge of the mechanisms involved in the chosen model is a crucial step forward demonstrating the mechanisms that the candidate drug would inhibit because the mechanisms triggered in that model will be addressed. Herein, it was investigated the role of spinal mitogen-activated protein (MAP) kinases ERK (extracellular signal-regulated kinase), JNK (Jun N-terminal Kinase) and p38, PI3K (phosphatidylinositol 3-kinase) and microglia in the writhing response induced by acetic acid and phenyl-p-benzoquinone, and flinch induced by formalin in mice. Acetic acid and phenyl-p-benzoquinone induced significant writhing response over 20 min. The nociceptive response in these models were significantly and in a dose-dependent manner reduced by intrathecal pre-treatment with ERK (PD98059), JNK (SB600125), p38 (SB202190) or PI3K (wortmannin) inhibitors. Furthermore, the co-treatment with MAP kinase and PI3K inhibitors, at doses that were ineffective as single treatment, significantly inhibited acetic acid- and phenyl-p-benzoquinone-induced nociception. The treatment with microglia inhibitors minocycline and fluorocitrate also diminished the nociceptive response. Similar results were obtained in the formalin test. Concluding. MAP kinases and PI3K are important spinal signaling kinases in acetic acid and phenyl-p-benzoquinone models of overt pain-like behavior and there is also activation of spinal microglia indicating that it is also important to determine whether drugs tested in these models also modulate such spinal mechanisms. Kaurenoic acid [ent-kaur-16-en-19-oic acid (1)] is a diterpene present in several plants including Sphagneticola trilobata. The only documented evidence for its antinociceptive effect is that it inhibits the writhing response induced by acetic acid in mice. Therefore, the analgesic effect of 1 in different models of pain and its mechanisms in mice were investigated further. Intraperitoneal and oral treatment with 1 dose-dependently inhibited inflammatory nociception induced by acetic acid. Oral treatment with 1 also inhibited overt nociception-like behavior induced by phenyl-p-benzoquinone, complete Freund's adjuvant (CFA), and both phases of the formalin test. Compound 1 also inhibited acute carrageenin- and PGE(2)-induced and chronic CFA-induced inflammatory mechanical hyperalgesia. Mechanistically, 1 inhibited the production of the hyperalgesic cytokines TNF-alpha and IL-1 beta. Furthermore, the analgesic effect of 1 was inhibited by L-NAME, ODQ, KT5823, and glybenclamide treatment, demonstrating that such activity also depends on activation of the NO-cyclic GMP-protein kinase G-ATP-sensitive potassium channel signaling pathway, respectively. These results demonstrate that 1 exhibits an analgesic effect in a consistent manner and that its mechanisms involve the inhibition of cytokine production and activation of the NO-cyclic GMP-protein lcinase G-ATP-sensitive potassium channel signaling pathway.

Spiranthera odoratissima A. St.-Hil., 'manaca', is a medicinal species used in Brazil, especially in central region, for the treatment of several diseases such as pain and inflammation. In this study, the methanol/aqueous phase of the ethanol extract of the leaves of 'manaca' (MAP), at the doses of 50, 150 and 500 mg/kg was used to evaluate the anti-inflammatory and/or antinociceptive effects and the possible anti-inflammatory mechanism. The antinociceptive and anti-inflammatory activities of MAP were assessed using formalin test, carrageenan-induced paw oedema. The myeloperoxidase activity, capillary permeability, leukocyte migration and tumour necrosis factor alpha (TNF-alpha) levels were evaluated in pleural exudate. The MAP reduced the licking time only in the later phase of formalin test, and showed anti-inflammatory activity by reducing the paw oedema, migration cell, myeloperoxidase activity, capillary permeability and TNF-alpha levels. In conclusion, we confirmed the inflammatory activity of MAP and affirm that this effect involves the reduction of TNF-alpha level.

A ligand-based drug design study was performed to acetaminophen regioisomers as analgesic candidates employing quantum chemical calculations at the DFT/B3LYP level of theory and the 6-31G* basis set. To do so, many molecular descriptors were used such as highest occupied molecular orbital, ionization potential, HO bond dissociation energies, and spin densities, which might be related to quench reactivity of the tyrosyl radical to give N-acetyl-p-benzosemiquinone-imine through an initial electron withdrawing or hydrogen atom abstraction. Based on this in silico work, the most promising molecule, orthobenzamol, was synthesized and tested. The results expected from the theoretical prediction were confirmed in vivo using mouse models of nociception such as writhing, paw licking, and hot plate tests. All biological results suggested an antinociceptive activity mediated by opioid receptors. Furthermore, at 90 and 120 min, this new compound had an effect that was comparable to morphine, the standard drug for this test. Finally, the pharmacophore model is discussed according to the electronic properties derived from quantum chemistry calculations.

Ethnopharmacological relevance: The pharmacological activity of geopropolis collected by stingless bees (important and threatened pollinators), a product widely used in folk medicine by several communities in Brazil, especially in the Northeast Region, needs to be studied. Objective: The aim of this study was to evaluate the antinociceptive activity of Melipona scutellaris geopropolis (stingless bee) using different models of nociception. Material and methods: The antinociceptive activity of the ethanolic extract of geopropolis (EEGP) and fractions was evaluated using writhing induced by acetic acid, formalin test, carrageenan-induced hypernociception, and quantification of IL-1 beta and TNF-alpha. The chemical composition was assessed by quantification of total flavonoids and phenolic compounds. Results: EEGP and its hexane and aqueous fractions showed antinociceptive activity. Both EEGP and its aqueous fraction presented activity in the mechanical inflammatory hypernociception induced by the carrageenan model, an effect mediated by the inhibition of IL-1 beta and TNF-alpha. The chemical composition of EEGP and its hexane and aqueous fractions showed a significant presence of phenolic compounds and absence of flavonoids. Conclusion: Our data indicate that geopropolis is a natural source of bioactive substances with promising antinociceptive activity. (C) 2012 Elsevier Ireland Ltd. All rights reserved.

Abstract Background Guava pomace is an example of the processing waste generated after the manufacturing process from the juice industry that could be a source of bioactives. Thus, the present investigation was carried out in order to evaluate the anti-inflammatory and antinociceptive potential and determinate the main phenolic compounds of a guava pomace extract (GPE). Methods The anti-inflammatory activity was evaluated by carrageenan, dextran, serotonin, histamine-induced paw edema and neutrophils migration in the peritoneal cavity models. Acetic acid-induced abdominal writhing and formalin test were performed to investigate the antinociceptive effects. In addition, the content of total phenolic and of individual phenolic compounds was determined by GC/MS. Results GPE showed anti-inflammatory activity by carrageenan, dextran, serotonin, histamine-induced paw edema and neutrophils migration in the peritoneal cavity models (p < 0.05). GPE also demonstrated antinociceptive activity by acetic acid-induced abdominal writhing and formalin test (p < 0.05). The total phenolic value was 3.40 ± 0.09 mg GAE/g and epicatechin, quercetin, myricetin, isovanilic and gallic acids were identified by GC/MS analysis. Conclusions The presence of bioactive phenolic compounds as well as important effects demonstrated in animal models suggest that guava pomace could be an interesting source of anti-inflammatory and analgesic substances.

Oggetto di studio in questa tesi è stato il ruolo modulatorio svolto dal neuropeptide nocicettina/orfanina FQ a carico della trasmissione nocicettiva. A scopo introduttivo, sono state illustrate le conoscenze attuali sul sistema nocicettina-NOP; sono state descritte le funzioni, la struttura e la distribuzione del recettore NOP, le azioni farmacologiche finora note e la distribuzione della nocicettina stessa al livello del S.N.C. e in periferia. Lo studio è stato condotto principalmente con due approcci differenti A) E’ stata studiata la capacità della nocicettina esogena o di suoi analoghi agonisti e antagonisti, di modificare la trasmissione nocicettiva. B) Sono state studiate le variazioni a carico del sistema endogeno nocicettina/recettore NOP in seguito a trattamenti di tipo farmacologico. A) E’ stata indagata la capacità della nocicettina e degli analoghi sintetici [Arg14, Lys15]N/OFQ e UFP-101 di modificare la soglia nocicettiva nel ratto, rilevata con il test del tail-flick, a seguito di somministrazione diretta nello spazio subaracnoideo, in confronto con la nocicettina stessa. La somministrazione intratecale del neuropeptide nocicettina (10 nmol/ratto) ha determinato un innalzamento statisticamente significativo delle latenze di risposta al test del tail-flick. L’analogo [Arg14, Lys15]N/OFQ è stato somministrato alla dose di 1 nmole/ratto i.t. provocando un innalzamento massimale delle soglie di latenza per tutto il periodo di osservazione, mentre alla dose 0,2 nmoli/ratto i.t ha provocato un effetto antinocicettivo sottomassimale pur dimostrandosi significativo rispetto ai controlli (p < 0,05 vs controlli a tutti i tempi di rilevazione). Il composto antagonista UFP-101 è risultato capace di antagonizzare l’azione sulla soglia analgesica sia della nocicettina sia dell’analogo [Arg14, Lys15]N/OFQ nel suo dosaggio minore, mentre contro la dose di 1 nmole/ratto i.t ha prodotto solamente una riduzione di effetto. Anche la somministrazione intratecale di MAP-N/OFQ si è dimostrata in grado di modificare la soglia nocicettiva determinata mediante il test del tail-flick, nel ratto, in modo dose dipendente. differentementeuna seconda somministrazione di MAP-N/OFQ dopo 24 ore, si è dimostrata totalmente inefficace nel modificare la soglia nocicettiva nei ratti precedentemente trattati, pur permanendo la loro suscettibilità all’azione analgesica della morfina, mostrando quindi il rapido sviluppo di tolerance al potente peptide nocicettinergico somministrato per via i.t.. Inoltre l’antagonista UFP-101 oltre ad essere ingrado di antagonizzare l’effetto della MAP-N/OFQ, ha mostrato la capacità di ridurre la tolerance sviluppata nei confronti del dendrimero. La somministrazione di MAP-N/OFQ per via i.c.v. ha prodotto variazione della soglia nocicettiva, producendo un innalzamento del volore soglia, dato contrastante con la maggior parte dei dati riguardanti la nocicettina in letteratura. Ha invece replicato l’effetto di antagonismo funzionale nei confronti della morfina, la quale dopo somministrazione di MAP-N/OFQ è risultata essere incapace di modificare la soglia nocicettiva nel ratto. Tale effetto perdura dopo 24 ore, quando una somministrazione di morfina produce un effetto analgesico inversamente proporzionale alla dose ricevuta di MAP-N/OFQ 24 ore prima. E’stato indagato il possibile ruolo neuromodulatorio del neuropeptide nocicettina esogeno, nell’analgesia prodotta da un farmaco di natura non oppiacea. In tal senso si è proceduto ad indagare l’eventuale capacità della nocicettina esogena, somministrata per via intracerebroventricolare e del suo analogo [Arg14, Lys15]N/OFQ, di antagonizzare l’analgesia prodotta dal farmaco paracetamolo. La nocicettina ha evidenziato la capacità di antagonizzare il potere antinocicettivo del paracetamolo fino a bloccarne completamente l’effetto al dosaggio più elevato, mostrando quindi proprietà antagonista dose-dipendente. Inoltre l’UFP-101, che di per se non altera l’analgesia indotta da paracetamolo, è ingrado di antagonizzare l’effetto della nocicettina sul paracetamolo in maniera dose-dipendente. Medesimo è risultato il comportamento dell’analogo della nocicettina, la Arg-Lys nocicettina. B) Sono state indagate le relazioni tra il sistema nocicettina/NOP e le proprietà farmacologiche di un noto farmaco oppiaceo quale la buprenorfina, le cui peculiari caratteristiche farmacodinamiche sano state recentemente collegate alla sua capacità di agire come agonista diretto al recettore NOP. In tal senso si è proceduto ad osservare l’effetto della somministrazione di buprenorfina sull’ assetto recettoriale di NOP, inseguito ad un trattamento prolungato con somministrazione sottocutanea mediante minipompe osmotiche nel ratto, rilevando successivamente, tramite uno studio di binding, le variazioni della densità recettoriale di NOP in alcune aree di interesse per la trasmissione nocicettiva. Sia nell’ippocampo che nel talamo e nella frontal cortex, la somministrazione prolungata di buprenorfina ha causato una riduzione significativa della densità recettoriale di NOP. Come ultimo aspetto indagato, al fine di determinare la presenza del neuropeptide nel liquido cerebrospinale e le sue eventuali modificazioni a seguito di manipolazioni farmacologiche e non farmacologiche, è stata messa a punto una metodica di perfusione dello spazio subaracnoideo nel ratto, che consentisse di ottenere materiale biologico su cui compiere la ricerca e quantificazione della presenza di nocicettina mediante dosaggio radioimmunologico. La perfusione di CSF artificiale arricchito di ione potassio ad una concentrazione pari a 60 mM ha evidenziato la possibilità di stimolare la liberazione della nocicettina nel liquido cerebrospinale di ratto, suggerendo quindi una sua provenienza da elementi eccitabili. E’ stato quindi possibile osservare l’andamento dei livelli di peptide a seguito della stimolazione nocicettiva prodotta da due agenti irritanti con caratteristiche differenti, la carragenina e la formalina. La somministrazione sottocutanea di carragenina (100 µl al 3 %) nella regione subplantare di entrambe le zampe posteriori del ratto non ha determinato alterazioni significative dei livelli di neuropeptide. Invece, la somministrazione di formalina (50 µl al 5 %), dopo un iniziale periodo di 30 minuti, ha causato un incremento significativo della liberazione di N/OFQ a partire dal terzo intervallo di raccolta seguente la somministrazione della sostanza. Questo rispecchia l’andamento di risposta al formalin test ottenuto anche mediante test di natura differente dagli analgesimetrici (es. comportamentale, elettrofisiologico), in quest’ottica l’aumento di nocicettina può essere interpretato come un evento dovuto alla sensibilizzazione centrale all’effetto pronocicettivo.

The widespread plant volatile beta-caryophyllene (BCP) was recently identified as a natural selective agonist of the peripherally expressed cannabinoid receptor 2 (CB2). It is found in relatively high concentrations in many spices and food plants. A number of studies have shown that CB2 is critically involved in the modulation of inflammatory and neuropathic pain responses. In this study, we have investigated the analgesic effects of BCP in animal models of inflammatory and neuropathic pain. We demonstrate that orally administered BCP reduced inflammatory (late phase) pain responses in the formalin test in a CB2 receptor-dependent manner, while it had no effect on acute (early phase) responses. In a neuropathic pain model the chronic oral administration of BCP attenuated thermal hyperalgesia and mechanical allodynia, and reduced spinal neuroinflammation. Importantly, we found no signs of tolerance to the anti-hyperalgesic effects of BCP after prolonged treatment. Oral BCP was more effective than the subcutaneously injected synthetic CB2 agonist JWH-133. Thus, the natural plant product BCP may be highly effective in the treatment of long lasting, debilitating pain states. Our results have important implications for the role of dietary factors in the development and modulation of chronic pain conditions.

Oxidative refolding is one of the key challenges hampering the development of peptide based compounds as therapeutics. The correct refolding for three disulfide peptide like w-Conotoxi n MVIIA is difficult and crucial for biological activity. This work advanced knowledge of chemical and biological for improve oxidative refolding of synthetic w-Conotoxi n MVIIA in base of Conus magus venom. The present study aimed to set up an appropriate and effective protocols for refolding of disulfide-rich w-Conotoxin MVIIA. In this study, the crude peptide was protected with Acm group, according to the right amino acid sequences (Synthesized by Australian Company). The crude peptide was purified by H PLC. To prepare the peptide to refolding, innovative deprotection applied molar ratio (AMR) method was performed based on mercury. Accuracy of deprotection was approved by reverse phase chromatography. The deprotected target peptide (omega-conotoxin) was determined by SDS-PAGE. Then the Oxidative refolding of target peptide was performed in six protocol based on Guanidinium chloride and oxidized and reduced Glutathione. Analgesic effect of refolded peptide was surveyed with formalin test in mice Balb/c. Non neurotoxic effects of target peptides were survey with ICV injection in mice model (C57/BL6). The innovative deprotection protocol performed based on the best ratio of mercury/2-mercaptoethanol adjusted to 1mg/10p1 in 90 minute. The results showed the yield and purity of omega-conotoxin MVIIA as 93 and 95%, respectively. Refolding of 40 mg omega Conotoxin with GSSG and GSH on ratio of 10:1 and 20 mM ammonium acetate showed the best analgesic effect compared with the other methods. The result showed 95.5% yield and 98% purity of omega-conotoxin MVIIA in this refolding method. Related refolding method reduced 85% pain in experimented mice using 7 ng of the peptide. That was 71.5 fold stronger than morphine and 2 times than standard Prialt®. And it was not neurotoxic in mice. In this study, refolding method for omega-conotoxin MVIIA was optimized in the fourth factor including: reducing the time, amount and number of reagent and increase the efficiency. We introduced new method for deprotection of omega-conotoxin MVIIA. Effective, economic and applied refolding and deprotecti on method was performed in this research may al so be applied to similar omega conotoxin peptides.

The aim of this study was to evaluate the application of PCR technique for the detection of BoHV-5 in routinely formalin-fixed, paraffin-embedded brain tissues in 20 naturally infected calves affected by fatal meningoencephalitis. Brains were divided into two halves, one kept fresh for virus isolation and PCR assay, targeting the glycoprotein C gene from BoHV-5 genome. The other half brain, corresponding to posterior cortex region, was submitted to formalin fixation and embedded into paraffin blocks for microscopic evaluation and total DNA isolation. Most of the slides showed severe multifocal non-supurative encephalitis with neuronal degeneration, neurophagia, and no acidophilic intranuclear inclusions could be found in neurons and glial. The 20 fresh samples were confirmed, by virus isolation and PCR assay, as having the BoHV-5 virus and, respective glicoprotein C sequence, while 15 of 20 formalin-fixed, paraffin-embedded samples were considered positive for the same analysis. The results revealed the first description of PCR efficiency, applied to formalin-fixed, paraffin-embedded brain collected from naturally infected calves, improving the detection of BoHV-5 from archival samples in South America.
