The State of Paraíba is one of the most dynamic states of Brazil, strategically located in the northeast, is notable for the excellent potential for integration of different transportation modes forming the states of Rio Grande do Norte, Pernambuco and Alagoas. The dynamic that occurs with port activity causes changes in the space where it is installed. And the elements of this space are always more than suffering direct or indirect influences as the flow in the port is expanded. Therefore, this region became subject to the accidental spillage of oil, because it presents a heavy traffic of ships of various sizes that can run aground or collide with oil causing accidental events. The study of geomorphological and sedimentological compositions of seafloor becomes important as more is known about the relationships between these parameters and associated fauna, and can identify their preferred habitats. The database background, acoustically collected along the proposed study area, is a wealth of information, which were duly examined, cataloged and made available. Such information can serve as an important tool, providing a geomorphological survey of the sedimentary area studied, and come to subsidize, in a flexible, future decision making. With the study area Port of Cabedelo, Paraíba - Brazil, this research aimed to evaluate the influence of the tidal surface and background in modeling the seabed, including the acquisition of information about the location of submerged rocky bodies and the depth of these bodies may turn out to be natural traps for the trapping of oil in case of leaks, and obtain the relationship between types of bed and the hydrodynamic conditions present in the region. In this context, for this study were collected bathymetric data (depth) and physical oceanographic (height of water column, water temperature, intensity and direction of currents, waves and turbidity), meteorological (rainfall, air temperature, humidity, winds and barometric pressure) of the access channel to the Port of Cabedelo / PB and its basin evolution (where the cruise ships dock), and includes tools of remote sensing (Landsat 7 ETM +, 2001), so that images and the results are integrated into Geographic Information Systems and used in the elaboration of measures aimed at environmental protection areas under the influence of this scale facilities, serving as a grant to prepare a contingency plan in case of oil spills in the region. The main findings highlight the techniques of using hydroacoustic data acquisition together bathymetric surveys of high and low frequency. From there, five were prepared in bathymetric pattern of Directorate of Hydrography and Navigation - DHN, with the depth in meters, on a scale of 1:2500 (Channel and Basin Evolution of Access to Port of Cabedelo), where there is a large extent possible beachrocks that hinder the movement of vessels in the port area, which can cause collisions, running aground and leaking oil. From the scatter diagram of the vectors of currents, it can be seen as the tidal stream and undergoes a channeling effect caused by the bidirectional effect of the tide (ebb and flood) in the basin of the Port of Cabedelo evolution in NW-direction SE and the highest speed of the currents occurs at low tide. The characterization weather for the period from 28/02 to 04/07/2010 values was within the expected average for the region of study. The multidisciplinary integration of products (digital maps and remote sensing images), proved to be efficient for the characterization of underwater geomorphological study area, reaching the aim to discriminate and enhance submerged structures, previously not visible in the images

In Fazenda Belém oil field (Potiguar Basin, Ceará State, Brazil) occur frequently sinkholes and sudden terrain collapses associated to an unconsolidated sedimentary cap covering the Jandaíra karst. This research was carried out in order to understand the mechanisms of generation of these collapses. The main tool used was Ground Penetrating Radar (GPR). This work is developed twofold: one aspect concerns methodology improvements in GPR data processing whilst another aspect concerns the geological study of the Jandaíra karst. This second aspect was strongly supported both by the analysis of outcropping karst structures (in another regions of Potiguar Basin) and by the interpretation of radargrams from the subsurface karst in Fazenda Belém. It was designed and tested an adequate flux to process GPR data which was adapted from an usual flux to process seismic data. The changes were introduced to take into account important differences between GPR and Reflection Seismic methods, in particular: poor coupling between source and ground, mixed phase of the wavelet, low signal-to-noise ratio, monochannel acquisition, and high influence of wave propagation effects, notably dispersion. High frequency components of the GPR pulse suffer more pronounced effects of attenuation than low frequency components resulting in resolution losses in radargrams. In Fazenda Belém, there is a stronger need of an suitable flux to process GPR data because both the presence of a very high level of aerial events and the complexity of the imaged subsurface karst structures. The key point of the processing flux was an improvement in the correction of the attenuation effects on the GPR pulse based on their influence on the amplitude and phase spectra of GPR signals. In low and moderate losses dielectric media the propagated signal suffers significant changes only in its amplitude spectrum; that is, the phase spectrum of the propagated signal remains practically unaltered for the usual travel time ranges. Based on this fact, it is shown using real data that the judicious application of the well known tools of time gain and spectral balancing can efficiently correct the attenuation effects. The proposed approach can be applied in heterogeneous media and it does not require the precise knowledge of the attenuation parameters of the media. As an additional benefit, the judicious application of spectral balancing promotes a partial deconvolution of the data without changing its phase. In other words, the spectral balancing acts in a similar way to a zero phase deconvolution. In GPR data the resolution increase obtained with spectral balancing is greater than those obtained with spike and predictive deconvolutions. The evolution of the Jandaíra karst in Potiguar Basin is associated to at least three events of subaerial exposition of the carbonatic plataform during the Turonian, Santonian, and Campanian. In Fazenda Belém region, during the mid Miocene, the Jandaíra karst was covered by continental siliciclastic sediments. These sediments partially filled the void space associated to the dissolution structures and fractures. Therefore, the development of the karst in this region was attenuated in comparison to other places in Potiguar Basin where this karst is exposed. In Fazenda Belém, the generation of sinkholes and terrain collapses are controlled mainly by: (i) the presence of an unconsolidated sedimentary cap which is thick enough to cover completely the karst but with sediment volume lower than the available space associated to the dissolution structures in the karst; (ii) the existence of important structural of SW-NE and NW-SE alignments which promote a localized increase in the hydraulic connectivity allowing the channeling of underground water, thus facilitating the carbonatic dissolution; and (iii) the existence of a hydraulic barrier to the groundwater flow, associated to the Açu-4 Unity. The terrain collapse mechanisms in Fazenda Belém occur according to the following temporal evolution. The meteoric water infiltrates through the unconsolidated sedimentary cap and promotes its remobilization to the void space associated with the dissolution structures in Jandaíra Formation. This remobilization is initiated at the base of the sedimentary cap where the flow increases its abrasion due to a change from laminar to turbulent flow regime when the underground water flow reaches the open karst structures. The remobilized sediments progressively fill from bottom to top the void karst space. So, the void space is continuously migrated upwards ultimately reaching the surface and causing the sudden observed terrain collapses. This phenomenon is particularly active during the raining season, when the water table that normally is located in the karst may be temporarily located in the unconsolidated sedimentary cap

Oriented LiNbO3 thin films were prepared using a polymeric precursor solution deposited on (0001) sapphire substrate by spin coating and crystallized in a microwave oven. Crystallization of the films was carried out in a domestic microwave oven. The influence of this type of heat treatment on the film orientation was analyzed by X-ray diffraction and electron channeling patterns, which revealed epitaxial growth of films crystallized at 550 and 650 degreesC for 10 min. A microstructural study indicated that the films treated at temperatures below 600 degreesC were homogeneous and dense, and the optical properties confirmed the good quality of these films. The indwelling urethral catheter has an important role for patients with urinary retention, bladder obstruction, neurological damage and other diseases. Urine contains minerals which precipitate in alkaline pH, crystallize and block the urological catheter. The crystallization of the ionic components of urine occurs in the presence of urease, an enzyme produced by Proteus mirabilis. This bacterium adheres to inanimate surfaces and forms biofilms. The aim of this study was to investigate the formation of crystalline biofilm on the luminal surface of siliconized latex catheters by means of scanning electron microscope, after channeling artificial urine infected with Proteus mirabilis. The experiment was performed in vitro using a dynamic flow system. The artificial urine compounds were salts of calcium, magnesium, phosphates, urea and egg albumin, and it was infected with Proteus mirabilis ATCC 25933. The urine flow was stopped after crystallization of the ionic components. Crystallization was observed after alkalinization of urine. Scanning electron microscopy showed the presence of crystals and morphologies typical of bacilli embedded in an amorphous mass on the internal lumen of the catheter. The present study showed that catheter encrustation may limit the use of long-term indwelling catheter. © SBEB - Sociedade Brasileira de Engenharia Biomédica.

Comunidades vulneráveis estão sujeitas aos impactos causados por eventos climáticos e aos efeitos nocivos que eles provocam como perturbar o funcionamento da sociedade e a possível ocorrência de perdas materiais, econômicas e sociais. Quanto maior a capacidade de adaptar-se, menores as vulnerabilidades. Em Belém, a presença de cursos hídricos dentro da área urbana e a ocupação irregular ao longo das faixas marginais evidenciam os fatores que levam às frequentes enchentes na região. Provavelmente, as enchentes tenham aumentado a partir da postura das instituições públicas, que favoreceram a ocupação de áreas alagáveis da cidade através dos aterramentos e canalização de igarapés. Dessa forma, o processo de periferização foi intensificado nas áreas de várzea e o espaço, profundamente alterado pela ocupação das áreas não edificáveis, como as bordaduras dos cursos d’água urbanos. Em função disso, a pesquisa objetivou definir a vulnerabilidade e a adaptação autônoma aos impactos das inundações considerando indicadores sociais, habitacionais e de saneamento. Foram analisadas as formas estruturais de adaptação, a partir da observação das edificações, suas tipologias e técnicas construtivas. Para tal, a metodologia para a realização do trabalho foi dividida em três etapas, que compreendem a delimitação da área de estudo; a descrição das medidas autônomas contra inundações e a análise da população como produtora destas medidas. Os resultados retratam a tentativa da comunidade de diminuir as perdas sobre algo que a representa valor, ou seja, as tomadas de decisão de adaptação quanto à capacidade e ao tipo de ação da população, onde as medidas adaptativas refletem não somente as habilidades da comunidade, mas uma série de outros indicadores sociais, econômicos e cognitivos.

La catena respiratoria mitocondriale è principalmente costituita da proteine integrali della membrana interna, che hanno la capacità di accoppiare il flusso elettronico, dovuto alle reazioni redox che esse catalizzano, al trasporto di protoni dalla matrice del mitocondrio verso lo spazio intermembrana. Qui i protoni accumulati creano un gradiente elettrochimico utile per la sintesi di ATP ad opera dell’ATP sintasi. Nonostante i notevoli sviluppi della ricerca sulla struttura e sul meccanismo d’azione dei singoli enzimi della catena, la sua organizzazione sovramolecolare, e le implicazioni funzionali che ne derivano, rimangono ancora da chiarire in maniera completa. Da questa problematica trae scopo la presente tesi volta allo studio dell’organizzazione strutturale sovramolecolare della catena respiratoria mediante indagini sia cinetiche che strutturali. Il modello di catena respiratoria più accreditato fino a qualche anno fa si basava sulla teoria delle collisioni casuali (random collision model) che considera i complessi come unità disperse nel doppio strato lipidico, ma collegate funzionalmente tra loro da componenti a basso peso molecolare (Coenzima Q10 e citocromo c). Recenti studi favoriscono invece una organizzazione almeno in parte in stato solido, in cui gli enzimi respiratori si presentano sotto forma di supercomplessi (respirosoma) con indirizzamento diretto (channeling) degli elettroni tra tutti i costituenti, senza distinzione tra fissi e mobili. L’importanza della comprensione delle relazioni che si instaurano tra i complessi , deriva dal fatto che la catena respiratoria gioca un ruolo fondamentale nell’invecchiamento, e nello sviluppo di alcune malattie cronico degenerative attraverso la genesi di specie reattive dell’ossigeno (ROS). E’ noto, infatti, che i ROS aggrediscono, anche i complessi respiratori e che questi, danneggiati, producono più ROS per cui si instaura un circolo vizioso difficile da interrompere. La nostra ipotesi è che, oltre al danno a carico dei singoli complessi, esista una correlazione tra le modificazioni della struttura del supercomplesso, stress ossidativo e deficit energetico. Infatti, la dissociazione del supercomplesso può influenzare la stabilità del Complesso I ed avere ripercussioni sul trasferimento elettronico e protonico; per cui non si può escludere che ciò porti ad un’ulteriore produzione di specie reattive dell’ossigeno. I dati sperimentali prodotti a sostegno del modello del respirosoma si riferiscono principalmente a studi strutturali di elettroforesi su gel di poliacrilammide in condizioni non denaturanti (BN-PAGE) che, però, non danno alcuna informazione sulla funzionalità dei supercomplessi. Pertanto nel nostro laboratorio, abbiamo sviluppato una indagine di tipo cinetico, basata sull’analisi del controllo di flusso metabolico,in grado di distinguere, funzionalmente, tra supercomplessi e complessi respiratori separati. Ciò è possibile in quanto, secondo la teoria del controllo di flusso, in un percorso metabolico lineare composto da una serie di enzimi distinti e connessi da intermedi mobili, ciascun enzima esercita un controllo (percentuale) differente sull’intero flusso metabolico; tale controllo è definito dal coefficiente di controllo di flusso, e la somma di tutti i coefficienti è uguale a 1. In un supercomplesso, invece, gli enzimi sono organizzati come subunità di una entità singola. In questo modo, ognuno di essi controlla in maniera esclusiva l’intero flusso metabolico e mostra un coefficiente di controllo di flusso pari a 1 per cui la somma dei coefficienti di tutti gli elementi del supercomplesso sarà maggiore di 1. In questa tesi sono riportati i risultati dell’analisi cinetica condotta su mitocondri di fegato di ratto (RLM) sia disaccoppiati, che accoppiati in condizioni fosforilanti (stato 3) e non fosforilanti (stato 4). L’analisi ha evidenziato l’associazione preferenziale del Complesso I e Complesso III sia in mitocondri disaccoppiati che accoppiati in stato 3 di respirazione. Quest’ultimo risultato permette per la prima volta di affermare che il supercomplesso I+III è presente anche in mitocondri integri capaci della fosforilazione ossidativa e che il trasferimento elettronico tra i due complessi possa effettivamente realizzarsi anche in condizioni fisiologiche, attraverso un fenomeno di channeling del Coenzima Q10. Sugli stessi campioni è stata eseguita anche un analisi strutturale mediante gel-elettroforesi (2D BN/SDS-PAGE) ed immunoblotting che, oltre a supportare i dati cinetici sullo stato di aggregazione dei complessi respiratori, ci ha permesso di evidenziare il ruolo del citocromo c nel supercomplesso, in particolare per il Complesso IV e di avviare uno studio comparativo esteso ai mitocondri di cuore bovino (BHM), di tubero di patata (POM) e di S. cerevisiae.

This study deals with the internationalization behavior of a new and specific type of e-business company, namely the network managing e-business company (NM-EBC). The business model of such e-business companies is based on providing a platform and applications for users to connect and interact, on gathering and channeling the inputs provided by the users, and on organizing and managing the cross-relationships of the various participants. Examples are online communities, matching platforms, and portals. Since NM-EBCs internationalize by replicating their business model in a foreign market and by building up and managing a network of users, who provide input themselves and interact with each other, they have to convince users in foreign markets to join the network and hence to adopt their platform. We draw upon Rogers’ Diffusion of Innovations Theory and Network Theory to explain the internationalization behavior of NM-EBCs. These two theories originate from neighboring disciplines and have not yet been used to explain the internationalization of firms. We combine both theories and formulate hypotheses about which strategies NM-EBCs may choose to expand abroad. To test the applicability of our theory and to gain rich data about the internationalization behavior of these firms, we carried out multiple case studies with internationally active Germany-based NM-EBCs.

Evidence accumulated in the last ten years has demonstrated that a large proportion of the mitochondrial respiratory chain complexes in a variety of organisms is arranged in supramolecular assemblies called supercomplexes or respirasomes. Besides conferring a kinetic advantage (substrate channeling) and being required for the assembly and stability of Complex I, indirect considerations support the view that supercomplexes may also prevent excessive formation of reactive oxygen species (ROS) from the respiratory chain. Following this line of thought we have decided to directly investigate ROS production by Complex I under conditions in which the complex is arranged as a component of the supercomplex I1III2 or it is dissociated as an individual enzyme. The study has been addressed both in bovine heart mitochondrial membranes and in reconstituted proteoliposomes composed of complexes I and III in which the supramolecular organization of the respiratory assemblies is impaired by: (i) treatment either of bovine heart mitochondria or liposome-reconstituted supercomplex I-III with dodecyl maltoside; (ii) reconstitution of Complexes I and III at high phospholipids to protein ratio. The results of this investigation provide experimental evidence that the production of ROS is strongly increased in either model; supporting the view that disruption or prevention of the association between Complex I and Complex III by different means enhances the generation of superoxide from Complex I . This is the first demonstration that dissociation of the supercomplex I1III2 in the mitochondrial membrane is a cause of oxidative stress from Complex I. Previous work in our laboratory demonstrated that lipid peroxidation can dissociate the supramolecular assemblies; thus, here we confirm that preliminary conclusion that primary causes of oxidative stress may perpetuate reactive oxygen species (ROS) generation by a vicious circle involving supercomplex dissociation as a major determinant.

Kalorimetrische Tieftemperatur-Detektoren (Calorimetric Low Temperature Detectors, CLTDs) wurden erstmals in Messungen zur Bestimmung des spezifischen Energieverlustes (dE/dx) niederenergetischer Schwerionen beim Durchgang durch Materie eingesetzt. Die Messungen wurden im Energiebereich unterhalb des Bragg-Peaks, mit 0.1 - 1.4 MeV/u 238U-Ionen in Kohlenstoff und Gold sowie mit 0.05 - 1.0 MeV/u 131Xe-Ionen in Kohlenstoff, Nickel und Gold, durchgeführt. Die Kombination der CLTDs mit einem Flugzeitdetektor ermöglichte dabei, kontinuierliche dE/dx-Kurven über größere Energiebereiche hinweg simultan zu bestimmen. Im Vergleich zu herkömmlichen Meßsystemen, die Ionisationsdetektoren zur Energiemessung verwenden, erlaubten die höhere Energieauflösung und -linearität der CLTDs eine Verringerung der Kalibrierungsfehler sowie eine Erweiterung des zugänglichen Energiebereiches der dE/dx-Messungen in Richtung niedriger Energien. Die gewonnen Daten können zur Anpassung theoretischer und semi-empirischer Modelle und somit zu einer Erhöhung der Präzision bei der Vorhersage spezifischer Energieverluste schwerer Ionen beitragen. Neben der experimentellen Bestimmung neuer Daten wurden das alternative Detektionsprinzip der CLTDs, die Vorteile dieser Detektoren bezüglich Energieauflösung und -linearität sowie der modulare Aufbau des CLTD-Arrays aus mehreren Einzeldetektoren genutzt, um diese Art von Messung auf potentielle systematische Unsicherheiten zu untersuchen. Unter anderem wurden hierbei unerwartete Channeling-Effekte beim Durchgang der Ionen durch dünne polykristalline Absorberfolien beobachtet. Die koinzidenten Energie- und Flugzeitmessungen (E-ToF) wurden weiterhin genutzt, um das Auflösungsvermögen des Detektor-Systems bei der direkten in-flight Massenbestimmung langsamer und sehr schwerer Ionen zu bestimmen. Durch die exzellente Energieauflösung der CLTDs konnten hierbei Massenauflösungen von Delta-m(FWHM) = 1.3 - 2.5 u für 0.1 - 0.6 MeV/u 238U-Ionen erreicht werden. In einer E-ToF-Messung mit Ionisationsdetektoren sind solche Werte in diesem Energie- und Massenbereich aufgrund der Limitierung der Energieauflösung durch statistische Schwankungen von Verlustprozessen beim Teilchennachweis nicht erreichbar.
