Up to 80% of avocados on the retail shelf have defects in the flesh which reduces consumer satisfaction. Flesh bruising is the single most important contributor. Avocados also develop skin spotting during harvesting and packing which can reduce domestic and international customer confidence. This project will identify where bruising occurs, develop decision aid tools to help industry reduce flesh bruising in ripe fruit, and understand the commercial impacts of skin spotting. The project will include a PhD student with stipend coming from an international scholarship and in kind support from the University of Queensland.

Sorghum grown in India in the post-rainy season (Rabi) relies on residual soil moisture, and the crop is commonly exposed to terminal drought stress. But there is a ready market for its high-quality grain and stover (used as fodder on dairy farms). Steps to improve productivity while maintaining quality offer an attractive opportunity for sorghum farmers to improve their incomes. Genetically improving the efficiency of using stored soil moisture is a prime target to maximise grain/stover production and quality of Rabi sorghum. This project aims to achieve this through the application of DNA sequences known as quantitative trait loci (QTLs). The project scientists will introduce marker-assisted introgression of stay-green QTLs into sorghum lines, enhancing both the quality and the quantity of grain/stover of postrainy sorghum. They will also use modelling to identify the key physiological traits involved in a higher, more stable yield across water-limited environments of India and Australia, and the key stay-green QTLs contributing to these traits. The publicly available QTL isolines lines developed in this project will be the basis of new varieties to be bred in a subsequent phase.

This full application seeks ongoing funding for the Agri-Science Queensland's forage oat breeding program through the Meat and Livestock Australia Partners in Innovation program with Heritage Seeds as the Donor Company and the commercial collaborator. The focus of the breeding program will shift to the use of minor genes that confer partial resistance or adult plant resistance to leaf rust. Accumulation of several minor genes in a single variety should produce a high level of effective field resistance, and better durability of resistance, with less likelihood of a mutational change in the pathogen.

The invasive liana cat’s claw creeper, Macfadyena unguis-cati, native to tropical Central and South America, is a major environmental weed in Queensland and New South Wales (NSW). Two morphologically distinct cat’s claw creeper varieties occur in Australia, a ‘short-pod’ variety that is widespread through Queensland and NSW and a ‘long-pod’ variety restricted to a few sites in southeast Queensland. In this study we report the differences in the above-ground morphological, phenological and reproductive traits between the two varieties. The ‘long-pod’ variety has significantly larger leaves, larger pods, and larger number of seeds per pod than the ‘short-pod’ variety. The ‘short-pod’ variety has a slightly wider pods, and thicker leaves than the ‘long-pod’ variety. Both varieties have a yellow trumpet shaped flower, but the flower of the ‘long-pod’ variety has a deeper hue of yellow than the ‘short-pod’ flower. The fruits of the ‘short-pod’ variety mature in late summer to early autumn while the fruits of ‘long-pod’ variety mature in late winter to early spring. The more widespread nature of the ‘short-pod’ variety could potentially be due to a preference for this variety as an ornamental plant, due to its more presentable foliage characteristics and shorter pods, in contrast to the ‘long-pod’ variety.

Quality and safety evaluation of agricultural products has become an increasingly important consideration in market/commercial viability and systems for such evaluations are now demanded by customers, including distributors and retailers. Unfortunately, most horticultural products struggle with delivering adequate and consistent quality to the consumer. Removing inconsistencies and providing what the consumer expects is a key factor for retaining and expanding both domestic and international markets. Most commercial quality classification systems for fruit and vegetables are based on external features of the product, for example: shape, colour, size, weight and blemishes. However, the external appearance of most fruit is generally not an accurate guide to the internal or eating quality of the fruit. Internal quality of fruit is currently subjectively judged on attributes such as volatiles, firmness, and appearance. Destructive subjective measures such as internal flesh colour, or objective measures such as extraction of juice to measure sweetness (oBrix) or assessment of dry matter (DM) content are also used, although obviously not for every fruit – just a sample to represent the whole consignment. For avocado fruit, external colour is not a maturity characteristic, and its smell is too weak and appears later in its maturity stage (Gaete-Garreton et al., 2005). Since maturity is a major component of avocado quality and palatability, it is important to harvest mature fruit, so as to ensure that fruit will ripen properly and have acceptable eating quality. Currently, commercial avocado maturity estimation is based on destructive assessment of the %DM, and sometimes percent oil, both of which are highly correlated with maturity (Clark et al., 2003; Mizrach & Flitsanov, 1999). Avocados Australia Limited (AAL (2008)) recommend a minimum maturity standard for its growers of 23 %DM (greater than 10% oil content) for the ‘Hass’ cultivar, although consumer studies indicate a preference for at least 25 %DM (Harker et al., 2007).

Introduced as an ornamental vine, cat's claw creeper Dolichandra unguis-cati (syn. Macfadyena unguis-cati) has invaded coastal and subcoastal areas of subtropical eastern Australia. Two varieties have been indentified, one of which ('short-pod') is found throughout south-eastern Australia, while the other ('long-pod') appears to be restricted to several sites in south-eastern Queensland. We compared the growth and biomass allocation patterns of the two varieties in the field over a 22-month period to determine if a higher growth rate and/or more efficient allocation of biomass may contribute to this disparity in distribution. The long-pod variety produced greater aboveground and total biomass than the short-pod variety in both riparian and non-riparian zones. Belowground the two varieties produced a similar number of tubers and overall biomass, though the long-pod variety allocated a smaller portion of its carbon belowground. High growth rates and greater biomass allocation aboveground are characteristic of invasive species, allowing them to outcompete and crowd out existing vegetation. There was no significant site by variety interaction, an indication of consistency in variety performance across riparian and non-riparian sites. Results from our study suggest that differences in growth and biomass allocations are unlikely to have contributed to the disparity in distribution of the two varieties. Despite currently occupying a relatively small range, the long-pod variety may be a more adept invader than the short-pod variety, and could become more prevalent in the future. © 2012 CSIRO.

Fourier Transform (FT)-near infra-red spectroscopy (NIRS) was investigated as a non-invasive technique for estimating percentage (%) dry matter of whole intact 'Hass' avocado fruit. Partial least squares (PLS) calibration models were developed from the diffuse reflectance spectra to predict % dry matter, taking into account effects of seasonal variation. It is found that seasonal variability has a significant effect on model predictive performance for dry matter in avocados. The robustness of the calibration model, which in general limits the application for the technique, was found to increase across years (seasons) when more seasonal variability was included in the calibration set. The R-v(2) and RMSEP for the single season prediction models predicting on an independent season ranged from 0.09 to 0.61 and 2.63 to 5.00, respectively, while for the two season models predicting on the third independent season, they ranged from 0.34 to 0.79 and 2.18 to 2.50, respectively. The bias for single season models predicting an independent season was as high as 4.429 but <= 1.417 for the two season combined models. The calibration model encompassing fruit from three consecutive years yielded predictive statistics of R-v(2) = 0.89, RMSEP = 1.43% dry matter with a bias of -0.021 in the range 16.1-39.7% dry matter for the validation population encompassing independent fruit from the three consecutive years. Relevant spectral information for all calibration models was obtained primarily from oil, carbohydrate and water absorbance bands clustered in the 890-980, 1005-1050, 1330-1380 and 1700-1790 nm regions. These results indicate the potential of FT-NIRS, in diffuse reflectance mode, to non-invasively predict the % dry matter of whole 'Hass' avocado fruit and the importance of the development of a calibration model that incorporates seasonal variation. Calonectria ilicicola, Gliocladiopsis sp. and Ilyonectria liriodendri were isolated from diseased roots of young avocado trees. Pathogenicity studies with seedlings of three avocado cultivars, Velvick, Hass and Reed, demonstrated that Calonectria ilicicola is a severe root rot pathogen, reducing the biomass of healthy roots, and reducing plant height over time. Calonectria ilicicola was re-isolated from diseased roots. Ilyonectria liriodendri and Gliocladiopsis sp. were not pathogenic and plant height was increased after Gliocladiopsis sp. amendment compared to all other treatments in trials with cvs Velvick and Hass.

The development of a more efficient in vitro regeneration system for somatic embryos (SEs) of avocado (Persea americana) would facilitate the development of new superior cultivars for this valuable horticultural crop. In this study, we report a new and efficient method for maintenance and regeneration of avocado SEs. Avocado SEs of four cultivars remained healthy and viable in vitro for 11 months on a medium used for mango somatic embryogenesis, compared with 3-4 months on Murashige and Skoog medium. Various supplements and media modifications were investigated to improve the low conversion rate of regenerated plants from avocado SEs reported previously. The one-step system for regeneration of white-opaque somatic embryos (WOSEs) used solid medium only over a period of 12-14 weeks (sub-culturing every 6 weeks). Addition of praline and glutamine improved the total regeneration from 0 to 17.5% and 10.5%, and plant/shoot recovery from 0 to 12.5% and 5%, respectively. A two-step culture system involving the transfer of WOSEs of cultivar 'Reed' after 6 weeks on solid to liquid medium for 12-15 days as an intermediate step, followed by subculturing again onto solid medium for 6 weeks improved total regeneration to 29% and plant/shoot recovery to 18.3 from 0% when regenerated by subculturing on solid medium only. Supplementation with proline in the solid as well as liquid medium in the two-step culture system at 0.4 g/L increased total regeneration to 35% and plant/shoot recovery to 20%. We were able to achieve highest regeneration using glutamine at 1 g/L in the two-step culture system in terms of both total regeneration (58.3%, including 43.3% bipolar regeneration) and plant/shoot recovery (36.7%) rates, which were significantly higher than in any other treatment investigated.
