Grains and legume seeds are foods that form the basis of the diets of many cultures around the world, winch contritbute to the daily nutrient requirements of humans. Vicilins (7S globulin) are storage proteins found in legume seeds, and may have an additional function constitutive defense of the embryo against pests and pathogens. In this work the vicilin from Anadenanthera macrocarpa - AmV (red-angico), was purified and partially characterized, its effect on development and larval survival and adult emergence of Callosobruchus maculatus was evaluated by determination of LD50, WD50 and ED50 in system bioassay. Purification of vicilin was initiated by the chitin affinity chromatography and then gel filtration (Superdex 75 Tricorn 10x300 mm) FPLC system followed by reverse phase chromatography (C8 phenomenex) on HPLC system. Bioassays WD50 and LD50 for larvae were 0.32% and 0.33% (w:w) respectively, since the ED50 for adults was 0.096%. The probable mechanism of action was evaluated by testing digestibility of AmV in vitro, and observed for the involvement of two fragments vicilins immunoreactive against polyclonal Anti-vicilin from Erythrina velutina (Anti-EvV) about of 22 and 13 kDa chitin binding. The AmV in its native form has been recognized by the anti-EvV, indicating that there is a conserved region in the vicilin and is probably corresponding to the chitin binding domains. These results point to a new vicilin chitin binding that can subsequently be used as a possible biopesticide protein source, in order to control insect pest C. maculatus and confirm literature findings that demonstrate vicilin in the presence of different kinds of ligands to conserved regions chitin not yet characterized

This work examined the possibility of using mussel Mytella falcata as bioindicator sample to detect metal ions in several estuaries potiguares, since species substances that accumulate in their tissues due to its characteristics filter feeders have been used for environmental monitoring. The chemometrics by principal components analysis was used to reduce the size of the original data in order to establish a pattern of distribution of metal ion. Samples were collected at three different points in the estuaries Curimataú, Guaraíra-Papeba, Potengi, Galinhos-Guamaré and Piranhas-Assu having been marked with the location using GPS (Global Positioning System). The determination of humidity content and digestion of the samples were performed using methods described in the Compendium of analytical standards of the Institute Adofo Lutz (2005) and the determination of metal ions of the elements Al, Ba, Cd, Cr, Cu, Mn, Ni, Pb, Sn and Zn were performed by optical emission spectrometry with inductively coupled plasma as described by USEPA method 6010C. The results show that it is possible to use this molluscum Mytella falcata in the estuaries of Rio Grande do Norte for the determination of metal ions. The data were subjected to principal components analysis (PCA) which enabled us to verify the distribution pattern of the metal ions studied in several estuaries potiguares and group them according to the metal ions in common with and relate them to the activities in each region

The impact of pollutants in an organism can be observed by changes in functional complexity at different levels. Bivalve gills are suitable for histopathological analysis because of their structure and function. This study aimed at examining the morphology of Mytella falcata gill filaments from three sites in the Santos estuary (São Paulo, Brazil) with different levels of environmental degradation to identify possible changes in gill structure and discuss the significance of these alterations. For this purpose, histological, histochemical and ultrastructural techniques were used. The filaments of animals from site A (less impacted site) were intact, while in sites B and C, pathological changes were observed, such as: detachment of the epithelium in the intermediate zone, morphological changes of this epithelium, inflammatory process, increase in the number of mucous cells and cell turnover processes. These results suggest that the related changes are an attempt to prevent the entrance of pollutants through gill filaments into the entire organism and that cell turnover is the final way to compensate cell injury. Bivalve filter feeders are sessile animals that live in constant contact with water and its pollutants. Their gill is an organ highly exposed to these conditions due to its large surface and its involvement in gas exchanges and feeding. The bivalve Mytella falcata is found in estuaries of Latin America, on the Atlantic as well as the Pacific Coast. It is commonly consumed, and sometimes is the only source of protein of low-income communities. In this study, gill filaments of M. falcata were characterized using histology, histochemistry and transmission electron microscopy for future comparative studies among animals exposed to environmental pollutants. Gill filaments may be divided into abfrontal, intermediate and frontal zones. Filaments are interconnected by ciliary discs. In the center of filaments, haemocytes circulate through a haemolymph vessel internally lined by an endothelium and supported by an acellular connective tissue rich in polysaccharides and collagen. The abfrontal zone contains cuboidal cells, while the intermediate zone consists of a simple squamous epithelium. The frontal zone is composed of five columnar cell types: one absorptive, mainly characterized by the presence of pinocytic vesicles in the apical region of the cell; one secretory, rarely observed and three ciliated with abundant mitochondria. All cells lining the filament exhibit numerous microvilli and seem to absorb substances from the environment. PAS staining was observed in mucous cells in the frontal and abfrontal zones. Bromophenol blue allowed the distinction of haemocytes and detection of a glycoprotein secretion in the secretory cells of the frontal region. The characteristics of M. falcata gill filaments observed in this study were very similar to those of other bivalves, especially other Mytilidae, and are suitable for histopathological studies on the effect of water-soluble pollutants.

The objective of this study was to evaluate the allelophatic potential ethanol extract of leaves of Anadenanthera macrocarpa (Benth.) Brenan (angico) and Astronium graveolens Jacq (aroeira) on the biomonitor germination of Brassica chinensis (Malaysia's cabbage) and Lactuca sativa (lettuce). For the test of allelopathy was the bioassay of germination of B. chinensis and L. sativa using solutions of ethanol extracts in different concentrations (5, 10 and 20 mg / mL) with continuous light for 11 days. The experimental and control groups were divided into the Petri dish containing 50 seeds of B. chinensis and L. sativa each. Were performed six replicates for each concentration of extract of leaves of A. macrocarpa and A. graveolens and the negative control (water). The rolling action of the treatments was performed by quantitative observation and monitoring of germination every six hours. The results show that the ethanol extract of A. macrocarpa and A. graveolens presents allelopathic potential, regardless of the concentration used, but with answers dose-dependent, and thus able to interfere directly in germination, average speed and time of germination biomonitor of B. chinensis and L. sativa.

The experiment had the aim of quantifying seasonal chlorophyll variation on leaves of Anadenanthera colubrina and Tabebuia avellanedae. The experiment used a randomized block design established as a 2 x 3 factorial (two species and three sampling seasons) with 5 replications. The concentrations of chlorophyll in leaves were performed at the end of winter, late spring, and summer. The content of chlorophyll a changed only for A. colubrina, bringing up from spring (117.52 mu g mL(-1)) to summer (151.13 mu g mL(-1)). For the two species, the proportion of chlorophyll b was higher between periods at the end of spring (average of 29.31 mu g mL(-1)) to the end of summer (average of 74.96 mu g mL(-1)) with close relationship with the decrease in global solar irradiance. The chlorophyll a/b relationship increased in late spring due to reduction of chlorophyll b content. A. colubrina showed greater adaptation to the studied environment.

Anadenanthera macrocarpa (Benth.) Brenan is a tree species belonging to the family Leguminosae-Mimosoideae which is popularly known as mimosa-black. It features characteristics of early successional, natural occurrence in Brazil and can form clusters almost homogeneous, with great potential in the recovery of degraded areas. This study was conducted at the Center for Agricultural Sciences, University Federal of Alagoas state, aiming at the physical and morphological characterization of seeds, describing the various stages of post-seminal development, and to evaluate various conditions of temperature and substrate to perform the test germination. The seeds were manually extracted, then homogenized, where two samples of 50 seeds were used to determine the initial moisture. Another sample, consisting of eight repetitions of 100 seeds was used to measure the biometry and the number of seeds per fruit. The morphological characterization and the seeds were immersed in distilled water to allow the cuts to the observed structure in microscopes. In the post-seminal study, it was observed the daily processes of seedling growth in rolled paper filter and constant temperature of 30 °C. To assess the germination behavior, the constant temperatures of 15 °C, 25 °C, 30 °C, 40 °C and 20-30 °C as well as the paper and the sand substrates were tested and it was evaluated the percentage, the rate of germination, the relative frequency distribution, and the tests conducted in a randomized design in factorial 5 x 2 (x substrate temperature) with four replicates of 25 seeds, and the averages compared by Duncan 5 % probability. The fruits of black angico show great variation in the seed number per fruit. The embryonic axis occupies part of the central region of the seed with axial and linear positions. The germination is epigeal and the seedlings are fanerocotylar. The temperature determines the 30 °C and the filter paper substrate provided higher average percentage and germination rate.
