Hydrothiolation of 1-organylbuta-1,3-diynes and 1,4-diorganylbuta-1,3-diynes with the sodium organylthiolate anions, which were generated in situ by reacting diphenyl and dibutyl disulfide with NaBH(4) in ethanol, results in the regio-, stereo-, and chemoselective formation of (Z)-1-organylthio-4-organylbut-1-en-3-ynes and (Z)-1-organylthio-1,4-diorganylbut-1-en-3-ynes, respectively.

The objective of this study is to determine if digital vasculitis (DV), a clinical manifestation with a high systemic lupus erythematosus disease activity index (SLEDAI) score, is associated with lupus severity. DV and other clinical manifestations defined according to the SLEDAI were evaluated in 168 consecutive patients with systemic lupus erythematosus (SLE). Two groups were defined according to presence (DV+, n = 27) or absence of DV (DV-, n = 141) at the time of evaluation. The exclusion criterion was the presence of antiphospholipid syndrome (Sapporo`s criteria). The two groups were comparable with regard to age (P = 0.09), gender (P = 1.00), white race (P = 0.81), and disease duration (P = 0.78). Compared to the DV-group, the DV+ group had a significantly higher frequency of mucocutaneous manifestations (66.7 vs. 39.0%, P = 0.01), haematological abnormalities (22.2 vs. 6.4%, P = 0.02) and constitutional symptoms (11.1 vs. 0.7%, P = 0.01). Renal and neurological involvements were similar in both groups ( P = 0.57 and P = 1.00, respectively). The evaluation of each SLEDAI parameter confirmed that the DV+ group had higher frequencies of mild manifestations, such as new rash (P = 0.02), alopecia (P = 0.02), oral ulcers (P = 0.045), fever (P = 0.01) and leucopenia (P = 0.005). In contrast, both groups had similarly increased anti-dsDNA (P = 0.78) and decreased complement levels (P = 0.29). In conclusion, DV in patients with SLE identifies a subgroup of a mild disease. The high `weighted` index attributed to this alteration in the SLEDAI score should therefore be revised. Lupus (2009) 18, 990-993.

Context Pheochromocytomas and paragangliomas are genetically heterogeneous neural crest-derived neoplasms. We recently identified germline mutations of the novel transmembrane-encoding gene FP/TMEM127 in familial and sporadic pheochromocytomas consistent with a tumor suppressor effect. Objectives To examine the prevalence and spectrum of FP/TMEM127 mutations in pheochromocytomas and paragangliomas and to test the effect of mutations in vitro. Design, Setting, and Participants We sequenced the FP/TMEM127 gene in 990 individuals with pheochromocytomas and/or paragangliomas, including 898 previously unreported cases without mutations in other susceptibility genes from 8 independent worldwide referral centers between January 2009 and June 2010. A multiplex polymerase chain reaction-based method was developed to screen for large gene deletions in 545 of these samples. Confocal microscopy of 5 transfected mutant proteins was used to determine their subcellular localization. Main Outcome Measures The frequency and type of FP/TMEM127 mutation or deletion was assessed and correlated with clinical variables; the subcellular localization of 5 overexpressed mutants was compared with wild-type FP/TMEM127 protein. Results We identified 19 potentially pathogenic FP/TMEM127 germline mutations in 20 independent families, but no large deletions were detected. All mutation carriers had adrenal tumors, including 7 bilateral (P=2.7 x 10(-4)) and/or with familial disease (5 of 20 samples; P=.005). The median age at disease onset in the FP/TMEM127 mutation group was similar to that of patients without a mutation (41.5 vs 45 years, respectively; P=.54). The most common presentation was that of a single benign adrenal tumor in patients older than 40 years. Malignancy was seen in 1 mutation carrier (5%). Expression of 5 novel FP/TMEM127 mutations in cell lines revealed diffuse localization of the mutant proteins in contrast with the discrete multiorganelle distribution of wild-type TMEM127. Conclusions Germline mutations of FP/TMEM127 were associated with pheochromocytoma but not paraganglioma and occured in an age group frequently excluded from genetic screening algorithms. Although cigarette smoking and alcohol consumption increase risk for head and neck cancers, there have been few attempts to model risks quantitatively and to formally evaluate cancer site-specific risks. The authors pooled data from 15 case-control studies and modeled the excess odds ratio (EOR) to assess risk by total exposure (pack-years and drink-years) and its modification by exposure rate (cigarettes/day and drinks/day). The smoking analysis included 1,761 laryngeal, 2,453 pharyngeal, and 1,990 oral cavity cancers, and the alcohol analysis included 2,551 laryngeal, 3,693 pharyngeal, and 3,116 oval cavity cancers, with over 8,000 controls. Above 15 cigarettes/day, the EOR/pack-year decreased with increasing cigarettes/day, suggesting that greater cigarettes/day for a shorter duration was less deleterious than fewer cigarettes/day for a longer duration. Estimates of EOR/pack-year were homogeneous across sites, while the effects of cigarettes/day varied, indicating that the greater laryngeal cancer risk derived from differential cigarettes/day effects and not pack-years. EOR/drink-year estimates increased through 10 drinks/day, suggesting that greater drinks/day for a shorter duration was more deleterious than fewer drinks/day for a longer duration. Above 10 drinks/day, data were limited. EOR/drink-year estimates varied by site, while drinks/day effects were homogeneous, indicating that the greater pharyngeal/oral cavity cancer risk with alcohol consumption derived from the differential effects of drink-years and not drinks/day.

Background/Aim: Some studies have identified an association of kidney stone formation with vitamin D receptor (VDR) or calcium-sensing receptor (CaSR) polymorphisms. We aimed to evaluate the association between these polymorphisms with urinary calcium excretion (uCa) in calcium-stone-forming patients. Methods: VDR polymorphism, detected by BsmI digestion, and 3 CaSR polymorphisms (G/T at codon 986, G/A at codon 990 and C/G at codon 1011), detected by direct sequencing, were evaluated in 100 hypercalciuric (HCa) and 101 normocalciuric (NCa) calcium-stone-forming patients. Results: The total allelic frequency of VDR polymorphism was: 16% BB, 49% Bb and 35% bb. The prevalence of bb genotype was significantly higher in the HCa when compared to the NCa group (43 vs. 27%). With respect to CaSR polymorphisms, 986S, 990G and 1011E variant alleles were detected, respectively, in 5, 4 and 3% of the whole sample and 5 CaSR haplotypes were identified: 94% ARQ (wildtype), 3% SRQ, 1.5% AGQ, 1.0% ARE and 0.5% AGE. No statistical differences have been observed between NCa and HCa with respect to these CaSR haplotypes. Conclusions: The present study suggested that bb homozygous for VDR polymorphism was overrepresented in hypercalciuric stone formers. To test a mathematical model for measuring blinking kinematics. Spontaneous and reflex blinks of 23 healthy subjects were recorded with two different temporal resolutions. A magnetic search coil was used to record 77 blinks sampled at 200 Hz and 2 kHz in 13 subjects. A video system with low temporal resolution (30 Hz) was employed to register 60 blinks of 10 other subjects. The experimental data points were fitted with a model that assumes that the upper eyelid movement can be divided into two parts: an impulsive accelerated motion followed by a damped harmonic oscillation. All spontaneous and reflex blinks, including those recorded with low resolution, were well fitted by the model with a median coefficient of determination of 0.990. No significant difference was observed when the parameters of the blinks were estimated with the under-damped or critically damped solutions of the harmonic oscillator. On the other hand, the over-damped solution was not applicable to fit any movement. There was good agreement between the model and numerical estimation of the amplitude but not of maximum velocity. Spontaneous and reflex blinks can be mathematically described as consisting of two different phases. The down-phase is mainly an accelerated movement followed by a short time that represents the initial part of the damped harmonic oscillation. The latter is entirely responsible for the up-phase of the movement. Depending on the instantaneous characteristics of each movement, the under-damped or critically damped oscillation is better suited to describe the second phase of the blink. (C) 2010 Elsevier B.V. Glioblastoma is the most frequent and malignant brain tumor, characterized by an elevated capacity for cellular proliferation and invasion. Recently, it was demonstrated that podoplanin membrane sialo-glycoprotein encoded by PDPN gene is over-expressed and related to cellular invasion in astrocytic tumors; however the mechanisms of regulation are still unknown. MicroRNAs are noncoding RNAs that regulate gene expression and several biological processes and diseases, including cancer. Nevertheless, their roles in invasion, proliferation, and apoptosis of glioblastoma are not completely understood. In this study, we focused on miR-29b and miR-125a, which were predicted to regulate PDPN, and demonstrated that these microRNAs directly target the 30 untranslated region of PDPN and inhibit invasion, apoptosis, and proliferation of glioblastomas. Furthermore, we report that miR-29b and miR-125a are downregulated in glioblastomas and also in CD133-positive cells. Taken together, these results suggest that miR-29b and miR-125a represent potential therapeutic targets in glioblastoma. (C) 2010 Wiley-Liss, Inc.

Objectives: To compare the circulating levels of matrix metalloproteinase (MMP)-8, pro-MMP-2, pro-MMP-9, and total MMP-9, their endogenous inhibitors, the tissue inhibitors of metalloproteinases (TIMP)-1 and TIMP-2, and the MMP-8/TIMP-1, MMP-9/TIMP-1, and MMP-2/TIMP-2 ratios in normotensive obese children and adolescents with those found in non obese children and adolescents. Design and methods: We studied 40 obese and 40 non obese (controls) children and adolescents in this cross-sectional study. MMP and TIMP concentrations were measured in plasma samples by gelatin zymography and ELISA. Results: Obese children and adolescents had higher circulating MMP-8 concentrations, lower plasma TIMP-1 concentrations, and higher MMP-8/TIMP-1 ratios than non obese controls (P < 0.05). We found no differences in pro-MMP-9 or total MMP-9 levels, or in MMP-9/TIMP-1 ratios between groups (P > 0.05). While we found no significant differences in pro-MMP-2 levels (P > 0.05) obese Subjects had higher TIMP-2 concentrations and lower pro-MMP-2/TIMP-2 ratios (P < 0.05) than non obese controls. Conclusions: In conclusion, we found evidence indicating higher net MMP-8 (but not MMP-9 and MMP-2) activity in childhood obesity. The increased MMP-8 levels found in obese children suggest a possibly relevant pathophysiological mechanism that may be involved in the increase of cardiovascular risk associated with childhood obesity. Este trabalho objetivou avaliar o crescimento e a produtividade de grãos da cultura do milho, submetida a diferentes doses de nitrogênio. O experimento foi conduzido com a cultura do milho híbrido Pioneer 30F35, no período de 25/06 a 28/10/2009, na região de Rio Largo, AL. O delineamento experimental foi em blocos casualizados e os tratamentos consistiram em seis doses de nitrogênio (0, 50, 100, 150, 200 e 250 kg ha-1 de N), com a fonte ureia, e cinco repetições. Os modelos logístico e pico log normal apresentaram ajustes estatísticos significativos (p<0,05) das variáveis de crescimento e coeficientes de determinação máximo de 0,994 e 0,990, respectivamente. Os valores observados e estimados pelos modelos apresentaram alta associação pelo índice de concordância de Willmott, com valores superiores a 0,953. As plantas apresentaram diferenças estatísticas significativas pelo teste F (p<0,05), entre os tratamentos, para altura e índice de área foliar, com crescimento máximo para a dose de 100 kg ha-1 de N. Os tratamentos mostraram diferenças estatísticas significativas (p<0,05) para a produtividade, sobressaindo-se a dose de 200 kg ha-1, que proporcionou maior produtividade de grãos (5,45 t ha-1). Os modelos de crescimento podem ser utilizados para auxiliar a análise de crescimento.

While fluoroscopy is still the most widely used imaging modality to guide cardiac interventions, the fusion of pre-operative Magnetic Resonance Imaging (MRI) with real-time intra-operative ultrasound (US) is rapidly gaining clinical acceptance as a viable, radiation-free alternative. In order to improve the detection of the left ventricular (LV) surface in 4D ultrasound, we propose to take advantage of the pre-operative MRI scans to extract a realistic geometrical model representing the patients cardiac anatomy. This could serve as prior information in the interventional setting, allowing to increase the accuracy of the anatomy extraction step in US data. We have made use of a real-time 3D segmentation framework used in the recent past to solve the LV segmentation problem in MR and US data independently and we take advantage of this common link to introduce the prior information as a soft penalty term in the ultrasound segmentation algorithm. We tested the proposed algorithm in a clinical dataset of 38 patients undergoing both MR and US scans. The introduction of the personalized shape prior improves the accuracy and robustness of the LV segmentation, as supported by the error reduction when compared to core lab manual segmentation of the same US sequences.

Compara-se o crescimento do peso-idade e da altura-idade de 7.990 crianças de 0-11 anos, aleatoriamente selecionadas e medidas em 1981-1982 no Estado da Paraíba (nordeste do Brasil), com o de estudo global realizado no nordeste em 1974-1975, com as curvas de crescimento das referências nacional e internacional. Os resultados colocam em evidência um déficit de crescimento muito acentuado, sobretudo com relação à referência internacional. A 11 anos nítidas diferenças se manifestam: 8 ou 9 quilos para o peso, segundo o sexo, e em torno de 11 cm para a altura. Os déficits de crescimento observados poderiam ser atribuídos à grande seca de 1978-1983, em particular o déficit de crescimento do peso em função da idade. No entanto, o déficit de crescimento da altura em função da idade mostra a existência de alterações mais profundas e mais insidiosas, ao longo do crescimento, manifestando-se por má nutrição crônica, atribuída sobretudo a fatores de ordem estrutural socioeconômica. Constata-se, portanto, no decorrer da última década, que eles têm sido cada vez mais deteriorados, como testemunham as alterações do modelo de crescimento - ele mesmo exprimindo o agravamento da situação nutricional da região.

Digital X-ray detector technologies provide several advantages when compared with screen-film (SF) systems: better diagnostic quality of the radiographic image, increased dose efficiency, better dynamic range and possible reduction of radiation exposure to the patient. The transition from traditional SF systems to digital technology-based systems highlights the importance of the discussion around technical factors such as image acquisition, themanagement of patient dose and diagnostic image quality. Radiographers should be aware of these aspects concerning their clinical practice regarding the advantages and limitations of digital detectors. Newdigital technologies require an up-to-date of scientific knowledge concerning their use in projection radiography. This is the second of a two-part review article focused on a technical overview of digital radiography detectors. This article provides a discussion about the issues related to the image acquisition requirements and advantages of digital technologies, the management of patient dose and the diagnostic image quality.

OBJETIVO: Conhecer a paridade, o desenvolvimento ovariano e a razão de sobrevivência da espécie Anopheles albitarsis, visando a estimar o potencial de transmissão malárica. MÉTODOS: Duas populações de Anopheles albitarsis, denominadas A e B, foram capturadas na Fazenda Experimental do Instituto Agronômico de Campinas, situada no Município de Pariquera-Açu, Estado de São Paulo. As capturas foram feitas no período crepuscular vespertino das 17h às 20h, utilizando-se armadilha tipo Shannon. As dissecções foram feitas utilizando-se a técnica de Polovodova, e a avaliação do desenvolvimento folicular segundo o critério de Christophers e Mer. Adotou-se o método de Vercrusse para se estimar a sobrevivência diária e o método de Davidson para se determinar a duração do ciclo gonotrófico. RESULTADOS: Foram dissecados 2.612 exemplares, sendo 237 da população A e 2.375 da B. As sobrevivências diárias para as populações A e B foram de 0,5339±0,047 e 0,5566±0,015, respectivamente, e a duração do ciclo gonotrófico para a população A foi de 1.990 dias e para a B de 2.046 dias. CONCLUSÕES: Os resultados contribuem para a avaliação do potencial de transmissão malárica na região.

OBJETIVO: Traçar um perfil de estudantes em relação ao consumo de álcool e comportamentos de risco. MÉTODOS: Participaram do estudo 1.990 alunos, com idade entre 11 e 21 anos, de ambos os sexos, matriculados em escolas públicas e privadas de Paulínia, SP, 2004. Um questionário de auto-preenchimento foi respondido em sala de aula, sem a presença do professor. Analisou-se a percepção da disponibilidade e facilidade de acesso às bebidas alcoólicas, contexto do beber e conseqüências do consumo. RESULTADOS: A prevalência de uso de álcool na vida foi de 62,2%. Em relação aos últimos 30 dias, 17,3% dos alunos relataram pelo menos um episódio de abuso agudo. Os adolescentes reportaram que adquiram facilmente bebidas alcoólicas de estabelecimentos comerciais e também em contextos sociais com parentes e amigos. Apenas 1% dos menores de idade relatou que tentou, mas não conseguiu comprar bebida alcoólica. Como conseqüências negativas do consumo nos últimos 12 meses, os estudantes relataram ter passado mal por ter bebido (17,9%), arrependimento por algo que fizeram sob o efeito do álcool (11%), blackout (9,8%) e ter brigado após beber (5%). Mais da metade (55%) dos estudantes conhecia alguém que sofreu acidente de trânsito provocado por motorista embriagado. CONCLUSÕES: Os dados revelaram alta prevalência de consumo de álcool entre os adolescentes estudados e fácil acesso às bebidas alcoólicas, inclusive por menores de idade. Os jovens se colocaram em risco e apresentaram conseqüências negativas do consumo de álcool. Há necessidade de ações imediatas em relação às políticas públicas para o consumo de álcool no Brasil.
