Atmospheric monitoring of high northern latitudes (> 40°N) has shown an enhanced seasonal cycle of carbon dioxide (CO2) since the 1960s but the underlying mechanisms are not yet fully understood. The much stronger increase in high latitudes compared to low ones suggests that northern ecosystems are experiencing large changes in vegetation and carbon cycle dynamics. Here we show that the latitudinal gradient of the increasing CO2 amplitude is mainly driven by positive trends in photosynthetic carbon uptake caused by recent climate change and mediated by changing vegetation cover in northern ecosystems. Our results emphasize the importance of climate-vegetation-carbon cycle feedbacks at high latitudes, and indicate that during the last decades photosynthetic carbon uptake has reacted much more strongly to warming than carbon release processes.

Calculated and measured estimations of biomass of small (<3 mm), large (3-30 mm), and total zooplankton were verified (compared). These integral parameters of epipelagic communities were estimated by two methods. We used previously obtained regression equations, which correlate these parameters with water transparency. Measured values of aforesaid parameters were compared with their mean values in waters of different productivity estimated from NASA satellite maps. We compared data collected at fifteen stations in September-December in regions of different productivity in the North Atlantic. In warm regions (to the south of 40°N) measured and calculated values coincide well. In boreal regions in autumn bulk of mesozooplankton descends to deep layers due to seasonal migrations; hence correlation between measured and calculated values is disrupted. It is evident that correlation between water transparency and mesozooplankton biomass (integral index of water productivity) obtained before should be corrected for seasonal variations.

Detailed records of the carbon and oxygen isotopic ratios of Neogloboquadrina pachyderma are compared between nine high-latitude sediment cores, from the Northern and Southern Hemispheres, covering the last 140000 yrs. The strong analogies between the delta13C records permit to define a delta13C stratigraphic scale, with three clear cut transitions simultaneous with the oxygen isotopic transitions 6/5 (125 kyrs.), 5/4 (65 kyrs.), and 2/1 (13 kyrs.). The delta13C records of N. pachyderma in the high-latitude cores, which follow the changes in delta13C of the surface water TCO2 near areas of deep water formation present trends similar to the benthic foraminifera delta13C records in cores V19-30 and M12-392, although amplitudes of the isotopic shifts are different. This implies that a large part of the observed variations represents global changes in the carbon distribution between biosphere and ocean. The 13C/12C ratios of N. pachyderma in the North Atlantic cores display larger regional variations at 18 kyrs. B.P. than at present. To explain these differences, we have plotted the 18 kyrs. B.P. delta13C values of N. pachyderma from 17 cores distributed N of 40°N. Comparison with published surface water temperature distribution at 18 kyrs. B.P. indicates that a strong divergent cyclonic cell, centered approximatively 55°N and 15°W, was active during most of the last ice-age maximum. This hydrology, analogous to the present Weddell Sea, explains the published evidences of bottom water formation, if located on the northern flank of the gyre, and the strong polar front on the southern flank, probable location of intermediate water formation.

Short-term changes in sea surface conditions controlling the thermohaline circulation in the northern North Atlantic are expected to be especially efficient in perturbing global climate stability. Here we assess past variability of sea surface temperature (SST) in the northeast Atlantic and Norwegian Sea during Marine Isotope Stage (MIS) 2 and, in particular, during the Last Glacial Maximum (LGM). Five high-resolution SST records were established on a meridional transect (53°N-72°N) to trace centennial-scale oscillations in SST and sea-ice cover. We used three independent computational techniques (SIMMAX modern analogue technique, Artificial Neural Networks (ANN), and Revised Analog Method (RAM)) to reconstruct SST from planktonic foraminifer census counts. SIMMAX and ANN reproduced short-term SST oscillations of similar magnitude and absolute levels, while RAM, owing to a restrictive analog selection, appears less suitable for reconstructing "cold end" SST. The SIMMAX and ANN SST reconstructions support the existence of a weak paleo-Norwegian Current during Dansgaard-Oeschger (DO) interstadials number 4, 3, 2, and 1. During the LGM, two warm incursions of 7°C water to occurred in the northern North Atlantic but ended north of the Iceland Faroe Ridge. A rough numerical estimate shows that the near-surface poleward heat transfer from 53° across the Iceland-Faroe Ridge up to to 72° N dropped to less than 60% of the modern value during DO interstadials and to almost zero during DO stadials. Summer sea ice was generally confined to the area north of 70°N and only rarely expanded southward along the margins of continental ice sheets. Internal LGM variability of North Atlantic (>40°N) SST in the GLAMAP 2000 compilation (Sarnthein et al., 2003, doi:10.1029/2002PA000771; Pflaumann et al., 2003, doi:10.1029/2002PA000774) indicates maximum instability in the glacial subpolar gyre and at the Iberian Margin, while in the Nordic Seas, SST was continuously low.

Present day hydrographic conditions along the western Iberian margin are characterized by seasonal upwelling with filaments that can penetrate more than 200 km into the open ocean and constitute areas of cold and highly productive waters. In order to investigate spatial and temporal gradients in temperature and productivity conditions during the last 150 ky, high-resolution proxy records were generated in 3 cores (SU92-03, MD95-2040, MD95-2042), located along the Iberian coast between 43°12'N and 37°48'N and forming a N-S profile. In all cores, planktonic foraminifera census counts are used to reconstruct summer sea surface temperature (SSTsu) and export productivity (Pexpsu) using the modern analog technique SIMMAX 28. SSTsu and Pexpsu values similar to the present are observed throughout the Holocene and MIS 5e periods for each site, respectively, indicating fairly stable conditions equivalent to the modern ones. On glacial/interglacial timescales, SSTsu increases by 2-3 °C from the northern to southernmost site. Pexpsu, on the other hand, shows a decrease of 30-40 gC/m**2/yr from North to South at present time and during interglacial periods, and no significant variation (90-100 gC/m**2/yr) during glacial periods. The northernmost core SU92-03 reveals the coldest conditions with records more similar to MD95-2040 than to MD95-2042, the later of which is, as at present, more affected by subtropical waters. Core SU92-03 shows higher interglacial productivity similar to open ocean mid- to high latitude sites, while the other two cores monitor higher glacial productivity conform with other upwelling sites off NW Africa. A boundary between differences in glacial/interglacial productivity appears to be present in our study between 43°12'N and 40°35'N. Especially north of 40°N, coldest SSTsu and lowest Pexpsu are found during Heinrich events (H)1-H8 and H10-H11. In contrast, lowest Pexpsu do not coincide with these events at site MD95-2042, but appear to be related to the presence of relatively warm and nutrient-poor subtropical Eastern North Atlantic Central Water advected with the Azores Current.

With ever increasing demands to strengthen existing reinforced concrete structures to facilitate higher loading due to change of use and to extend service lifetime, the use of fibre reinforced polymers (FRPs) in structural retrofitting offers an opportunity to achieve these aims. To date, most research in this area has focussed on the use of glass fibre reinforced polymer (GFRP) and carbon fibre reinforced polymer (CFRP), with relatively little on the use of basalt fibre reinforced polymer (BFRP) as a suitable strengthening material. In addition, most previous research has been carried out using simply supported elements, which have not considered the beneficial influence of in-plane lateral restraint, as experienced within a framed building structure. Furthermore, by installing FRPs using the near surface mounted (NSM) technique, disturbance to the existing structure can be minimised.
This paper outlines BFRP NSM strengthening of one third scale laterally restrained floor slabs which reflect the inherent insitu compressive membrane action (CMA) in such slabs. The span-to-depth ratios of the test slabs were 20 and 15 and all were constructed with normal strength concrete (~40N/mm2) and 0.15% steel reinforcement. 0.10% BFRP was used in the retrofitted samples, which were compared with unretrofitted control samples. In addition, the bond strength of BFRP bars bonded into concrete was investigated over a range of bond lengths with two different adhesive thicknesses. This involved using an articulated beam arrangement in order to establish optimum bond characteristics for use in strengthening slab samples.

O objetivo deste trabalho foi avaliar as características físicas e químicas de queijo Serra da Estrela de pasta semi mole e correlacionar estes resultados com os resultados da análise sensorial. Foram alvo de estudo 29 amostras de queijo Serra da Estrela produzidas com 6 ecótipos de cardo diferentes. Os queijos foram produzidos em várias queijarias da região demarcada para a produção do queijo Serra da Estrela, mais propriamente dos concelhos de Penalva do Castelo, Oliveira do Hospital e Fornos de Algodres. Procedeu-se à avaliação das propriedades químicas como a humidade, o pH, cloretos, cinzas e proteínas, de propriedades físicas, como a textura e a cor e uma avaliação sensorial. Dos resultados obtidos foi possível verificar que os valores da humidade variaram entre 34, 9% e 49,8%. As cinzas e a proteína encontram-se abaixo dos valores na literatura (3,4%–5,5% e 13,0%–21,9%, respetivamente). Os cloretos variaram entre 1,1% e 3,0% e o pH entre 4,8 e 5,7. Verificou-se que com mais dias de maturação, e portanto menor humidade, as amostras apresentam, maior firmeza, menos adesividade e menos pegamento da pasta. Os valores da dureza da casca e da pasta variam entre 1,56N e 9,40N e 0,4 N e 3,4 N, respetivamente. A adesividade é bastante elevada (de -26,29 N.sec a -2,21 N.sec), o que traduz o caráter amanteigado destes queijos. Nas coordenadas de cor os valores de L* variaram entre 53,57 a 64,34, os de a* entre -4,53 e 1,29 e os de b* entre 15,56 e 29,03, revelando uma cor amarelada. Os resultados da análise sensorial não apresentam grande variabilidade entre os diferentes ecótipos e verificou-se que, em geral, as análises instrumentais realizadas acabaram por confirmar a percepção dos provadores.

Purpose: To develop processes for effective isolation and purification of recombinant human plasminogen activator (rhPA) from transgenic rabbit milk. Methods: Immunoaffinity chromatography was selected and improved by a special polyol-responsive monoclonal antibody (PR-mAb). Alteplase was used as immunogen because of its similarity to rhPA in terms of structure. The PR-mAb was prepared by hybridoma technology and screened by ELISA-elution assay. Screening antibody was performed using rhPA milk in an ELISA-elution assay. The antibody clone C4-PR-mAb was selected for immunoaffinity chromatography. The rhPA was effectively bound to immobilized C4-PR-mAb on the column and was eluted with Tris buffer comprising 0.75 mol/L ammonium sulfate and 40n% propanediol (pH7.9). The rhPA was further purified by passing through Chromdex75 gel filtration column. Results: There were 12 hybridoma strains selected into the polyol-responsive mAbs screen step and three hybridoma strains were superior for producing PR-mAbs (C1, C4, C8). The rhPA can be purified from transgenic rabbit milk and maintained a higher thrombolytic activity in vitro by FAPA. Conclusion: The results demonstrate the suitability of the alternative approach used in this study. Using immunoaffinity chromatography and gel filtration column is feasible and convenient for extracting rhPA from milk, and should be useful for purifying other tPA mutants or other novel recombinant milkderived proteins.
