Chromosomes of Didelphis albiventris, D. marsupialis, Philander opossum and Lutreolina crassicaudata, four species of marsupials with very similar karyotypes and 2n=22 were studied. All the chromosomes were acrocentrics except the X in L. crassicaudata, which is a metacentric. The G-band patterns of these species are similar but the distribution of constitutive heterochromatin differs among them as shown by C-banding. The hypothesis that the X in L. crassicaudata might be an isochromosome derived from the acrocentric X in the other species is discarded since G-and C-banding patterns differ in the two arms. In D. marsupialis the Ag-NORs are terminal and located in both arms of one pair and in the long arms of two pairs of medium-sized autosomes. In P. opossum the NOR-bearing chromosomes could be precisely identified through simultaneous silver staining and G-banding. The Ag-NORs are terminal and located at the short arm of pair 5 and the long arm of pair 7. © 1982 Dr W. Junk Publishers.

O rotavírus (RV) é o principal agente viral associado às gastrenterites, ocasionando em média 39% dos casos diarreicos que culminam em hospitalizaões, sendo responsável por cerca de 520.000 óbitos entre crianças menores de cinco anos de idade a cada ano. Pertencem à família Reoviridae, gnero Rotavirus, possui RNA de dupla fita (dsRNA) com 11 segmentos codificando 12 proteínas, sendo seis estruturais (VPs) e seis não estruturais (NSPs). A proteína VP4, juntamente com a VP7, compem a camada externa do RV, designando os getipos P e G, respectivamente. Até o momento foram descritos 23 tipos G e 31 tipos P. O getipo G9 emergiu em escala global e é possivelmente associado a manifestação clínica mais grave, estando geralmente acompanhado do getipo P[8]. O getipo G9 possui 6 linhagens distintas e o P[8] 4 linhagens. Este estudo objetivou caracterizar os genes VP7 e VP4 de RV do getipo G9, circulantes na região metropolitana de Belém, Pará, no período de 1999 a 2007. O dsRNA viral de 38 amostras selecionadas foi extrado a partir das suspensões fecais e submetido à eletroforese em gel de poliacrilamida para determinaão dos eletroferotipos, seguido da reaão de seqüenciamento. Na presente investigação, foi possível a análise de 32 amostras selecionadas, sendo todas getipo G9P[8] associadas ao eletroferotipo longo. A análise filogetica do gene VP7 demonstrou que as amostras G9 agruparam na linhagem 3 com elevados índices de similaridade, apresentando 8 substituições nucleotdicas. Contudo, apenas três modificaões aminoacídicas foram observadas nas posições 43 (I→V), 66 (AV) e 73 (Q→R), sendo estes resíduos 43 e 73 exclusivos das amostras do ano de 2007. A análise do gene VP4 demonstrou que as amostras P[8] agruparam na linhagem 3, identificando-se 15 substituições nucleotdicas, as quais ocasionaram quatro modificaões aminoacídicas nos resíduos 108 (V→I), 172 (R→K), 173 (I→V) e 275 (K→R). As modificaões nos resíduos 172 e 275 são exclusivos das amostras dos anos de 1999 a 2002. As amostras do presente estudo apresentaram elevada similaridade ao longo do tempo estudado. As amostras de 2007 foram as mais divergentes, tanto para o gene VP4 quanto para o gene VP7. É importante se proceder ao contnuo monitoramento do getipo G9 na região metropolitana de Belém, a fim de detectar possíveis variantes emergentes que possam representar um desafio as estratégias de imunizaão atuais.

Background: Acute respiratory infections (ARI) are the leading cause of infant mortality in the world, and human respiratory syncytial virus (HRSV) is one of the main agents of ARI. One of the key targets of the adaptive host immune response is the RSV G-protein, which is responsible for attachment to the host cell. There is evidence that compounds such as flavonoids can inhibit viral infection in vitro. With this in mind, the main purpose of this study was to determine, using computational tools, the potential sites for interactions between G-protein and flavonoids. Results: Our study allowed the recognition of an hRSV G-protein model, as well as a model of the interaction with flavonoids. These models were composed, mainly, of -helix and random coil proteins. The docking process showed that molecular interactions are likely to occur. The flavonoid kaempferol-3-O-α-L-arabinopyranosil-(2 → 1)-α-L-apiofuranoside-7-O-α-L-rhamnopyranoside was selected as a candidate inhibitor. The main forces of the interaction were hydrophobic, hydrogen and electrostatic. Conclusions: The model of G-protein is consistent with literature expectations, since it was mostly composed of random coils (highly glycosylated sites) and -helices (lipid regions), which are common in transmembrane proteins. The docking analysis showed that flavonoids interact with G-protein in an important ectodomain region, addressing experimental studies to these sites. The determination of the G-protein structure is of great importance to elucidate the mechanism of viral infectivity, and the results obtained in this study will allow us to propose mechanisms of cellular recognition and to coordinate further experimental studies in order to discover effective inhibitors of attachment proteins.

Let G be a group, W a nonempty G-set and M a Z2G-module. Consider the restriction map resG W : H1(G,M) → Pi wi∈E H1(Gwi,M), [f] → (resGG wi [f])i∈I , where E = {wi, i ∈ I} is a set of orbit representatives in W and Gwi = {g G | gwi = wi} is the G-stabilizer subgroup (or isotropy subgroup) of wi, for each wi ∈ E. In this work we analyze some results presented in Andrade et al [5] about splittings and duality of groups, using the point of view of Dicks and Dunwoody [10] and the invariant E'(G,W) := 1+dimkerresG W, defined when Gwi is a subgroup of infinite index in G for all wi in E, andM = Z2 (where dim = dimZ2). We observe that the theory of splittings of groups (amalgamated free product and HNN-groups) is inserted in the combinatory theory of groups which has many applications in graph theory (see, for example, Serre [12] and Dicks and Dunwoody [10]).

Background: Cytokines secreted by the adipose tissue influence inflammation and insulin sensitivity, and lead to metabolic disturbances. How certain single-nucleotide polymorphisms (SNPs) interfere on lifestyle interventions is unclear. We assessed associations of selected SNPs with changes induced by a lifestyle intervention. Methods: This 9-month intervention on diet and physical activity included 180 Brazilians at high cardiometabolic risk, genotyped for the TNF-alpha -308 G/A, IL-6 -174 G/C and AdipoQ 45 T/G SNPs. Changes in metabolic and inflammatory variables were analyzed according to these SNPs. Individuals with at least one variant allele were grouped and compared with those with the reference genotype. Results: In the entire sample (66.7% women; mean age 56.5 +/- 11.6 years), intervention resulted in lower energy intake, higher physical activity, and improvement in anthropometry, plasma glucose, HOMA-IR, lipid profile and inflammatory markers, except for IL-6 concentrations. After intervention, only variant allele carriers of the TNF-alpha -308 G/A decreased plasma glucose, after adjusting for age and gender (OR 2.96, p = 0.025). Regarding the IL-6 -174 G/C SNP, carriers of the variant allele had a better response of lipid profile and adiponectin concentration, but only the reference genotype group decreased plasma glucose. In contrast to individuals with the reference genotype, carriers of variant allele of AdipoQ 45 T/G SNP did not change plasma glucose, apolipoprotein B, HDL-c and adiponectin concentrations in response to intervention. Conclusion: The TNF alpha -308 G/A SNP may predispose a better response of glucose metabolism to lifestyle intervention. The IL-6 -174 G/C SNP may confer a beneficial effect on lipid but not on glucose metabolism. Our findings reinforce unfavorable effects of the AdipoQ 45 T/G SNP in lipid profile and glucose metabolism after intervention in Brazilians at cardiometabolic risk. Further studies are needed to direct lifestyle intervention to subsets of individuals at cardiometabolic risk.

BACKGROUND: Caring for a spouse with Alzheimer's disease is associated with increased psychological distress, impaired immunity, and heightened cardiovascular risk. Hyperreactivity of sympathetic and platelet activation responses to acute psychological stress, or the failure to recover quickly from stressful events, may constitute an important pathway linking stress and negative affect with cardiovascular disease (CVD). OBJECTIVES: (1) To evaluate associations between negative affect (i.e., depressive and anxious symptoms) with increased norepinephrine and P-selectin responses to an acute psychological stress task. (2) To establish whether these associations are augmented among elderly spousal caregivers (CG) compared to non-caregivers (NC). METHODS: Depressive (DEP) and anxious (ANX) symptoms from the Brief Symptom Inventory were assessed among 39 CG and 31 NC. Plasma norepinephrine levels (NE) and percent platelet P-selectin (PSEL) expression were assayed at three time-points: rest, immediately following a laboratory speech test (reactivity), and after 14 min of recovery. Results: Among CG, but not NC, increased symptoms of depression and anxiety were associated with delayed NE recovery (DEP: beta=.460, p=.008; ANX: beta=.361, p=.034), increased PSEL reactivity (DEP: beta=.703, p<.001; ANX: beta=.526, p=.002), and delayed PSEL recovery (DEP: beta=.372, p=.039; ANX: beta=.295, p=.092), while controlling for age, gender, aspirin use, antidepressant use, and preexisting CVD. Bivariate correlations showed delayed NE recovery was also associated with increased PSEL reactivity (r=.416) and delayed PSEL recovery (r=.372; all ps<.05) among CG but not NC. DISCUSSION: Among chronically stressed caregivers, increased levels of depressive and anxious symptoms are associated with prolonged sympathetic activation and pronounced platelet activation. These changes may represent one pathway linking caregiving stress to cardiovascular risk.
