Cell exclusion is the phenomenon whereby the hematocrit and viscosity of blood decrease in areas of high stress. While this is well known in naturally occurring Poiseuille flow in the human body, it has never previously been shown in Couette flow, which occurs in implantable devices including blood pumps. The high-shear stresses that occur in the gap between the boundaries in Couette flow are known to cause hemolysis in erythrocytes. We propose to mitigate this damage by initiating cell exclusion through the use of a spiral-groove bearing (SGB) that will provide escape routes by which the cells may separate themselves from the plasma and the high stresses in the gap. The force between two bearings (one being the SGB) in Couette flow was measured. Stained erythrocytes, along with silver spheres of similar diameter to erythrocytes, were visualized across a transparent SGB at various gap heights. A reduction in the force across the bearing for human blood, compared with fluids of comparable viscosity, was found. This indicates a reduction in the viscosity of the fluid across the bearing due to a lowered hematocrit because of cell exclusion. The corresponding images clearly show both cells and spheres being excluded from the gap by entering the grooves. This is the first time the phenomenon of cell exclusion has been shown in Couette flow. It not only furthers our understanding of how blood responds to different flows but could also lead to improvements in the future design of medical devices.

Electromyography readings (EMGs) from quadriceps of fifteen subjects were recorded during whole body vibration treatment at different frequencies (10-50 Hz). Additional electrodes were placed on the patella to monitor the occurrence of motion artifact, triaxial accelerometers were placed onto quadriceps to monitor motion. Signal spectra revealed sharp peaks corresponding to vibration frequency and its harmonics, in accordance with the accelerometer data. EMG total power was compared to that associated with vibration harmonics narrow bands, before and during vibration. On average, vibration associated power resulted in only 3% (±0.9%) of the total power prior to vibration and 29% (±13.4%) during vibration. Often, studies employ surface EMG to quantitatively evaluate vibration evoked muscular activity and to set stimulation frequency. However, previous research has not accounted for motion artifacts. The data presented in this study emphasize the need for the removal of motion artifacts, as they consistently affect RMS estimation, which is often used as a concise muscle activity index during vibrations. Such artifacts, rather unpredictable in amplitude, might be the cause of large inter-study differences and must be eliminated before analysis. Motion artifact filtering will contribute to thorough and precise interpretation of neuromuscular response to vibration treatment. © 2008 Elsevier Ltd. All rights reserved.

We present data on the development a new type of optical fibre polariser and the characterisation of its wavelength properties. The device is fashioned using a two step process. Firstly, a standard UV long period grating (LPG) with a period of 330μm is inscribed into hydrogenated SMF-28, followed by femtosecond laser ablation of a groove parallel to the fibre axis. The UV inscribed LPGs have inherently low birefringence. However, the removal of the cladding layer parallel to the location of the LPG within the fibre core (as a result the ablation) modifies the cladding modes that couple with the LPG. Furthermore, the groove breaks the fibre symmetry introducing a non-uniform stress profile across the fibre cross section leading to significant birefringence. We show that increasing the depth of the groove increases the birefringence, and this behaviour coupled with the ability to control the wavelength location of the LPGs attenuations peaks results in a polariser able to operate at almost any wavelength and birefringence. The maximum birefringence reported here as polarisation mode splitting was approximately 39±0.1nm with a polarisation loss of 10dB. © 2011 Copyright Society of Photo-Optical Instrumentation Engineers (SPIE).

The exponential growth of studies on the biological response to ocean acidification over the last few decades has generated a large amount of data. To facilitate data comparison, a data compilation hosted at the data publisher PANGAEA was initiated in 2008 and is updated on a regular basis (doi:10.1594/PANGAEA.149999). By January 2015, a total of 581 data sets (over 4 000 000 data points) from 539 papers had been archived. Here we present the developments of this data compilation five years since its first description by Nisumaa et al. (2010). Most of study sites from which data archived are still in the Northern Hemisphere and the number of archived data from studies from the Southern Hemisphere and polar oceans are still relatively low. Data from 60 studies that investigated the response of a mix of organisms or natural communities were all added after 2010, indicating a welcomed shift from the study of individual organisms to communities and ecosystems. The initial imbalance of considerably more data archived on calcification and primary production than on other processes has improved. There is also a clear tendency towards more data archived from multifactorial studies after 2010. For easier and more effective access to ocean acidification data, the ocean acidification community is strongly encouraged to contribute to the data archiving effort, and help develop standard vocabularies describing the variables and define best practices for archiving ocean acidification data.

We analyze the physical-chemical surface properties of single-slit, single-groove subwavelength-structured silver films with high-resolution transmission electron microscopy and calculate exact solutions to Maxwell’s equations corresponding to recent far-field interferometry experiments using these structures. Contrary to a recent suggestion the surface analysis shows that the silver films are free of detectable contaminants. The finite-difference time-domain calculations, in excellent agreement with experiment, show a rapid fringe amplitude decrease in the near zone (slit-groove distance out to 3–4 wavelengths). Extrapolation to slit-groove distances beyond the near zone shows that the surface wave evolves to the expected bound surface plasmon polariton (SPP). Fourier analysis of these results indicates the presence of a distribution of transient, evanescent modes around the SPP that dephase and dissipate as the surface wave evolves from the near to the far zone.

Nucleic acids (DNA and RNA) play essential roles in the central dogma of biology for the storage and transfer of genetic information. The unique chemical and conformational structures of nucleic acids – the double helix composed of complementary Watson-Crick base pairs, provide the structural basis to carry out their biological functions. DNA double helix can dynamically accommodate Watson-Crick and Hoogsteen base-pairing, in which the purine base is flipped by ~180° degrees to adopt syn rather than anti conformation as in Watson-Crick base pairs. There is growing evidence that Hoogsteen base pairs play important roles in DNA replication, recognition, damage or mispair accommodation and repair. Here, we constructed a database for existing Hoogsteen base pairs in DNA duplexes by a structure-based survey from the Protein Data Bank, and structural analyses based on the resulted Hoogsteen structures revealed that Hoogsteen base pairs occur in a wide variety of biological contexts and can induce DNA kinking towards the major groove. As there were documented difficulties in modeling Hoogsteen or Watson-Crick by crystallography, we collaborated with the Richardsons’ lab and identified potential Hoogsteen base pairs that were mis-modeled as Watson-Crick base pairs which suggested that Hoogsteen can be more prevalent than it was thought to be. We developed solution NMR method combined with the site-specific isotope labeling to characterize the formation of, or conformational exchange with Hoogsteen base pairs in large DNA-protein complexes under solution conditions, in the absence of the crystal packing force. We showed that there are enhanced chemical exchange, potentially between Watson-Crick and Hoogsteen, at a sharp kink site in the complex formed by DNA and the Integration Host Factor protein. In stark contrast to B-form DNA, we found that Hoogsteen base pairs are strongly disfavored in A-form RNA duplex. Chemical modifications N1-methyl adenosine and N1-methyl guanosine that block Watson-Crick base-pairing, can be absorbed as Hoogsteen base pairs in DNA, but rather potently destabilized A-form RNA and caused helix melting. The intrinsic instability of Hoogsteen base pairs in A-form RNA endows the N1-methylation as a functioning post-transcriptional modification that was known to facilitate RNA folding, translation and potentially play roles in the epitranscriptome. On the other hand, the dynamic property of DNA that can accommodate Hoogsteen base pairs could be critical to maintaining the genome stability.

Grazing mollusks are used as a food resource worldwide, and limpets are harvested commercially for both local consumption and export in several countries. This study describes a field experiment to assess the effects of simulated human exploitation of limpets Patella vulgata on their population ecology in terms of protandry (age-related sex change from male to female), growth, recruitment, migration, and density regulation. Limpet populations at two locations in southwest England were artificially exploited by systematic removal of the largest individuals for 18 months in plots assigned to three treatments at each site: no (control), low, and high exploitation. The shell size at sex change (L50: the size at which there is a 50:50 sex ratio) decreased in response to the exploitation treatments, as did the mean shell size of sexual stages. Size-dependent sex change was indicated by L50 occurring at smaller sizes in treatments than controls, suggesting an earlier switch to females. Mean shell size of P. vulgata neuters changed little under different levels of exploitation, while males and females both decreased markedly in size with exploitation. No differences were detected in the relative abundances of sexual stages, indicating some compensation for the removal of the bigger individuals via recruitment and sex change as no migratory patterns were detected between treatments. At the end of the experiment, 0–15 mm recruits were more abundant at one of the locations but no differences were detected between treatments. We conclude that sex change in P. vulgata can be induced at smaller sizes by reductions in density of the largest individuals reducing interage class competition. Knowledge of sex-change adaptation in exploited limpet populations should underpin strategies to counteract population decline and improve rocky shore conservation and resource management.

Coastal defences are proliferating in response to climate change, leading to the creation of more vertical substrata. Efforts are being made to mitigate their impacts and create novel habitats to promote biodiversity. Little is known about the effect of aspect (i.e. north–south directionality) and inclination on intertidal biodiversity in artificial habitats. Artificial and natural habitats were compared to assess the role of aspect and substratum inclination in determining patterns of biodiversity at two tidal heights (high and mid). We also compared grazing activity between north- and south-facing surfaces in natural habitats to examine the potential for differential grazing pressure to affect community structure and functioning. Results were variable but some clear patterns emerged. Inclination had no effect on biodiversity or abundance. There was a general trend towards greater taxon richness and abundance on north-facing than south-facing substrata in natural and artificial habitats. On natural shores, the abundance and grazing activity of ‘southern’ limpets (i.e. Patella depressa) was greater on south-facing than north-facing substrata, with possible implications for further range-expansion. These results highlight the importance of incorporating shaded habitats in the construction of artificial habitats. These habitats may represent an important refuge from grazing pressure and thermal and desiccation stress in a warming climate.

Starting from the relationship between urban planning and mobility management, TeMA has gradually expanded the view of the covered topics, always remaining in the groove of rigorous scientific in-depth analysis. During the last two years a particular attention has been paid on the Smart Cities theme and on the different meanings that come with it. The last section of the journal is formed by the Review Pages. They have different aims: to inform on the problems, trends and evolutionary processes; to investigate on the paths by highlighting the advanced relationships among apparently distant disciplinary fields; to explore the interaction’s areas, experiences and potential applications; to underline interactions, disciplinary developments but also, if present, defeats and setbacks. Inside the journal the Review Pages have the task of stimulating as much as possible the circulation of ideas and the discovery of new points of view. For this reason the section is founded on a series of basic’s references, required for the identification of new and more advanced interactions. These references are the research, the planning acts, the actions and the applications, analysed and investigated both for their ability to give a systematic response to questions concerning the urban and territorial planning, and for their attention to aspects such as the environmental sustainability and the innovation in the practices. For this purpose the Review Pages are formed by five sections (Web Resources; Books; Laws; Urban Practices; News and Events), each of which examines a specific aspect of the broader information storage of interest for TeMA.

On the national scene, soybean crop occupies a prominent position in cultivated area and volume production, being cultivated largely in the no tillage system. This system, due to the intense traffic of machines and implements on its surface has caused soil compaction problems, which has caused the yield loss of crops. In order to minimize this effect the seeder-drill uses the systems to opening the furrow by shank or the double disc type. The use of the shank has become commonplace for allowing the disruption of the compacted surface layer, however requires greater energy demand and may cause excessive tillage in areas where there is not observed high levels of compaction. Thus, this study aimed to evaluate the effects of furrowers mechanisms and levels of soil compacting on traction requirement by a seeder-drill and on the growing and productivity of soybean in an Oxisol texture clay, in a two growing seasons. The experimental design consisted of randomized blocks with split plots with the main plots composed of four levels of soil compaction (N0 – no tillage without additional compaction, N1, N2 and N3 – no tillage subjected to compaction through two, four and six passes with tractor, respectively) corresponding to densities of soil 1.16, 1.20, 1.22 and 1.26 g cm-3, and subplots by two furrowers mechanisms (shank and double disc) with four replicates. To evaluate the average, maximum and specific traction force requested by the seeder-drill, was used a load cell, with capacity of 50 kN and sensitivity of 2 mV V-1, coupled between the tractor and seeder-drill, whose data are stored in a datalogger system model CR800 of Campbell Scientific. In addition, were evaluated the bulk density, soil mechanical resistance to penetration, sowing depth, depth and groove width, soil area mobilized, emergence speed index, emergence operation, final plant stand, stem diameter, plant height, average number of seeds per pod, weight of 1,000 seeds, number of pods per plant and crop productivity. Data were subjected to analysis of variance, the mean of furrowers were compared by Tukey test (p≤0.05), while for the factor soil compaction, polynomial regression analysis was adopted, selected models by the criterion of greater R2 and significance (p≤0.05) of equation parameters. Regardless of the crop season, penetration resistance increase as soil compaction levels up to around 0.20 m deep, and bulk density influenced the sowing quality parameters, however, did not affect the crop yield. In the first season, there was a higher productivity with the use of the shank type. In the second crop season, the shank demanded greater energetic requirement with the increase of bulk density and opposite situation with the double disc. The locking of sowing lines allow better performance of the shank to break the compacted layer.
