Germline mutations in TP53 gene are associated with Li-Fraumeni syndrome (LFS) and its variants Li-Fraumeni-like (LFL). They predispose carriers to a wide variety of early onset tumors. In Brazil, there is a high frequency of a germline mutation in this gene (NC_000017.9: c.1010G>A; p.R337H) in Southern and Southeastern regions, due to a founder effect. It is estimated to be present in 0,3% ofthe local population, but only few families have been detected. Due to this significant divergence, the purpose of this study was to verify the effectiveness of wider criteria for detection of these individuals. Herein, clinical criteria were established, DNA samples were collected, analyzed by Restriction Fragment Length Polymorphism (RFLP) and sequenced. Thus, assessing the prevalence of this mutation in families with multiple cases of cancer. Based on our proposed criteria, one out of 31 patients (3,22%) was found to carry p.R337H mutation. The patient developed ductal invasive breast cancer at age 47, invasive adenocarcinoma of the lung at age 48 and soft-tissue sarcoma at age 49. In addition, an extensive cancer family history was referred, atypical for LFS, including a case of Ewing’s sarcoma. These outcomes indicate that the proposed criteria may detect probable carriers who did not fit previous LFS criteria. Nevertheless, additional studies, which might include a larger number of families and more stringent parameters, will be useful to improve screening sensibility

1. Hydatid cysts are found in more than 30 per cent of all cattle, sheep and goats in Kenya, but the disease is prevalent in man only in the semi-desert area of Turkana. Up to the time of the present investigation the life-cycle of the parasite in East Africa had not been studied, but it was suggested that wild carnivores, such as hyenas and jackals, might be the main hosts of the adult worms. 2. One hundred and forty-three carnivores, representing 23 species, have been examined. Echinococcus adults were found in 27 out of 43 domestic dogs (Canis familiaris), in three out of four hunting dogs (Lycaon pictus), in one out of nine jackals (Thos mesomelas), and in three out of 19 hyaenas (Crocuta crocuta). 3. A detailed morphological study was made of the Kenya material. After comparison with specimens from other parts of the world, it was concluded that the only species occurring in Kenya was E. granulosus, but it is possible that the minor morphological and biological differences are evidence of distinct strains. Further laboratory studies are necessary to compare the parasite from man and animals in different parts of Kenya with material from elsewhere. 4. A search was made for larval hydatids in 92 ungulates representing 18 species, and in a miscellaneous collection of nearly 2,000 rodents and primates representing a further 31 species. Only one animal was positive, a wildebeest (Gorgon taurinus). 5. The infections in the wild carnivores were all very light; only domestic dogs were heavily infected. It is concluded that the main cycle of transmission in Kenya is between dogs and domestic livestock. 6. Turkana tribesmen are the most heavily infected people in Kenya, either because the strain of parasite is more pathogenic to man in that area, or, more probably, because of the intimate contact between children and the large number of infected dogs. A particularly dangerous custom in the area is the use of dogs to clean the face and anal regions of babies when they vomit or have diarrhea. No satisfactory explanation can be given for the rarity of the disease in man in many of the other areas of Kenya where hydatids are very common in domestic animals. 7. The control of the disease will depend upon an active health-education campaign, together with the destruction of all unregistered dogs and improvement in meat hygiene.

Color vision was examined in subjects with long-term occupational exposure to mercury (Hg) vapor. The color vision impairment was assessed by employing a quantitative measure of distortion of individual and group perceptual color spaces. Hg subjects (n = 18; 42.1 +/- 6.5 years old; exposure time = 10.4 +/- 5.0 years; time away from the exposure source = 6.8 +/- 4.6 years) and controls (n = 18; 46.1 +/- 8.4 years old) were examined using two arrangement tests, D-15 and D-15d, in the traditional way, and also in a triadic procedure. From each subject`s `odd-one-out` choices, matrices of inter-cap subjective dissimilarities were derived and processed by non-metric multidimensional scaling (MDS). D-15d results differed significantly between the Hg-group and the control group (p < 0.05), with the impairment predominantly along the tritan axis. 2D perceptual color spaces, individual and group, were reconstructed, with the dimensions interpreted as the red-green (RG) and the blue-yellow (BY) systems. When color configurations from the Hg-group were compared to those of the controls, they presented more fluctuations along both chromatic dimensions, indicating a statistically significant difference along the BY axis. In conclusion, the present findings confirm that color vision impairments persist in subjects that have received long-term occupational exposure to Hg-vapor although, at the time of testing, they were presenting mean urinary concentration within the normal range for non-exposed individuals. Considering the advantages of the triadic procedure in clinical evaluation of acquired color vision deficiencies, further studies should attempt to verify and/or improve its efficacy.

A data set of a commercial Nellore beef cattle selection program was used to compare breeding models that assumed or not markers effects to estimate the breeding values, when a reduced number of animals have phenotypic, genotypic and pedigree information available. This herd complete data set was composed of 83,404 animals measured for weaning weight (WW), post-weaning gain (PWG), scrotal circumference (SC) and muscle score (MS), corresponding to 116,652 animals in the relationship matrix. Single trait analyses were performed by MTDFREML software to estimate fixed and random effects solutions using this complete data. The additive effects estimated were assumed as the reference breeding values for those animals. The individual observed phenotype of each trait was adjusted for fixed and random effects solutions, except for direct additive effects. The adjusted phenotype composed of the additive and residual parts of observed phenotype was used as dependent variable for models' comparison. Among all measured animals of this herd, only 3160 animals were genotyped for 106 SNP markers. Three models were compared in terms of changes on animals' rank, global fit and predictive ability. Model 1 included only polygenic effects, model 2 included only markers effects and model 3 included both polygenic and markers effects. Bayesian inference via Markov chain Monte Carlo methods performed by TM software was used to analyze the data for model comparison. Two different priors were adopted for markers effects in models 2 and 3, the first prior assumed was a uniform distribution (U) and, as a second prior, was assumed that markers effects were distributed as normal (N). Higher rank correlation coefficients were observed for models 3_U and 3_N, indicating a greater similarity of these models animals' rank and the rank based on the reference breeding values. Model 3_N presented a better global fit, as demonstrated by its low DIC. The best models in terms of predictive ability were models 1 and 3_N. Differences due prior assumed to markers effects in models 2 and 3 could be attributed to the better ability of normal prior in handle with collinear effects. The models 2_U and 2_N presented the worst performance, indicating that this small set of markers should not be used to genetically evaluate animals with no data, since its predictive ability is restricted. In conclusion, model 3_N presented a slight superiority when a reduce number of animals have phenotypic, genotypic and pedigree information. It could be attributed to the variation retained by markers and polygenic effects assumed together and the normal prior assumed to markers effects, that deals better with the collinearity between markers. Information about rainfall erosivity is important during soil and water conservation planning. Thus, the spatial variability of rainfall erosivity of the state Mato Grosso do Sul was analyzed using ordinary kriging interpolation. For this, three pluviograph stations were used to obtain the regression equations between the erosivity index and the rainfall coefficient EI30. The equations obtained were applied to 109 pluviometric stations, resulting in EI30 values. These values were analyzed from geostatistical technique, which can be divided into: descriptive statistics, adjust to semivariogram, cross-validation process and implementation of ordinary kriging to generate the erosivity map. Highest erosivity values were found in central and northeast regions of the State, while the lowest values were observed in the southern region. In addition, high annual precipitation values not necessarily produce higher erosivity values.

[EN]Gender recognition has achieved impressive results based on the face appearance in controlled datasets. Its application in the wild and large datasets is still a challenging task for researchers. In this paper, we make use of classical techniques to analyze their performance in controlled and uncontrolled condition respectively with the LFW and MORPH datasets. For both sets the benchmarking protocol follows the 5-fold cross-validation proposed by the BEFIT challenge.

The main goals of this Ph.D. study are to investigate the regional and global geophysical components related to present polar ice melting and to provide independent cross validation checks of GIA models using both geophysical data detected by satellite mission, and geological observations from far field sites, in order to determine a lower and upper bound of uncertainty of GIA effect. The subject of this Thesis is the sea level change from decades to millennia scale. Within ice2sea collaboration, we developed a Fortran numerical code to analyze the local short-term sea level change and vertical deformation resulting from the loss of ice mass. This method is used to investigate polar regions: Greenland and Antarctica. We have used mass balance based on ICESat data for Greenland ice sheet and a plausible mass balance for Antarctic ice sheet. We have determined the regional and global fingerprint of sea level variations, vertical deformations of the solid surface of the Earth and variations of shape of the geoid for each ice source mentioned above. The coastal areas are affected by the long wavelength component of GIA process. Hence understanding the response of the Earth to loading is crucial in various contexts. Based on the hypothesis that Earth mantle materials obey to a linear rheology, and that the physical parameters of this rheology can be only characterized by their depth dependence, we investigate the Glacial Isostatic Effect upon the far field sites of Mediterranean area using an improved SELEN program. We presented new and revised observations for archaeological fish tanks located along the Tyrrhenian and Adriatic coast of Italy and new RSL for the SE Tunisia. Spatial and temporal variations of the Holocene sea levels studied in central Italy and Tunisia, provided important constraints on the melting history of the major ice sheets.

Die Entstehung und Aufrechterhaltung von Knorpel- und Knochengewebe wird durch eine Vielzahl von hemmenden oder fördernden Faktoren hoch komplex reguliert, wobei die dabei involvierten physiologischen Prozesse bisher nur teilweise verstanden werden. Auch die Ursachen sowohl degenerativer Erkrankungen, aber auch durch Mutationen im FGFR3-Gen verursachter Chondrodysplasien sind in ihrer Ätiopathogenese noch nicht vollständig erforscht. In dieser Arbeit wurden verschiedene experimentelle Ansätze verfolgt, die zur weiteren Aufklärung der Pathophysiologie zweier unterschiedlicher Skeletterkrankungen beitragen sollten.rnEin relevantes Charakteristikum der degenerativen Gelenkserkrankung Osteoarthrose ist der Verlust an Aggrekan, hauptverantwortlich verursacht durch die Aggrekanase ADAMTS5. Es wurde ein Tiermodell generiert, bei dem gezielt mittels des Tet-ON-Systems die Aggrekanase mAdamts-5 überexprimiert werden kann. Nach Konstruktherstellung und Generierung als auch Charakterisierung des in vitro-Modells wurde das Tiermodell hergestellt, um die Folgen der Überexpression im Hinblick auf einen verstärkten Aggrekanabbau im Knorpel der Mäuse zu analysieren. Nach initialer Charakterisierung auf Induzierbarkeit zeigte eine Gründerlinie eine induzierbare transgene mAdamts5-Expression. Die Überprüfung auf Knorpelspezifität zeigte, sowohl embryonal als auch im adulten Tier, dass sich der verwendete, zusammengesetzte Kollagen-Typ II Promotor wie der endogene verhielt und somit funktional war. Nach Doxyzyklininduktion wurde bei der optimalen Dosis von 1 mg/ml im Vergleich zum induzierten Wildtyp-Tier eine 15%ige Abnahme des Gesamt-Glykosamino-glykan(GAG)-Gehaltes und eine um 120% erhöhte GAG-Abgabe ins Medium detektiert, was eine verstärkte Spaltung von Aggrekan bedeutete. Die transgene Aggrekanase wurde überexprimiert und spaltete verstärkt Aggrekan. Da aufgrund der histologischen Untersuchungen jedoch keine Knorpelerosionen feststellbar waren, konnte im Umkehrschluss gefolgert werden, dass der Knorpel einen Verlust an Glykosaminoglykanen bis zu einer gewissen Grenze tolerieren kann. Mit dem generierten und charakterisierten Tiermodell konnte mit dem Verlust an GAG eine Osteoarthrose-ähnliche Situation simuliert werden, insbesondere im Hinblick auf frühe Stadien der Erkrankung, bei denen noch keine makroskopisch eindeutig sichtbare Knorpelerosionen vorliegen. rnIm zweiten Teil der Arbeit wurden Zellkulturexperimente zur weiteren Aufklärung FGFR3-regulierter Prozesse durchgeführt. Nach Generierung und Verifizierung der stabilen Zelllinien, die mittels des Tet-ON-Systems das FGFR3-Gen mit jeweils einer Chondrodysplasie-assoziierten Mutation (Achondroplasie-Mutation G380R, Thanatophore Dysplasie Typ II-Mutation K650E) induzierbar überexprimieren, wurden die Auswirkungen der zwei verschiedenen Mutationen anhand bereits beschriebener Signalwege untersucht. Über die Rekrutierung des ERK-Signalweges konnte bei beiden Zelllinien die Funktionalität nachgewiesen werden, wobei die Zelllinie mit der einen schwereren Phänotyp beim Menschen verursachenden TDII-Mutation eine stärkere Aktivierung zeigte. Bei der Aktivierung von STAT1 wies nur die TDII-Zelllinie eine Phosphorylierung auf, nicht jedoch die ACH-Zelllinie; dies deckte sich mit bereits publizierten Untersuchungen. Beide Kaskaden zeigten eine unterschiedliche Signalantwort aufgrund der verschiedenen Mutationen. Des Weiteren konnte eine unterschiedliche MMP13-Zielgenexpression nachgewiesen werden, wobei lediglich die ACH-Zelllinie eine erhöhte MMP13-Expression (6-fach) zeigte. Zur Identifizierung neuer involvierter FGFR3-Zielgene wurde die differentielle Genexpression der TDII-Zelllinie im Vergleich induziert/nicht induziert mittels Microarray-Hybridisierung untersucht. Als interessantes Zielgen fiel STC1 auf, welches ebenfalls eine Rolle in der Chondrogenese spielt und bislang nicht mit FGFR3 in Verbindung gebracht wurde. Es konnte jedoch nur auf RNA-Ebene eine Regulation nachgewiesen werden. Nachfolgend durchgeführte transiente Experimente zeigten, dass die Wildtyp-Variante von FGFR3 möglicherweise eine Funktion in der Sekretion des Proteins STC1 hat und dass durch die beiden eingefügten Mutationen (ACH, TDII) diese aufgehoben ist. Der Einfluss von FGFR3 auf die Sekretion von STC1 stellt ein neues Ergebnis dar, insbesondere auch die Auswirkungen der beiden für die unterschiedlichen Krankheitsbilder stehenden Mutationen. Welche Relevanz allerdings die STC1-Sekretion im Rahmen FGFR3-assoziierter Erkrankungen hat, kann nicht eindeutig beurteilt werden. Weitere Faktoren aus dem hoch komplexen Zusammenspiel während der Knorpel/Knochenentwicklung müssen untersucht werden, um eine definitive Einordnung zu ermöglichen.

Il presente lavoro di tesi si pone nell'ambito dell'analisi dati attraverso un metodo (QDanet_PRO), elaborato dal Prof. Remondini in collaborazine coi Dott. Levi e Malagoli, basato sull'analisi discriminate a coppie e sulla Teoria dei Network, che ha come obiettivo la classificazione di dati contenuti in dataset dove il numero di campioni è molto ridotto rispetto al numero di variabili. Attraverso questo studio si vogliono identificare delle signature, ovvero un'insieme ridotto di variabili che siano in grado di classificare correttamente i campioni in base al comportamento delle variabili stesse. L'elaborazione dei diversi dataset avviene attraverso diverse fasi; si comincia con una un'analisi discriminante a coppie per identificare le performance di ogni coppia di variabili per poi passare alla ricerca delle coppie più performanti attraverso un processo che combina la Teoria dei Network con la Cross Validation. Una volta ottenuta la signature si conclude l'elaborazione con una validazione per avere un'analisi quantitativa del successo o meno del metodo.

To check the effectiveness of campaigns preventing drug abuse or indicating local effects of efforts against drug trafficking, it is beneficial to know consumed amounts of substances in a high spatial and temporal resolution. The analysis of drugs of abuse in wastewater (WW) has the potential to provide this information. In this study, the reliability of WW drug consumption estimates is assessed and a novel method presented to calculate the total uncertainty in observed WW cocaine (COC) and benzoylecgonine (BE) loads. Specifically, uncertainties resulting from discharge measurements, chemical analysis and the applied sampling scheme were addressed and three approaches presented. These consist of (i) a generic model-based procedure to investigate the influence of the sampling scheme on the uncertainty of observed or expected drug loads, (ii) a comparative analysis of two analytical methods (high performance liquid chromatography-tandem mass spectrometry and gas chromatography-mass spectrometry), including an extended cross-validation by influent profiling over several days, and (iii) monitoring COC and BE concentrations in WW of the largest Swiss sewage treatment plants. In addition, the COC and BE loads observed in the sewage treatment plant of the city of Berne were used to back-calculate the COC consumption. The estimated mean daily consumed amount was 107 ± 21 g of pure COC, corresponding to 321 g of street-grade COC.
