Recent laboratory studies have demonstrated that Prunus necrotic ringspot virus (PNRSV) (family Bromoviridae) can be readily transmitted when thrips and virus-bearing pollen are placed together on to test plants. For this transmission mechanism to result in stonefruit tree infection in the field, PNRSV-bearing pollen must be deposited onto surfaces of stonefruit trees on which thrips also occur. In a previous paper, we demonstrated that almost all pollen in a PNRSV-infected Japanese plum orchard in southeastern Queensland was deposited onto flowers, whereas few grains occurred on leaves and none on stems. Here, we present results of our investigation of thrips species composition, distribution and abundance on stonefruit trees in the same study area as our previous pollen deposition study. We collected a total of 2010 adult thrips from 13 orchards during the 1989, 1991 and 1992 flowering seasons of which all but 14 were in the suborder Terebrantia. Most (97.4%) terebrantian thrips were of three species, Thrips imaginis, Thrips australis and Thrips tabaci. Thrips tabaci as well as species mixtures that included T imaginis, T australis and T tabaci have been shown to transmit PNRSV via infected pollen in laboratory tests. Adult thrips were frequently collected from flowers but rarely from leaves and never from stems. Large and significant differences in numbers of T imaginis, T australis and T tabaci adults in flowers occurred among orchards and between seasons. No factor was conclusively related to thrips numbers but flowers of late-flowering stonefruit varieties tended to hold more thrips than those of early-flowering varieties. Our results indicate that the common thrips species present on stonefruit trees in the Granite Belt are also ones previously shown to transmit PNRSV via infected pollen in the laboratory and that these thrips are concentrated in stonefruit flowers where most stonefruit pollen is deposited. These results contribute to mounting circumstantial evidence that stonefruit flowers may be inoculated with PNRSV via an interaction of thrips with virus-bearing pollen and that this transmission mechanism may be an important cause of new tree infections in the field.

Ergot, caused by Claviceps africana, has emerged as a serious threat to sorghum hybrid seed production worldwide. In the absence of gene-for-gene-based qualitative resistance in commercial cultivars, varieties with high pollen production that can escape ergot infection are preferred. Recent demonstration of differences in ergot susceptibility among male-sterile lines has indicated the presence of partial resistance. Using chitin-specific fluorescin-isothiocyanate-conjugated wheat germ agglutin and callose-specific aniline blue, this study investigated the process of sorghum ovary colonization by C. africana. Conidia germinated within 24 h after inoculation (a.i.); the pathogen was established in the ovary by 79 h a.i., and at least half of the ovary was converted into sphacelial tissue by 120 h a.i. Changes in fungal cell wall chitin content and strategic callose deposition in the host tissue were associated with penetration and invasion of the ovary. The rate of ovary colonization differed in three male-sterile lines that also differed in ergot susceptibility. This work demonstrates a possible histological basis for partial resistance in male-sterile sorghum lines that could lay the foundation for variety improvement through further breeding and selection.

Approaching the fiftieth year since its original description, primary aldosteronism is now thought to be the commonest potentially curable and specifically treatable form of hypertension. Correct identification of patients with primary aldosteronism requires that the effects of time of day, posture, dietary sodium intake, potassium levels and medications on levels of aldosterone and renin be carefully considered. Accurate elucidation of the subtype is essential for optimal treatment, and adrenal venous sampling is the only reliable means of differentiating aldosterone-producing adenoma from bilateral adrenal hyperplasia. With genetic testing already available for one inherited form, making more cumbersome biochemical testing for that subtype virtually obsolete and bringing about improvements in treatment approach, an intense search is underway for genetic mutations causing other, more common familial varieties of primary aldosteronism.

Two varieties of adzuki beans (Vigna angularis), Bloodwood and Erimo, were stored at temperatures of 10, 20 or 30degreesC, and relative humidities (RH) 40 or 65%, and samples were analysed at 0, 1.5, 3 and 6 months. Storage at 30degreesC for > 1.5 months caused a significant decrease in the a(star) and b(star) colour values and darkening of the seed coat. Beans stored at 65% RH had lower L-star but higher a(star) and b(star) colour values than those stored at 40% RH. Bloodwood and Erimo samples showed similar trends in colour during storage. The best storage conditions for the preservation of the adzuki colour were 10degreesC and 65% RH. The Australian beans had lower L-star, a(star) and b(star) colour values than Japanese Erimo-shouzu beans and storage increased the difference.

Two varieties of adzuki grown in Australia, Bloodwood and Erimo, were stored for up to 6 months at three temperatures (10, 20 and 30 degreesC), and two relative humidities (RH; 40 and 65%). The amount of cell wall material increased with time under all storage conditions. This increase was greatest at 30 degreesC and 40% RH. Storage time and conditions did not affect the total pectin levels in the cell wall. Erimo constantly exhibited a higher total pectin level than Bloodwood. The Bloodwood soluble pectin, Ca++ and Mg++ and Erimo Ca++ in the cell wall remained stable during storage, while the Erimo soluble pectin and Mg++ exhibited a slight decrease at 20 and 30 degreesC after 3 months of storage. Functional knowledge of the physiological basis of crop adaptation to stress is a prerequisite for exploiting specific adaptation to stress environments in breeding programs. This paper presents an analysis of yield components for pearl millet, to explain the specific adaptation of local landraces to stress environments in Rajasthan, India. Six genotypes, ranging from high-tillering traditional landraces to low-tillering open-pollinated modern cultivars, were grown in 20 experiments, covering a range of nonstress and drought stress patterns. In each experiment, yield components (particle number, grain number, 100 grain mass) were measured separately for main shoots, basal tillers, and nodal tillers. Under optimum conditions, landraces had a significantly lower grain yield than the cultivars, but no significant differences were observed at yield levels around 1 ton ha(-1). This genotype x environment interaction for grain yield was due to a difference in yield strategy, where landraces aimed at minimising the risk of a crop failure under stress conditions, and modem cultivars aimed at maximising yield potential under optimum conditions. A key aspect of the adaptation of landraces was the small size of the main shoot panicle, as it minimised (1) the loss of productive tillers during stem elongation; (2) the delay in anthesis if mid-season drought occurs; and (3) the reduction in panicle productivity of the basal tillers under stress. In addition, a low investment in structural panicle weight, relative to vegetative crop growth rate, promoted the production of nodal tillers, providing a mechanism to compensate for reduced basal tiller productivity if stress occurred around anthesis. A low maximum 100 grain mass also ensured individual grain mass was little affected by environmental conditions. The strategy of the high-tillering landraces carries a yield penalty under optimum conditions, but is expected to minimise the risk of a crop failure, particularly if mid-season drought stress occurs. The yield architecture of low-tillering varieties, by contrast, will be suited to end-of-season drought stress, provided anthesis is early. Application of the above adaptation mechanisms into a breeding program could enable the identification of plant types that match the prevalent stress patterns in the target environments. Este estudo teve como foco inicial de investigação o modo como livros didáticos de alfabetização propõem o estudo das relações entre sons e letras e letras e sons, e como essa dimensão se articula (ou não) a uma concepção de alfabetização que toma o texto como unidade de ensino. Caracteriza-se como uma pesquisa de cunho documental, trazendo para estudo produções acadêmico-científicas acerca da temática de investigação, tendências teóricas no estudo da alfabetização, o processo histórico da política de avaliação de livros didáticos no Brasil, o Guia de livros didáticos – PNLD 2010 – letramento e alfabetização – língua portuguesa – 2009 e duas coleções de livros didáticos de alfabetização, quais sejam: A Escola é Nossa – Letramento e Alfabetização Linguística; e Porta Aberta – Letramento e Alfabetização Linguística, avaliadas e selecionadas pelo Programa Nacional do Livro Didático (PNLD), na edição de 2009, para o ano letivo de 2010. Assume a hipótese de que a proposta de trabalho com as relações sons e letras e letras e sons trazida pelos livros didáticos de alfabetização, no contexto do letramento, por não tomar o texto como unidade de ensino, acaba por criar obstáculos para a própria compreensão dessas relações pelos estudantes. Toma como princípios teóricos e metodológicos a abordagem bakhtiniana de linguagem, bem como a concepção de alfabetização que baliza este estudo (GONTIJO; SCHWARTZ, 2009). Conclui que, ao não trazer os textos (gêneros discursivos) como enunciados, indiferentes à alternância dos sujeitos do discurso, os livros analisados, não obstante as poucas diferenças existentes entre um e outro, que se referem mais especificamente a informações que tangem à linguística, vão ao encontro de uma concepção de linguagem como um sistema de normas que devem ser anteriormente internalizadas pelo estudante para que este possa proceder à leitura e à escrita. Tratam, pois, a língua materna como uma língua estrangeira ou morta, como se esta fosse estática, permanecendo imune à evolução histórica. O estudo corroborou a hipótese de investigação, uma vez que, desconsiderando e/ou desconhecendo o aspecto dialógico do enunciado, os livros analisados minimizam a possibilidade da instauração de uma abordagem discursiva de linguagem, o que incide no tratamento das relações sons e letras e letras e sons que acabam por apresentarem-se dicotomizadas do texto e seu contexto discursivo e, dessa forma, sua reflexão e sistematização pelos estudantes distancia-se de um estudo dessas relações no bojo dos aspectos sócio-históricos, ideológicos, linguísticos, estilísticos, dentre outros que perpassam seu ensino. Logo, por não propiciarem um tratamento discursivo da linguagem, pouco contribuem para um tratamento “linguístico” adequado, acabando por criar obstáculos para a compreensão dessas relações pelos estudantes. Entende que conhecimentos linguísticos, principalmente referentes às variedades linguísticas e dialetais, tornam-se importantes quando da abordagem dessas relações, entretanto, estes por si sós não garantem sua apropriação. Ressalta o necessário conhecimento por parte dos professores (e autores) acerca da abordagem linguística tomada pelo livro didático de alfabetização e o resgate da autoria docente diante do ensino da língua materna, instaurando um processo autoral-dialógico da produção de conhecimentos junto aos estudantes.

A presente dissertação investiga o atual estágio de manutenção das variedades dialetais da Itália setentrional no município de Santa Teresa, localizado na região serrana do Espírito Santo. Este trabalho se justifica porque, após 141 anos da chegada dos primeiros italianos a esse município, ainda não existem estudos que abordem questões relacionadas aos dialetos italianos da localidade. Considerando esse cenário, o objetivo deste estudo é oferecer um panorama da situação bilíngue português-dialeto italiano no município, com a identificação das áreas de maior ou menor uso do dialeto e ainda os fatores determinantes para a escolha linguística, os domínios de uso e as atitudes linguísticas dos falantes. Um segundo objetivo do estudo foi documentar algumas tradições orais italianas ainda presentes em Santa Teresa. Os dados foram coletados por meio de observação participante, questionário sociolinguístico e 146 entrevistas semiestruturadas, nas quais os informantes foram divididos por local de residência (zona rural e urbana) e em três faixas etárias (entre 08-30 anos, 31-60 e acima de 60 anos de idade). Os resultados encontrados revelam que o termo taliàn, que significa italiano nos dialetos da Itália setentrional (cf. BOERIO, 1856; RICCI, 1906 etc.), é usado pela maior parte dos falantes da faixa etária acima de 60 anos das zonas rural e urbana. Analisando diacronicamente o processo de uso do dialeto italiano através dos diferentes domínios, no período da infância dos informantes e na atualidade, é possível verificar a perda do dialeto no trajeto de vida dos falantes das faixas etárias de 31-60 anos e dos acima de 60 anos. Entre os informantes da faixa etária de 08-30 anos, verifica-se um quase completo monolinguismo português. Entre os informantes da faixa etária de 31-60 anos, o uso do dialeto italiano é fortemente influenciado pela idade do interlocutor: usam-no mais com seus avós do que com seus pais, e com seus pais mais do que com seus irmãos. Entretanto, nenhum informante desta faixa etária relatou usar o dialeto italiano com os filhos. Em resumo, o uso do dialeto italiano somente entre os membros mais idosos indica o processo de sua substituição pelo português e aponta que sua transmissão às gerações mais jovens está seriamente ameaçada. A análise das atitudes linguísticas dos informantes acima de 60 anos permitiu constatar o desprestígio e o preconceito em relação ao uso do dialeto no período da infância dos informantes. Por outro lado, os relatos em relação ao uso do dialeto na atualidade referem-se à associação da língua e da cultura de origem italiana com elementos positivos; à vontade explícita de manutenção do dialeto pelos adultos e idosos, à recuperação da língua de imigração pelos informantes de 08-30 anos. Aliás, entre os mais jovens, percebe-se uma tentativa de retorno às origens, de valorização da cultura e da língua dos antepassados.

The combined use of precision agriculture and the Diagnosis and Recommendation Integrated System (DRIS) allows the spatial monitoring of coffee nutrient balance to provide more balanced and cost-effective fertilizer recommendations. The objective of this work was to evaluate the spatial variability in the nutritional status of two coffee varieties using the Mean Nutritional Balance Index (NBIm) and its relationship with their respective yields. The experiment was conducted in eastern Minas Gerais in two areas, one planted with variety Catucaí and another with variety Catuaí. The NBIm of the two varieties and their yields were analyzed through geostatistics and, based on the models and parameters of the variograms, were interpolated to obtain their spatial distribution in the studied areas. Variety Catucai, with grater spatial variability, was more nutritional unbalanced than variety Catuai, and consequently produced lower yields. Excess of Fe and Mn makes these elements limiting yield factors.

In recent years, several new coffee cultivars recommended for different regions have been released. However, the performance of these varieties in many traditionally producing regions is unknown. Difference of climate and soil may jeopardize the productivity of the new cultivars and cause losses to farmers. The objective of this study is to evaluate the vegetative growth and productive genotypes of C. arabica in the conditions of the Northwestern Rio de Janeiro State, Brazil. The experiment was settled in 2007, in Panorama 1 Farm, located in the municipality of Varre Sai, RJ. Twenty-five genotypes of C. arabica were planted in a spacing of 2.5 × 0.8 m, using a completely randomized design with five replications and eight plants per plot. There were eight measurements of vegetative growth represented by plant height, stem diameter and number of plagiotropic branches. Assessments of productivity were also performed in years 2009 and 2010. There was a positive phenotypic correlation among vegetative characteristics and between vegetative characteristics and yield in the first harvest, while in the second harvest only the number of plagiotropic branches was positively correlated with yield. Up to date, the genotypes Catucaí amarelo 2 SL, Catiguá MG 02, Acauã, Palma II, Sabiá 398, IPR 103/ Iapar, IPR 100/Iapar, H 419-10-6-2-12-1, Catucaí amarelo 24 / 137, Iapar 59, Catucaí amarelo 20/15, H 419-10-6-2-5-10-1 and H 419-10-6-2-5-1 had the highest average yield after two harvests.

In vitro propagation has become an effective practice for large-scale production of strawberry plants. The objective of this study was to evaluate the hyperhydricity and the multiplication capacity of two strawberry varieties (Fragaria x ananassa Duch. 'Dover' and 'Burkley') propagated in vitro. Plants maintained in MS medium supplemented with 1.0 mg L-1 BA were individualized and transferred to the same medium solidified with Agar (6.5 g L-1) or Phytagel® (2.5 g L-1) and BA at different concentrations (0; 0.5; 1.0; 2.0 and 3.0 mg L-1). Biochemical and anatomical analyses were carried out, as well as the analysis of the morphological hyperhydricity characteristics. The analysis of data showed: a) the increase in cytokinin concentration increased hyperhydricity frequency in both varieties; b) at concentrations up to 2.0 mg L-1 BA, the replacement of Agar by Phytagel® induced a higher formation of hyperhydric shoots; and c) the addition of BA induced oxidative stress, which is characterized by increased antioxidant activity and lipid peroxidation, as well as alterations at the cellular level, such as malformation of stomata and epidermal cells. In conclusion, the culture medium containing 0.5 mg L-1 BA solidified with Agar provided lower hyperhydricity percentages in association with higher rates of shoot proliferation in strawberry.

The deficiency of essential micronutrients and excess of toxic metals in cereals, an important food items for human nutrition, can cause public health risk. Therefore, before their consumption and adoption of soil supplementation, concentrations of essential micronutrients and metals in cereals should be monitored. This study collected soil and two varieties of wheat samples–Triticum aestivum L. (Jordão/bread wheat), and Triticum durum L. (Marialva/durum wheat) from Elvas area, Portugal and analyzed concentrations of As, Cr, Co, Fe, K, Na, Rb and Zn using Instrumental Neutron Activation Analysis (INAA) to focus on the risk of adverse public health issues. The low variability and moderate concentrations of metals in soils indicated a lower significant effect of environmental input on metal concentrations in agricultural soils. The Cr and Fe concentrations in soils that ranged from 93–117 and 26,400–31,300 mg/kg, respectively, were relatively high, but Zn concentration was very low (below detection limit <22 mg/kg) indicating that soils should be supplemented with Zn during cultivation. The concentrations of metals in roots and straw of both varieties of wheat decreased in the order of K>Fe>Na>Zn>Cr>Rb>As>Co. Concentrations of As, Co and Cr in root, straw and spike of both varieties were higher than the permissible limits with exception of a few samples. The concentrations of Zn in root, straw and spike were relatively low (4–30 mg/kg) indicating the deficiency of an essential micronutrient Zn in wheat cultivated in Portugal. The elemental transfer from soil to plant decreases with increasing growth of the plant. The concentrations of various metals in different parts of wheat followed the order: Root>Straw>Spike. A few root, straw and spike samples showed enrichment of metals, but the majority of the samples showed no enrichment. Potassium is enriched in all samples of root, straw and spike for both varieties of wheat. Relatively to the seed used for cultivation, Jordão presented higher transfer coefficients than Marialva, in particular for Co, Fe, and Na. The Jordão and Marialva cultivars accumulated not statistically significant different concentrations of different metals. The advantages of using INAA are the multielementality, low detection limits and use of solid samples (no need of digestion).

Consumer awareness, pesticide and fertilizer contaminations and environmental concerns have resulted in significant demand for organically grown farm produce. Consumption of berries has become popular among health-conscious consumers due to the high levels of valuable antioxidants, such as anthocyanins and other phenolic compounds. The present study evaluated the influence that organic farming (OF) and integrated pest management (IPM) practise exert on the total phenolic content in 22 strawberry samples from four varieties. Postharvest performance of OF and IPM strawberries grown in the same area in the centre of Portugal and harvested at the same maturity stage were compared. Chemical profiles (phenolic compounds) were determined with the aid of HPLC-DAD/MS. Total phenolic content was higher for OF strawberry extracts. This study showed that the main differences in bioactive phytochemicals between organically and IPM grown strawberries concerned their anthocyanin levels. Organically grown strawberries were significantly higher in antioxidant activity than were the IPM strawberries, as measured by DPPH and FRAP assays.

High salinity causes remarkable losses in rice productivity worldwide mainly because it inhibits growth and reduces grain yield. To cope with environmental changes, plants evolved several adaptive mechanisms, which involve the regulation of many stress-responsive genes. Among these, we have chosen OsRMC to study its transcriptional regulation in rice seedlings subjected to high salinity. Its transcription was highly induced by salt treatment and showed a stress-dose-dependent pattern. OsRMC encodes a receptor-like kinase described as a negative regulator of salt stress responses in rice. To investigate how OsRMC is regulated in response to high salinity, a salt-induced rice cDNA expression library was constructed and subsequently screened using the yeast one-hybrid system and the OsRMC promoter as bait. Thereby, two transcription factors (TFs), OsEREBP1 and OsEREBP2, belonging to the AP2/ERF family were identified. Both TFs were shown to bind to the same GCC-like DNA motif in OsRMC promoter and to negatively regulate its gene expression. The identified TFs were characterized regarding their gene expression under different abiotic stress conditions. This study revealed that OsEREBP1 transcript level is not significantly affected by salt, ABA or severe cold (5 °C) and is only slightly regulated by drought and moderate cold. On the other hand, the OsEREBP2 transcript level increased after cold, ABA, drought and high salinity treatments, indicating that OsEREBP2 may play a central role mediating the response to different abiotic stresses. Gene expression analysis in rice varieties with contrasting salt tolerance further suggests that OsEREBP2 is involved in salt stress response in rice.
