The effect of deacetylated xanthan gum, additives (sucrose, soybean oil, sodium phosphate and propylene glycol) and pH modifications on mechanical properties, hydrophilicity and water activity of cassava starch-xanthan gum films has been studied. Sucrose addition resulted in the highest effect observed on cassava starch films elongation at break. The deacetylated xanthan gum had higher effect on elongation at break when comparing to the acetylated gum, although both gums presented an inferior effect in relation to the obtained with sucrose. However, when comparing to the control and PVC films, lower tensile strength resistance values were observed when adding sucrose. Increased water activity was observed for films added with sucrose, thus, increasing the material biodegradation. Sucrose and deacetylated xanthan gum addition resulted in a slight hydrophilicity increase. (C) 2004 Elsevier Ltd. All rights reserved.

A series of studies was conducted to establish a methodology for the accurate and efficient determination of betaine in different feed ingredients. The final methodology involves an extraction step in which the feed sample is heated for 3h in a methanolic KOH solution using a Goldfisch apparatus. Impurities are removed by the addition of activated charcoal and concentrated (36%) HCl. After centrifugation the extractant is passed through a strong cation exchange resin (Dowex 50W-X12, H+). The betaine retained in the column is eluted with 1.5 N HCl. A 2 nil aliquot of the elute is air dried and reconstituted with 1 ml of deionised water. HPLC separation with a cation exchange column (Partisil SCX-10) is used for the separation of betaine from other compounds. The mobile phase is kept constant at 50mm KH2PO4 in water, and eluted compounds are detected by UV absorbance (200nm). The flow rate is maintained at 1.5ml min(-1). This assay is very accurate over the range of betaine concentrations from 15 to 650 mug ml(-1), with a lower detection limit in feeds of approximately 500 mug g(-1) when 4g of sample is extracted. Recovery assays done with standard betaine hydrochloride and hard red wheat resulted in a consistent recovery of 80%. Betaine content was quantified in several feed ingredients, including alfalfa (1.77 mg kg(-1)), wheat (3.96 mg kg(-1)), wheat middlings (4.98 mg kg(-1)) and poultry meal (0.77 mg kg(-1)). Betaine in corn and soybean meal was not detectable by this method, even when 16g of sample was used (<125 mg kg(-1)). Betaine present in several feed ingredients should influence choline supplementation to animal feeds and may have implications for human health. (C) 2002 Society of Chemical Industry.

Nitrogen (N) mineralization dynamics in no-till systems is affected, among other factors, by N amount and quality in the mulch and by climatic conditions. Leaching of NO3-N and NH4-N from six plant species used as soil cover crops in tropical environments were evaluated when the straw was submitted to rainfall after chemical desiccation. Millet (Pennisetum glaucum), guinea sorghum (Sorghum vulgare), black oat (Avena strigosa), triticale (Triticum secale), Indian hemp (Crotalaria juncea), and brachiaria (Brachiaria decumbens) were grown in a greenhouse, in Botucatu-SP, Brazil. Forty-five days after emergence, the plants were cut at the root collar, oven-dried, and submitted to simulated rainfalls of 4.4, 8.7, 17.04, 34.9, and 69.8 mm, considering an amount of straw equivalent to 8 t ha(-1) of dry matter. The amounts of N-NO3- extracted from the straw by rainwater were very small. However, accumulated rainfall around 70 mm caused ammonium leaching ranging from 2.5 to 9.5kg ha(-1), depending on the species. Plant residues of triticale and black oat (grasses) and Indian hemp (legume) showed high N leaching intensity with the first rains after chemical desiccation. The amount of N leached from straw was highly correlated with N tissue content.

Soil compaction has a negative effect and Ca was shown to enhance root growth. The effects of soil subsurface compaction and liming on root growth and nutrient uptake by soybean were studied at the Department of Agriculture and Plant Breeding, São Paulo State University, Brazil. A Dark Red Latosol, sandy loam (Haplortox) was limed to raise base saturations to 40.1, 52.4 and 66.7%. The experimental pots were made of PVC tubes with 100 mm of diameter. Three rings with 150, 35 and 150 mm long were fixed one on the top of the other. In the central ring of 35 mm, the soil was compacted to bulk densities of 1.06, 1.25, 1.43 and 1.71 g.cm(-3). There was no effect of base saturation on soybean root and shoot growth and nutrition. Subsurface compaction led to an increase in root growth in the superficial layer of the pots with a correspondent quadratic decrease in the compacted layer. There was no effect of subsoil compaction on total root length and surface, soybean growth and nutrition. Soybean root growth was decreased by 10% and 50% when the soil penetrometer resistances were 0.52 MPa (bulk density of 1.45 g.cm(-1)) and 1.45 MPa (bulk density of 1.69 g.cm(-3)), respectively. In spite of the poor root growth in the compacted layer, once it nas overcome the root system showed an almost complete recovery.

Current knowledge of the bioavailability of lycopene in humans is limited due to the inability to distinguish newly administered lycopene from the body reserves of lycopene. A quantitative method to assess the absorption and relative bioavailability of newly absorbed synthetic or natural lycopene was developed using two deuterated lycopene sources, in conjunction with an advanced LC/APCI-MS (liquid chromatography/atmospheric pressure chemical ionization-mass spectrometry) to analyze newly absorbed lycopene in blood samples of study subjects. Two subjects (1 male and 1 female) consumed hydroponically grown tomatoes containing deuterium-enriched lycopene (8084 g wet weight tomato containing 16.3 and 17.4 mu mol lycopene, respectively) and two subjects (1 male, and 1 female) consumed 11 mu mol synthetic H-2(10) lycopene in 6 g of corn oil. Tomatoes were steamed and pureed. The doses were given together with a liquid formulated drink with 25% energy from fat. Our results showed that up to 34 days after taking an oral 2 1110 lycopene dose (synthetic or from tomato) with a liquid formula drink, the area under the curve of the average serum percent enrichment response of synthetic lycopene reached 33.9 (+/- 1.7) nmol-day/mu mol lycopene in the dose, whereas that of lycopene from the tomato dose was 11.8 (+/- 0.3) nmol-day/mu mol lycopene in the dose. Our study provides evidence that the absorption of physiological levels of lycopene in intrinsically labeled tomatoes can be studied in humans. From these preliminary investigations, we find that the bioavailability of synthetic lycopene in oil appears to be about three times higher than that of lycopene from steamed and pureed tomatoes. Sensitive immunologic techniques for the detection of alterations that occur in protein antigens were used to evaluate the immunogenicity of soybean glycinin after isolation, heat denaturation and pH alteration. The objective was to determine the effect of these agents on the immunogenic ability of this protein fraction. Immunologic assays performed on heat-denatured glycinin up to 80 degrees C in the presence of antinative glycinin serum demonstrated that glycinin retains its immunogenic properties. Above 90 degrees C this biological property begins to disappear, with protein insolubilization and epitope modification due to the conformational changes imposed by temperature. A reduction in immunogenicity also occurred when glycinin was taken to pH 2.0 (below its pi) and pH 11.00 (above its pi) and exposed to high temperatures in the presence of native antiglycinin serum. From these data one can conclude that, at extreme pH values, intramolecular reactions may occur which, in combination with the structural disorganization caused by high temperatures, may contribute to the reduction of immunogenicity.

Asiatic citrus canker, caused by Xanthomonas smithii ssp. citri, formerly X. axonopodis pv. citri, is one of the most serious phytosanitary problems in Brazilian citrus crops. Experiments were conducted under controlled conditions to assess the influence of temperature and leaf wetness duration on infection and subsequent symptom development of citrus canker in sweet orange cvs Hamlin, Natal, Pera and Valencia. The quantified variables were incubation period, disease incidence, disease severity, mean lesion density and mean lesion size at temperatures of 12, 15, 20, 25, 30, 35, 40 and 42 degrees C, and leaf wetness durations of 0, 4, 8, 12, 16, 20 and 24 h. Symptoms did not develop at 42 degrees C. A generalized beta function showed a good fit to the temperature data, severity being highest in the range 30-35 degrees C. The relationship between citrus canker severity and leaf wetness duration was explained by a monomolecular model, with the greatest severity occurring at 24 h of leaf wetness, with 4 h of wetness being the minimum duration sufficient to cause 100% incidence at optimal temperatures of 25-35 degrees C. Mean lesion density behaved similarly to disease severity in relation to temperature variation and leaf wetness duration. A combined monomolecular-beta generalized model fitted disease severity, mean lesion density or lesion size as a function of both temperature and duration of leaf wetness. The estimated minimum and maximum temperatures for the occurrence of disease were 12 degrees C and 40 degrees C, respectively.

The in vitro gas production of four single roughages and their paired combinations (1:1 on dry matter basis) were evaluated. Two roughage samples (100 mg) per treatment were fermented with ruminal fluid during a 48 h incubation period. Total 48 h gas volumes of fermentation dry matter (DM), neutral detergent fiber (NDF) and soluble compounds in neutral detergent (NDS) were for sugarcane = 16.8, 11.2, 6.9 mL; sugarcane + corn silage = 20.1, 12.6, 9.1 mL; sugarcane + 60-day elephantgrass = 16.5, 17.6 mL; sugarcane + 180-day elephantgrass = 13.8, 8.2, 5.9 mL; corn silage = 18.8, 16.8, 4.7 mL; corn silage + 60-day elephantgrass = 16.3, 15.4, 2.4 mL; corn silage + 180-day elephantgrass = 16.1, 11.8, 4.2 mL; 60-day elephantgrass = 16.9, 19.0 mL and 180-day elephantgrass = fermented 10.7, 12.2 mL, respectively. The NDS gas production was not possible to estimate for sugarcane + 60-day elephantgrass, 60-day elephantgrass and 180-day elephantgrass. The present data shows that the curves subtraction method can be an option to evaluate the contribution of the soluble fractions in roughages to digestion kinetics. However, this method underestimates the NDS gas contribution when roughages are low in crude protein and soluble carbohydrates. It is advisable to directly apply the two-compartmental mathematical model to the digestion curves for roughage DM, when determining the NDS gas volume and the digestion rate. This method is more straightforward and accurate when compared to the curve subtraction method. Non-structural carbohydrates combined with fiber and protein promoted a positive associative effect in sugarcane + corn silage (50:50) mixture. Therefore, it can be concluded that the soluble fraction of roughages greatly contributes to gas production. (C) 2004 Elsevier B.V. All rights reserved.

Occurrence of adults and biological aspects of Geniates borelli Camerano (Coleoptera, Scarabaeidae, Rutelinae) in Aquidauana, Mato Grosso do Sul, Brazil. Due to the importance of Geniates borelli Camerano as a pest in many crops, studies were developed at Universidade Estadual de Mato Grosso do Sul (UEMS). Aquidauana campus, MS, Brazil. Adults were collected with a light trap from January 2006 to December 2007. 3,320 adults were collected, and the highest quantities were obtained in October 2006 and November 2007, with 1,548 and 802 adults recorded, respectively. Collected adults were kept in plastic containers with soil and Brachiaria decumbens seedlings for oviposition. 535 eggs measuring 2.30 x 1.60 mm were obtained. As the embryonic development progressed, eggs increased in size to 3.00 x 2.70 mm, and this change occurred between 6 and 10 days after oviposit ion. The embryonic period lasted 17.9 days. The first, second, and third instars lasted 37.6, 49.7, and 74 days, respectively The prepupal stage lasted 65.9 days and the pupal stage lasted an average of 18.5 days. The biological cycle is completed in 315.8 days, which characterizes the species as univoltine. The average longevity of females was 35.4 days and 28.5 days for males.

Corn starch, partially hydrolyzed by fungal alpha-amylase was investigated by using thermal analysis, microscopy and X-ray diffraction. After enzymatic treatment lower degradation onset temperatures were observed. DSC analysis showed almost similar range of gelatinization temperature, however, the enthalpies of gelatinization increased for the partially hydrolyzed starch granules. According to the X-ray diffraction analysis, stronger cereal pattern peaks were recognized after enzymatic digestion. The results suggested that the hydrolysis was more pronounced in the amorphous part of the starch granules.

Although research on the environmental impacts of using waste as a fertilizer is of great importance, the basic principle for using a product as fertilizer is that it should provide nutrients for plants without causing any harm to them. The objective of this study was to evaluate the agronomic traits (number of nodes, plant height, leaf number, yield, and protein content of grains) and the nutritional status of corn treated with sewage sludge. The experiment was conducted in the municipality of Jaboticabal in a Red Latosol. A randomized block design with four treatments (0, 55, 110, and 167.5 Mg ha(-1) of sewage sludge) and five repetitions was used. At 30 days after emergence (DAE), the dose of 110 Mg ha(-1) dry weight presented greater values for plant height, leaf number and stem diameter. At 60 DAE, the treatments did not affect the agronomic traits. No influence from the treatments tested was observed for protein content of grains and yield. The dose of 167.5 Mg ha(-1) showed greater weight of 100 seeds. All treatments showed nutritional imbalances. This study confirmed the agricultural potential of sewage sludge as a source of nutrients.

Vegetated riparian buffer strips have been established in Southern Quebec (Canada) in order to intercept nutrients such as nitrate (NO(3)(-)) and protect water quality near agricultural fields. Buffer strips may also favour denitrification through a combination of high soil moisture, NO(3)(-) and carbon supply, which could lead to the production of nitrous oxide (N(2)O), a greenhouse gas. Denitrification could be further amplified by the presence of earthworms, or by plant species that promote earthworm and bacterial activity in soils. Soils from four farms, comprising maize fields and adjacent buffer strips, were sampled in the fall of 2008. A total of six earthworm species were found, but average earthworm biomass did not differ between buffer strips and maize agroecoecosystems. Nitrate concentrations and net nitrification rates were higher in the maize fields than in the buffer strips: there was no difference in N(2)O production in soils collected from the two sampling locations. Potential denitrification, measured by acetylene inhibition, varied by two orders of magnitude, depending on experimental conditions: when amended with H(2)O or with H(2)O + NO3-, potential denitrification was higher (P < 0.05) in soils from buffer strips than from maize fields. Potential denitrification was highest in soils amended with H(2)O+glucose, or with H(2)O+ NO(3)(-) + glucose. Using microcosms, we tested the effect of litter-soil mixtures on earthworm growth, and the effect of earthworm-litter-soil mixtures on potential denitrification. Based on four categories of chemical assays, litters of woody species (oak, apple, Rhododendron) were generally of lower nutritional quality than litter from agronomic species (alfalfa, switchgrass, corn stover). Alfalfa litter had the most positive effect, whereas apple litter had the most negative effect, on earthworm growth. Potential denitrification was 2-4 times higher in earthworm-litter-soil mixtures than in plain soil. Litter treatments that included corn stover had lower potential denitrification than those that included alfalfa or switchgrass, whereas litter treatments that included oak had lower potential denitrification than those that included apple or Rhododendron. Results suggest that potential N(2)O emissions may be higher in riparian buffer strips than in adjacent maize fields, that N(2)O emissions in buffer strips may be amplified by comminuting earthworms, and that plant litters that reduce earthworm growth may not be best at mitigating N(2)O emissions. The work described was part of the programme, Innovative biological indicators to improve the efficiency of water and nitrogen use and the fruit quality in tree crops Project, a partnership between ISA and INRA. Field studies were conducted in Portugal on different irrigated plots of nectarine trees; a fully irrigated (unstressed plot) and a plot that was not irrigated for some days (stressed plot). The aim of this work was to investigate the effects of plant water stress on canopy temperature, to determine the nonwater-stressed baseline and to observe diurnal and seasonal variations of Crop Water Stress Index (CWSI). Canopy temperature, psychrometric and wind speed data were taken each half-hour, between 9:30 and 15:30 h. Results showed that canopy temperature was higher during the daytime, for both unstressed and stressed plots. A linear regression of canopy-air temperature differential and the vapor pressure deficit (non-water-stress baseline) showed a r2= 0.65. During the stress period, the average canopy temperature of the stressed plot was up to 5.4°C higher than the unstressed plot. Diurnal and seasonal average of CWSI values showed differences between unstressed and stressed plots, during the stress period.
