BAFF, APRIL and their receptors play important immunological roles, especially in the B cell arm of the immune system. A number of splice isoforms have been described for both ligands and receptors in this subfamily, some of which are conserved between mouse and human, while others are species-specific. Structural and mutational analyses have revealed key determinants of receptor-ligand specificity. BAFF-R has a strong selectivity for BAFF; BCMA has a higher affinity for APRIL than for BAFF, while TACI binds both ligands equally well. The molecular signaling events downstream of BAFF-R, BCMA and TACI are still incompletely characterized. Survival appears to be mediated by upregulation of Bcl-2 family members through NF-kappaB activation, degradation of the pro-apototic Bim protein, and control of subcellular localization of PCKdelta. Very little is known about other signaling events associated with receptor engagement by BAFF and APRIL that lead for example to B cell activation or to CD40L-independent Ig switch.

El objetivo principal de este trabajo es determinar si las comunidades virtuales de práctica son un espacio adecuado para la autoformación de los docentes. Con este fin, primero se especifican algunos conceptos clave que ayuden a contextualizar el tema de la formación; en segundo lugar se recogen los rasgos que se consideran hoy necesarios para llevar a cabo la labor docente, dibujándose así el perfil “ideal” del profesor en el contexto actual desde distintos ángulos de vista; y por último, se narran algunas experiencias personales en las redes sociales para ilustrar el funcionamiento de este nuevo paradigma social.

Inhalation of fungal particles is a ubiquitous way of exposure to microorganisms during human life; however, this exposure may promote or exacerbate respiratory diseases only in particular exposure conditions and human genetic background. Depending on the fungal species and form, fungal particles can induce symptoms in the lung by acting as irritants, aeroallergens or pathogens causing infection. Some thermophilic species can even act in all these three ways (e.g. Aspergillus, Penicillium), mesophilic species being only involved in allergic and/or non-allergic airway diseases (e.g. Cladosporium, Alternaria, Fusarium). The goal of the present review is to present the current knowledge on the interaction between airborne fungal particles and the host immune system, to illustrate the differences of immune sensing of different fungal species and to emphasise the importance of conducting research on non-conventional mesophilic fungal species. Indeed, the diversity of fungal species we inhale and the complexity of their composition have a direct impact on fungal particle recognition and immune system decision to tolerate or respond to those particles, eventually leading to collateral damages promoting airway pathologies.

Nilotinib, a novel tyrosine kinase inhibitor (TKI) that inhibits BCR-ABL, the stem cell factor receptor (KIT), and platelet-derived growth factor receptor-alpha (PDGFRα), is approved for the treatment of patients with newly diagnosed Philadelphia chromosome-positive chronic myelogenous leukemia (CML) and those with CML that is imatinib-resistant or -intolerant. Due to its potent inhibition of KIT and PDGFRα--the two tyrosine kinases that are the central oncogenic mechanisms of gastrointestinal stromal tumors (GIST)--nilotinib also has been investigated for potential efficacy and safety in patients with GIST who have progressed on other approved treatments. Initial results have been encouraging, as nilotinib has demonstrated clinical efficacy and safety in a phase I trial as either a single agent or in combination with imatinib, as well as in heavily pretreated patients with GIST in a compassionate use program. In addition, the phase III trial of nilotinib versus best supportive care (with or without a TKI at the investigator's discretion) indicated that nilotinib may have efficacy in some third-line patients. Furthermore, the Evaluating Nilotinib Efficacy and Safety in Clinical Trials (ENEST g1 trial), a phase III randomized, open-label study comparing the safety and efficacy of imatinib versus nilotinib in the first-line treatment of patients with GIST, is currently under way. Other studies with nilotinib either have been initiated or are in development. Based on published and accruing clinical data, nilotinib shows potential as a new drug in the clinician's armamentarium for the management of GIST.

Despite numerous discussions, workshops, reviews and reports about responsible development of nanotechnology, information describing health and environmental risk of engineered nanoparticles or nanomaterials is severely lacking and thus insufficient for completing rigorous risk assessment on their use. However, since preliminary scientific evaluations indicate that there are reasonable suspicions that activities involving nanomaterials might have damaging effects on human health; the precautionary principle must be applied. Public and private institutions as well as industries have the duty to adopt preventive and protective measures proportionate to the risk intensity and the desired level of protection. In this work, we present a practical, 'user-friendly' procedure for a university-wide safety and health management of nanomaterials, developed as a multi-stakeholder effort (government, accident insurance, researchers and experts for occupational safety and health). The process starts using a schematic decision tree that allows classifying the nano laboratory into three hazard classes similar to a control banding approach (from Nano 3 - highest hazard to Nano1 - lowest hazard). Classifying laboratories into risk classes would require considering actual or potential exposure to the nanomaterial as well as statistical data on health effects of exposure. Due to the fact that these data (as well as exposure limits for each individual material) are not available, risk classes could not be determined. For each hazard level we then provide a list of required risk mitigation measures (technical, organizational and personal). The target 'users' of this safety and health methodology are researchers and safety officers. They can rapidly access the precautionary hazard class of their activities and the corresponding adequate safety and health measures. We succeed in convincing scientist dealing with nano-activities that adequate safety measures and management are promoting innovation and discoveries by ensuring them a safe environment even in the case of very novel products. The proposed measures are not considered as constraints but as a support to their research. This methodology is being implemented at the Ecole Polytechnique de Lausanne in over 100 research labs dealing with nanomaterials. Information and Communication Technologies provide public administrations new ways to meet their users' needs. At the same time, e-Government practices support the public sector in improving the quality of service provision and of its internal operations. In this paper we discuss the impacts of digitization on the management of administrative procedures. The theoretical framework and the research model that we will use in this study help us tackle the question of how digitization transforms administrative procedures as, for example, in terms of time and roles. The multiplicity of institutions involved in issuing building permits led us to consider this administrative procedure as a very interesting case study. An online survey was first addressed to Swiss civil servants to explore the field, and here we present some of its results. We are currently undertaking an in-depth case study of the building permit procedures in three Swiss Cantons, which we will also present in this paper. We will conclude with a discussion and the future steps of this project.

Spondylo-megaepiphyseal-metaphyseal dysplasia (SMMD; OMIM 613330) is a dysostosis/dysplasia caused by recessive mutations in the homeobox-containing gene, NKX3-2 (formerly known as BAPX1). Because of the rarity of the condition, its diagnostic features and natural course are not well known. We describe clinical and radiographic findings in six patients (five of which with homozygous mutations in the NKX3-2 gene) and highlight the unusual and severe changes in the cervical spine and the neurologic complications. In individuals with SMMD, the trunk and the neck are short, while the limbs, fingers and toes are disproportionately long. Radiographs show a severe ossification delay of the vertebral bodies with sagittal and coronal clefts, missing ossification of the pubic bones, large round "balloon-like" epiphyses of the long bones, and presence of multiple pseudoepiphyses at all metacarpals and phalanges. Reduced or absent ossification of the cervical vertebrae leads to cervical instability with anterior or posterior kinking of the cervical spine (swan neck-like deformity, kyknodysostosis). As a result of the cervical spine instability or deformation, five of six patients in our series suffered cervical cord injury that manifested clinically as limb spasticity. Although the number of individuals observed is small, the high incidence of cervical spine deformation in SMMD is unique among skeletal dysplasias. Early diagnosis of SMMD by recognition of the radiographic pattern might prevent of the neurologic complications via prophylactic cervical spine stabilization. © 2012 Wiley Periodicals, Inc.

Os microrganismos realizam processos imprescindíveis para a sustentabilidade dos ecossistemas e para a vida no planeta; apesar disso, a sociedade de modo geral ignora esse papel e os considera apenas do ponto de vista de patógenos. Este trabalho foi realizado com os objetivos de analisar o conteúdo sobre os microrganismos do solo nos livros didáticos de Biologia adotados nas escolas de ensino médio de Lavras, MG; caracterizar estudantes e professores de três escolas desta cidade; e avaliar a percepção sobre Microbiologia do Solo de 334 estudantes do ensino médio dessas escolas após a apresentação do assunto em diversos formatos atrativos (palestras, workshops e aulas práticas). A coleta de dados, a priori e posteriori, foi realizada por questionários; avaliou-se ainda a viabilidade de trabalhar esse conteúdo no ensino médio. Apesar da importância dos microrganismos do solo, os materiais didáticos para o ensino de Biologia no ensino médio os abordam superficialmente e não destacam sua relevância nos contextos agrícola e ambiental. Após apresentação do assunto, houve diferenças significativas na percepção por estudantes das escolas tanto públicas como privadas. Concluiu-se ser possível ensinar e melhorar esse conteúdo da disciplina de Biologia por meio de recursos acessíveis e constatou-se a necessidade de levar os avanços científicos conhecidos na universidade para atualizar o ensino médio.

Quantitative approaches in ceramology are gaining ground in excavation reports, archaeological publications and thematic studies. Hence, a wide variety of methods are being used depending on the researchers' theoretical premise, the type of material which is examined, the context of discovery and the questions that are addressed. The round table that took place in Athens on November 2008 was intended to offer the participants the opportunity to present a selection of case studies on the basis of which methodological approaches were discussed. The evaluation’s overarching question was “Did the activities undertaken through the state’s LSTA plan achieve results related to priorities identified in the Act?” The evaluation was conducted and is organized according to the six LSTA priorities. The research design employed two major methodologies: 1. Data sources from Iowa Library Services / State Library of Iowa2 as well as U.S and state sources were indentified for quantitative analysis. These sources, which primarily reflect outputs for various projects, included: Statistics from the Public Library Annual Survey Statistics collected internally by Iowa Library Services such as number of libraries subscribing to sponsored databases, number of database searches, attendance at continuing education events, number of interlibrary loan transactions Evaluation surveys from library training sessions, professional development workshops and other programs supported by LSTA funds Internal databases maintained by Iowa Library Services Impact results from post training evaluations conducted by Iowa Library Services 2010 Iowa census data from the U.S. Census Bureau LSTA State Program Reports for the grant period 2. Following the quantitative analysis, the evaluator gathered qualitative data through interviews with key employees, a telephone focus group with district library consultants and two surveys: LSTA Evaluation Survey (Public Libraries) and LSTA Evaluation Survey (Academic Libraries). Both surveys provided sound samples with 43 representatives of Iowa’s 77 academic libraries and 371 representatives of Iowa’s 544 public libraries participating. Respondents represented libraries of all sizes and geographical areas. Both surveys included multiple choice and rating scale items as well as open-ended questions from which results were coded to identify trends, issues and recommendations.

QUESTIONS UNDER STUDY: The starting point of the interdisciplinary project "Assessing the impact of diagnosis related groups (DRGs) on patient care and professional practice" (IDoC) was the lack of a systematic ethical assessment for the introduction of cost containment measures in healthcare. Our aim was to contribute to the methodological and empirical basis of such an assessment. METHODS: Five sub-groups conducted separate but related research within the fields of biomedical ethics, law, nursing sciences and health services, applying a number of complementary methodological approaches. The individual research projects were framed within an overall ethical matrix. Workshops and bilateral meetings were held to identify and elaborate joint research themes. RESULTS: Four common, ethically relevant themes emerged in the results of the studies across sub-groups: (1.) the quality and safety of patient care, (2.) the state of professional practice of physicians and nurses, (3.) changes in incentives structure, (4.) vulnerable groups and access to healthcare services. Furthermore, much-needed data for future comparative research has been collected and some early insights into the potential impact of DRGs are outlined. CONCLUSIONS: Based on the joint results we developed preliminary recommendations related to conceptual analysis, methodological refinement, monitoring and implementation.

Con la finalidad de adaptarnos al EEES, desarrollamos una herramienta que nos permitiera realizar un proceso de evaluación continua de la asignatura troncal de Toxicología. En el presente trabajo presentamos los resultados de este modelo en el que utilizamos los seminarios como elementos básicos de este proceso. Describimos cómo se estructuran y desarrollan estos seminarios, así como el modelo de evaluación de los mismos. Los seminarios fueron evaluados con una puntuación máxima del 30 % sobre la nota final de la signatura, y la participación en los mismos con un máximo del 10 %. Algunos de estos seminarios incorporaban evaluaciones realizadas antes del desarrollo de los mismos, que denominábamos «pre», y otras justo al final del desarrollo de los mismos, que denominábamos «post». Esta herramienta de evaluación continua se ha mostrado muy eficaz en lo que respecta al grado de participación y preparación de los alumnos. Además, ha supuesto un cambio significativo en el grado de implicación de los profesores, y una mejora de la comunicación alumno-profesor.

En el presente trabajo presentamos los resultados de este modelo en el que utilizamos los seminarios como elementos básicos de este proceso. Describimos cómo se estructuran y desarrollan estos seminarios, así como el modelo de evaluación de los mismos.
