The Quinone outside Inhibitors (QoI) are one of the most important and recent fungicide groups used in viticulture and also allowed by Integrated Pest Management. Azoxystrobin, kresoxim-methyl and trifloxystrobin are the main active ingredients for treating downy and powdery mildews that can be present in grapes and wines. In this paper, a method is reported for the analysis of these three QoI-fungicides in grapes and wine. After liquid–liquid extraction and a clean-up on commercial silica cartridges, analysis was by isocratic HPLC with diode array detection (DAD) with a run time of 13 min. Confirmation was by solid-phase micro-extraction (SPME), followed by GC/MS determination. The main validation parameters for the three compounds in grapes and wine were a limit of detection up to 0.073mg kg-1, a precision not exceeding 10.0% and an average recovery of 93% ±38.

An optimised version of the Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS) method for simultaneous determination of 14 organochlorine pesticides in carrots was developed using gas chromatography coupled with electron-capture detector (GC-ECD) and confirmation by gas chromatography tandem mass spectrometry (GC-MS/MS). A citrate-buffered version of QuEChERS was applied for the extraction of the organochlorine pesticides, and for the extract clean-up, primary secondary amine, octadecyl-bonded silica (C18), magnesium sulphate (MgSO4) and graphitized carbon black were used as sorbents. The GC-ECD determination of the target compounds was achieved in less than 20 min. The limits of detection were below the EUmaximum residue limits (MRLs) for carrots, 10–50 μg kg−1, while the limit of quantification did exceed 10 μg kg−1 for hexachlorobenzene (HCB). The introduction of a sonication step was shown to improve the recoveries. The overall average recoveries in carrots, at the four tested levels (60, 80, 100 and 140 μg kg−1), ranged from 66 to 111% with relative standard deviations in the range of 2– 15 % (n03) for all analytes, with the exception of HCB. The method has been applied to the analysis of 21 carrot samples from different Portuguese regions, and β-HCH was the pesticide most frequently found, with concentrations oscillating between less than the limit of quantification to 14.6 μg kg−1. Only one sample had a pesticide residue (β-HCH) above the MRL, 14.6 μg kg−1. This methodology combines the advantages of both QuEChERS and GC-ECD, producing a very rapid, sensitive and reliable procedure which can be applied in routine analytical laboratories.

Four Cynara cardunculus clones, two from Portugal and two from Spain were studied for biomass production and their lignin was characterized. The clones differed in biomass partitioning: Spanish clones produced more capitula (54.5% vs. 43.9%), and Portuguese clones more stalks (37.2% vs. 25.6%). The heating values (HHV0) of the stalks were similar, ranging from 17.1 to 18.4 MJ/kg. Lignin was studied by analytical pyrolysis (Py-GC/MS(FID)), separately in depithed stalks (stalksDP) and pith. StalksDP had in average higher relative proportions of lignin derived compounds than pith (23.9% vs. 21.8%) with slightly different lignin monomeric composition: pith samples were richer in syringyl units as compared to stalksDP (64% vs. 53%), with S/G ratios of 2.1 and 1.3, respectively. The H:G:S composition was 7:40:53 in stalksDP and 7:29:64 in pith. The lignin content ranged from 18.8% to 25.5%, enabling a differentiation between clones and provenances. © 2015 Elsevier Ltd. This study analysed 22 strawberry and soil samples after their collection over the course of 2 years to compare the residue profiles from organic farming with integrated pest management practices in Portugal. For sample preparation, we used the citrate-buffered version of the quick, easy, cheap, effective, rugged, and safe (QuEChERS) method. We applied three different methods for analysis: (1) 27 pesticides were targeted using LC-MS/MS; (2) 143 were targeted using low pressure GC-tandem mass spectrometry (LP-GC-MS/MS); and (3) more than 600 pesticides were screened in a targeted and untargeted approach using comprehensive, two-dimensional gas chromatography time-of-flight mass spectrometry (GC × GC-TOF-MS). Comparison was made of the analyses using the different methods for the shared samples. The results were similar, thereby providing satisfactory confirmation of both similarly positive and negative findings. No pesticides were found in the organic-farmed samples. In samples from integrated pest management practices, nine pesticides were determined and confirmed to be present, ranging from 2 μg kg−1 for fluazifop-pbutyl to 50 μg kg−1 for fenpropathrin. Concentrations of residues in strawberries were less than European maximum residue limits.

Pyrethroids are pesticides very used in agriculture, which tend to replace organophosfate and carbamate insecticides. These pesticides have shown to exhibit cardiotoxicity. The aim of this study was to assess if cardiotoxicity is due to direct or indirect effects (metabolites) of permethrin on hearts. There were studies 8 rats; three of them were sacrificed after 24 of the end of treatment with permethrin and the other four were sacrificed after 14 days of the end of treatment. Afterwards hearts and urines were collected. The amounts of permethrin and its main metabolite (3-PBA) were evaluated on hearts and urines of female rats which were treated with permethrin and sacrificed the day after and 14 days after the treatment. Moreover has been highlighted the difference of amount of permethrin and its metabolite between rats sacrificed immediately at the end of treatment and those sacrificed after 14 days. The study of permethrin was accomplished by liquid-liquid extraction and GC-ECD. The evaluation of 3-PBA was performed by SPE procedure with 2-PBA as internal standard and gas-chromatography GC-MS. The concentration of permethrin in hearts is basically the same in 24h and 14 days. The 3-PBA concentration in urines decreased 50 times from 24h to 14 days. In hearts the 3-PBA level also decrease but only 2.24 times and a high variation of results were achieved in rats after 14 days.

Synthetic pyrethroids are pesticides derived from naturally occurring pyrethrins, taken from pyrethrum of dried Chrysanthemum flowers. Pyrethroid insecticides are one of the most commonly used residential and agricultural insecticides. Many pyrethroids can significantly harm the nervous system. Permethrin could be one of the factors involved in the onset of neurodegenerative diseases. The present study aims to evaluate in brain, the effect that can induce the exposure to permethrin, during early life of female rats (from 6 to 21 days of life). Therefore, have been examined the concentrations of permethrin and its main metabolite (3-PBA) in the brain and urine in female rats sacrificed the day after and 14 days after treating. The different concentrations of permethrin and 3-PBA (after 24h and after 14 days in the end of treatment) were obtained using two different methods. The evaluation of permethrin by liquid-liquid extraction and GC-ECD was performed. The levels of the 3-PBA (in urine and brains) were obtained by SPE procedure and GC-MS using the 2- PBA as internal standard.

O creme de pasteleiro é uma das preparações clássicas mais utilizadas em pastelaria e a principal base para recheios de diversos produtos. Apesar de ser profusamente utilizado, este creme tem um conjunto de limitações que a serem ultrapassadas permitiriam obter produtos com melhores propriedades organolépticas e um maior tempo de vida útil em prateleira. Tem ainda o inconveniente de não poder ser submetido a processos de congelamento e descongelamento devido à ocorrência de sinérese. O objectivo do presente trabalho foi desenvolver um creme com características funcionais idênticas às do creme pasteleiro clássico, mas sem as limitações referidas, nomeadamente um creme com uma melhor textura e libertação de aromas, que não forme um filme à superfície ao arrefecer e que possa ser submetido a processos de congelamento/descongelamento. Através de análises de textura e reologia, avaliou-se o efeito da substituição do amido por goma xantana e goma xantana de rápida dispersão (RD). Tendo-se verificado que a goma xantana RD permitia obter resultados mais compatíveis com a funcionalidade pretendida. Para avaliação do efeito do texturizante usado na libertação de aromas, prepararam-se cremes de pasteleiro aromatizados com maracujá. Usando como técnica a microextração em fase sólida do headspace (HS-SPME) e a cromatografia gasosa-espectrometria de massa (GC-MS) levou-se a cabo uma comparação do perfil de libertação de compostos voláteis dos cremes com amido de milho e goma xantana RD. Finalmente, para avaliar a aceitação por parte do consumidor, foram realizados testes de análise sensorial. Os resultados obtidos permitem concluir que o produto desenvolvido tem características reológicas adequadas para poder ser usado em substituição do creme de pasteleiro clássico, apresentando ainda vantagens, nomeadamente: uma boa performance quando submetido a processos de congelamento/descongelamento, um maior tempo de prateleira, melhores características organolépticas, nomeadamente uma textura mais agradável e uma melhor libertação de aromas. Estes resultados levam-nos a sugerir que este novo produto pode ter um impacto significativo na confeção de pastelaria do ponto de vista da qualidade e gestão de trabalho.

Se realizará el estudio y caracterización química de la fracción orgánica volátil (COVs) presente en plantas Medicinales y Aromáticas. Dicho estudio se llevará a cabo mediante la utilización de la Microextracción en Fase Sólida (SPME) y posterior análisis cuali-cuantitativo por Cromatografía Gaseosa adosada a un Espectrómetro de Masas (GC-MS). Mediante el estudio y caracterización de los compuestos orgánicos volátiles presentes, se tratará de establecer similitudes y diferencias en especímenes de flora autóctona perteneciente al mismo género pero de diferentes especies. Además de esto, se tratarán de establecer las mejores condiciones de conservación y almacenamiento de aquellas plantas aromáticas que posean importancia económica reconocida (estabilidad de los COVs responsables de las características aromáticas, durante el proceso de conservación y almacenamiento). Por otra parte, se realizará la formación de recursos humanos en el área de la química orgánica analítica.

El factor de crecimiento similar a insulina-I (IGF-I) es una hormona peptídica que participa en el crecimiento animal, desarrollo de los tejidos y diferenciación. Hay un creciente cuerpo de evidencias que indican que esta hormona es también importante en la regulación del desarrollo del sistema nervioso. (...) Este factor de crecimiento ejerce sus acciones interaccionando con un receptor heterotetramérico similar al receptor de insulina. b-gc es una subunidad b novedosa del receptor de IGF-I, típica del sistema nervioso central y enriquecida en conos de crecimiento neural. Los objetivos del presente proyecto consisten en: a) Caracterización molecular de b-gc mediante clonado y secuenciado del cDNA que codifica su biosíntesis; b) Caracterización bioquímica de las vesículas que transportan bgc en neuronas en desarrollo desde el aparato de Golgi hacia el cono de crecimiento neural; c) Estudio de la expresión y distribución de bgc en células nerviosas no neuronales, como oligodendrocitos y astrocitos. (...) La caracterización molecular de los receptores de IGF-I conteniendo b-gc y que presenta una distribución muy interesante durante el desarrollo del sistema nervioso central, puede aportar datos fundamentales para la comprensión de la función de estos receptores y su correspondiente hormona durante el desarrollo del sistema nervioso y su transporte específico a los conos de crecimiento neural. La caracterización bioquímica de las vesículas conteniendo b-gc puede aportar importantes datos sobre la presencia de diferentes tipos de vesículas de transporte no sinápticas, como surge de resultados obtenidos recientemente.

Además de esto, se trata de establecer las mejores condiciones de conservación y almacenamiento de aquellas plantas aromáticas que poseen importancia económica, evaluando así la estabilidad de los COVs responsables de las características aromáticas, durante el proceso de conservación y almacenamiento

Durant les darreres dècades, i degut, principalment, a un canvi en els hàbits alimentaris, hi ha hagut un augment a nivell mundial de malalties cròniques (l’obesitat, malalties cardiovasculars, etc.). En els països mediterranis hi ha menys incidència d’aquestes malalties i sembla ser que això es deu a l’anomenada dieta mediterrània. La dieta mediterrània es caracteritza per una combinació d’oli d’oliva com a grassa principal, verdures, hortalisses i fruites en abundància, lleguminoses, fruits secs, formatges i iogurt, peix, pa, pasta, cereals i els seus derivats i un consum moderat de vi i carns. Aquest model alimentari, ric en tocoferols, fitosterols i fitoestanols que ajuden a reduir el contingut de colesterol en sang, fa que en les poblacions mediterrànies hi hagi menys incidència de malalties cardiovasculars. Aquests compostos inhibeixen el deteriorament oxidatiu dels olis, actuen com agent antipolimerització per olis de fregir. Tenen capacitat de reduir els nivells de colesterol, evitant la incidència de malalties cardiovasculars. Els fitoesterols y fitoestanols es poden trobar en forma lliure o esterificada amb àcids grassos, àcids fenòlics i glucosa. Els objectius d’ aquest treball han estat, primer en el desenvolupament de mètodes d'anàlisi ràpids, fiables i robusts dels tocoferols, fitoesterols i fitoestanols i la seva aplicació en fruits sec, oli de segó, oli de pinyol de raïm i productes que els continguin. El primer mètode va estar basat en la cromatografía líquida (HPLC-DAD) amb extracció en fase sòlida (SPE) com tècnica alternativa a la saponificació para la determinació de fitoesterols lliures. Aquest mètode va estar aplicada a mostres de bombons que contenia fitoesterols. El segon mètode va estar basat en la cromatografia de gasos (GCFID) amb aponificació i SPE per quantificar fitoesterols i fitoestanols lliures, esterificats i totals. En els documents annexos es descriuen a profunditat els mètodes desenvolupats.

Gas chromatography (GC) is an analytical tool very useful to investigate the composition of gaseous mixtures. The different gases are separated by specific columns but, if hydrogen (H2 ) is present in the sample, its detection can be performed by a thermal conductivity detector or a helium ionization detector. Indeed, coupled to GC, no other detector can perform this detection except the expensive atomic emission detector. Based on the detection and analysis of H2 isotopes by low-pressure chemical ionization mass spectrometry (MS), a new method for H2 detection by GC coupled to MS with an electron ionization ion source and a quadrupole analyser is presented. The presence of H2 in a gaseous mixture could easily be put in evidence by the monitoring of the molecular ion of the protonated carrier gas. Copyright © 2013 John Wiley & Sons, Ltd.

Drug abuse is a widespread problem affecting both teenagers and adults. Nitrous oxide is becoming increasingly popular as an inhalation drug, causing harmful neurological and hematological effects. Some gas chromatography-mass spectrometry (GC-MS) methods for nitrous oxide measurement have been previously described. The main drawbacks of these methods include a lack of sensitivity for forensic applications; including an inability to quantitatively determine the concentration of gas present. The following study provides a validated method using HS-GC-MS which incorporates hydrogen sulfide as a suitable internal standard allowing the quantification of nitrous oxide. Upon analysis, sample and internal standard have similar retention times and are eluted quickly from the molecular sieve 5Å PLOT capillary column and the Porabond Q column therefore providing rapid data collection whilst preserving well defined peaks. After validation, the method has been applied to a real case of N2O intoxication indicating concentrations in a mono-intoxication.

A study od Typanosomatidae GC distribution and codon usage is presented. The codon usage patterns in coincidence with the phylogenetical data are similar in Crithidia and Leishmania, whereas they are more divergent in Trypanosoma brucei and T. cruzi. The analyisis of the GC mutational pressure in these organisms reveals that T. brucei, and to a lesser extent T. cruzi, have envolved towards a more balanced use of all bases, whereas Leishmania and Crithidia retain features of a primeval genetic apparatus. Tables with approximated GC mutational pressure in homologous genes, and codon usage in Trypanosomatidae are presented.
