In this study, 21 small beam specimens with a pre-set through-crack were tested with FRP strips bonded to the sides to study the bond behavior between shear-strengthening FRP and concrete (referred to as "the interface" for simplicity hereafter). The test parameters included the bond length, width, thickness of FRP strip and the angle between fiber orientation and crack opening direction (referred to as "fiber tensile angle" θ hereafter) on the bond behavior of the interface. Test results showed that: 1) the small beam test setup with a pre-set crack is suitable for studying the bond behavior between FRP and concrete; 2) the bond length, width, and thickness of the FRP as well as the fiber angle have significant effects on the bond strength; 3) the distribution of FRP strains along the direction perpendicular to the fiber orientation (FRP width direction) is non-uniform; this is mainly attributed to the progressive debonding of the FRP strips in the width direction, with the FRP at the location of larger crack width debonding earlier.

It seems unlikely that Moscow can hope for an outright victory in Syria’s civil war, so some kind of political compromise with the moderate opposition is in the offing. This, however, is at best a long shot given the hostility to Assad in the West and the intensity of the conflict in Syria.
Instead, the immediate priority seems to be to ensure a survival of the Syrian state and military institutions in the areas it can control, what one Russian observer called an “Alawite Israel” – a strip of land from the Mediterranean coast to Damascus, able to at least contain IS with some external support.
The Kremlin has consistently prioritised stability over revolutionary change and sovereign rights over humanitarian intervention. In fact, from the Russian point of view, the Western interventionist agenda of democratisation, which ignored local conditions, has made the situation in the Middle East worse – from Iraq to Libya and Syria.

Introduction: Protease activity is essential for the progression of periodontal disease and several studies have shown that gingival crevicular fluid (GCF) proteases are associated with the attachment loss and bone destruction associated with periodontial disease. In addition to measuring protease levels using ELISA, it is also important to consider enzyme activity which can be measured using appropriate substrates. Aim: The aim of this work was to measure the proteolyitc activity in gingival crevicular fluid (GCF) from periodontitis patients using zymography and a fluorogenic protease substrate. Materials and Methods: Twenty four GCF samples were collected from patients with established periodontitis who had not received any periodontal treatment in the previous six months. A strip of perio-paper was inserted into the gingival crevice until light resistance was felt. After 30 seconds the perio-paper was removed and placed into 500 ul ice cold 0.01M sodium phosphate buffer, pH 7.2, containing 0.15M sodium chloride, vortex mixed for 30 seconds and stored at -80°C until required. GCF samples (10 ul) were electrophoresed on 4-16% Blue casein zymogram gels at 125V constant voltage for 90 min. Following electrophoresis the gel was washed in renaturation buffer for 30 min and then placed in developing buffer overnight. Areas of protease activity appeared as clear bands against a blue background. The total caseinolytic activity of each GCF sample was measured using a fluorescent assay with resorufin-labelled casein as the substrate. Results: The results showed that both casein zymography and fluorogenic assay methods were suitable for analysing caseinolytic activity in GCF samples from periodontitis patients. Caseinolytic activity was variable in the periodontitis samples studied and may reflect the episodic nature of the disease. Conclusion: Casein zymography and fluorogenic assay methods may be useful in future attempts to measure active episodes of periodontal disease.

Objectives: To determine whether neuropeptide Y (NPY) is present in gingival crevicular fluid (GCF) in both periodontal health and disease and to study the relationship of NPY with periodontal inflammation. Methods: GCF samples (30 s) were collected from one site with both pocket depth (>4mm) and loss of periodontal attachment (>4mm) in 20 patients with chronic periodontitis (mean age 41.4, SD 9.6 yrs; 10 m, 10 f). GCF was also collected from clinically healthy sites (< 3mm, no bleeding on probing) in 20 subjects with no periodontitis (mean age 37.4, SD 11.7; 10 m, 10 f). GCF was collected using the periopaper strip method, diluted in 500 ul of phosphate-buffered saline and stored at –70°C. Samples were analysed in duplicate for NPY by radioimmunoassay. NPY levels were compared using the Mann-Whitney test. Results: Measurable NPY was present in all the GCF samples collected from healthy subjects. NPY was below the level of detection in 4 (20%) of the diseased subjects. There was considerable variability in the amount of NPY collected from both groups. There were no differences between the levels of NPY measured in males compared with females in either the healthy or diseased groups. Significantly more (P< 0.0001) NPY (pg) was collected from healthy subjects (Median 165, IQR 80; mean 161, SD 64) than diseased subjects (Median 37.5, IQR 56.3; mean 39.8, SD 35.1). There was more variability in the NPY concentration (pg/ul) which was also significantly higher in healthy (Median 575.7, IQR 562.3; mean 645.7, SD 416.7) compared with diseased subjects (Median 43.6, IQR 117.4; mean 96.4, SD 124.5). Pine wilt disease (PWD) is perhaps the most serious threat to pine forests worldwide. Since it´s discovery in the early XXth century by Japanese forest researchers, and the relationship with its causative agent, the pinewood nematode (PWN) Bursaphelenchus xylophilus, in the 1970s, PWD has wreaked havoc wherever it appears. Firstly in the Far East (Japan, China and Korea) and now, more recently in 1999, in the EU (Portugal). The forest sector in Portugal plays a major role in the Portuguese economy with a 12% contribution to the industrial gross domestic product, 3.2% of the gross domestic product, 10% of foreign trade and 5% of national employment. Maritime pine (Pinus pinaster) is one of the most important pine productions, and industrial activity, such as the production of wood and resin, as well as coastal protection associated with sand dunes. Also, stone pine (Pinus pinea) plays an important role in the economy with a share derived from the exports of high-quality pineon seed. Thus, the tremendous economical and ecological impact of the introduction of a pest and pathogen such as the PWN, although as far as is known, the only species susceptible to the nematode is maritime pine. Immediately following detection, the research team involved (Univ. Évora, INIAP) informed the national plant quarantine and forest authorities, which relayed the information to Brussels and the appropriate EU authorities. A task force (GANP), followed by a national program (PROLUNP) was established. Since then, national surveys have been taking place, involving MADRP (Ministry of Agriculture), the University of Évora and several private corporations (e.g. UNAC). Forest growers in the area are particularly interested and involved since the area owned by the growers organizations totals 700 000 ha, largely affected by PWD. Detection of the disease has led to serious consequences and restrictions regarding exploration and commercialization of wood. A precautionary phytosanitary strip, 3 km-wide, has been recently (2007) established surrounding the affected area. The Portuguese government, through its national program PROLUNP, has been deeply involved since 1999, and in conjunction with the EU (Permanent Phytosanitary Committee, and FVO) and committed to controlling this nematode and the potential spread to the rest of the country and to the rest of the EU. The global impact of the presence of Bursaphelenchus xylophilus or the threat of its introduction and the resulting pine wilt disease in forested areas in different parts of the world is of increasing concern economically. The concern is exacerbated by the prevailing debate on climate change and the putative impact this could have on the vulnerability of the world’s pine forests to this disease. The scientific and regulatory approach taken in different jurisdictions to the threat of pine wilt disease varies from country to country depending on the perceived vulnerability of their pine forests to the disease and/or to the economic cost due to lost trade in wood products. Much of the research surrounding pine wilt disease has been located in the northern hemisphere, especially in southern Europe and in the warmer, coastal, Asian countries. However, there is an increased focus on this problem also in those countries in the southern hemisphere where plantations of susceptible pine have been established over the years. The forestry sector in Australia and New Zealand are on “high alert” for this disease and are practicing strict quarantine procedures at all ports of entry for wood products. As well, there is heightened awareness, as there is worldwide, for the need to monitor wood packaging materials for all imported goods. In carrying out the necessary monitoring and assessment of products for B. xylophilus and its vectors substantial costs are incurred especially when decisions have to be made rapidly and regardless of whether the outcome is positive or negative. Australia’s response recently to the appearance of some dying pines in a plantation illustrated the high sensitivity of some countries to this disease. Some $200,000 was spent on the assessment in order to save a potential loss of millions of dollars to the disease. This rapid, co-ordinated response to the report was for naught, because once identified it was found not to be B. xylophilus. This illustrates the particular importance of taking the responsibility at all levels of management to secure the site and the need of a rapid, reliable diagnostic method for small nematode samples for use in the field. Australia is particularly concerned about the vulnerability of its 1million hectares of planted forests, 80% of which are Pinus species, to attack from incursions of one or more species of the insect vector. Monochamus alternatus incursions in wood pallets have been reported from Brisbane, Queensland. The climate of this part of Australia is such that the Pinus plantations are particularly vulnerable to the potential outcome of such incursions, and the state of Queensland is developing a risk management strategy and a proactive breeding programme in response to this putative threat. New Zealand has 1.6 million hectares of planted forests and 89% of the commercial forest is Pinus radiata. Although the climate where these forests are located tends to be somewhat cooler than that in Australia the potential for establishment and development of the disease in that country is believed to be high. The passage alone of 200,000 m³/year of wood packaging through New Zealand ports is itself sufficient to require response. The potential incursion of insect vectors of pinewood nematode through the port system is regarded as high and is monitored carefully. The enormous expansion of global trade and the continued use of unprocessed/inadequately-processed wood for packaging purposes is a challenge for all trading nations as such wood packaging material often harbours disease or pest species. The extent of this problem is readily illustrated by the expanding economies and exports of countries in south-east Asia. China. Japan and Korea have significant areas of forestland infested with B. xylophilus. These countries too are among the largest exporting countries of manufactured goods. Despite the attempts of authorities to ensure that only properly treated wood is used in the crating and packaging of goods B. xylophilus and/or its insect vector infested materials is being recorded at ports worldwide. This reminds us, therefore, of the ease with which this nematode pest can gain access to forest lands in new geographic locations through inappropriate use, treatment or monitoring of wood products. It especially highlights the necessity to find an alternative to using low-grade lumber for packaging purposes. Lest we should believe that all wood products are always carriers of B. xylophilus and its vectors, it should be remembered that international trade of all kinds has occurred for thousands of years and that lumber-born pests and diseases do not have worldwide distribution. Other physico-biological factors have a significant role in the occurrence, establishment and sustainability of a disease. The question is often raised as to why the whole of southern Europe doesn’t already have B. xylophilus and pine wilt disease. European countries have traded with countries that are infested with B. xylophilus for hundreds of years. Turkey is an example of a country that appears to be highly vulnerable to pine wilt disease due to its extensive forests in the warm, southern region where the vector, Monochamus galloprovincialis, occurs. However, there is no record of the presence of B. xylophilus occurring there despite the importation of substantial quantities of wood from several countries In many respects, Portugal illustrates both the challenge and the dilemma. In recent times B. xylophilus was discovered there in the warm coastal region. The research, administrative and quarantine authorities responded rapidly and B. xylophilus appears to have been confined to the region in which it was found. The rapid response would seem to have “saved the day” for Portugal. Nevertheless, it raises again the long-standing questions, how long had B. xylophilus been in Portugal before it was found? If Lisbon was the port of entry, which seems very likely, why had B. xylophilus not entered Lisbon many years earlier and established populations and the pine wilt disease? Will the infestation in Portugal be sustainable and will it spread or will it die out within a few years? We still do not have sufficient understanding of the biology of this pest to know the answers to these questions.

A Micro-Hole & Strip Plate (MHSP) é uma microestrutura desenvolvida recentemente em instituições universitárias portuguesas. Neste trabalho, o seu desempenho como detector para imagiologia é explorado, tendo como objectivo a imagiologia não só de neutrões, mas também de raios X. A aplicação do método da divisão resistiva de carga é aplicada a uma MHSP especialmente desenhada para sensibilidade em posição. Várias abordagens em termos da electrónica de aquisição de sinal são testadas, sem perder de vista a relação desempenho-preço. Resoluções espaciais abaixo de 1 mm foram obtidas com a MHSP a operar em xénon e tetrafluorometano, com um sistema de detecção a um preço modesto quando comparado com as alternativas, e apropriado para inúmeras aplicações em imagiologia de neutrões e de raios X.

Uma descrição detalhada do processo de electroluminescência é um prérequisito na optimização de detectores gasosos para sistemas de imagiologia, astrofísica, física de altas energias e experiências de eventos raros. Neste trabalho, é apresentada e caracterizada uma nova e versátil plataforma de simulação da emissão de luz durante a deriva de electrões em gases nobres, desenvolvida usando os programas Magboltz e Garfield. Propriedades intrínsecas da electroluminescência em gases nobres são calculadas e apresentadas em função do campo eléctrico aplicado, nomeadamente eficiências, rendimento e flutuações estatísticas associadas. São obtidos resultados em grande concordância com dados experimentais e simulações Monte Carlo anteriores. A plataforma é usada para determinar as condições óptimas de funcionamento de detectores como o NEXT (Neutrino Experiment with a Xenon TPC) e outros baseados nas micro-estruturas GEM (Gas Electron Multiplier) e MHSP (Micro- Hole & Strip Plate).

Este trabalho descreve o desenvolvimento e aplicação de sistemas baseados em detetores gasosos microestruturados, para imagiologia de fluorescência de raios-X por dispersão em energia (EDXRF). A técnica de imagiologia por fluorescência de raios-X assume-se como uma técnica poderosa, não-destrutiva, em análises da distribuição espacial de elementos em materiais. Os sistemas para imagiologia de EDXRF desenvolvidos são constituídos por: um tubo de raios-X, usado para excitar os elementos da amostra; um detetor gasoso microestruturado; e uma lente pinhole que foca a radiação de fluorescência no plano do detetor formando assim a imagem e permitindo a sua ampliação. Por outro lado é estudada a influência do diâmetro da abertura do pinhole bem como do fator de ampliação obtido para a imagem, na resolução em posição do sistema. Foram usados dois conceitos diferentes de detetores gasosos microestruturados. O primeiro, baseado na microestrutura designada por 2D-Micro-Hole & Strip Plate (2D-MHSP) com uma área ativa de 3 3 cm2, enquanto que o segundo, baseado na estrutura 2D-Thick-COBRA (2D-THCOBRA) apresenta uma área ativa de deteção de 10 10 cm2. Estes detetores de raios-X de baixo custo têm a particularidade de funcionar em regime de fotão único permitindo a determinação da energia e posição de interação de cada fotão que chega ao detetor. Deste modo permitem detetar a energia dos fotões X de fluorescência, bem como obter imagens 2D da distribuição desses fotões X para o intervalo de energias desejado. São por isso adequados a aplicações de imagiologia de EDXRF. Os detetores desenvolvidos mostraram resoluções em energia de 17% e 22% para fotões incidentes com uma energia de 5.9 keV, respectivamente para o detetor 2D-MHSP e 2D-THCOBRA e resoluções em posição adequadas para um vasto número de aplicações. Ao longo deste trabalho é detalhado o desenvolvimento, o estudo das características e do desempenho de cada um dos detetores, e sua influência na performance final de cada sistema proposto. Numa fase mais avançada apresentam-se os resultados correspondentes à aplicação dos dois sistemas a diversas amostras, incluindo algumas do nosso património cultural e também uma amostra biológica.

The development of computed tomography systems with energy resolving detectors is a current challenge in medical physics and biomedical engineering. A computed tomography system of this kind allows getting complementary informations relatively to conventional systems, that can help the medical diagnosis, being of great interest in medicine. The work described in this thesis is related to the development of a computed tomography system using micropattern gaseous detectors, which allow storing, simultaneously, information about the interaction position and the energy of each single photon that interacts with the detector. This kind of detectors has other advantages concerning the cost and characteristics of operation when compared with solid state detectors. Tomographic acquisitions were performed using a MicroHole & Strip Plate based detector, which allowed reconstructing cross-sectional images using energy windows, applying the energy weighting technique and performing multi-slice and tri-dimensional reconstructions. The contrast-to-noise ratio was improved by 31% by applying the energy weighting technique, comparing with the corresponding image obtained with the current medical systems. A prototype of a computed tomography with flexibility to change the detector was developed, making it possible to apply different detectors based on Thick-COBRA. Several images acquired with these detectors are presented and demonstrate their applicability in X-ray imaging. When operating in NeCH4, the detector allowed a charge gain of 8 104, an energy resolution of 20% (full width at half maximum at 8 keV), a count rate of 1 106 Hz/mm2, a very stable operation (gain fluctuations below 5%) and a spacial resolution of 1.2 mm for an energy photon of 3.6 keV. Operating the detector in pure Kr allowed increasing the detection efficiency and achieving a charge gain of 2 104, an energy resolution of 32% (full width at half maximum at 22 keV), a count rate of 1 105 Hz/mm2, very stable operation and a spatial resolution of 500 m. The software already existing in the group was improved and tools to correct geometric misalignments of the system were also developed. The reconstructions obtained after geometrical correction are free of artefacts due to the referred misalignments.

Recent studies have shown that, besides the well-recognized T3 and T4 hormones, there are other relevant thyroid hormones circulating in the human body. In particular, this is the case for 3-iodothyronamine (T1AM) and thyronamine (T0AM). One of the reasons for the lack of studies showing their precise importance is the absence of analytical methodologies available. Herein, for the first time, T1AM and T0AM are electrochemically characterized. T0AM was sensed by means of a glassy carbon electrode; furthermore, T1AM was sensed both with a graphitic surface (oxidatively) as well as with mercury (reductively). For both compounds, after oxidation, it was possible to observe the reversible redox reaction concerning the benzoquinone/hydroquinone couple, thus increasing the specificity of the electroanalysis. Therefore, this work provides the basis for an ‘at-point-of-use’ electrochemical strip test for T1AM and T0AM.

Guilhotinas são máquinas robustas, de corte rectilíneo, normalmente associadas a equipamentos de baixo custo, devido à pequena quantidade de dispositivos tecnológicos incorporados. No entanto, esta situação pode ser alterada através da criatividade dos projetistas deste tipo de equipamento. Analisando algumas operações específicas, pode-se observar que algumas ferramentas, quando associadas ao equipamento, podem aumentar substancialmente a produtividade do processo de corte e a qualidade do produto final. Em relação ao processo de corte de chapas finas de metal, pode-se observar que na fase final de corte, o peso do material a cortar é suspenso por uma pequena porção de material que ainda não foi sujeita ao corte. Este facto leva a uma deformação plástica nesta última zona, causando problemas de qualidade no produto final, que não ficará completamente plano. Este trabalho foi desenvolvido em torno deste problema, estudando a melhor solução para desenvolver uma nova ferramenta, capaz de evitar a falta de nivelamento da placa, após corte. Um novo equipamento foi concebido, capaz de ser facilmente incorporado na guilhotina, permitindo o acompanhamento da inclinação da lâmina durante a operação de corte. O sistema é totalmente automatizado, sendo operado por uma única instrução de corte dada pelo operador da máquina. Este sistema permite à empresa fabricante aumentar o valor agregado de cada máquina, oferecendo aos clientes soluções avançadas, contribuindo desta forma para a sustentabilidade do negócio da empresa.

O presente trabalho teve como objectivo a minimização do impacto ambiental do processo de curtume da pele de bovino. A indústria de curtumes transforma a pele, material putrescível, em couro, material nobre, termicamente estável e imputrescível. A transformação da pele em couro origina uma carga poluente apreciável quer quanto a efluentes líquidos quer quanto a resíduos sólidos. O fluxo produtivo da indústria de curtumes pode dividir-se em quatro sectores: ribeira, curtume, tinturaria e acabamento, sendo que os três primeiros geram efluentes líquidos com elevada carga poluente. Neste trabalho, foram avaliadas as fases do processo que geram efluentes líquidos: molho, caleiro, curtume e tinturaria. Na avaliação do processo do molho testou-se uma protease e uma lipase contra um molhante e um desengordurante tradicional, agentes químicos normalmente utilizados no molho mas menos biodegradáveis que as enzimas testadas. Salienta-se o bom resultado obtido quanto à eficiência do molho Na avaliação do processo de caleiro testaram-se várias alternativas no sentido da redução da quantidade de sulfureto de sódio utilizada e da minimização da carga poluente. Entre as alternativas, depilação por oxidação, depilação enzimática com e sem destruição do pêlo, elegeu-se a depilação enzimática sem destruição do pêlo que conduziu a resultados com menor impacto ambiental, nomeadamente a redução da % da quantidade de sulfureto de sódio, sendo a redução da carga poluente de 4,19 % de sulfureto, 32.80% de sólidos suspensos totais (SST),27.09% de sólidos totais (ST) e de 76.90% da carência química de oxigénio (CQO) no efluente de caleiro. No processo do curtume da pele é utilizado crómio como agente de curtume em cerca de 80% das peles tratadas, sendo este metal problemático em termos ambientais. No sentido de reduzir a quantidade de crómio utilizada no processo foi realizado um planeamento factorial onde as variáveis a estudar foram a concentração de crómio e a temperatura, tendo este como objectivo observar qual a quantidade mínima de crómio necessária para termos um produto final nas condições desejadas e gerando um menor impacto ambiental. Concluiu-se desenvolvendo um processo que mostra ser possível reduzir a quantidade de sal de crómio de 7% para 5%, além de ter um impacto ambiental claramente menos agressivo nos efluentes de curtume gerado. Este processo quando comparado com o processo tradicional permite a redução de 27% na CQO, 79% nos SST, 11% nos ST e 38% no teor de crómio Na avaliação do processo de tinturaria foi estudado um processo compacto contra o processo tradicional, tendo-se concluído pelo menor impacto ambiental do processo estudado, nomeadamente ao nível da redução do consumo de água e da carga poluente gerada. A comparação dos dois processos, no que respeita à carga poluente gerada, permitiu concluir por uma redução de 39% da CQO, 50% dos SST, e 12% dos ST quando aplicado o processo compacto Por fim foi feita uma avaliação do impacto ambiental do efluente global gerado pelos processos considerados com menor impacto ambiental contra o processo tradicional, normalmente aplicado na indústria. A aplicação do conjunto dos processos desenvolvidos, quando comparada com a aplicação do conjunto dos processos tradicionais, mostra uma redução de 1% no sulfureto, 40% na CQO, 60 % nos SST, 42 % no crómio e 11% nos ST, mostrando claramente que é possível reduzir a carga poluente da indústria de curtumes atuando no processo. Este trabalho mostrou a importância de atuar no processo para minimizar os custos de tratamento e mesmo de investimento dos efluentes da indústria de curtumes. Importa salientar que os processos desenvolvidos necessitam de validação a uma escala semi-industrial.

A explosão de Macau nos últimos dez anos deve-se a um conjunto de factores, que este texto procura descrever, propondo uma necessária reflexão. Depois da liberalização do jogo em 2001/2002, o Sands Macau inaugura, em 2004, como o primeiro casino da nova vaga. Em 2005, Macau é Património Mundial da Humanidade pela UNESCO. O Cotai é inicialmente um projecto da administração portuguesa como uma nova cidade para 150 000 habitantes. O Cotai Strip nasce de uma epifania de Sheldon Adelson, chairman da Las Vegas Sands Corporation, em 2003. Em 2007, o Venetian inaugura como “flagship” do Cotai Strip e, em 2011, o Galaxy amplia a dimensão delirante da arquitectura do jogo. No pós-Portugal, a condição de Macau como espaço híbrido, colonizado, transitório, aumenta exponencialmente enquanto “Região Administrativa Especial” da República Popular da China. Macau é hoje uma distopia suave, corporizada nos casinos; uma vida subterrânea que se expressa no skyline.
