The aim of this work is to evaluate the influence of S. pombe and T. delbrueckii species on the sensory quality of red wine when used in sequential and mixed fermentations with S. cerevisiae.

Two experiments were conducted to estimate the standardized ileal digestible (SID) Trp:Lys ratio requirement for growth performance of nursery pigs. Experimental diets were formulated to ensure that lysine was the second limiting AA throughout the experiments. In Exp. 1 (6 to 10 kg BW), 255 nursery pigs (PIC 327 × 1050, initially 6.3 ± 0.15 kg, mean ± SD) arranged in pens of 6 or 7 pigs were blocked by pen weight and assigned to experimental diets (7 pens/diet) consisting of SID Trp:Lys ratios of 14.7%, 16.5%, 18.4%, 20.3%, 22.1%, and 24.0% for 14 d with 1.30% SID Lys. In Exp. 2 (11 to 20 kg BW), 1,088 pigs (PIC 337 × 1050, initially 11.2 kg ± 1.35 BW, mean ± SD) arranged in pens of 24 to 27 pigs were blocked by average pig weight and assigned to experimental diets (6 pens/diet) consisting of SID Trp:Lys ratios of 14.5%, 16.5%, 18.0%, 19.5%, 21.0%, 22.5%, and 24.5% for 21 d with 30% dried distillers grains with solubles and 0.97% SID Lys. Each experiment was analyzed using general linear mixed models with heterogeneous residual variances. Competing heteroskedastic models included broken-line linear (BLL), broken-line quadratic (BLQ), and quadratic polynomial (QP). For each response, the best-fitting model was selected using Bayesian information criterion. In Exp. 1 (6 to 10 kg BW), increasing SID Trp:Lys ratio linearly increased (P < 0.05) ADG and G:F. For ADG, the best-fitting model was a QP in which the maximum ADG was estimated at 23.9% (95% confidence interval [CI]: [<14.7%, >24.0%]) SID Trp:Lys ratio. For G:F, the best-fitting model was a BLL in which the maximum G:F was estimated at 20.4% (95% CI: [14.3%, 26.5%]) SID Trp:Lys. In Exp. 2 (11 to 20 kg BW), increasing SID Trp:Lys ratio increased (P < 0.05) ADG and G:F in a quadratic manner. For ADG, the best-fitting model was a QP in which the maximum ADG was estimated at 21.2% (95% CI: [20.5%, 21.9%]) SID Trp:Lys. For G:F, BLL and BLQ models had comparable fit and estimated SID Trp:Lys requirements at 16.6% (95% CI: [16.0%, 17.3%]) and 17.1% (95% CI: [16.6%, 17.7%]), respectively. In conclusion, the estimated SID Trp:Lys requirement in Exp. 1 ranged from 20.4% for maximum G:F to 23.9% for maximum ADG, whereas in Exp. 2 it ranged from 16.6% for maximum G:F to 21.2% for maximum ADG. These results suggest that standard NRC (2012) recommendations may underestimate the SID Trp:Lys requirement for nursery pigs from 11 to 20 kg BW.

The Agrobacterium tumefaciens VirB7 lipoprotein contributes to the stabilization of VirB proteins during biogenesis of the putative T-complex transport apparatus. Here, we report that stabilization of VirB7 itself is correlated with its ability to form disulfide cross-linked homodimers via a reactive Cys-24 residue. Three types of beta-mercaptoethanol-dissociable complexes were visualized with VirB7 and/or a VirB7::PhoA41 fusion protein: (i) a 9-kDa complex corresponding in size to a VirB7 homodimer, (ii) a 54-kDa complex corresponding in size to a VirB7/VirB7::PhoA41 mixed dimer, and (iii) a 102-kDa complex corresponding to a VirB7::PhoA41 homodimer. A VirB7C24S mutant protein was immunologically undetectable, whereas the corresponding VirB7C24S::PhoA41 derivative accumulated to detectable levels but failed to form dissociable homodimers or mixed dimers with wild-type VirB7. We further report that VirB7-dependent stabilization of VirB9 is correlated with the ability of these two proteins to dimerize via formation of a disulfide bridge between reactive Cys-24 and Cys-262 residues, respectively. Two types of dissociable complexes were visualized: (i) a 36-kDa complex corresponding in size to a VirB7/VirB9 heterodimer and (ii) an 84-kDa complex corresponding in size to a VirB7/VirB9::PhoA293 heterodimer. A VirB9C262S mutant protein was immunologically undetectable, whereas the corresponding VirB9C262S::PhoA293 derivative accumulated to detectable levels but failed to form dissociable heterodimers with wild-type VirB7. Taken together, these results support a model in which the formation of disulfide cross-linked VirB7 dimers represent critical early steps in the biogenesis of the T-complex transport apparatus.

Rap phosphatases are a recently discovered family of protein aspartate phosphatases that dephosphorylate the Spo0F--P intermediate of the phosphorelay, thus preventing sporulation of Bacillus subtilis. They are regulators induced by physiological processes that are antithetical to sporulation. The RapA phosphatase is induced by the ComP-ComA two-component signal transduction system responsible for initiating competence. RapA phosphatase activity was found to be controlled by a small protein, PhrA, encoded on the same transcript as RapA. PhrA resembles secreted proteins and the evidence suggests that it is cleaved by signal peptidase I and a 19-residue C-terminal domain is secreted from the cell. The sporulation deficiency caused by the uncontrolled RapA activity of a phrA mutant can be complemented by synthetic peptides comprising the last six or more of the C-terminal residues of PhrA. Whether the peptide controls RapA activity directly or by regulating its synthesis remains to be determined. Complementation of the phrA mutant can also be obtained in mixed cultures with a wild-type strain, suggesting the peptide may serve as a means of communication between cells. Importation of the secreted peptide required the oligopeptide transport system. The sporulation deficiency of oligopeptide transport mutants can be suppressed by mutating the rapA and rapB genes or by introduction of a spo0F mutation Y13S that renders the protein insensitive to Rap phosphatases. The data indicate that the sporulation deficiency of oligopeptide transport mutants is due to their inability to import the peptides controlling Rap phosphatases.

Disulfide bond formation is catalyzed in the periplasm of Escherichia coli. This process involves at least two proteins: DsbA and DsbB. Recent evidence suggests that DsbA, a soluble periplasmic protein directly catalyzes disulfide bond formation in proteins, whereas DsbB, an inner membrane protein, is involved in the reoxidation of DsbA. Here we present direct evidence of an interaction between DsbA and DsbB. (Kishigami et al. [Kishigami, S., Kanaya, E., Kikuchi, M. & Ito, K. (1995) J. Biol. Chem. 270, 17072-17074] have described similar findings.) We isolated a dominant negative mutant of dsbA, dsbAd, where Cys-33 of the DsbA active site is changed to tyrosine. Both DsbAd and DsbA are able to form a mixed disulfide with DsbB, which may be an intermediate in the reoxidation of DsbA. This complex is more stable with DsbAd. The dominance can be suppressed by increasing the production of DsbB. By using mutants of DsbB in which one or two cysteines have been changed to alanine, we show that only Cys-104 is important for complex formation. Therefore, we suggest that in vivo, reduced DsbA forms a complex with DsbB in which Cys-30 of DsbA is disulfide-bonded to Cys-104 of DsbB. Cys-104 is rapidly replaced by Cys-33 of DsbA to generate the oxidized form of this protein.

Odortypes--namely, body odors that distinguish one individual from another on the basis of genetic polymorphism at the major histocompatibility complex and other loci--are a fundamental element in the social life and reproductive behavior of the mouse, including familial imprinting, mate choice, and control of early pregnancy. Odortypes are strongly represented in urine. During mouse pregnancy, an outcrossed mother's urine acquires fetal major histocompatibility complex odortypes of paternal origin, an observation that we took as the focus of a search for odortypes in humans, using a fully automated computer-programmed olfactometer in which trained rats are known to distinguish precisely the odortypes of another species. Five women provided urine samples before and after birth, which in each case appropriately trained rats were found to distinguish in the olfactometer. Whether this olfactory distinction of mothers' urine before and after birth reflects in part the odortype and hence genotype of the fetus, and not just the state of pregnancy per se, was tested in a second study in which each mother's postpartum urine was mixed either with urine from her own infant or with urine of a different, same-aged infant. Responses of trained rats were more positive with respect to the former (congruous) mixtures than to the latter (incongruous) mixtures, implying that, as in the mouse, human fetal odortypes of paternal genomic origin are represented in the odortype of the mother, doubtless by circulatory transfer of the pertinent odorants.

Pouco se sabe sobre o efeito do substrato e a interação entre as leveduras selvagens e bactérias do gênero Lactobacillus na fermentação alcoólica, pois os estudos tem se concentrado na avaliação dos efeitos da contaminação por um ou outro contaminante separadamente. Diante disso, este trabalho teve como objetivos estudar o efeito do substrato e das condições de tratamento do fermento sobre as fermentações contaminadas com ambos os micro-organismos, leveduras S. cerevisiae selvagens (três linhagens apresentando colônias rugosas e células dispostas em pseudohifas) e Lactobacillus fermentum, tendo a linhagem industrial de S. cerevisiae PE-2 como levedura do processo. Foram realizadas fermentações em batelada em mosto de caldo e de melaço, sem reciclo e com reciclo celular, utilizando tanto a cultura pura da linhagem PE-2 quanto as culturas mistas com as linhagens rugosas e ou L. fermentum. Foram avaliadas modificações no tratamento ácido do fermento, visando o controle do crescimento dos contaminantes sem afetar a levedura do processo. Em seguida, foram conduzidas fermentações contaminadas e não contaminadas submetidas ao tratamento ácido combinado com adição de etanol, tanto em caldo quanto em melaço, utilizando-se PE-2, uma das linhagens rugosas e L. fermentum. A atividade da invertase extracelular foi também avaliada em ambos os substratos para os micro-organismos estudados, em condições de crescimento. Concluiu-se que o tipo de substrato de fermentação, caldo de cana ou melaço, influenciou o desempenho da linhagem industrial PE-2 assim como afetou o desenvolvimento das contaminações com as leveduras rugosas S. cerevisiae na presença ou ausência da bactéria L. fermentum, em fermentações sem reciclo celular. O efeito da contaminação foi mais evidente quando se utilizou caldo de cana do que melaço como substrato, no caso da contaminação com leveduras rugosas, e o inverso no caso da contaminação com L. fermentum. O efeito da contaminação sobre a eficiência fermentativa foi maior na presença da levedura rugosa do que com a bactéria, e a contaminação dupla (tanto com a levedura rugosa quanto com a bactéria) não teve efeito maior sobre a eficiência fermentativa do que a contaminação simples, por um ou por outro micro-organismo isoladamente, especialmente na fermentação em batelada com reciclo celular, independentemente do substrato. Nas fermentações com reciclo de células, o efeito do substrato foi menos evidente. O controle do crescimento das linhagens rugosas pode ser realizado modificando o tratamento ácido normalmente realizado na indústria, seja pela adição de etanol à solução ácida ou pelo abaixamento do pH, dependendo da linhagem rugosa. O tratamento combinado baixo pH (2,0) + 13% etanol afetou a fisiologia da linhagem industrial, trazendo prejuízos à fermentação com reciclo celular, com pequeno controle sobre o crescimento da levedura rugosa e causando morte celular à L. fermentum. A diferença na atividade invertásica entre as linhagens rugosas e industrial de S. cerevisiae pode ser a responsável pela fermentação lenta apresentada pelas linhagens rugosas quando presentes na fermentação, sendo não significativa a influência do substrato sobre a atividade dessa enzima.

Seventy-five years ago, Walter Benjamin showed us that the line between "production" and "reproduction" had begun to blur. Reproduction was no longer optional, consequential and degrading (the shredding of the original’s aura), but was instead being transformed into a principle of production itself: something was produced bearing in mind how it was to be reproduced. No longer did the original exist (in photography, film, music recordings), but instead diffusion, exhibition. The work existed precisely at the time and place of its enjoyment. Today, the cultural pirates of the new digital era take this principle to the extreme, with a certain characteristic also foreseen by Benjamin: a yearning to participate, to post-produce something captured in order to later return it to the Internet, modified in some way and made available to others. This postproduction is what is now often mixed up with reception, just as production and reproduction used to be in Benjamin’s day. Postproduction on the receiver’s side, which somehow augments and extends the received work, in other words creates an etymologically rigorous author-ization (auctor as the root of both author and augmentation). The cultural pirate only deserves redemption thanks to this creative augmentation.

A series of CeO2–Nb2O5 mixed oxides with different Nb content, as well as the pure oxides, have been synthesized by co-precipitation with excess urea. These materials have been used as supports for platinum catalysts, with [Pt(NH3)4](NO3)2 as precursor. Both supports and catalysts have been characterized by several techniques: N2 physisorption at 77 K, X-ray diffraction, Raman spectroscopy, X-ray photoelectron spectroscopy, UV–vis spectroscopy, scanning electron microscopy, transmission electron microscopy, temperature-programmed reduction and temperature-programmed desorption (CO and H2), and their catalytic behaviour has been determined in the PROX reaction, both with an ideal gas mixture (CO, O2 and H2) and in simulated reformate gas containing CO2 and H2O. Raman spectroscopy analysis has shown the likely substitution of some Ce4+ cations by Nb5+ to some extent in supports with low niobium contents. Moreover, the presence of Nb in the supports hinders their ability to adsorb CO and to oxidize it to CO2. However, an improvement of the catalytic activity for CO oxidation is obtained by adding Nb to the support, although the Pt/Nb2O5 catalyst shows very low activity. The best results are found with the Pt/0.7CeO2–0.3Nb2O5 catalyst, which shows a high CO conversion (85%) and a high yield (around 0.6) after a reduction treatment at 523 K. The effect of the presence of CO2 and H2O in the feed has also been determined.

Este artículo aborda la investigación, realizada con los estudiantes del primer semestre de la titulación de Informática de la Facultad de Filosofía, Letras y Ciencias de la Educación de la Universidad Central del Ecuador, cuyo propósito ha sido analizar el uso de entornos de programación no mediados simbólicamente como herramienta didáctica para el desarrollo del pensamiento computacional. Se pretende establecer las posibles ventajas de aplicar este tipo de entorno para que los estudiantes desarrollen habilidades del pensamiento computacional tales como la creatividad, modelación y abstracción, entre otras, consideradas relevantes dentro de la programación. La metodología en que se apoyó la investigación es mixta, con investigación de campo y documental a nivel descriptivo. Se utilizó como instrumento un cuestionario para la recolección de datos entre el alumnado de la titulación. Finalmente, con la información recopilada se procedió al procesamiento de datos a partir de la estadística descriptiva para, así, obtener resultados que permitiesen alcanzar las pertinentes conclusiones y recomendaciones.

Trata-se de um estudo descritivo de série histórica, com os dados coletados do Sistema de Informação de Mortalidade, através de relatórios do TABWIN, gerados em Excel, com banco de dados e análise estatística feitas no programa estatístico SPSS. Tem por objetivo determinar a mortalidade por suicídio na população do município de Arapiraca-AL, no período de 2007 a 2013, caracterizar o perfil epidemiológico e verificar a existência da associação entre a exposição a agrotóxicos e o suicídio. Identificamos 76 óbitos por suicidio no periodo, porém 14 óbitos foram eliminados por apresentarem informações ignoradas. Dos 53 óbitos, que apresentou uma média de 7,6 óbitos/ano, 40 foram praticados por homens e 13 por mulheres, a faixa etária inferior a 30 anos teve 45% dos óbitos, concentrados entre 20 a 29 anos, com 32,1%; os solteiros representaram 64,1%; pessoas de cor parda, com 41 mortos, (77,3%); a escolaridade apresentou 50,9% com um a 7 anos de estudos; 73,6% das pessoas residiam na zona urbana; 37,7% eram trabalhadores da agricultura, e o meio mais utilizado foi o enforcamento com 28 dos óbitos, (52,9%), seguido do uso de pesticidas com 16 óbitos, (30,2%). O Teste Exato de Fisher utilizado para avaliar a associação entre os fatores que poderiam influenciar a prática do suicídio, mostrou não existir diferença estatística na prática do suicídio entre os trabalhadores com agrotóxicos (p=0,64), bem como em relação a ocupação (p=0,76). Porém, foram encontradas associações significativas entre o desfecho suicídio e a existência de quadro depressivo (p=0,001) e a utilização de medicamentos (p=0,03). Novos estudos devem ser realizados com o intuíto de melhor explicar a ocorrência de depressão e do suicídio e principalmente determinar a sua relação causal com a exposição aos agrotóxicos. Samples from sutface sediments of the shell, continental slope, and adjacent deep sea regions off West Africa between 27° N and 15° N were investigated with respect to grain size distribution of the total samples, sand contents of the acid insoluble residues, carbonate content of the total samples, and the separate grain size fractions, specific surfaces, colours and mineralogical composition of the clay fractions. The grain size distributions of the total samples of the sediments of the shelf and the continental slope off Spanish-Sahara are controlled mainly by biogenic components. The supply of terrigeneous material in this area is very low. At deeper parts of the continental slope and in the deep sea areas, the relative amounts of carbonate minerals in the sediments are considerably reduced. The prevailing sand contents of the upper slope changes into clay dominance. On the shelf of Mauritania - represented by profiles extending down to 200 m water depth - the grain size is also controlled mainly by biogenic carbonates. Nevertheless, the admixture of fossil silicate components is important, too. The southern parts of the area is investigated are located in a region influenced by sediments of the Senegal River, which especially control the contents of silt and clay. The silicate sands, predominately of quartz, are fossil and form a mixed sediment with younger deposits. The carbonate contents of the different grain size fractions are formed either by sedimentation of carbonate and silicate particles of the respective grain size or by autochtonous disintegration of coarser sediment particles, as shown by the occurence of Mg-rich calcite and especially aragonite in the clay sized fraction. In the northern parts of the area investigated, which have very minute terrigeneous supply, the latter mechanism is the dominant factor, controlling the carbonate contents of the fine grain sized fractions. In the vicinity of the mouth of the Senegal the carbonate contents are influenced by extremely high dilution with terrigencous silicates. Mg-rich calcite and aragonite are produced preferentially in shallow slope and shelf areas up to 500 m of water depth. The specific surfaces of the carbonate-free clay fractions indicate that the clay fractions of the shelfareas with little terrigenous supply consits of relatively coarser particles. Very fine particles are removed and transported towards the deep sea. Lateral differentiation of this kind was not observed in the area off Senegal. The high surface areas, characterizing the clay fractions of this region, are thought to be due to high montmorillonite contents as was found for deep seas sediments. The mineralogical composition of the clay fraction from the southern parts of the area is characterized by high kaolinite and montmorillonite contents, while in the northern illite is predominating. At least two types of montmorillionites are present: in areas influenced by the Senegal mostly one type was found, which could swell to 17; on the shelves and slopes of the other regions the montmorillonite-group is represented by a montmorillonite-mica-type mixed-layer mineral. A "glauconite", found in the sand fraction, which had very similar properties to those of the montmorillonite-mica mixed-layer, is believed to be the source of this mixed-layer-type mineral. Palygorskite is present in all samples out of range of the Senegal supply. It may be an indicator of eolian transported material. The occurence of rich palygorskit deposits in the arid hinterlands emphasizes the terrestrial origin.
