Cyanobacteria are the only prokaryotic organisms performing oxygenic photosynthesis. They comprise a diverse and versatile group of organisms in aquatic and terrestrial environments. Increasing genomic and proteomic data launches wide possibilities for their employment in various biotechnical applications. For example, cyanobacteria can use solar energy to produce H2. There are three different enzymes that are directly involved in cyanobacterial H2 metabolism: nitrogenase (nif) which produces hydrogen as a byproduct in nitrogen fixation; bidirectional hydrogenase (hox) which functions both in uptake and in production of H2; and uptake hydrogenase (hup) which recycles the H2 produced by nitrogenase back for the utilization of the cell. Cyanobacterial strains from University of Helsinki Cyanobacteria Collection (UHCC), isolated from the Baltic Sea and Finnish lakes were screened for efficient H2 producers. Screening about 400 strains revealed several promising candidates producing similar amounts of H2 (during light) as the ΔhupL mutant of Anabaena PCC 7120, which is specifically engineered to produce higher amounts of H2 by the interruption of uptake hydrogenase. The optimal environmental conditions for H2 photoproduction were significantly different between various cyanobacterial strains. All suitable strains revealed during screening were N2-fixing, filamentous and heterocystous. The top ten H2 producers were characterized for the presence and activity of the enzymes involved in H2 metabolism. They all possess the genes encoding the conventional nitrogenase (nifHDK1). However, the high H2 photoproduction rates of these strains were shown not to be directly associated with the maximum capacities of highly active nitrogenase or bidirectional hydrogenase. Most of the good producers possessed a highly active uptake hydrogenase, which has been considered as an obstacle for efficient H2 production. Among the newly revealed best H2 producing strains, Calothrix 336/3 was chosen for further, detailed characterization. Comparative analysis of the structure of the nif and hup operons encoding the nitrogenase and uptake hydrogenase enzymes respectively showed minor differences between Calothrix 336/3 and other N2-fixing model cyanobacteria. Calothrix 336/3 is a filamentous, N2-fixing cyanobacterium with ellipsoidal, terminal heterocysts. A common feature of Calothrix 336/3 is that the cells readily adhere to substrates. To make use of this feature, and to additionally improve H2 photoproduction capacity of the Calothrix 336/3 strain, an immobilization technique was applied. The effects of immobilization within thin alginate films were evaluated by examining the photoproduction of H2 of immobilized Calothrix 336/3 in comparison to model strains, the Anabaena PCC 7120 and its ΔhupL mutant. In order to achieve optimal H2 photoproduction, cells were kept under nitrogen starved conditions (Ar atmosphere) to ensure the selective function of nitrogenase in reducing protons to H2. For extended H2 photoproduction, cells require CO2 for maintenance of photosynthetic activity and recovery cycles to fix N2. Application of regular H2 production and recovery cycles, Ar or air atmospheres respectively, resulted in prolongation of H2 photoproduction in both Calothrix 336/3 and the ΔhupL mutant of Anabaena PCC 7120. However, recovery cycles, consisting of air supplemented with CO2, induced a strong C/N unbalance in the ΔhupL mutant leading to a decrease in photosynthetic activity, although total H2 yield was still higher compared to the wild-type strain. My findings provide information about the diversity of cyanobacterial H2 capacities and mechanisms and provide knowledge of the possibilities of further enhancing cyanobacterial H2 production.

2-Hydroxybutyric acid appears at high concentrations in situations related to deficient energy metabolism (e.g., birth asphyxia) and also in inherited metabolic diseases affecting the central nervous system during neonatal development, such as "cerebral" lactic acidosis, glutaric aciduria type II, dihydrolipoyl dehydrogenase (E3) deficiency, and propionic acidemia. The present study was carried out to determine the effect of 2-hydroxybutyric acid at various concentrations (1-10 mM) on CO2 production and lipid synthesis from labeled substrates in cerebral cortex of 30-day-old Wistar rats in vitro. CO2 production was significantly inhibited (30-70%) by 2-hydroxybutyric acid in cerebral cortex prisms, in total homogenates and in the mitochondrial fraction. We also demonstrated a significant inhibition of lipid synthesis (20-45%) in cerebral cortex prisms and total homogenates in the presence of 2-hydroxybutyric acid. However, no inhibition of lipid synthesis occurred in homogenates free of nuclei and mitochondria. The results indicate an impairment of mitochondrial energy metabolism caused by 2-hydroxybutyric acid, a fact that may secondarily lead to reduction of lipid synthesis. It is possible that these findings may be associated with the neuropathophysiology of the situations where 2-hydroxybutyric acid is accumulated.

In the present study, we examined the relationship between cell phenotype and cell survival of three human non-small cell lung carcinoma cell lines (A549, NCI-H596 and NCI-H520). Cells in exponential growth at various densities were incubated for 24 h at 37ºC in a 5% CO2 humidified atmosphere and then exposed to UV radiation for 1 min (256 nm, 40 W, source-to-target distance 100 cm). After two days the surviving cells were quantified by sulforhodamine ß staining and DNA fragmentation assay. The differences in UV sensitivity at 60 x 10³ cells/cm² among the cell lines were not related to the proliferative state of the cells but to the extent of intercellular contact. In contrast to A549 and NCI-H596, irradiated NCI-H520 cells presented lower DNA fragmentation and an aggregated cell culture phenotype even prior to confluence, suggesting that a contact-effect mechanism provides further protection against UV radiation.

Various categories of food packaging indicators namely; VTT, Ageless Eye, Mocon, Åbo Akademi and Impak were selected and incorporated into food trays manufactured at LUT packaging laboratory. Each of these food packaging indicators was used to investigate (visually and qualitatively) the transmission of oxygen through the seal, and tray material, as well as to detect microbial activity within the content of the package. Applications of different methods of gas flushing, content variation and introduction of two distinct levels of oxygen scavengers were employed as treatments to evaluate the packaging performance of the food packaging indicators. Ease of handling of each food packaging indicator was also taken into considerations. Findings showed that for packages, which contained chicken product, the amount of oxygen in the package, measured immediately after the sealing operation on the first day gradually decreased to zero percent by the third day of the storage period. The oxygen level remained at this point throughout the duration of storage for the chicken packages. Besides, level of oxygen in the packages without product continued to increase with the storage time, at moderate rate of 0.1% for 100%N2 and 0.3% for 30%CO2/70%N2 empty packages. More carbon dioxide gas was recorded for packages flushed with 30%CO2/70%N2. Results also revealed that visual analysis of one of the color indicators for example Ageless Eye, conformed to the data derived from the luminescence food-packaging indicator. This shows that packaging operation of the packaging line was considerably stable, and efficient with negligible exception. However, it was found that most of the food packaging indicators investigated in this research study exhibited considerable packaging challenges, such as, reaction with the content of the package (Impak); over sensitivity (Åbo Akademi and Impak); ease of handling problem (Åbo Akademi); and ease of activation problem (VTT indicators). In this study, the strengths and limitations of different indicators were analyzed. This study demonstrates the applicability of various indicators in MAP using chicken package application.

This work focuses on the study of the determination on the possibilities of controlling the required moisture within the inside of film sealed packages. The task is based on the challenges faced by fresh food producers in actualizing a longer product shelf-life coupled with the growing complex desires coming from consumers in the aspect of quality. One way to realize this is by proper evaluation on the use of the flexible plastic films through permeation measurements on the required amount of moisture penetrating through the plastic film with the application of microperforation. A packaging material requires proper interaction on moisture transmission, between the product and the outside environment. The plastic film material that stands between, fresh fruits, vegetables and the outside environment could have appropriate respiration rates through possible micro holes. This work simulates similar process with the aid of water vapor transmission rate (WVTR) experiment using anhydrous CaCl2 as the desiccant, in studying the WVTR values of various perforated film materials at different conditions of storage (standard, fridge, and tropical conditions). However, the results showed absorption rates of water vapor at various conditions in grams of H2O/m2/24h.

Climatic changes threaten the planet. Most articles related to the subject present estimates of the disasters expected to occur, but few have proposed ways to deal with the impending menaces. One such threat is the global warming caused by the continuous increase in CO2 emissions leading to rising ocean levels due to the increasing temperatures of the polar regions. This threat is assumed to eventually cause the death of hundreds of millions of people. We propose to desalinize ocean water as a means to reduce the rise of ocean levels and to use this water for populations that need good quality potable water, precisely in the poorest regions of the planet. Technology is available in many countries to provide desalinated water at a justifiable cost considering the lives threatened both in coastal and desertified areas.

Este estudo constatou a possibilidade de extrairem-se ácidos graxos livres de óleos vegetais com CO2 supercrítico. Utilizou-se neste trabalho óleos de soja e de castanha do Pará. A pesquisa desenvolveu-se em duas etapas, inicialmente investigou-se a possibilidade de se extrairem os ácidos graxos destes óleos em condições de extração que variaram de 50-140 bar e de 40-80oC durante 40-160 minutos. Concluiu-se que é possível realizar a desacidificação de óleos vegetais. A eficiência da extração foi de aproximadamente 30% a 140 bar e 80oC para ambos os óleos. Após esta primeira etapa, o próximo passo foi a tentativa de obter-se um aumento na eficiência do processo promovendo a pré-degomagem nos óleos brutos como uma etapa anterior ao processo de desacidificação com CO2 supercrítico. A degomagem foi realizada através de dois métodos diferentes, um para extrair as gomas hidratáveis e outro para extrair tanto as hidratáveis como as não-hidratáveis. Os resultados experimentais foram obtidos a 140 bar e 80oC durante 40-160 minutos, mostrando que a pré-degomagem é realmente necessária, pois a eficiência da extração aumentou para 57% para o óleo de soja e 42% para o de castanha do Pará totalmente degomados, sendo que o método escolhido deve extrair tanto as gomas hidratáveis como as não-hidratáveis. Finalmente, utilizou-se cossolvente (etanol a 1-5% do peso do óleo) para auxiliar a extração, observando-se um aumento na eficiência para aproximadamente 65% para o óleo de soja e 56% para o de castanha do Pará totalmente degomados a 140 bar e 80oC por 40-160 minutos.

Residual fibers from palm oil production are a good source of carotene, since they contain more than 5% of the original oil, with about 5000 ppm of carotenoids. As carotenoids are thermosensitive molecules, supercritical CO2 can be used for oil recovery, because this technique employs low temperatures. In this work results of oil extraction experiments from pressed palm oil fibers are shown. Fibers were from AGROPALMA, an industry which is located in Tailândia (Pará, Brazil). Extractions were carried out at 200, 250 and 300 bar and at temperatures of 45 and 55oC. Oil was analyzed by UV/vis spectrophotometry for total carotene determination. Results showed a large increase in extraction rate from 200 to 250 bar and a small variation from 250 to 300 bar. The total amount of carotenes did not increase in the course of extraction at 300 bar, but it showed a large increase at 200 and at 250 bar. Free fatty acids are present in amounts larger than those found in commercial oils.

The effective diffusivity of clove essential oil in subcritical liquid CO2 was estimated. The experimental apparatus employed was a fixed-bed extractor. The fixed bed was formed with grounded (mesh -32 + 65) and compacted clove buds which were considered a solid element. The effective diffusion coefficient was evaluated by fitting the experimental concentration profile to the unsteady state mass balance equation for unidirectional diffusion in a finite solid medium. The diffusion coefficient was related to the concentration of oil in the solid by an exponential function. The estimated values of the effective diffusion coefficient varied from 3.64 to 5.22x10-10 m2/s. The average relative errors were lower than 3.1%.

Dentre as fontes de corantes naturais mais utilizadas na indústria de alimentos, encontra-se a cúrcuma (Curcuma longa L), um rizoma do qual podem ser obtidas substâncias como a curcumina, demetoxicurcumina e bis-demetoxicurcumina. Estes pigmentos possuem coloração amarela e capacidade de substituir corantes artificiais. Com a finalidade de verificar a influência do pré-tratamento de secagem na extração, foram realizados experimentos de extração de oleoresina de cúrcuma com CO2 supercrítico, na unidade de extração do Laboratório de Engenharia Química da Universidade Federal do Pará, submetendo-se a matéria-prima a uma secagem nas temperaturas de 70 e 105oC. As extrações foram feitas a pressões de 200, 250 e 300 bar, e na temperatura de 45oC. Os resultados estão apresentados em tabelas e gráficos, em termos de rendimentos totais e teor de curcumina presente na oleoresina. A secagem a 70oC favoreceu a extração de oleoresina em termos de tempo de extração, e contribuiu para a manutenção de curcumina na matéria-prima.

Nos últimos anos, vem crescendo o interesse em alimentos com baixo índice de calorias. Nos métodos tradicionais de extração de lipídeos com solventes orgânicos, devido as condições de extração, a integridade dos compostos extraídos e da matriz pode ser afetada pela decomposição térmica ou por contaminação pelo solvente. A extração com fluido supercrítico (EFSC) pode proporcionar um método alternativo para remover lipídeos, sem que haja redução significativa nas propriedades organolépticas do produto. O objetivo deste trabalho é extrair parcialmente o óleo contido na amêndoa da castanha de caju (Anacardium occidentale) com dióxido de carbono (CO2) supercrítico em condições de pressão de 100-170 bar e temperatura de 40-80 (C, para obter um produto de valor calórico reduzido que poderá ser usado como substituto para o amendoim e outras amêndoas na indústria de confeitos. Os experimentos foram realizados em um extrator de 300 ml, dispondo de sistemas de agitação magnética e variação de temperatura. Os resultados experimentais mostraram que o aumento da pressão acarretou um aumento na eficiência de extração. O efeito da temperatura foi o mesmo porém menos significativo.

Ginger (Zingiber officinale Roscoe) belongs to the Zingiberacea family. It is a spice of great commercial importance. In this work ginger oleoresin was obtained with ethanol, isopropanol and liquid carbon dioxide. The chemical compositions of the extract were compared with each other. All oleoresin samples had monoterpenes and sesquiterpenes. Carboxylic acids were found in organic solvent extracts for an extraction time of 2 hours. The component responsible the for pungent characteristic of the oleoresin, gingerois, were detected in samples obtained with organic solvent for extraction times of 6 hours and in samples obtained with CO2 liquid for extraction times of 2 hours.

O projeto do processo de desterpenação do óleo da casca de laranja com CO2 supercrítico exige o bom conhecimento do comportamento de fases da mistura envolvida. Neste trabalho, faz-se uma análise termodinâmica preliminar desse processo, calculando-se a seletividade a partir da modelagem do equilíbrio líquido-vapor (ELV) para o sistema CO2-limoneno-linalol, com base em dados ternários medidos recentemente. Utilizou-se uma modificação da equação de Peng-Robinson e, com dados binários de ELV, avaliou-se a sua capacidade preditiva considerando-se 2 aspectos: a predição do equilíbrio a uma certa temperatura usando-se os parâmetros estimados em outra temperatura e a predição do comportamento de fases do sistema ternário com os parâmetros estimados dos sistemas binários. Foram determinados também os parâmetros de interação entre limoneno e linalol a partir dos dados experimentais do ternário.

A desterpenação do óleo da casca de laranja com CO2 supercrítico foi investigada através da modelagem e simulação da separação de uma mistura sintética de limoneno (90 % em peso) e linalol (10 %), em um extrator operando em modo semi-contínuo. A modelagem matemática da extração supercrítica foi realizada por analogia com a destilação de uma mistura binária, em batelada, expressando-se a composição das fases em equilíbrio numa base molar livre de CO2. O cálculo das variáveis do processo foi feito por integração numérica da equação de Rayleigh empregando-se o método de Runge-Kutta de quarta ordem. Para a determinação da relação de equilíbrio entre as fases, adotou-se a equação de Peng-Robinson modificada, com os parâmetros de interação obtidos de dados de ELV dos sistemas binários CO2+limoneno e CO2+linalol e do ternário CO2+limoneno+linalol.

Dados de equilíbrio de fases a pressões elevadas dos sistemas binários CO2-Limoneno e CO2-Citral e do sistema ternário CO2-Limoneno-Citral foram coletados da literatura e usados na modelagem termodinâmica que emprega a equação de estado de PENG-ROBINSON [1] com: 1) Regra de mistura clássica; 2) Regra de mistura dependente da composição de STRYJEK & VERA [2]. Os parâmetros de interação binária entre CO2-Limoneno e CO2-Citral foram obtidos pelo ajuste dos modelos a dados experimentais, fazendo uso de dois programas computacionais, os quais envolvem a minimização de uma função objetivo, pelo método Simplex de NELDER & MEAD (3), que foi escrita em termos dos desvios relativos entre os pontos experimentais e os calculados pelos modelos. O equilíbrio de fases do sistema ternário foi calculado utilizando-se os parâmetros de interação binária ajustados previamente e considerando parâmetros nulos entre os componentes Limoneno e Citral. A análise dos resultados indica, para as condições supercríticas, que os dois modelos foram capazes de predizer qualitativamente o sistema ternário, fornecendo resultados compatíveis, em ordem de grandeza, com os valores experimentais.
