An analytical procedure using supercritical fluid extraction (SFE) and capillary gas chromatography with electron-capture detection was developed to determine simultaneously residues of different pesticides (organochlorine, organophosphorus, organonitrogen and pyrethroid) in honey samples. Fortification experiments were conducted to test conventional extraction (liquid-liquid) and optimize the extraction procedure in SFE by varying the CO2-modifier, temperature, extraction time and pressure. Best efficiency was achieved at 400 bar using acetonitrile as modifier at 90 degreesC. For the clean-up step, Florisil cartridges were used for both methods LLE and SFE. Recoveries for majority of pesticides from fortified samples of honey at fortification level of 0.01-0.10 mg/kg ranged 75-94% from both methods. Limits of detection found were less than 0.01 mg/kg for ECD and confirmation of pesticide identity was performed by gas chromatography-mass spectrometry in selected-ion monitoring mode. The multiresidue methods in real honey samples were applied and the results of developed methods were compared. (C) 2004 Elsevier B.V. All rights reserved.

The objective this work was to evaluate the effect of microbial and chemical insecticides on the fall armyworm Spodoptera frugiperda in the development initial phase of crop corn. The work was lead in the experimental area of the State University of Mato Grosso do sul, in Cassilandia (MS) city. The treatments had been: 1) control; 2) fipronil; 3) thiametoxan; 4) Beauveria bassiana (0,5 kg of p.c/ha), 5) Beauveria bassiana (1 kg of p.c/ha), 6) teflubenzuron and 7) spinosad, in the doses and recommended way of application. After the tenth second after the emergency of the plants (DAE), had been day applied the treatments in foliar spraying (PF), being evaluated to the 2, 9, 15 and 21 days after the application (DAA), the number of small caterpillars (lesser that 1,5 cm) for parcel in 20 showed plants. The applied results show that teflubenzuron and spinosad in PF to the 12 DAE had been most efficient (71 and 83% respectively). The microbial's insecticides had not differed significantly from the chemical insecticides in the percentage of plants attacked for the pest and the 15 and 21 DAA.

This paper aims to describe the familiar cotton culture in the districts of Tangará and Triunfo Potiguar, located in the state of Rio Grande do Norte. It relates specific problems that small cotton agriculturists face in the cotton production and sale as well as their perception in relation to public policies that have been put into practice in these districts. The research revealed soils with potentiality for the cotton culture, but that are not being cultivated for anything else but subsistence, not being able to become productive, specifically through the cotton culture. That activity should be practiced so that it preserves the environment and the familiar farmers themselves, since it was verified they are constant1y exposed to health problems due to insecticides use. In what refers to fomentation policies it was observed that bureaucracy delays the liberation of the money destined to production, seeds delivery and other actions related to production and commercialization. Some procedures such as information and training about productivity development and attention with the environrnent and infrastructure are recommended. The results indicate lack of investments, rural credit and technical help, main1y in Triunfo Potiguar. There is unanimity, from the rural farmers of that district in what refers to the desire of improving the production and remaining in that activity. In this sense, the contribution of this research for the most appropriate culture production can be materialized through the technique of tissue's culture, aimed at obtaining plants that are resistant to diseases. The data indicated that the hormonal supplementations used induced regenerative calluses making in vitro morphogenesis possible in alI tested varieties. The use of the activated coal antioxidant was efficient, reducing oxidation, however not suppressing it. The results lead to an opportunity for the familiar tàrmers from Rio Grande do Norte enlarge the cotton culture, because to obtain plants that are resistant to diseases implies in insecticides reduction, propitiating smaller impact to the atmosphere and less cost

Societal concerns about environmental sustainability has lead to the development of ecologically-friendly alternatives to chemical insecticides for crop protection. One such alternative is biological pest control. In particular, baculoviruses are well suited as insect biopesticides due to their narrow host specificity and relative ease of propagation. In Brazil, the baculovirus Anticarsia gemmatalis nucleopolyhedrovirus (AgMNPV) is the main biological control agent employed for the soybean pest, Anticarsia gemmatalis. This baculovirus biopesticide is currently produced using caterpillars, but increasing market demand for the product has encouraged the development of an in vitro manufacturing process, which can be scaled up to much higher virus productivities. In this study, three wild-type AgMNPV isolates (AgMNPV-2D, AgMNPV-MP2 and AgMNPV-MP5) and a recombinant form (vAgEGT-LacZ) were characterised in terms of occlusion body (OB) production and infection kinetics, to enable future optimisation of the in vitro production process. These viruses were propagated using a Spodoptera frugiperda (IPLB-SF21) insect cell line grown in shaker-flask batch cultures. Among the virus isolates tested, AgMNPV-MP5 was found to be the best producer, yielding (5.3±0.85)x108 OB/mL after 8 days post infection. The characterisation of vAgEGT-LacZ propagation in suspension cell cultures has not been previously reported in the literature; hence it became the main focus for this thesis. In particular, it was carried out a study on the effect of the multiplicity of infection (MOI) on OB production. Five successive batches were performed getting a final production (8.9±1.42)x1014 occlusion bodies, considering that production is related for a bioreactor with final volume of 10m3. A low MOI associated with a fed-batch process for vAgEGT-LacZ production was found to support a 3-fold higher OB yield when compared to the default batch process (1.8x107 and 5.3x107 OB/mL, respectively). This yield is competitive with regards to the production process.

Dengue and dengue hemorrhagic fever, vector-borne diseases transmitted by the mosquito Aedes aegypti, are presently important public health problems in Brazil. As the strategy for disease control is based on vector control through the use of insecticides, the development of resistance is a threat to programs efficacy. The objective of this study was to compare the Aedes aegypti susceptibility in nine vector populations from the state of São Paulo and seven from Northeast region of Brazil, since there was a difference on group of insecticide used between the areas. Bioassays with larvae and adult were performed according to the World Health Organization methods. The results showed higher resistance levels to organophosphates group in populations from the Northeast region where this group was used for both larvae and adult control than in São Paulo where organophosphates were used for larvae and pyretroids for adult control. Resistance to pyretroids in adults was widespread in São Paulo after ten years of use of cypermethrin while in vector populations from the Northeast region it was punctual. The difference in resistance profile between the areas is in accordance to the group of insecticide used.

Background Head lice constitute a problem in children. Each year, numerous cases of pediculosis occur worldwide. Little work has been performed to evaluate the understanding, opinions, and actions of populations regarding head lice. These areas are important as they enable clinicians and educators to alert parents on how to avoid treatments that are innocuous or of high risk to patients. Methods A cross-sectional study was performed by interviewing the heads of households of 100 randomly chosen residences within the study area. Results The results obtained showed that 13% were infested during the first week of the survey, and 86% in the 24 weeks prior to the study. The number of positive cases increased with increasing resident number, and decreased in families with parents with a higher educational level. Itching was the principal clinical manifestation and caused sleep compromise in 65% of respondents. Innocuous and unhealthy practices to combat infestation, such as the use of inflammables and home insecticides, were common. Conclusion The results showed that certain beliefs generated worry and confusion in parents, who blamed head lice as the cause of various health problems which were not due to this insect.
