Tanulmányunk célja a versenyképesség közösségi beágyazottságának elemzéséhez alkalmas elemzési keretek bemutatása és a versenyképesség fogalmának elemzése a fogalom intézményi, normatív tartalma szempontjából. Célunk a közösségi versenyképesség fogalmának és az ezt elemezni képes megközelítés kidolgozása. A feladat kettős: 1. A versenyképesség értelmezése a döntések közösségi keretei szempontjából 2. A közösségi keretek versenyképességre gyakorolt hatásának elemzése Ennek érdekében a tanulmány első fejezetében az egyéni döntést meghatározó tényezőket és az egyéni döntések jövőbeli interakciók környezetére gyakorolt hatását elemző keretet vázoljuk fel. Megközelítésünk szerint az egyéni döntést négy tényező határozza meg. a közösségi környezet, a természeti környezet, s személyes jellemzők és az interakciós partnerek. Az ez alapján születő döntések formálják a jövőbeli döntési környezetet. A döntések hatásának elemzéséhez a környezetre gyakorolt hatások értékelését orientáló fogalomra van szükség. Elemzésünk esetében ez a fogalom az értékteremtés, amit a következőképp határozunk meg: az értékteremtő tevékenységek során valaki arra törekszik, hogy saját személyes céljait a másokkal való kölcsönösen előnyös együttműködések lehetőségeinek bővítésével, hozamainak növelésével szolgálja. A második fejezetben az egyéni döntések közösségi kereteit és az egyéni döntések közösségre gyakorolt hatását elemezzük részletesen. A formális és informális intézmények világát, a közösségi magatartásokat szabályozó normák és konvenciók rendszerét a következő öt – a valóságban gyakran keveredő - alapelemre bontjuk értékrend, konvenció, közösségi szabály, hivatalos előírás, egyének közötti megállapodás. Ezek közül a magánszereplők együttműködésének az érintett szereplők által módosítható intézményi elemeinek (konvenció, megállapodás) alkalmazkodása a leggyorsabb, a közösség egészét irányító formális intézmények a status quo iránti elfogultságuk miatt lomhábbak, míg a közösség életét informálisan befolyásoló normák a legstabilabb intézményi elemek. A közösségek változása általában lassú, legtöbbször nem szándékolt hatások következménye. Mindezek mellett a közösségi intézmények tudatos alakításában komoly szerepe van (1) a konvenciókat megújító intézményi innovátoroknak, (2) a szerződéses formulákon módosító vállalkozóknak és (3) a hivatalos előírások formálásába bekapcsolódó politikai szereplőknek politikai vállalkozóként, tisztviselőként, vagy közéleti résztvevőként. A harmadik fejezetben a versenyképesség fogalmát elemezzük, és ez alapján határozzuk meg a közösségi versenyképesség fogalmát. Megvizsgáljuk, milyen feltevésekkel él a fogalom a közösségi környezettel kapcsolatban, illetve milyen normatív elemei vannak a definíciónak. A vizsgálathoz a versenyképesség fogalmának egy lecsupaszított változatát használtuk. E szerint a versenyképesség valaki képessége értékteremtő módon bekapcsolódni a gazdasági munkamegosztásba úgy, hogy tevékenysége relatív hozama nem csökken. Az elemzés alapján a versenyképesség a közösségi környezet következő hét elemére épül: 1. A közösség tagjainak és a tagság tartalmának meghatározottsága; 2. a potenciális együttműködő felek közös múltja, jövője, konvenció- és normarendszere; 3. A gazdasági együttműködés intézményeinek (csere, vállalkozás, tulajdon, szerződés) működőképessége; 4. Az értékteremtés normatív koncepciója és az arra épülő részleteiben meghatározott, és részleteiben is közösségi legitimációval bíró szabályrendszer; 5. Az innovációt támogató és a kellően rugalmas értékrend és közösségi szabályok. 6. A gazdasági munkamegosztás igényeihez részleteiben és változásával is igazodó konvenciók, hivatalos előírások és szerződések; 7. A közösségi környezet tudatos alakításával foglalkozó szereplők (közösségi innovátorok, vállalkozók és politikai szereplők) motivációja és lehetősége a hozamok relatív szintjének tartását támogató intézményi környezet karbantartásában. A versenyképesség fogalmának intézményi elemzése rámutat, hogy a fogalom gazdag értéktartalommal és határozott közösségi intézményrendszer-képpel rendelkezik. A közösségi versenyképesség ez alapján a versenyképesség fogalmába kódolt közösségi környezetként határozható meg. A kutatás következő lépése a közösségi versenyképesség meghatározása, az azt befolyásoló mechanizmusok feltárása és javítását támogató elemzési eszközök, gyakorlati segédletek kidolgozása. Ezen feladatok előkészítése érdekében a tanulmány mellékletében két történelmi esettanulmányt mutatunk be, röviden áttekintjük a téma szempontjából releváns irodalom főbb eredményeit és bemutatunk egy praktikus alkalmazásra szánt normatív elemzési eszközt, mellyel az elemezhető, hogy az állami lépések mennyire bátorítják az értékteremtő vállalkozást. _________ This paper (1) introduces an analytical framework to study the impact of the community on competitiveness and (2) analyses the institutional and normative content in the concept of competitiveness. The goal is to elaborate an approach that supports the definition and analysis of the ‘competiveness of community’. This task has two main parts: 1. interpretation of competiveness from social choice perspective 2. assessing the impact of social settings on the competiveness of a community The first chapter of the study draws up an analytical framework to study the social factors of individual decisions and their impact on the environment of future interactions. We focus on four main factors that shape setting of future interactions: social environment, natural environment, personal characteristics and partners in interactions. We use the concept of value creation to assess the impact of individual decisions on these factors. The second chapter discusses the social factors of individual decisions and the impact of individual decisions on the community. Institutions are conceptualized as value systems, conventions, community rules, official rules and contracts in the study. The conventions and contracts can accommodate to the changes of environment more smoothly, formal institutions are less flexible due to their bias toward status quo. Informal rules and value systems resists change more frequently. The formation of social environment is usually slow and based on unintended effects. Altogether (1) innovators who revise social conventions, (2) entrepreneurs who reshape contracts and (4) political entrepreneurs who formulate formal rules have influential roles on the institutional setting. The third chapter discusses the social assumptions included into the definition of competitiveness and we give a definition for the competiveness of communities. A simplified definition of competitiveness is used for this analysis: competiveness is someone’s ability and motivation to participate in the economic division of labor in a way that is based on value creation and maintains the relative return of activities. Our analysis reveals that competitiveness assumes the following features of the community: 1. Defined membership of community: who are the members and what does membership mean. 2. Common past, future, convention and norm system of the potential participants of interactions 3. Functionality of institutions that facilitate division of labor (exchange, entrepreneurship, property, contract) 4. Existing normative concept on value creation and social accepted rules that govern interactions 5. Value system and rules that promote innovation 6. Conventions, official norms and contracts that fits to economic division of labor in a detailed and dynamic way 7. Motivation and potential of actors who shape social environment consciously to maintain institutions in order to sustain the relative return of economic activities This analysis shows that the concept of competitiveness assumes well established values and detailed expectations on institutional settings. Followingly, competiveness of community can be defined with these criteria of social environment. Two historical case studies and the draft of a policy oriented toolkit demonstrate the applicability of the introduced approach in the appendix. The core findings of the literature are also reviewed there.

Metagenomics is the culture-independent study of genetic material obtained directly from environmental samples. It has become a realistic approach to understanding microbial communities thanks to advances in high-throughput DNA sequencing technologies over the past decade. Current research has shown that different sites of the human body house varied bacterial communities. There is a strong correlation between an individual’s microbial community profile at a given site and disease. Metagenomics is being applied more often as a means of comparing microbial profiles in biomedical studies. The analysis of the data collected using metagenomics can be quite challenging and there exist a plethora of tools for interpreting the results. An automatic analytical workflow for metagenomic analyses has been implemented and tested using synthetic datasets of varying quality. It is able to accurately classify bacteria by taxa and correctly estimate the richness and diversity of each set. The workflow was then applied to the study of the airways microbiome in Chronic Obstructive Pulmonary Disease (COPD). COPD is a progressive lung disease resulting in narrowing of the airways and restricted airflow. Despite being the third leading cause of death in the United States, little is known about the differences in the lung microbial community profiles of healthy individuals and COPD patients. Bronchoalveolar lavage (BAL) samples were collected from COPD patients, active or ex-smokers, and never smokers and sequenced by 454 pyrosequencing. A total of 56 individuals were recruited for the study. Substantial colonization of the lungs was found in all subjects and differentially abundant genera in each group were identified. These discoveries are promising and may further our understanding of how the structure of the lung microbiome is modified as COPD progresses. It is also anticipated that the results will eventually lead to improved treatments for COPD.

Vascular cognitive impairment (VCI), including its severe form, vascular dementia (VaD), is the second most common form of dementia. The genetic etiology of sporadic VCI remains largely unknown. We previously conducted a systematic review and meta-analysis of all published genetic association studies of sporadic VCI prior to 6 July 2012, which demonstrated that APOE (ɛ4, ɛ2) and MTHFR (rs1801133) variants were associated with susceptibility for VCI. De novo genotyping was conducted in a new independent relatively large collaborative European cohort of VaD (nmax = 549) and elderly non-demented samples (nmax = 552). Where available, genotype data derived from Illumina's 610-quad array for 1210 GERAD1 control samples were also included in analyses of genes examined. Associations were tested using the Cochran-Armitage trend test: MTHFR rs1801133 (OR = 1.36, 95% CI 1.16-1.58, p = <0.0001), APOE rs7412 (OR = 0.62, 95% CI 0.42-0.90, p = 0.01), and APOE rs429358 (OR = 1.59, 95% CI 1.17-2.16, p = 0.003). Association was also observed with APOE epsilon alleles; ɛ4 (OR = 1.85, 95% CI 1.35-2.52, p = <0.0001) and ɛ2 (OR = 0.67, 95% CI 0.46-0.98, p = 0.03). Logistic Regression and Bonferroni correction in a subgroup of the cohort adjusted for gender, age, and population maintained the association of APOE rs429358 and ɛ4 allele.

BACKGROUND: The aim of this study was to investigate the association between statin use and survival in a population-based colorectal cancer (CRC) cohort and perform an updated meta-analysis to quantify the magnitude of any association.

METHODS: A cohort of 8391 patients with newly diagnosed Dukes' A-C CRC (2009-2012) was identified from the Scottish Cancer Registry. This cohort was linked to the Prescribing Information System and the National Records of Scotland Death Records (until January 2015) to identify 1064 colorectal cancer-specific deaths. Adjusted hazard ratios (HRs) and 95% confidence intervals (CIs) for cancer-specific mortality by statin use were calculated using time dependent Cox regression models. The systematic review included relevant studies published before January 2016. Meta-analysis techniques were used to derive combined HRs for associations between statin use and cancer-specific and overall mortality.

RESULTS: In the Scottish cohort, statin use before diagnosis (HR=0.84, 95% CI 0.75-0.94), but not after (HR=0.90, 95% CI 0.77-1.05), was associated with significantly improved cancer-specific mortality. The systematic review identified 15 relevant studies. In the meta-analysis, there was consistent (I(2)=0%,heterogeneity P=0.57) evidence of a reduction in cancer-specific mortality with statin use before diagnosis in 6 studies (n=86,622, pooled HR=0.82, 95% CI 0.79-0.86) but this association was less apparent and more heterogeneous (I(2)=67%,heterogeneity P=0.03) with statin use after diagnosis in 4 studies (n=19,152, pooled HR=0.84, 95% CI 0.68-1.04).

CONCLUSION: In a Scottish CRC cohort and updated meta-analysis there was some evidence that statin use was associated with improved survival. However, these associations were weak in magnitude and, particularly for post-diagnosis use, varied markedly between studies.

We evaluate whether society can adequately be conceptualized as a component of social-ecological systems, given social theory and the current outputs of systems-based research. A mounting critique from the social sciences posits that resilience theory has undertheorized social entities with the concept of social-ecological systems. We trace the way that use of the term has evolved, relating to social science theory. Scientometic and network analysis provide a wide range of empirical data about the origin, growth, and use of this term in academic literature. A content analysis of papers in Ecology and Society demonstrates a marked emphasis in research on institutions, economic incentives, land use, population, social networks, and social learning. These findings are supported by a review of systems science in 18 coastal assessments. This reveals that a systems-based conceptualization tends to limit the kinds of social science research favoring quantitative couplings of social and ecological components and downplaying interpretive traditions of social research. However, the concept of social-ecological systems remains relevant because of the central insights concerning the dynamic coupling between humans and the environment, and its salient critique about the need for multidisciplinary approaches to solve real world problems, drawing on heuristic devices. The findings of this study should lead to more circumspection about whether a systems approach warrants such claims to comprehensiveness. Further methodological advances are required for interdisciplinarity. Yet there is evidence that systems approaches remain highly productive and useful for considering certain social components such as land use and hybrid ecological networks. We clarify advantages and restrictions of utilizing such a concept, and propose a reformulation that supports engagement with wider traditions of research in the social sciences.

Synthetic polymers constitute a wide class of materials which has enhanced the quality of human life, providing comforts and innovations. Anyway, the increasing production and the incorrect waste management, are leading to the occurrence of polymers in the environment, generating concern. To understand the extent of this issue, analytical investigation holds an essential position. Standardised methods have not established yet, and additional studies are required to improve the present knowledge. The main aim of this thesis was to provide comprehensive information about the potential of pyrolysis coupled with gas-chromatography and mass spectrometry (Py-GC-MS) for polymers investigation, from their characterisation to their identification and quantification in complex matrices. Water-soluble (poly(dimethylsiloxanes), PDMS bearing poly(ethylene glycol), PEG, side chains) and water-insoluble polymers (microplastics, MPs, and bioplastics) were studied. The different studies revealed the possibility to identify heterogeneous classes of polymers, fingerprinting the presence of PDMS copolymers and distinguishing chemically different polyurethanes (PURs). The occurrence of secondary reactions in pyrolysis of polymer mixtures was observed as possible drawback. Pyrolysis products indicative of secondary reactions and their reaction mechanisms were identified. Py-GC-MS also revealed its fundamental role for the identification of polymers composing commercial bioplastics items based. The results aided to identify chemicals that have the potential to migrate in sea waters. Investigations of environmental samples demonstrated the capability of Py-GC-MS to provide reliable, reproducible and comparable results about polymers in complex matrices (PEG-PDMS in sewage sludges and PURs and other MPs in road dusts and spider webs). Criticisms were especially found in quantitation, such as the retrieval reference materials, the construction of reliable calibration protocols and the occurrence of bias due to interferences between pyrolysis products. This thesis pursues the greater purpose to develop harmonised and standardised methods for environmental investigations of polymers, that is fundamental to assess the real state of the environment.

A rapid, sensitive and specific method for quantifying propylthiouracil in human plasma using methylthiouracil as the internal standard (IS) is described. The analyte and the IS were extracted from plasma by liquid-liquid extraction using an organic solvent (ethyl acetate). The extracts were analyzed by high performance liquid chromatography coupled with electrospray tandem mass spectrometry (HPLC-MS/MS) in negative mode (ES-). Chromatography was performed using a Phenomenex Gemini C18 5μm analytical column (4.6mm×150mm i.d.) and a mobile phase consisting of methanol/water/acetonitrile (40/40/20, v/v/v)+0.1% of formic acid. For propylthiouracil and I.S., the optimized parameters of the declustering potential, collision energy and collision exit potential were -60 (V), -26 (eV) and -5 (V), respectively. The method had a chromatographic run time of 2.5min and a linear calibration curve over the range 20-5000ng/mL. The limit of quantification was 20ng/mL. The stability tests indicated no significant degradation. This HPLC-MS/MS procedure was used to assess the bioequivalence of two propylthiouracil 100mg tablet formulations in healthy volunteers of both sexes in fasted and fed state. The geometric mean and 90% confidence interval CI of Test/Reference percent ratios were, without and with food, respectively: 109.28% (103.63-115.25%) and 115.60% (109.03-122.58%) for Cmax, 103.31% (100.74-105.96%) and 103.40% (101.03-105.84) for AUClast. This method offers advantages over those previously reported, in terms of both a simple liquid-liquid extraction without clean-up procedures, as well as a faster run time (2.5min). The LOQ of 20ng/mL is well suited for pharmacokinetic studies. The assay performance results indicate that the method is precise and accurate enough for the routine determination of the propylthiouracil in human plasma. The test formulation with and without food was bioequivalent to reference formulation. Food administration increased the Tmax and decreased the bioavailability (Cmax and AUC).

Ecosystem engineering is increasingly recognized as a relevant ecological driver of diversity and community composition. Although engineering impacts on the biota can vary from negative to positive, and from trivial to enormous, patterns and causes of variation in the magnitude of engineering effects across ecosystems and engineer types remain largely unknown. To elucidate the above patterns, we conducted a meta-analysis of 122 studies which explored effects of animal ecosystem engineers on species richness of other organisms in the community. The analysis revealed that the overall effect of ecosystem engineers on diversity is positive and corresponds to a 25% increase in species richness, indicating that ecosystem engineering is a facilitative process globally. Engineering effects were stronger in the tropics than at higher latitudes, likely because new or modified habitats provided by engineers in the tropics may help minimize competition and predation pressures on resident species. Within aquatic environments, engineering impacts were stronger in marine ecosystems (rocky shores) than in streams. In terrestrial ecosystems, engineers displayed stronger positive effects in arid environments (e.g. deserts). Ecosystem engineers that create new habitats or microhabitats had stronger effects than those that modify habitats or cause bioturbation. Invertebrate engineers and those with lower engineering persistence (<1 year) affected species richness more than vertebrate engineers which persisted for >1 year. Invertebrate species richness was particularly responsive to engineering impacts. This study is the first attempt to build an integrative framework of engineering effects on species diversity; it highlights the importance of considering latitude, habitat, engineering functional group, taxon and persistence of their effects in future theoretical and empirical studies.

Frailty and anemia in the elderly appear to share a common pathophysiology associated with chronic inflammatory processes. This study uses an analytical, cross-sectional, population-based methodology to investigate the probable relationships between frailty, red blood cell parameters and inflammatory markers in 255 community-dwelling elders aged 65 years or older. The frailty phenotype was assessed by non-intentional weight loss, fatigue, low grip strength, low energy expenditure and reduced gait speed. Blood sample analyses were performed to determine hemoglobin level, hematocrit and reticulocyte count, as well as the inflammatory variables IL-6, IL-1ra and hsCRP. In the first multivariate analysis (model I), considering only the erythroid parameters, Hb concentration was a significant variable for both general frailty status and weight loss: a 1.0g/dL drop in serum Hb concentration represented a 2.02-fold increase (CI 1.12-3.63) in an individual's chance of being frail. In the second analysis (model II), which also included inflammatory cytokine levels, hsCRP was independently selected as a significant variable. Each additional year of age represented a 1.21-fold increase in the chance of being frail, and each 1-unit increase in serum hsCRP represented a 3.64-fold increase in the chance of having the frailty phenotype. In model II reticulocyte counts were associated with weight loss and reduced metabolic expenditure criteria. Our findings suggest that reduced Hb concentration, reduced RetAbs count and elevated serum hsCRP levels should be considered components of frailty, which in turn is correlated with sarcopenia, as evidenced by weight loss.

Chlorophenylpiperazines (CPP) are psychotropic drugs used in nightclub parties and are frequently used in a state of sleep deprivation, a condition which can potentiate the effects of psychoactive drugs. This study aimed to investigate the effects of sleep deprivation and sleep rebound (RB) on anxiety-like measures in mCPP-treated mice using the open field test. We first optimized our procedure by performing dose-effect curves and examining different pretreatment times in naïve male Swiss mice. Subsequently, a separate cohort of mice underwent paradoxical sleep deprivation (PSD) for 24 or 48h. In the last experiment, immediately after the 24h-PSD period, mice received an injection of saline or mCPP, but their general activity was quantified in the open field only after the RB period (24 or 48h). The dose of 5mgmL(-1) of mCPP was the most effective at decreasing rearing behavior, with peak effects 15min after injection. PSD decreased locomotion and rearing behaviors, thereby inhibiting a further impairment induced by mCPP. Plasma concentrations of mCPP were significantly higher in PSD 48h animals compared to the non-PSD control group. Twenty-four hours of RB combined with mCPP administration produced a slight reduction in locomotion. Our results show that mCPP was able to significantly change the behavior of naïve, PSD, and RB mice. When combined with sleep deprivation, there was a higher availability of drug in plasma levels. Taken together, our results suggest that sleep loss can enhance the behavioral effects of the potent psychoactive drug, mCPP, even after a period of rebound sleep.

Mapping of elements in biological tissue by laser induced mass spectrometry is a fast growing analytical methodology in life sciences. This method provides a multitude of useful information of metal, nonmetal, metalloid and isotopic distribution at major, minor and trace concentration ranges, usually with a lateral resolution of 12-160 µm. Selected applications in medical research require an improved lateral resolution of laser induced mass spectrometric technique at the low micrometre scale and below. The present work demonstrates the applicability of a recently developed analytical methodology - laser microdissection associated to inductively coupled plasma mass spectrometry (LMD ICP-MS) - to obtain elemental images of different solid biological samples at high lateral resolution. LMD ICP-MS images of mouse brain tissue samples stained with uranium and native are shown, and a direct comparison of LMD and laser ablation (LA) ICP-MS imaging methodologies, in terms of elemental quantification, is performed.

Few articles deal with lead and strontium isotopic analysis of water samples. The aim of this study was to define the chemical procedures for Pb and Sr isotopic analyses of groundwater samples from an urban sedimentary aquifer. Thirty lead and fourteen strontium isotopic analyses were performed to test different analytical procedures. Pb and Sr isotopic ratios as well as Sr concentration did not vary using different chemical procedures. However, the Pb concentrations were very dependent on the different procedures. Therefore, the choice of the best analytical procedure was based on the Pb results, which indicated a higher reproducibility from samples that had been filtered and acidified before the evaporation, had their residues totally dissolved, and were purified by ion chromatography using the Biorad® column. Our results showed no changes in Pb ratios with the storage time.

The present research deals with two mural paintings made in 1947 with the fresco technique by Fulvio Pennacchi in the Catholic Chapel of the Hospital das Clínicas (São Paulo City, Brazil), namely the Virgin Annunciation and the Supper at Emmaus. This study regards the materials and painting techniques used by the artist, based on historical research,on in situ observations and laboratory analytical techniques (stereomicroscopy,scanning electron microscopy with an energy dispersive spectrometer, X-ray diffractometry, electron microprobe, images obtained with UV-light), aiming to improve the methods of characterization of objects of our cultural heritage, and to enhance its preservation accordingly. The results lead to the identification of the plaster components and of distinct layers in the frescoes, besides further information on grain size, impurities and textures, composition of pigments, and features of deterioration, such as efflorescences. The degree of degradation of the murals painting was assessed by this way. Our data suggest that a single layer of plaster was used by Pennacchi, as a common mortar with fine- and medium-grained aggregates. Differences in texture were obtained by adding gypsum to the plaster.
