The EU-funded research project ALARM will develop and test methods and protocols for the assessment of large-scale environmental risks in order to minimise negative human impacts. Research focuses on the assessment and forecast of changes in biodiversity and in the structure, function, and dynamics of ecosystems. This includes the relationships between society, the economy and biodiversity.

Agri-environment schemes (AES) are widely used policy instruments intended to combat widespread biodiversity declines across agricultural landscapes. Here, using a light trapping and mark-release-recapture study at a field-scale on nine common and widespread larger moth species, we investigate the effect of wide field margins (a popular current scheme option) and the presence of hedgerow trees (a potential scheme option in England) on moth abundance. Of these, we show that wide field margins positively affected abundances, although species did not all respond in the same way. We demonstrate that this variation can be attributed to species-specific mobility characteristics. Those species for which the effect of wide margins was strongest covered shorter distances, and were more frequently recaptured at their site of first capture. This demonstrates that the standard, field-scale uptake of AES may be effective only for less mobile species. We discuss that a landscape-scale approach, in contrast, could deliver significant biodiversity gains, as our results indicate that such an approach (perhaps delivered through targeting farmers to join AES) would be effective for the majority of wider countryside species, irrespective of their mobility level. A detailed spore investigation of spore release and dispersal from an isolated colony of Phascum cuspidatum Hedw. indicated that approximately 98% of the spores originally present remained within the colony. The spatial distribution of colonies of P.cuspidatum and Pottia truncata (Hedw.) Fürer. in relation to those of the previous year was investigated by mapping the occurrence of colonies in five permanent quadrats for each species during two successive years. Phascum cuspidatum reoccurred in three quadrats during the second year, and P. truncata in only one, in the latter case apparently due to invasion by other mosses, principally Barbula hornschuchiana Schultz. A substantial proportion of the second year colonies overlapped in position with the first year colonies, particularly in P.cuspidatum. The results are discussed in relation to data on spore dispersal and other aspects of the life-history of these annual or short-lived shuttle mosses.

Background: The large-scale production of G-protein coupled receptors (GPCRs) for functional and structural studies remains a challenge. Recent successes have been made in the expression of a range of GPCRs using Pichia pastoris as an expression host. P. pastoris has a number of advantages over other expression systems including ability to post-translationally modify expressed proteins, relative low cost for production and ability to grow to very high cell densities. Several previous studies have described the expression of GPCRs in P. pastoris using shaker flasks, which allow culturing of small volumes (500 ml) with moderate cell densities (OD600 similar to 15). The use of bioreactors, which allow straightforward culturing of large volumes, together with optimal control of growth parameters including pH and dissolved oxygen to maximise cell densities and expression of the target receptors, are an attractive alternative. The aim of this study was to compare the levels of expression of the human Adenosine 2A receptor (A(2A)R) in P. pastoris under control of a methanol-inducible promoter in both flask and bioreactor cultures. Results: Bioreactor cultures yielded an approximately five times increase in cell density (OD600 similar to 75) compared to flask cultures prior to induction and a doubling in functional expression level per mg of membrane protein, representing a significant optimisation. Furthermore, analysis of a C-terminally truncated A2AR, terminating at residue V334 yielded the highest levels (200 pmol/mg) so far reported for expression of this receptor in P. pastoris. This truncated form of the receptor was also revealed to be resistant to C-terminal degradation in contrast to the WT A(2A)R, and therefore more suitable for further functional and structural studies. Conclusion: Large-scale expression of the A(2A)R in P. pastoris bioreactor cultures results in significant increases in functional expression compared to traditional flask cultures.

The efficacy of family interventions in psychosis is well documented. UK and USA schizophrenia treatment guidelines advocate the practice of family interventions within routine clinical services. However, less attention has been paid to the study of treatment fidelity and the tools used in its assessment. This study reports the inter-rater reliability of a new scale: Family Intervention in Psychosis-Adherence Scale (FIPAS). This measure is designed to assess therapist adherence to the Kuipers et al. (2002) family intervention in psychosis treatment manual. Reliability ratings were based on a sample of thirteen audiotapes drawn from a randomized controlled trial of family intervention. The results indicated that the majority of items of the FIPAS had acceptable levels of inter-rater reliability. The findings are discussed in terms of their implications for the training and monitoring of the effectiveness of practitioners for family interventions in psychosis.

The factor structure of the Edinburgh Postnatal Depression scale (EPDS) and similar instruments have received little attention in the literature. The researchers set out to investigate the construct validity and reliability of the EPDS amongst impoverished South African women. The EPDS was translated into isiXhosa (using Brislin's back translation method) and administered by trained interviewers to 147 women in Khayelitsha, South Africa. Responses were subjected to maximum likelihood confirmatory factor analysis. A single factor structure was found, consistent with the theory on which the EPDS was based. Internal consistency was satisfactory (a = 0.89).
