La Piana di foce del Garigliano (al confine tra Lazio e Campania) è caratterizzata, fino ad epoche recenti, dalla presenza di aree palustri e umide. Lo studio in corso cerca di ricostruire l’evoluzione dell’ambiente costiero mettendolo in relazione alla presenza dell’uomo, alla gestione del territorio, alle vicende storiche e alle variazioni climatiche utilizzando molteplici metodologie tipiche della geoarcheologia. Si tratta di un approccio multidisciplinare che cerca di mettere insieme analisi tipiche dell’archeologia, della topografia antica, della geomorfologia, della geologia e della paleobotanica. Fino all’età del Ferro l’unica traccia di popolamento viene da Monte d’Argento, uno sperone roccioso isolato lungo la costa, posto al limite occidentale di un ambiente sottostante che sembra una palude chiusa e isolata da apporti sedimentari esterni. Con il passaggio all’età del ferro si verifica un mutamento ambientale con la fine della grande palude e la formazione di una piccola laguna parzialmente comunicante con il mare. L’arrivo dei romani alla fine del III secolo a.C. segna la scomparsa dei grandi centri degli Aurunci e la deduzione di tre colonie (Sessa Aurunca, Sinuessa, Minturno). Le attività di sistemazione territoriale non riguardarono però le aree umide costiere, che non vennero bonificate o utilizzate per scopi agricoli, ma mantennero la loro natura di piccoli laghi costieri. Quest’epoca è dunque caratterizzata da una diffusione capillare di insediamenti, basati su piccole fattorie o installazioni legate allo sfruttamento agricolo. Poche sono le aree archeologiche che hanno restituito materiali successivi al II-III secolo d.C. La città resta comunque abitata fino al VI-VII secolo, quando l’instabilità politica e l’impaludamento dovettero rendere la zona non troppo sicura favorendo uno spostamento verso le zone collinari. Un insediamento medievale è attestato solo a Monte d’Argento e una frequentazione saracena dell’inizio del IX secolo è riportata dalle fonti letterarie, ma non vi è ancora nessuna documentazione archeologica.

This work comprises three different types of unconventional correlated systems.rnChapters 3-5 of this work are about the open shell compounds Rb4O6 and Cs4O6. These mixed valent compounds contain oxygen in two different modifications: the closed-shell peroxide anion is nonmagnetic, whereas the hyperoxide anion contains an unpaired electrons in an antibonding pi*-orbital. Due to this electron magnetic ordering is rendered possible. In contrast to theoretical predictions, which suggested half-metallic ferromagnetism for Rb4O6,rndominating antiferromagnetic interactions were found in the experiment. Besidesrna symmetry reduction due to the mixed valency, strong electronic correlations of this highly molecular system determine its properties; it is a magnetically frustrated insulator. The corresponding Cs4O6 was found to show similar properties.rnChapters 6-9 of this work are about intermetallic Heusler superconductors. rnAll of these superconductors were rationally designed using the van Hove scenario as a working recipe. A saddle point in the energy dispersion curve of a solid leads to a van Hove singularity in the density of states. In the Ni-based and Pd-based Heusler superconductors presented in this work this sort of a valence instability occurs at the high-symmetry L point and coincides or nearly coincides with the Fermi level. The compounds escape the high density of states at the Fermi energy through a transition into the correlated superconducting state.rnChapter 10 of this work is about the tetragonally distorted ferrimagnetic DO22 phase of Mn3Ga. This hard-magnetic modification is technologically useful for spin torque transfer applications. The phase exhibits two different crystallographic sites that are occupied by Mn atoms and can thus be written as Mn2MnGa. The competition between the mainly itinerant moments of the Mn atoms at the Wyckoff position 4d and the localized moments of the Mn atoms at the Wyckoff position 2b leads to magnetic correlations. The antiferromagnetic orientation of these moments determines the compound to exhibit a resulting magnetic moment of approximately 1 µB per formula unit in a partially compensated ferrimagnetic configuration.

La ricerca ha preso le mosse da tre ipotesi fondamentali: 1) Esiste un legame tra processi cognitivi di basso ed alto livello; 2) Lo spazio senso-motorio è una percezione soggettiva; 3) Lo spazio senso-motorio varia in funzione delle diverse modalità di interazione sociale. La tesi sostiene che lo spazio senso-motorio si lascia modulare dalla semplice co-presenza di un altro agente umano e da interazioni cooperative e non cooperative. I capitoli I, II, III, hanno lo scopo di scomporre e spiegare il significato della prima, seconda e terza ipotesi; giungendo a formulare la tesi centrale che sarà poi dimostrata sperimentalmente nel capitolo IV. Il capitolo V introduce future linee di ricerca nell’ambito dell’etica proponendo una nuova ipotesi sul legame che potrebbe sussistere tra la percezione dello spazio durante l’interazione sociale e i giudizi morali. Il lavoro svolto chiama ad operare insieme diverse discipline che concorrono a formare le scienze cognitive: la storia della filosofia, la filosofia della mente contemporanea, la neuropsicologia sperimentale ed alcuni temi della psicologia sociale.

La tesi indaga i temi fondamentali, i limiti e le implicazioni della relazione tra le filosofie di Nietzsche e di Spinoza, sia analizzando storiograficamente il ruolo di Spinoza nei testi di Nietzsche (I parte), sia sviluppando una proposta teoretica a partire dai due autori (II, III e IV parte). L’indagine storiografica procede presentando le riflessioni che Nietzsche dedica a Spinoza secondo un criterio tematico-cronologico. Durante l’esposizione sono discusse le fonti utilizzate da Nietzsche e il significato che le menzioni di Spinoza ricoprono nell’economia generale del pensiero nietzschiano. La seconda parte della ricerca indaga le ragioni per le quali la connessione tra Nietzsche e Spinoza può risultare teoreticamente convincente, in particolare a partire dall’istanza propriamente critica della loro riflessione, che si manifesta come domanda “empia” sul mondo delle credenze condivise. In particolare, è la nozione di “causa” a fungere da rifugio per la credenza metafisica nel soggetto anche all’interno dell’universo deterministico in cui opera la scienza. Come va invece pensato un determinismo che sia filosoficamente coerente? Le ultime due parti di questa ricerca sviluppano alcune ipotesi in proposito, considerando singolarmente le filosofie di Spinoza e di Nietzsche.

Diese Arbeit unterstreicht das Potential von Hybridfunktionalen (B3LYP) für die Untersuchung einer großen Bandbreite von Systemen. Durch die Einbeziehung der exakten Hartree-Fock Austauschenergie kann B3LYP für molekulare und kristalline Systeme eingesetzt werden. Zum Beispiel können stark korrelierte Systeme mit B3LYP erfolgreich erforscht werden. Die elektronische Struktur von PAHs wurde mit B3LYP Hybriddichtefunktionalen untersucht. Mit der ∆SCF-Methode wurden Elektronenbindungsenergien bestimmt, welche die mit UPS gewonnenen experimentellen Resultate bestätigen und ergänzen. Symmetrieeigenschaften der molekularen Orbitale wurden analysiert, um eine Zuordnung und Einschätzung der zugehörigen Signalstärke zu ermöglichen. Während σ-artige Orbitale nur schwer durch UPS-Messungen an dünnen Filmen detektiert werden können, bieten Rechnungen eine detaillierte Einsicht in die verborgenen Teile der Spektren.rnWeiterhin wurden π−π-Komplexe untersucht, welche von verschiedenen Donor- und Akzeptor-Molekülen gebildet werden. Die Moleküle basieren auf polyzyklischen, aromatischen Kohlenwasserstoffen. Für Ladungstransferkomplexe finden DFT Rechnungen ein Minimum in der Oberfläche der potentiellen Energie. Diese attraktive Wechselwirkung wird durch Coulombanziehung verursacht. Allerdings ist die Coulombanziehung nicht die stärkste Wechselwirkung in Ladungstransferkomplexen. Die Einbeziehung von van der Waals-Korrekturen verbessert den intermolekularen Abstand und die Bindungsenergie.rnEine Verkleinerung der intermolekularen Abstände führt zu einer großen Verschiebung der HOMO- und LUMO-Energie.rnAus der Klasse der kristallinen korrelierten Systeme wurden Rb4O6 und FeSe untersucht. Im Falle von Rb4O6 führen Ladungsordnung und Korrelationen zu einem isolierenden Grundzustand. Das hypothetische druckabhängige Phasendiagramm wurde untersucht. Eine Erhöhung des Drucks führt zu einer vergrößerten Bandlücke. Bei etwa 75 GPa wird die Bandbreite W größer als der Bandabstand U und das System nimmt einen homogen gemischt valenten Zustand mit teilweise besetzten π−π-Orbitalen an. Für Drücke ab 160 GPa wird W sehr viel größer als U und das System wird metallisch.rnIm Fall von FeSe finden wir eine korrelierte und isolierende Phase bei hohen Drücken, während das System bei niedrigen Drücken supraleitendes Verhalten zeigt. Die Berechnungen der Elektronenstruktur mit dem Hybridfunktional B3LYP führt zum korrekten halbleitenden Grundzustand in der NiAs- und MnP-Struktur von FeSe. Die Rolle der Korrelationen, der Stöchiometrie und der Nähe zum Magnetismus wird besprochen. Im Speziellen wird gezeigt, dass die Phase mit NiAs-Struktur starke lokale Korrelationen aufweist, was zu einem halbleitenden Zustand in einem weiten Druckbereich führt.

Long-term potentiation in the neonatal rat rnbarrel cortex in vivo rnLong-term potentiation (LTP) is important for the activity-dependent formation of early cortical circuits. In the neonatal rodent barrel cortex LTP has been so far only studied in vitro. I combined voltage-sensitive dye imaging with extracellular multi-electrode recordings to study whisker stimulation-induced LTP for both the slope of field potential and the number of multi-unit activity in the whisker-to-barrel cortex pathway of the neonatal rat barrel cortex in vivo. Single whisker stimulation at 2 Hz for 10 min induced an age-dependent expression of LTP in postnatal day (P) 0 to P14 rats with the strongest expression of LTP at P3-P5. The magnitude of LTP was largest in the stimulated barrel-related column, smaller in the surrounding septal region and no LTP could be observed in the neighboring barrel. Current source density analyses revealed an LTP-associated increase of synaptic current sinks in layer IV / lower layer II/III at P3-P5 and in the cortical plate / upper layer V at P0-P1. This study demonstrates for the first time an age-dependent and spatially confined LTP in the barrel cortex of the newborn rat in vivo. These activity-dependent modifications during the critical period may play an important role in the development and refinement of the topographic map in the barrel cortex. (An et al., 2012)rnEarly motor activity triggered by gamma and spindle bursts in neonatal rat motor cortexrnSelf-generated neuronal activity generated in subcortical regions drives early spontaneous motor activity, which is a hallmark of the developing sensorimotor system. However, the neuronal activity patterns and functions of neonatal primary motor cortex (M1) in the early movements are still unknown. I combined voltage-sensitive dye imaging with simultaneous extracellular multi-electrode recordings in the neonatal rat S1 and M1 in vivo. At P3-P5, gamma and spindle bursts observed in M1 could trigger early paw movements. Furthermore, the paw movements could be also elicited by the focal electrical stimulation of M1 at layer V. Local inactivation of M1 could significantly attenuate paw movements, suggesting that the neonatal M1 operates in motor mode. In contrast, the neonatal M1 can also operate in sensory mode. Early spontaneous movements and sensory stimulations of paw trigger gamma and spindle bursts in M1. Blockade of peripheral sensory input from the paw completely abolished sensory evoked gamma and spindle bursts. Moreover, both sensory evoked and spontaneously occurring gamma and spindle bursts mediated interactions between S1 and M1. Accordingly, local inactivation of the S1 profoundly reduced paw stimulation-induced and spontaneously occurring gamma and spindle bursts in M1, indicating that S1 plays a critical role in generation of the activity patterns in M1. This study proposes that both self-generated and sensory evoked gamma and spindle bursts in M1 may contribute to the refinement and maturation of corticospinal and sensorimotor networks required for sensorimotor coordination.rn

Die vorliegende kumulative Arbeit umfasst Analysen zur Aufklärung der molekularen Grundlagen des humanen Usher-Syndroms (USH), der häufigsten Ursache kombinierter vererblicher Taub-Blindheit. Ziel dieser Arbeit war es, neue Erkenntnisse zur Funktion der USH-Proteine und den von ihnen organisierten Protein-Netzwerken in der Photorezeptorzelle zu erhalten. Dadurch sollten weitere Einsichten in die molekularen Ursachen des retinalen Phänotyps von USH gewonnen werden. Die Ergebnisse dieser Analysen wurden in einem Übersichtsartikel (I) und zwei Originalarbeiten (II, III) zusammengestellt.rn Im Übersichtsartikel (I) wurden die vorliegenden Hinweise zusammengefasst, die USH auf Grundlage der molekularen Verbindungen ebenfalls als Ciliopathien definiert. Zudem wird die Bedeutung des periciliären USH-Proteinnetzwerkes für das sensorische Cilium (Außensegment) der Photorezeptorzelle herausgestellt. rn In Publikation II wurde der Aufbau des USH1-USH2-Proteinnetzwerkes als Teil des periciliären Komplexes analysiert, der beim cargo handover von vesikulärer Fracht vom Innensegment- auf den ciliären Transport für die Photorezeptorzelle essentiell ist. Experimentell wurde Ush2a als neuer SANS-Interaktionspartner validiert. Des Weiteren wurde ein ternärer Komplex aus den USH-Proteinen SANS, Ush2a und Whirlin identifiziert, dessen Zusammensetzung durch die phosphorylierungsabhängige Interaktion zwischen SANS und Ush2a reguliert werden könnte. Dieser ternäre Komplex kann sowohl der Integrität der Zielmembran dienen als auch am Transfer von Molekülen ins Außensegment beteiligt sein.rn In Publikation III wurde das MAGUK-Protein Magi2 als neuer Interaktionspartner von SANS identifiziert und die Interaktion durch komplementäre Interaktionsassays validiert. Dabei wurde ein internes PDZ-Binde-Motiv in der SAM-Domäne von SANS identifiziert, das die Interaktion zur PDZ5-Domäne von Magi2 phosphorylierungsabhängig vermittelt. Dadurch wurde bestätigt, dass SANS durch post-translationale Modifizierung reguliert wird. Weiterführende Experimente zur Funktion des Magi2-SANS-Komplexes zeigen, dass Magi2 an Prozess der Rezeptor-vermittelten Endocytose beteiligt ist. Die Phosphorylierung von SANS durch die Kinase CK2 spielt bei der Endocytose ebenfalls eine wichtige Rolle. Der Phosphorylierungsstatus von SANS moduliert die Interaktion zu Magi2 und reguliert dadurch negativ den Prozess der Endocytose. In RNAi-Studien wurde die durch Magi2-vermittelte Endocytose darüber hinaus mit dem Prozess der Ciliogenese verknüpft. Die Analyse der subzellulären Verteilung der Interaktionspartner lokalisieren Magi2 im periciliären Komplex und assoziieren das periciliäre USH-Proteinnetzwerk dadurch mit dem Prozess der Endocytose in der ciliary pocket. Der SANS-Magi2-Komplex sollte demnach für Aufbau und Funktion des sensorischen Ciliums der Photorezeptorzelle eine wichtige Rolle spielen.rn Die Gesamtheit an Informationen, die aus den Publikationen dieser Dissertation und aus den Kooperationsprojekten (*) resultieren, haben die Kenntnisse zur zellulären Funktion der USH-Proteine und ihrer Interaktionspartner und damit über die pathogenen Mechanismen von USH erweitert. Dies bildet die Basis, um fundierte Therapiestrategien zu entwickeln.

Il presente lavoro ha come obiettivo quello di descrivere, alla scala dell'affioramento, 1) la sequenza di deformazione fragile: fratture di modo I (joints e vene) e di modo II-III (faglie) e 2) le modalità di circolazione dei fluidi in relazione alle strutture stesse. Da una parte i joint hanno agevolato la circolazione generando dei particolari fronti reattivi (diffusione e ossidazione). Dall'altra vene e faglie sembrano aver agito da barriera, disturbando la circolazione dei fluidi.

Meprin-α is a metalloprotease overexpressed in cancer cells, leading to the accumulation of this protease in a subset of colorectal tumors. The impact of increased meprin-α levels on tumor progression is not known. We investigated the effect of this protease on cell migration and angiogenesis in vitro and studied the expression of meprin-α mRNA, protein and proteolytic activity in primary tumors at progressive stages and in liver metastases of patients with colorectal cancer, as well as inhibitory activity towards meprin-α in sera of cancer patient as compared to healthy controls. We found that the hepatocyte growth factor (HGF)-induced migratory response of meprin-transfected epithelial cells was increased compared to wild-type cells in the presence of plasminogen, and that the angiogenic response in organ-cultured rat aortic explants was enhanced in the presence of exogenous human meprin-α. In patients, meprin-α mRNA was expressed in colonic adenomas, primary tumors UICC (International Union Against Cancer) stage I, II, III and IV, as well as in liver metastases. In contrast, the corresponding protein accumulated only in primary tumors and liver metastases, but not in adenomas. However, liver metastases lacked meprin-α activity despite increased expression of the corresponding protein, which correlated with inefficient zymogen activation. Sera from cancer patients exhibited reduced meprin-α inhibition compared to healthy controls. In conclusion, meprin-α activity is regulated differently in primary tumors and metastases, leading to high proteolytic activity in primary tumors and low activity in liver metastases. By virtue of its pro-migratory and pro-angiogenic activity, meprin-α may promote tumor progression in colorectal cancer.

Elevated rates of methicillin-resistant Staphylococcus aureus (MRSA) carriage have been reported in veterinary personnel, suggesting an occupational colonization risk. Hong Kong veterinary personnel (n = 150) were sampled for coagulase-positive staphylococci (CPS) nasal colonization. Risk factors for colonization were assessed by questionnaire. Isolates were identified and antibiotic susceptibility determined. All CPS isolates were investigated for mecA carriage, SCCmec type and PVL genes. Two subjects were colonized with methicillin-resistant CPS: one with MRSA (spa type t002 (CC5), SCCmec type II) and one with methicillin-resistant Staphylococcus pseudintermedius (MRSP) (MLST type ST71, SCCmec type II-III). MLST type ST71 S. pseudintermedius strain is the predominant MRSP clone circulating in dogs in Europe and in Hong Kong. The low MR-CPS colonization rate may be associated with low levels of large animal exposure or low rates of MRSA colonization of companion animals in Hong Kong. Colonization with non-aureus CPS, which may cause human infection, must also be considered in veterinary personnel.

OBJECTIVES: The aim of this study was to determine the phenotypic and genotypic resistance profiles of methicillin-resistant Staphylococcus pseudintermedius (MRSP) and to examine the clonal distribution in Europe and North America. METHODS: A total of 103 MRSP isolates from dogs isolated from several countries in Europe, the USA and Canada were characterized. Isolates were identified by PCR-restriction fragment length polymorphism (RFLP), antimicrobial susceptibility was determined by broth dilution or gradient diffusion, and antimicrobial resistance genes were detected using a microarray. Genetic diversity was assessed by multilocus sequence typing (MLST), PFGE and spa typing. Staphylococcal cassette chromosome mec (SCCmec) elements were characterized by multiplex PCR. RESULTS: Thirteen different sequence types (STs), 18 PFGE types and 8 spa types were detected. The hybrid SCCmec element II-III described in a MRSP isolate was present in 75 (72.8%) isolates. The remaining isolates either had SCCmec type III (n=2), IV (n=6), V (n=14) or VII-241 (n=4) or were non-typeable (n=2). The most common genotypes were ST71(MLST)-J(PFGE)-t02(spa)-II-III(SCCmec) (56.3%) and ST68-C-t06-V (12.6%). In addition to mecA-mediated beta-lactam resistance, isolates showed resistance to trimethoprim [dfr(G)] (90.3%), gentamicin/kanamycin [aac(6')-Ie-aph(2')-Ia] (88.3%), kanamycin [aph(3')-III] (90.3%), streptomycin [ant(6')-Ia] (90.3%), streptothricin (sat4) (90.3%), macrolides and/or lincosamides [erm(B), lnu(A)] (89.3%), fluoroquinolones (87.4%), tetracycline [tet(M) and/or tet(K)] (69.9%), chloramphenicol (cat(pC221)) (57.3%) and rifampicin (1.9%). CONCLUSIONS: Two major clonal MRSP lineages have disseminated in Europe (ST71-J-t02-II-III) and North America (ST68-C-t06-V). Regardless of their geographical or clonal origin, the isolates displayed resistance to the major classes of antibiotics used in veterinary medicine and thus infections caused by MRSP isolates represent a serious therapeutic challenge.
Cellular mechanisms of burst firing-mediated long-term depression in rat neocortical pyramidal cells

During wakefulness and sleep, neurons in the neocortex emit action potentials tonically or in rhythmic bursts, respectively. However, the role of synchronized discharge patterns is largely unknown. We have recently shown that pairings of excitatory postsynaptic potentials (EPSPs) and action potential bursts or single spikes lead to long-term depression (burst-LTD) or long-term potentiation, respectively. In this study, we elucidate the cellular mechanisms of burst-LTD and characterize its functional properties. Whole-cell patch-clamp recordings were obtained from layer V pyramidal cells in somatosensory cortex of juvenile rats in vitro and composite EPSPs and EPSCs were evoked extracellularly in layers II/III. Repetitive burst-pairings led to a long-lasting depression of EPSPs and EPSCs that was blocked by inhibitors of metabotropic glutamate group 1 receptors, phospholipase C, protein kinase C (PKC) and calcium release from the endoplasmic reticulum, and that required an intact machinery for endocytosis. Thus, burst-LTD is induced via a Ca2+- and phosphatidylinositol-dependent activation of PKC and expressed through phosphorylation-triggered endocytosis of AMPA receptors. Functionally, burst-LTD is inversely related to EPSP size and bursts dominate single spikes in determining the sign of synaptic plasticity. Thus burst-firing constitutes a signal by which coincident synaptic inputs are proportionally downsized. Overall, our data thus suggest a mechanism by which synaptic weights can be reconfigured during non-rapid eye movement sleep.

OBJECTIVES: To compare the outcome of arthroscopic lysis and lavage of TMJ with internal derangement of Wilkes stages II, III, IV, and V. STUDY DESIGN: Arthroscopic lysis and lavage was performed in 45 TMJ of 39 patients with internal derangement. The cases were divided into 4 groups corresponding to Wilkes stages II, III, IV, and V. Two parameters were compared pre- and postoperatively: pain and mouth opening. Statistical significance was determined using the chi(2) test. RESULTS: Overall success rate was 86.7% (Wilkes stage II 90.9%, Wilkes stage III 92.3%, Wilkes stage IV 84.6%, Wilkes stage V 75%). There were no statistically significant differences between the success rates for Wilkes stages II, III, IV, and V. CONCLUSION: Arthroscopic lysis and lavage should be performed as a standard operation for internal derangement of the TMJ after failure of conservative treatment in all Wilkes stages.

Scaphoid is one of the 8 carpal bones found adjacent to the thumb supported proximally by Radius bone. During the free fall, on outstretched hand, the impact load gets transferred to the scaphoid at its free anterior end. Unique arrangement of other carpal bones in the palm is also one of the reasons for the load to get transferred to scaphoid. About half of the total load acting upon carpal bone gets transferred to scaphoid at its distal pole. There are about 10 to 12 clinically observed fracture pattern in the scaphoid due to free fall. The aim of the study is to determine the orientation of the load, magnitude of the load and the corresponding fracture pattern. This study includes both static and dynamic finite element models validated by experiments. The scaphoid model has been prepared from CT scans of a 27 year old person. The 2D slices of the CT scans have been converted to 3D model by using MIMICS software. There are four cases of loading studied which are considered to occur clinically more frequently. In case (i) the load is applied at the posterior end at distal pole whereas in case (ii), (iii) and (iv), the load is applied at anterior end at different directions. The model is given a fixed boundary condition at the region which is supported by Radius bone during the impact. Same loading and boundary conditions have been used in both static and dynamic explicit finite element analysis. The site of fracture initiation and path of fracture propagation have been identified by using max principal stress / gradient and max principal strain / gradient criterion respectively in static and dynamic explicit finite element analysis. Static and dynamic impact experiments were performed on the polyurethane foam specimens to validate the finite element results. Experimental results such as load at fracture, site of fracture initiation and path of fracture propagation have been compared with the results of finite element analysis. Four different types of fracture patterns observed in clinical studies have been identified in this study.

Tenomodulin (Tnmd) is a member of a new family of type II transmembrane glycoproteins. It is predominantly expressed in tendons, ligaments, and eyes, whereas the only other family member, chondromodulin I (ChM-I), is highly expressed in cartilage and at lower levels in the eye and thymus. The C-terminal extracellular domains of both proteins were shown to modulate endothelial-cell proliferation and tube formation in vitro and in vivo. We analyzed Tnmd function in vivo and provide evidence that Tnmd is processed in vivo and that the proteolytically cleaved C-terminal domain can be found in tendon extracts. Loss of Tnmd expression in gene targeted mice abated tenocyte proliferation and led to a reduced tenocyte density. The deposited amounts of extracellular matrix proteins, including collagen types I, II, III, and VI and decorin, lumican, aggrecan, and matrilin-2, were not affected, but the calibers of collagen fibrils varied significantly and exhibited increased maximal diameters. Tnmd-deficient mice did not have changes in tendon vessel density, and mice lacking both Tnmd and ChM-I had normal retinal vascularization and neovascularization after oxygen-induced retinopathy. These results suggest that Tnmd is a regulator of tenocyte proliferation and is involved in collagen fibril maturation but do not confirm an in vivo involvement of Tnmd in angiogenesis.
