Stratum corneum (SC) desorption experiments have yielded higher calculated steady-state fluxes than those obtained by epidermal penetration studies. A possible explanation of this result is a variable diffusion or partition coefficient across the SC. We therefore developed the diffusion model for percutaneous penetration and desorption to study the effects of either a variable diffusion coefficient or variable partition coefficient in the SC over the diffusion path length. Steady-state flux, lag time, and mean desorption time were obtained from Laplace domain solutions. Numerical inversion of the Laplace domain solutions was used for simulations of solute concentration-distance and amount penetrated (desorbed)-time profiles. Diffusion and partition coefficients heterogeneity were examined using six different models. The effect of heterogeneity on predicted flux from desorption studies was compared with that obtained in permeation studies. Partition coefficient heterogeneity had a more profound effect on predicted fluxes than diffusion coefficient heterogeneity. Concentration-distance profiles show even larger dependence on heterogeneity, which is consistent with experimental tape-stripping data reported for clobetasol propionate and other solutes. The clobetasol propionate tape-stripping data were most consistent with the partition coefficient decreasing exponentially for half the SC and then becoming a constant for the remaining SC. Thin, piezoelectric circular plates are frequently used as active components in transducer and smart materials applications. This paper reports on the exact, explicit solution for the transient motion of a piezoelectric circular plate, built-in or simply supported on the edge and electrically grounded over the entire surface. Expressed by elementary Bessel functions and obtained via exact inverse Laplace transforms, the solution enables the efficient calculation of accurate system parameters. (C) 2004 Elsevier Ltd. The problem of regression under Gaussian assumptions is treated generally. The relationship between Bayesian prediction, regularization and smoothing is elucidated. The ideal regression is the posterior mean and its computation scales as O(n3), where n is the sample size. We show that the optimal m-dimensional linear model under a given prior is spanned by the first m eigenfunctions of a covariance operator, which is a trace-class operator. This is an infinite dimensional analogue of principal component analysis. The importance of Hilbert space methods to practical statistics is also discussed.

We consider the problem of assigning an input vector bfx to one of m classes by predicting P(c|bfx) for c = 1, ldots, m. For a two-class problem, the probability of class 1 given bfx is estimated by s(y(bfx)), where s(y) = 1/(1 + e-y). A Gaussian process prior is placed on y(bfx), and is combined with the training data to obtain predictions for new bfx points. We provide a Bayesian treatment, integrating over uncertainty in y and in the parameters that control the Gaussian process prior; the necessary integration over y is carried out using Laplace's approximation. The method is generalized to multi-class problems (m >2) using the softmax function. We demonstrate the effectiveness of the method on a number of datasets.
