We explore charge migration in DNA, advancing two distinct mechanisms of charge separation in a donor (d)–bridge ({Bj})–acceptor (a) system, where {Bj} = B1,B2, … , BN are the N-specific adjacent bases of B-DNA: (i) two-center unistep superexchange induced charge transfer, d*{Bj}a → d∓{Bj}a±, and (ii) multistep charge transport involves charge injection from d* (or d+) to {Bj}, charge hopping within {Bj}, and charge trapping by a. For off-resonance coupling, mechanism i prevails with the charge separation rate and yield exhibiting an exponential dependence ∝ exp(−βR) on the d-a distance (R). Resonance coupling results in mechanism ii with the charge separation lifetime τ ∝ Nη and yield Y ≃ (1 + δ̄ Nη)−1 exhibiting a weak (algebraic) N and distance dependence. The power parameter η is determined by charge hopping random walk. Energetic control of the charge migration mechanism is exerted by the energetics of the ion pair state d∓B1±B2 … BNa relative to the electronically excited donor doorway state d*B1B2 … BNa. The realization of charge separation via superexchange or hopping is determined by the base sequence within the bridge. Our energetic–dynamic relations, in conjunction with the energetic data for d*/d− and for B/B+, determine the realization of the two distinct mechanisms in different hole donor systems, establishing the conditions for “chemistry at a distance” after charge transport in DNA. The energetic control of the charge migration mechanisms attained by the sequence specificity of the bridge is universal for large molecular-scale systems, for proteins, and for DNA.

Functional expression of the multidrug resistance protein P-glycoprotein (P-gp) in Escherichia coli is providing an appropriate system for structure/function studies and might provide an invaluable tool to screen potential P-gp substrates and inhibitors. The major problem encountered in such studies, however, is the impermeability of the outer membrane of Gram-negative bacteria, which protects microorganisms against the cytotoxic effects of many lipophilic cancer drugs and blocks accessibility of P-gp reversal agents. In the present study we have constructed, by mutagenesis, a "leaky" (containing a permeable outer membrane) strain of E. coli, which is significantly more susceptible to the toxic effect of known P-gp substrates and cytotoxic agents. Expression of mouse Mdr1 in the mutant confers cross-resistance to daunomycin, quinidine, chloroquine, rhodamine 6G, and puromycin. Most importantly, reserpine and doxorubicin completely abolish Mdr1-mediated rhodamine resistance. The results provide strong support for previous observations, suggesting that Mdr1 can be expressed functionally in E. coli and indicate that the leaky mutant will be useful for further structure/function studies of the heterologously expressed eukaryotic drug efflux protein.

La necesidad que da origen al presente proyecto se relaciona con la ausencia de un tratamiento de la cuestión de la ciudadanía que haga interactuar distintos enfoques filosóficos -el principal indicador de esta carencia es la ausencia de producciones académicas que den cuenta de la complejidad que adquiere la temática si se la aborda desde los problemas que nos proporcionan otras perspectivas filosóficas y políticas-. En este sentido, el problema general del proyecto apunta a hacer discutir diferentes abordajes conceptuales para pensar la ciudadanía. Específicamente, trabajamos a partir de dos enfoques: 1) la discusión entre liberales y comunitaristas y sus actuales derivas y 2) la cuestión de la biopolítica y su relación con la temática de la ciudadanía. Se procura revisar la discusión liberales-comunitaristas propia de las ciencias políticas, interpelándola a partir de conceptos como los de dominación, relaciones de poder, control sobre la vida, disciplina, entre otros provenientes de la filosofía práctica, la teoría social, las ciencias de la educación, etc. Nuestra investigación parte de la hipótesis de que hacer discutir las problemáticas que se disputan liberales-comunitaristas, con la Teoría Crítica de la Escuela de Frankfurt y con los recientes fenómenos biopolíticos, permite un abordaje que atiende a la efectiva complejidad de las prácticas de ciudadanía en nuestra vida en común en las sociedades democráticas contemporáneas. Esto permitirá complejizar los presupuestos con los que tradicionalmente se ha pensado la ciudadanía, a partir sobre todo de los fenómenos socio-políticos más recientes, como los nuevos movimientos sociales, las discusiones acerca de la legislación del aborto y la eutanasia, los esfuerzos de los estados nacionales por incrementar medidas de seguridad que van desde la imposición de fuertes barreras a la inmigración hasta la realización de guerras preventivas. Entendemos que estos, entre otros fenómenos, desafían la hermenéutica tradicional sobre la ciudadanía. Es de esta manera que se buscará comprender los límites y alcances de las ideas de ciudadanía, entendiéndola como un concepto histórico formador de subjetividades. La metodología se basa en una perspectiva interdisciplinaria que proporciona las herramientas para un análisis conceptual de la temática de la ciudadanía. Esta metodología está orientada al desarrollo de un marco teórico que resulte productivo para investigaciones de campo en las ciencias sociales, así como también para la elaboración de un material bibliográfico destinado a docentes abocados a la ciudadanía. Otro de los propósitos fundamentales es el de formar una red entre diferentes equipos de investigación a nivel nacional a partir de las “I Jornadas Nacionales sobre Ciudadanía” y de la organización de un seminario especializado con un profesor visitante.

The age hardening, stress corrosion cracking (SCC) and hydrogen embrittlement (HE) of an Al-Zn-Mg-Cu 7175 alloy were investigated experimentally. There were two peak-aged states during ageing. For ageing at 413 K, the strength of the second peak-aged state was slightly higher than that of the first one, whereas the SCC susceptibility was lower, indicating that it is possible to heat treat 7175 to high strength and simultaneously to have high SCC resistance. The SCC susceptibility increased with increasing Mg segregation at the grain boundaries. Hydrogen embrittlement (HE) increased with increased hydrogen charging and decreased with increasing ageing time for the same hydrogen charging conditions. Computer simulations were carried out of (a) the Mg grain boundary segregation using the embedded atom method and (b) the effect of Mg and H segregation on the grain boundary strength using a quasi-chemical approach. The simulations showed that (a) Mg grain boundary segregation in Al-Zn-Mg-Cu alloys is spontaneous, (b) Mg segregation decreases the grain boundary strength, and (c) H embrittles the grain boundary more seriously than does Mg. Therefore, the SCC mechanism of Al-Zn-Mg Cu alloys is attributed to the combination of HE and Mg segregation induced grain boundary embrittlement. (C) 2004 Acta Materialia Inc. Grass pollen is an important risk factor for allergic rhinitis and asthma in Australia and is the most prevalent pollen component of the aerospora of Brisbane, accounting for 71.6% of the annual airborne pollen load. A 5-year (June 1994-May 1999) monitoring program shows the grass pollen season to occur during the summer and autumn months (December-April), however the timing of onset and intensity of the season vary from year to year. During the pollen season, Poaceae counts exceeding 30 grains m(-3) were recorded on 244 days and coincided with maximum temperatures of 28.1 +/- 2.0degreesC. In this study, statistical associations between atmospheric grass pollen loads and several weather parameters, including maximum temperature, minimum temperature and precipitation, were investigated. Spearman's correlation analysis demonstrated that daily grass pollen counts were positively associated (P < 0.0001) with maximum and minimum temperature during each sampling year. Precipitation, although considered a less important daily factor (P < 0.05), was observed to remove pollen grains from the atmosphere during significant periods of rainfall. This study provides the first insight into the influence of meteorological variables, in particular temperature, on atmospheric Poaceae pollen counts in Brisbane. An awareness of these associations is critical for the prevention and management of allergy and asthma for atopic individuals within this region.
