Dichloromethane extracts from twigs of Virola surinamensis (Myristicaceae) showed in vitro trypanosomicidal activity against trypomastigote form of Trypanosoma cruzi. The extract, fractionated by preparative circular chromatography and preparative high-performance liquid chromatography yielded two steroids, two lignans, five flavonoids, and one polyketide. Among the isolated compounds, the lignans presented the highest trypanosomicidal activity.

Two novel acylated flavonol glycosides, along with iridoids, triterpenes, steroids and alpha-tocopherolquinone, were isolated from the leaves of Alibertia sessilis (Rubiaceae). The determination of the structures of the new compounds was based mainly on H-1- and C-13-NMR.

Chromatographic fractionation of the dichloromethane extract from roots of Virola surinamensis yielded two new natural products, 3-epi-juruenolide C and 2'-hydroxy-7,4'-dimethoxyisoflavone, as well as various known steroids, lignans, isoflavones, flavonoids and diarylpropanes. of these, alpha,2'-dihydroxy-4,3'-dimethoxydihydrochal biochanin A and 2'-hydroxy-7,4'-dimethoxyisoflavone displayed antifungal activity against Cladosporium cladosporioides at a minimum amount of 5 mu g, whereas 7-hydroxyflavanone and 7-hydroxy-4'-methoxyisoflavone exhibited an antifungal activity 10-fold higher than the positive control Nystatin. The influence of fetal calf serum alone (FCS) or associated with proestrous (FCS+PCS), estrous (FCS+ECS) or metaestrous (FCS+MCS) cow serum added to the culture medium and of the steroids produced by co-cultured granulosa cells were evaluated in terms of the in vitro maturation (TVM) and fertilization (IVF) of bovine oocytes. Supplementation of the medium with FCS+ECS and FCS+MCS resulted in higher proportions of oocytes that reached metaphase II (96.0% and 93.3%, respectively) and in higher proportions of embryos that reached the four- and eight-cell/morula stages (51.9% and 65.6%, respectively), whereas the supplementation with FCS and FCS+PCS resulted in only 79.2% and 67.5%, respectively, of matured oocytes and 26.7% and 34.3%, respectively, of cleaved embryos. These findings show that the best IVM and IVF were obtained at lower concentrations of estradiol produced by co-cultured granulosa cells (supplementation with FCS+ECS: 10.3 ng/ml and FCS+MCS: 2.1 ng/ml), whereas the worst-results in IVM and IVF occurred at higher concentrations of estradiol that were obtained with FCS (33.1 ng/ml) and FCS+PCS (19.9 ng/ml) supplementation. These data suggest an inhibitory effect of estradiol on resumption of oocyte meiosis in vitro.

From stems of Aristolochia chamissonis Duch., five new diterpenes, one rearranged (4 --> 2)-abeo-clerodane and four ent-clerodanes, were isolated. Their structures were determined to be (+)-(4 --> 2)-abeo-kolavelool-3-oic acid, (-)-13-epi-2-oxokolavelool, (-)-2 beta-hydroxykolavelool, (-)-2 beta-hydroperoxykolavelool and (+)- 13-epi-2 alpha-hydroxykolavelool. A. suite of 10 different marine evaporitic oil samples from Sergipe-Alagoas Basin, Brazil was studied for its biomarker content, in particular its acidic constituents. The oils showed different molecular distributions and relative abundances of n-alkanoic, isoprenoid and hopanoic acids. The observed differences were assigned to the incorporation of immature organic matter in the oils and fractionation along the migration pathway. The diagenetic precursor functionality (alcohol/ether or acid) was proposed based on the comparison of the relative abundances of the neutral and acidic biomarkers (hopanoids, isoprenoids, alkyl-steranes, monoaromatic alkyl-steroids). In the acidic fraction, 3 series of steroid-alkanoic acids and monoaromatic steroid-alkanoic acids (steroid-methanoic, ethanoic and propanoic acids and monoaromatic steroid-methanoic, ethanoic and propanoic acids) were detected, while in the neutral fraction only 2 series of each corresponding class could be observed (methyl and ethyl-steranes and monoaromatic methyl and ethyl-steroids). These carbon shifts suggest that decarboxylation is an important process in the formation of the alkyrsteranes and monoaromatic alkyl-steroids, and we infer that carboxylic acids are the diagenetic precursors of these classes of compounds. When alcohol or ether are the diagenetic precursors (isoprenoids and hopanoids), no significant differences in the molecular distributions between neutral and acidic fractions were observed. Steroids hormones modify the hematological features of homozygous sickle cell disease, including the levels of fetal hemoglobin. We used semi-quantitative RT-PCR analysis of GATA-1, GATA-2, NF-E2, and gamma-globin mRNA levels in a two-phase liquid culture system of human adult erythroid cells in order to assay the effect of progesterone upon gene expression. The levels of expression of GATA-1 and gamma-globin mRNA were significantly increased in cells treated with progesterone compared to untreated cells (1.7- to 2.0-fold). Progesterone treatment did not produce any stimulatory effect upon GATA-2 and NF-E2 mRNA expression. Differences in the synthesis of HbF protein could not be detected by flow cytometry, although we observed a small difference in mean intensity fluorescence between cells treated and cells untreated with progesterone on days 7 and 9. Using anti-transferrin receptor and anti-glycophorin A antibodies, we verified that addition of progesterone did not cause any change in erythroid proliferation and differentiation. In conclusion, it is possible that the increased expression of gamma-globin mRNA after progesterone treatment observed in this study may be related to the increased GATA-1 mRNA expression. Interactions of the steroid receptors with the basal transcriptional machinery and with transcription factors might mediate their transcriptional effects. (C) 2002 Elsevier B.V. (USA).

The effects of gonadectomy on the secretion of prolactin, LH, TSH, and thyroxine were investigated. Blood serum hormone concentrations were analysed before and at 20, 120, and 180 min after a single iv TRH injection in each of eight healthy intact and castrated male beagle dogs before (control) and after 4-week treatment with the dopamine-2 receptor agonist cabergoline. Under control conditions the mean prolactin, TSH, and thyroxine concentrations were similar in intact and gonadectomised dogs, and administration of TRH provoked a significant (p < 0.01) increase in concentrations of the three hormones. The overall inhibitory effect of cabergoline treatment on prolactin secretion was more pronounced in the castrated dogs compared with the intact group. Cabergoline significantly suppressed the TRH-induced prolactin increase in each group (p < 0.01). Corresponding TRH-stimulated TSH concentrations were not affected by cabergoline. In the gonadectomised dogs, thyroxine concentrations before and at 120 and 180 min after TRH injection were significantly lower than under control conditions. LH concentrations were always higher (p < 0.01) in gonadectomised dogs compared with the intact dogs, but appeared to be affected neither by TRH nor by cabergoline administration. It can thus be concluded from the results, that gonadectomy does not result in hyperprolactinaemia in male dogs, while LH concentrations are significantly increased due to missing androgen feedback. Thyroid function remains unaffected by gonadectomy. Testicular steroids appear to interact with central dopaminergic and probably other neuroendocrine mechanisms regulating the secretion of prolactin, TSH, and thyroxine. Thus, long-term dopamine-2 receptor agonistic treatment may lead to a hypothyroid condition in castrated male dogs. (c) 2009 Elsevier B.V. All rights reserved.

The effects of androgenic deprivation induced by castration on the norepinephrine contractile response of vas deferens from rats, which have been submitted to acute swimming-stress were determined. Acute swimming-stress led to subsensitivity to norepinephrine in vas deferens excised from intact rats. Similarly, castration also induced subsensitivity to norepinephrine, but no further subsensitivity occurred in organs from castrated rats submitted to acute stress. The results indicate a different response to norepinephrine in terms of relative responsiveness ratio, when vas deferens was excised from castrated rats or castrated rats submitted to acute stress. It is suggested that androgenic steroids modulate the recovery of homeostasis in rat vas deferens during acute stress, and that this effect may involve mechanisms that affect both the sensitivity of adrenergic receptors and the system of neuronal uptake of catecholamines.

PURPOSE: To evaluate the therapeutic complications due to the use of immunosupressors in patients with nephropathy. METHODS: 76 patients who had used steroids and cyclophosphamide were retrospectively studied. The cases were divided into three groups: G1= 15 patients with Systemic Lupus Erythematosus without renal lesion; G2= 33 patients with lupus nephritis and G3= 28 patients with nephrotic syndrome owing to idiopathic glomerulopathy. RESULTS: There were no differences related to time of follow up (G1= 42.4 +/- 51, G2= 52.3 +/- 51, G3= 41.8 +/- 47.8 months), total used dosage of steroids (G1= 20, G2= 28, G3= 16 grams) and time of drug use (G1= 20, G2= 26, G3= 14.5 months). About cyclophosphamide use, there was no difference in the percentage of patients who used it (13% in G1, 51% in G2, 28% in G3), but the patients from G1 received lower total dosage than those from G2 (p<0.05). Cushingoid appearance, epigastric distress, psychiatric disorders, diabetes mellitus and ocular alterations occurred in all the three groups, with no statistically significant differences. The infections complications, those considered more severe clinically, were more frequent in G2 (G1= 6%, G2= 15%, G3= 0% - p<0.05), the same occurring with the deaths (7% in G1, 30% in G2, 0% in G3 - p<0.05). CONCLUSION: In patients with lupus nephritis there were more infections complications owing to prolonged immunosuppresion what may indicate a severity marker of this type of lesion.

Wilbrandia ebracteata (Cogn.) Cogn. is a medicinal plant belonging to the Cucurbitaceae family used popularly as an antiulcer and analgesic medicine. The hydromethanol extract of leaves was investigated to determine its anti-ulcerogenic (ethanol and indomethacin induced gastric damage) and analgesic (writhing and tail-flick tests) activities in mice (efficacy), its acute toxicity (safety), and its phytochemistry (quality control). Oral administration of leaf extract at a dose of 1000 mg/kg body wt. significantly reduced 73.3% of the total area of lesion in ethanol-induced gastric damage, but was inactive in an indomethacin-induced gastric damage test. The hydromethanol extract was also inactive in both analgesic tests. Oral administration of the leaf extract did not produce mortality in mice, while the LD50 value of the roots was 22.10 mg/kg body wt. in female mice and 58.31 mg/kg body wt. in male mice. Leaves of W. ebracteata reacted positively for steroids, flavonols, flavanones, saponins, tannins and xanthones and negative for other compounds, including cucurbitacins. Leaf extract of W. ebracteata was active as an anti-ulcerogenic, probably through increasing gastric defensive factors, and flavonoids might be the main constituent responsible for this activity.

BACKGROUND AND OBJECTIVES: Before epídural steroids were used in chronic lumbar pain, subarachnoid injection of these agents was the treatment of choice. Although still preconized by some authors, this technique may lead to severe complications with neurological sequelae. This report aimed at describing a case of accidental subarachnoid injection of steroid associated to local anesthetics during epidural puncture to treat lumbar pain. CASE REPORT: Male patient, 46 years old, followed byneuro-surgery for presenting right sciatic pain for 9 month, refractory to clinical treatment due to L 4-L 5 disk protrusion confirmed by CT scan, without neurological deficit. Epidural puncture for pain treatment was performed in L 4-L 5 with 17G needle and 10 mL solution were injected containing 4 mL of 0.25% bupivacaine, 80 mg methylprednisolone and 4 mL of 0.9% saline. Although there has not been CSF reflux, 5 minutes after injection there were sensory block in T 4 and motor block in T 6, associated to blood pressure and heart rate decrease. CONCLUSIONS: Accidental subarachnoid injections with the association of steroids for pain relief may cause adverse effects. There are several risks, varying from mild transient symptoms to nervous injuries, including spinal cord injuries. Our patient had no sequelae from the accidental subarachnoid injection, probably because it has been a single injection.
