A diversificação do uso dos produtos de cortiça exige uma adaptação constante dos processos produtivos utilizados para a transformação deste produto. Este trabalho tem como principal intuito a idealização e investigação de um novo processo de fabrico adaptado a um tipo muito particular de produto, os aglomerados puros de cortiça. Estes, não recorrem a qualquer tipo de aglutinante para assegurar a sua ligação, apenas as resinas contidas na própria cortiça são utilizadas para esse efeito. Analisando os processos de fabrico atuais, verifica-se que, a conceção de geometrias complexas com aglomerados puros de cortiça, só é possível com recurso à maquinação. Este processo gera desperdício e tem uma série de processos que o antecedem, destacando-se a aglomeração prévia numa forma mais simples (bloco paralelepípedo ou cilindro) e o corte numa pré forma. A ideia de se executar uma peça complexa sem recurso à maquinação é o objetivo primordial deste trabalho. Para isso foi construído um molde, com todos os mecanismos auxiliares necessários, à semelhança do que já se faz noutro tipo de indústrias, nomeadamente, do vidro e do plástico. Esse mesmo molde foi submetido a uma série de ensaios para determinação dos parâmetros de enchimento e moldação, tendo-se chegado a uma formulação que garante características atrativas na peça final. A aglutinação do granulado de cortiça revelou-se bastante dependente de três parâmetros: a temperatura aplicada, a humidade do granulado e o grau de compactação aplicado. A industrialização do processo revelou-se também plausível de execução, sendo apenas necessários alguns melhoramentos. A utilização deste tipo de processo revela-se, portanto, atrativa para grandes séries de peças, conseguindo-se elevada reprodutibilidade das mesmas com apenas um processo, sem necessidade de procedimentos adicionais de acabamento.

Durante os últimos anos tem-se assistido a um crescimento exponencial da utilização dos dispositivos móveis. Atualmente, a presença destes dispositivos está tão consolidada na sociedade em que nos encontramos que é praticamente impossível estar num espaço público sem encontrar uma pessoa a utilizar um exemplar deste tipo de dispositivos, na forma de um Smartphone ou um Tablet. Nesse sentido, um grande número de organizações de desenvolvimento de software tem vindo a apostar na criação de aplicações para servir este mercado com enorme potencial. De forma a iniciar o desenvolvimento aplicacional nesta área, cabe às organizações escolherem a metodologia de desenvolvimento que mais se adapta às suas necessidades e ao contexto que desejam explorar. Por vezes, a abordagem adotada acaba por ser tornar inadaptada a longo prazo, podendo as aplicações começar a apresentar níveis de desempenho e problemas comportamentais indesejados, de acordo com as alterações realizadas e as necessidades evidenciadas no seu processo de evolução, sendo necessário explorar outras alternativas metodológicas. No contexto apresentado surge a temática da presente dissertação, que se propõe à criação de um plano de migração tecnológica genérico direcionado a um conjunto aplicações móveis em estudo, desenvolvidas através de uma metodologia híbrida, adaptada ao desenvolvimento de aplicações compatíveis com as diversas plataformas móveis. O plano devia então ser construído com o intuito de efetuar a migração das aplicações para uma metodologia nativa, adaptada ao desenvolvimento de aplicações móveis específicas a uma determinada plataforma móvel, que no presente caso seria sob a plataforma Android. No decorrer do trabalho realizado foram desenvolvidos mecanismos e componentes genéricos que permitiram implementar um módulo de migração responsável por construir um projeto nativo base de Android através de um projeto híbrido, construído segundo as suas tecnologias específicas. Finalizada a implementação, foram realizados testes comparativos entre a aplicação alvo de migração e o protótipo nativo conseguido e foi apresentada uma série de resultados que permitiram concluir a adequação da metodologia nativa para ultrapassar a problemática evidenciada. Além disso, foi também possível concluir que o sistema de migração genérico resultante do trabalho desta dissertação permite reduzir significativamente o tempo de desenvolvimento necessário em migrações de outras aplicações que se enquadrem na mesma problemática.

To determine whether the slope of a maximal bronchial challenge test (in which FEV1 falls by over 50%) could be extrapolated from a standard bronchial challenge test (in which FEV1 falls up to 20%), 14 asthmatic children performed a single maximal bronchial challenge test with methacholin(dose range: 0.097–30.08 umol) by the dosimeter method. Maximal dose-response curves were included according to the following criteria: (1) at least one more dose beyond a FEV1 ù 20%; and (2) a MFEV1 ù 50%. PD20 FEV1 was calculated, and the slopes of the early part of the dose-response curve (standard dose-response slopes) and of the entire curve (maximal dose-response slopes) were calculated by two methods: the two-point slope (DRR) and the least squares method (LSS) in % FEV1 × umol−1. Maximal dose-response slopes were compared with the corresponding standard dose-response slopes by a paired Student’s t test after logarithmic transformation of the data; the goodness of fit of the LSS was also determined. Maximal dose-response slopes were significantly different (p < 0.0001) from those calculated on the early part of the curve: DRR20% (91.2 ± 2.7 FEV1% z umol−1)was 2.88 times higher than DRR50% (31.6 ± 3.4 DFEV1% z umol−1), and the LSS20% (89.1 ± 2.8% FEV1 z umol−1) was 3.10 times higher than LSS 50% (28.8 ± 1.5%FEV1 z umol−1). The goodness of fit of LSS 50% was significant in all cases, whereas LSS 20% failed to be significant in one. These results suggest that maximal dose-response slopes cannot be predicted from the data of standard bronchial challenge tests.

SUMMARYAIDS-related cryptococcal meningitis continues to cause a substantial burden of death in low and middle income countries. The diagnostic use for detection of cryptococcal capsular polysaccharide antigen (CrAg) in serum and cerebrospinal fluid by latex agglutination test (CrAg-latex) or enzyme-linked immunoassay (EIA) has been available for over decades. Better diagnostics in asymptomatic and symptomatic phases of cryptococcosis are key components to reduce mortality. Recently, the cryptococcal antigen lateral flow assay (CrAg LFA) was included in the armamentarium for diagnosis. Unlike the other tests, the CrAg LFA is a dipstick immunochromatographic assay, in a format similar to the home pregnancy test, and requires little or no lab infrastructure. This test meets all of the World Health Organization ASSURED criteria (Affordable, Sensitive, Specific, User friendly, Rapid/robust, Equipment-free, and Delivered). CrAg LFA in serum, plasma, whole blood, or cerebrospinal fluid is useful for the diagnosis of disease caused by Cryptococcusspecies. The CrAg LFA has better analytical sensitivity for C. gattii than CrAg-latex or EIA. Prevention of cryptococcal disease is new application of CrAg LFA via screening of blood for subclinical infection in asymptomatic HIV-infected persons with CD4 counts < 100 cells/mL who are not receiving effective antiretroviral therapy. CrAg screening of leftover plasma specimens after CD4 testing can identify persons with asymptomatic infection who urgently require pre-emptive fluconazole, who will otherwise progress to symptomatic infection and/or die.

After vaccination with the live PF strain of Trypanosoma cruzi, 194 blood cultures were performed in 143 mice, 9 dogs, 5 Cebus monkeys and 7 human subjects. Some of these blood cultures were simultaneously done with xenodiagnosis, subinoculation in baby mice and/or culture of viscerae. The trypanosomes isolated from the few positive cases (6,1%) were incapable of infecting baby mice were considered as cases of immunotolerance. All the other tests were negative.

RESUMO - Introdução: Segundo a Organização Mundial de Saúde, a evolução verificada nos últimos anos no consumo de álcool e tabaco e outras substâncias psicoactivas está a aumentar rapidamente e a contribuir de maneira importante para a carga das doenças em todo o mundo. A nível militar e nos elementos destacados para as missões de apoio à paz, é pertinente conhecer e caracterizar os padrões de consumo de álcool e tabaco a que estão sujeitos nestes contextos específicos de modo a compreender esta realidade e a sua importância e a poder intervir face às suas consequências. De ainda referir que existe escassez de estudos nesta área em Portugal. Objectivo: O objectivo deste estudo consiste em caracterizar os padrões de consumo de álcool e de tabaco dos militares das Forças Nacionais Destacadas, em missão de apoio à paz, no pré-deslocamento ainda em Portugal, no deslocamento em Teatro de Operações e no pós-deslocamento em território nacional. Metodologia: Apresenta-se o delineamento do estudo e o tipo de estudo que é longitudinal e analítico, a metodologia utilizada, bem como, as hipóteses colocadas, consolidadas na revisão de literatura efectuada previamente; faz-se a caracterização das variáveis sócio demográficas e instrumentais utilizadas. São definidos os critérios de inclusão e exclusão assim como aspectos éticos necessários. Definem-se ainda os testes estatísticos considerados apropriados para este tipo de estudo. Simulação de resultados, discussão e conclusões: Faz-se uma simulação do modo de apresentação dos resultados de acordo com os objectivos e hipóteses traçados tendo em conta a aplicação estatística considerada mais conveniente. Quanto ao ponto de discussão e conclusões dever-se-á após a realização do estudo comparar os resultados obtidos com os resultados de outros estudos análogos e no mesmo âmbito e tecer algumas considerações pertinentes sobre os mesmos. Assim seria importante sugerir novas orientações para outros estudos tendo em conta os resultados mais significativos que possam ser apurados e ainda ser reflectido e até apontado algumas respostas em termos de intervenção neste contexto.

BACKGROUND: The baseline susceptibility of primary HIV-2 to maraviroc (MVC) and other entry inhibitors is currently unknown. METHODS: The susceptibility of 19 HIV-2 isolates obtained from asymptomatic and AIDS patients and seven HIV-1 clinical isolates to the fusion inhibitors enfuvirtide (ENF) and T-1249, and to the coreceptor antagonists AMD3100, TAK-779 and MVC, was measured using a TZM-bl cell-based assay. The 50% inhibitory concentration (IC(50)), 90% inhibitory concentration (IC(90)) and dose-response curve slopes were determined for each drug. RESULTS: ENF and T-1249 were significantly less active on HIV-2 than on HIV-1 (211- and 2-fold, respectively). AMD3100 and TAK-779 inhibited HIV-2 and HIV-1 CXCR4 tropic (X4) and CCR5 tropic (R5) variants with similar IC(50) and IC(90) values. MVC, however, inhibited the replication of R5 HIV-2 variants with significantly higher IC(90) values (42.7 versus 9.7 nM; P<0.0001) and lower slope values (0.7 versus 1.3; P<0.0001) than HIV-1. HIV-2 R5 variants derived from AIDS patients were significantly less sensitive to MVC than variants from asymptomatic patients, this being inversely correlated with the absolute number of CD4(+) T-cells. CONCLUSIONS: T-1249 is a potent inhibitor of HIV-2 replication indicating that new fusion inhibitors might be useful to treat HIV-2 infection. Coreceptor antagonists TAK-779 and AMD3100 are also potent inhibitors of HIV-2 replication. The reduced sensitivity of R5 variants to MVC, especially in severely immunodeficient patients, indicates that the treatment of HIV-2-infected patients with MVC might require higher dosages than those used in HIV-1 patients, and should be adjusted to the disease stage.

BACKGROUND: Allergy to natural rubber latex is a well-recognized health problem, especially among health care workers and patients with spina bifida. Despite latex sensitization being acquired in health institutions in both health care workers and patients with spina bifida, differences in allergen sensitization profiles have been described between these two risk groups. OBJECTIVE: To investigate the in vivo reactivity of health care workers and patients with spina bifida to extracts of internal and external surfaces of latex gloves and also to specific extracts enriched in major allergens for these risk groups. METHODS: Gloves from different manufacturers were used for protein extraction, and salt precipitation and hydrophobic interaction chromatography (HIC) were applied to obtain the enriched latex extracts. The major latex allergens were quantified by an enzyme immunoassay. The extracts obtained were tested in 14 volunteers using skin prick tests (SPT). RESULTS: Latex glove extracts enriched in the hydrophobic allergens that are most often seen in patients with spina bifida were obtained by selective precipitation, whereas HIC produced extracts enriched in the hydrophilic allergens commonly found in health care workers. The health care workers had positive SPTs to glove extracts from internal surfaces and to the hydrophilic allergen-enriched extracts. By contrast, patients with spina bifida had larger skin reactions both to external glove extracts and to the extracts enriched with the hydrophobic major allergens for this risk group. Despite the protein concentration of these extracts being less than half the concentration of the commercial extract, the weal-and-flare reactions were of similar magnitude. CONCLUSION: Using novel latex extracts, our study showed a different in vivo reactivity pattern in health care workers and in patients with spina bifida to extracts of the internal and external surfaces of gloves, which suggests that sensitization may occur by different routes of exposure, and that this influences the allergen reactivity profiles of these risk groups

Clinical and serological follow-up of 7 patients submitted to renal transplantation and presenting positive serological reactions to Chagas 'disease before immunossupression did not show significant changes in indirect immunofluorescence and complement fixation titres for Chagas ' disease, or signs and symptoms indicating exacerbation of the disease during follow- up. In addition, 18 of 66 recipients of renal transplants considered to be non-chagasic before immunosuppression showed at least one positive result to the indirect immunofluorescence test for Chagas ' disease during the study period. The results suggest that the immunosuppression State induced in chagasic patients submitted to renal transplant did notpromoted exacerbation of the chronic infection in these patients and not interfere with the serological response of chronic chagasics, thus permitting the use of these serologic reactions for diagnostic purposes in these cases. However, the positive results ofthe indirect immunofluorescence test in non- chagasic patients indicate the needforjudicious interpretation ofthe indirect immunofluorescence test for the diagnosis of Chagas' disease in renal transplanted patients.

The liver abscess is the most frequent extraintestinal complication of intestinal amoebiasis: its diagnosis is suggested by the clinical picture but it must be confirmed by paraclinic tests. Themost stringent diagnosis requires identification of E. histolytica. But this is possible only in a few cases. Serological tests greatly improve the diagnosis of this severe complication of amoebiasis. We compared the Enzyme Linfed Immunosorbent Assay and the Counterimmunoeletrophoresis techniques. Both techniques were used to detect amoebic antibodies in 50 control patients, 30 patients with liver abscess and 30 patients with intestinal amoebiasis. All the sera from control patients gave negative results iin both techniques. When analysing the sera from patients with intestinal amoebiasis, 10% of them were positive by ELISA but non by CIE. The sera of patients with liver abscess, we found that 90% were positive by the ELISA method and 66.6% by the CIE technique. In patients with amoebic liver abscess, the results showed that the ELISA was more sensitive than the CIE, as it presented a higher sensitivity (100%) than that of the CIE technique (66%).

The human lymphotropic viruses type I (HTLV-I) and type II (HTLV-II) are members of a group of mammalian retroviruses with similar biological properties, and blood transfusion is an important route of transmission. HTLV-I is endemic in a number of different geographical areas and is associated with several clinical disorders. HTLV-II is endemic in several Indian groups of the Americas and intravenous drug abusers in North and South America, Europe and Southeast Asia. During the year of 1995, all blood donors tested positive to HTLV-I/II in the State Blood Bank (HEMOPA), were directed to a physician and to the Virus Laboratory at the Universidade Federal do Pará for counselling and laboratory diagnosis confirmation. Thirty-five sera were tested by an enzyme immune assay, and a Western blot that discriminates HTLV-I and HTLV-II infection. Two HTLV-II positive samples were submitted to PCR analysis of pX and env genomic region, and confirmed to be of subtype IIa. This is the first detection in Belém of the presence of HTLV-IIa infection among blood donors. This result emphasizes that HTLV-II is also present in urban areas of the Amazon region of Brazil and highlights the need to include screening tests that are capable to detect antibodies for both types of HTLV.

Minimum parking requirements are the norm for urban and suburban development in the United States (Davidson and Dolnick (2002)). The justification for parking space requirements is that overflow parking will occupy nearby street or off-street parking. Shoup (1999) and Willson (1995) provides cases where there is reason to believe that parking space requirements have forced parcel developers to place more parking than they would in the absence of parking requirements. If the effect of parking minimums is to significantly increase the land area devoted to parking, then the increase in impervious surfaces would likely cause water quality degradation, increased flooding, and decreased groundwater recharge. However, to our knowledge the existing literature does not test the effect of parking minimums on the amount of lot space devoted to parking beyond a few case studies. This paper tests the hypothesis that parking space requirements cause an oversupply of parking by examining the implicit marginal value of land allocated to parking spaces. This is an indirect test of the effects of parking requirements that is similar to Glaeser and Gyourko (2003). A simple theoretical model shows that the marginal value of additional parking to the sale price should be equal to the cost of land plus the cost of parking construction. We estimate the marginal values of parking and lot area with spatial methods using a large data set from the Los Angeles area non-residential property sales and find that for most of the property types the marginal value of parking is significantly below that of the parcel area. This evidence supports the contention that minimum parking requirements significantly increase the amount of parcel area devoted to parking.
