For an embedded singly periodic minimal surface (M) over tilde with genus rho >= 4 and annular ends, some weak symmetry hypotheses imply its congruence with one of the Hoffman-Wohlgemuth examples. We give a very geometrical proof of this fact, along which they come out many valuable clues for the understanding of these surfaces.

The activation parameters for the thermal decomposition of 13 acridinium-substituted 1,2-dioxetanes, bearing an aromatic moiety, were determined and their chemiluminescence emission quantum yields estimated, utilizing in situ photosensitized 1,2-dioxetane generation and observation of its thermal decomposition kinetics, without isolation of these highly unstable cyclic peroxides. Decomposition rate constants show linear free-energy correlation for electron-withdrawing substituents, with a Hammett reaction constant of rho = 1.3 +/- 0.1, indicating the occurrence of an intramolecular electron transfer from the acridinium moiety to the 1,2-dioxetane ring, as postulated by the intramolecular chemically initiated electron exchange luminescence (CIEEL) mechanism. Emission quantum yield behavior can also be rationalized on the basis of the intramolecular CIEEL mechanism, additionally evidencing its occurrence in this transformation. Both relations constitute the first experimental evidence for the occurrence of the postulated intramolecular electron transfer in the catalyzed and induced decomposition of properly substituted 1,2-dioxetanes.

We previously demonstrated that Bis[(2-oxindol-3-ylimino)-2-(2-aminoethyl) pyridine-N, N`] copper(II) [Cu(isaepy)(2)] was an efficient inducer of the apoptotic mitochondrial pathway. Here, we deeply dissect the mechanisms underlying the ability of Cu(isaepy)(2) to cause mitochondriotoxicity. In particular, we demonstrate that Cu(isaepy)(2) increases NADH-dependent oxygen consumption of isolated mitochondria and that this phenomenon is associated with oxy-radical production and insensitive to adenosine diphosphate. These data indicate that Cu(isaepy)(2) behaves as an uncoupler and this property is also confirmed in cell systems. Particularly, SH-SY5Y cells show: (i) an early loss of mitochondrial transmembrane potential; (ii) a decrease in the expression levels of respiratory complex components and (iii) a significant adenosine triphosphate (ATP) decrement. The causative energetic impairment mediated by Cu(isaepy)(2) in apoptosis is confirmed by experiments carried out with rho(0) cells, or by glucose supplementation, where cell death is significantly inhibited. Moreover, gastric and cervix carcinoma AGS and HeLa cells, which rely most of their ATP production on oxidative phosphorylation, show a marked sensitivity toward Cu(isaepy)(2). Adenosine monophosphate-activated protein kinase (AMPK), which is activated by events increasing the adenosine monophosphate: ATP ratio, is deeply involved in the apoptotic process because the overexpression of its dominant/negative form completely abolishes cell death. Upon glucose supplementation, AMPK is not activated, confirming its role as fuel-sensing enzyme that positively responds to Cu(isaepy)(2)-mediated energetic impairment by committing cells to apoptosis. Overall, data obtained indicate that Cu(isaepy)(2) behaves as delocalized lipophilic cation and induces mitochondrial-sited reactive oxygen species production. This event results in mitochondrial dysfunction and ATP decrease, which in turn triggers AMPK-dependent apoptosis.

Objective To investigate if a home environment test battery can be used to measure effects of Parkinson’s disease (PD) treatment intervention and disease progression. Background Seventy-seven patients diagnosed with advanced PD were recruited in an open longitudinal 36-month study at 10 clinics in Sweden and Norway; 40 of them were treated with levodopa-carbidopa intestinal gel (LCIG) and 37 patients were candidates for switching from oral PD treatment to LCIG. They utilized a mobile device test battery, consisting of self-assessments of symptoms and objective measures of motor function through a set of fine motor tests (tapping and spiral drawings), in their homes. Both the LCIG-naïve and LCIG-non-naïve patients used the test battery four times per day during week-long test periods. Methods Assessments The LCIG-naïve patients used the test battery at baseline (before LCIG), month 0 (first visit; at least 3 months after intraduodenal LCIG), and thereafter quarterly for the first year and biannually for the second and third years. The LCIG-non-naïve patients used the test battery from the first visit, i.e. month 0. Out of the 77 patients, only 65 utilized the test battery; 35 were LCIG-non-naïve and 30 LCIG-naïve. In 20 of the LCIG-naïve patients, assessments with the test battery were available during oral treatment and at least one test period after having started infusion treatment. Three LCIG-naïve patients did not use the test battery at baseline but had at least one test period of assessments thereafter. Hence, n=23 in the LCIG-naïve group. In total, symptom assessments in the full sample (including both patient groups) were collected during 379 test periods and 10079 test occasions. For 369 of these test periods, clinical assessments including UPDRS and PDQ-39 were performed in afternoons at the start of the test periods. The repeated measurements of the test battery were processed and summarized into scores representing patients’ symptom severities over a test period, using statistical methods. Six conceptual dimensions were defined; four subjectively-reported: ‘walking’, ‘satisfied’, ‘dyskinesia’, and ‘off’ and two objectively-measured: ‘tapping’ and ‘spiral’. In addition, an ‘overall test score’ (OTS) was defined to represent the global health condition of the patient during a test period. Statistical methods Change in the test battery scores over time, that is at baseline and follow-up test periods, was assessed with linear mixed-effects models with patient ID as a random effect and test period as a fixed effect of interest. The within-patient variability of OTS was assessed using intra-class correlation coefficient (ICC), for the two patient groups. Correlations between clinical rating scores and test battery scores were assessed using Spearman’s rank correlations (rho). Results In LCIG-naïve patients, mean OTS compared to baseline was significantly improved from the first test period on LCIG treatment until month 24. However, there were no significant changes in mean OTS scores of LCIG-non-naïve patients, except for worse mean OTS at month 36 (p<0.01, n=16). The mean scores of all subjectively-reported dimensions improved significantly throughout the course of the study, except ‘walking’ at month 36 (p=0.41, n=4). However, there were no significant differences in mean scores of objectively-measured dimensions between baseline and other test periods, except improved ‘tapping’ at month 6 and month 36, and ‘spiral’ at month 3 (p<0.05). The LCIG-naïve patients had a higher within-subject variability in their OTS scores (ICC=0.67) compared to LCIG-non-naïve patients (ICC=0.71). The OTS correlated adequately with total UPDRS (rho=0.59) and total PDQ-39 (rho=0.59). Conclusions In this 3-year follow-up study of advanced PD patients treated with LCIG we found that it is possible to monitor PD progression over time using a home environment test battery. The significant improvements in the mean OTS scores indicate that the test battery is able to measure functional improvement with LCIG sustained over at least 24 months.

The aim of this study was to investigate if a telemetry test battery can be used to measure effects of Parkinson’s disease (PD) treatment intervention and disease progression in patients with fluctuations. Sixty-five patients diagnosed with advanced PD were recruited in an open longitudinal 36-month study; 35 treated with levodopa-carbidopa intestinal gel (LCIG) and 30 were candidates for switching from oral PD treatment to LCIG. They utilized a test battery, consisting of self-assessments of symptoms and fine motor tests (tapping and spiral drawings), four times per day in their homes during week-long test periods. The repeated measurements were summarized into an overall test score (OTS) to represent the global condition of the patient during a test period. Clinical assessments included ratings on Unified PD Rating Scale (UPDRS) and 39-item PD Questionnaire (PDQ-39) scales. In LCIG-naïve patients, mean OTS compared to baseline was significantly improved from the first test period on LCIG treatment until month 24. In LCIG-non-naïve patients, there were no significant changes in mean OTS until month 36. The OTS correlated adequately with total UPDRS (rho = 0.59) and total PDQ-39 (0.59). Responsiveness measured as effect size was 0.696 and 0.536 for OTS and UPDRS respectively. The trends of the test scores were similar to the trends of clinical rating scores but dropout rate was high. Correlations between OTS and clinical rating scales were adequate indicating that the test battery contains important elements of the information of well-established scales. The responsiveness and reproducibility were better for OTS than for total UPDRS.

A fatoração de números inteiros é um assunto que, embora muito antigo, desperta cada vez mais interesse. Existem vários métodos de criptografia de chave pública, baseados não só em fatoração de inteiros, mas também em resolução de logarítmos discretos, por exemplo, cuja segurança depende da ineficiência dos métodos de fatoração conhecidos. Este trabalho tem como objetivo descrever os principais métodos de fatoração utillizados hoje em dia. Primeiramente, três métodos elementares serão estudados: o método de Fermat e os métodos Rho e p - 1 de Pollard. A seguir, os dois mais poderosos métodos de fatoração para inteiros sem forma especial: o método de curvas elípticas, e o método de peneira quadrática, os quais tomam como base os métodos p - 1 e de Fermat, respectivamente.

Com o crescente uso das teorias e métodos vindos das áreas de economia regional e urbana, economia e econometria espacial na fronteira entre as áreas de economia e políticas públicas, estudar esse arcabouço teórico e metodológico torna-se relevante no campo da análise de políticas públicas. O objetivo geral dessa dissertação é discutir de modo crítico aspectos da literatura de econometria espacial e sua aplicabilidade, com especial foco na comparação entre os diferentes modelos e matrizes de ponderação espacial. Esse estudo procura analisar diferentes formas de medidas que permitem a construção de uma matriz de ponderação espacial, bem como testar por meio de simulação experimental e em uma aplicação empírica a robustez de algumas dessas matrizes e de diferentes modelos. Dadas as indicações da literatura e as evidências mostradas nessa dissertação, a estratégia mais confiável para a busca de um modelo parece ser utilizar as estimativas dos modelos, em conjunto com as estatísticas de Moran, para identificar presença de algum tipo de correlação espacial, e do teste LM da variável dependente, para indicar esse tipo de correlação especificamente. Observa-se que, de modo geral, não há muita diferença em termos de esperança dos parâmetros não espaciais, quando comparadas a estimações tradicionais com as que utilizam as técnicas espaciais. Com isso, a indicação que se pode fazer é que o uso das ferramentas de econometria espacial deve ser aplicado quando o interesse da análise é justamente na estimação dos parâmetros de correlação espacial. Comparando o uso da matriz de contigüidade no lugar da matriz de distâncias, é possível notar que a matriz de contigüidade não é, de forma geral, uma boa substituição para a matriz de distâncias. Ela capta parcialmente o efeito de rho e lambda, além de captar erroneamente lambda, quando este inexiste.

A Doença do Xarope do Bordo (DXB) é um erro inato do metabolismo causado pela deficiência na atividade do complexo desidrogenase dos cetoácidos de cadeia ramificada, levando ao acúmulo de concentrações milimolares dos seguintes α-cetoácidos de cadeia ramificada (CACR): ácidos α-cetoisocapróico (CIC), α-ceto-β-metilvalérico (CMV), α-cetoisovalérico (CIV) e dos seus aminoácidos precursores, leucina, isoleucina e valina em tecidos de pacientes afetados. Essa doença é caracterizada por severos sintomas neurológicos que incluem edema e atrofia cerebral, entretanto, os mecanismos envolvidos na neuropatologia da DXB ainda não são bem estabelecidos. Neste trabalho utilizamos um modelo experimental de DXB com o objetivo de verificar os efeitos dos CACR que se acumulam nessa desordem neurodegenerativa sobre o citoesqueleto de células neurais de ratos. Nesse modelo fatias de córtex cerebral de ratos de diferentes idades, ou culturas de células neurais foram incubados com concentrações variando de 0,1 a 10 mM de cada metabólito. Inicialmente demonstramos que o CIC, CMV e CIV inibiram a captação de glutamato em fatias de córtex cerebral de ratos durante o desenvolvimento. O CIC inibiu a captação de glutamato em ratos de 9, 21 e 60 dias de idade, enquanto o CMV e o CIV inibiram a captação de glutamato em animais de 21 e 60 dias. Observamos que o CIC alterou a fosforilação de proteínas do citoesqueleto de um modo dependente do desenvolvimento através de receptores glutamatérgicos ionotrópicos em fatias de córtex cerebral de ratos. O metabólito causou diminuição da fosforilação dos filamentos intermediários (FI) em ratos de 9 dias de idade e aumento dessa fosforilação em animais de 21 dias de vida. Também demonstramos que em animais de 9 dias de idade o efeito do CIC foi mediado pelas proteínas fosfatases PP2A e principalmente pela PP2B, uma proteína fosfatase dependente de cálcio, enquanto que em animais de 21 dias de idade o efeito deste metabólito foi mediado pela proteína quinase dependente de AMP cíclico (PKA) e pela proteína quinase dependente de cálcio e calmodulina (PKCaMII). Além disso, verificamos que o CIC promoveu um aumento nos níveis intracelulares dos segundos mensageiros AMPc e Ca2+. O aumento do Ca2+ intracelular provocado pelo CIC foi demonstrado pelo uso de bloqueadores específicos de canais de cálcio dependentes de voltagem tipo L, por exemplo, nifedipina, canais de cálcio dependentes de ligantes, por exemplo, NMDA e de quelantes de cálcio intracelular, por exemplo, BAPTA-AM. Por outro lado, o CMV aumentou a fosforilação de FI somente em ratos de 12 dias de idade, sendo esse efeito mediado por receptores GABAérgicos do tipo A e B, desencadeando a ativação das proteínas quinases PKA e PKCaMII. É importante salientar que o CIV não alterou a atividade do sistema fosforilante em nenhuma das idades estudadas. Além dos efeitos causados pelos CACR que se acumulam na DXB sobre a atividade do sistema fosforilante associado aos FI em fatias de córtex cerebral de ratos, demonstramos que esses metabólitos foram capazes de alterar a fosforilação da proteína glial fibrilar ácida (GFAP) na linhagem de glioma C6. Essa alteração de fosforilação causou uma reorganização do citoesqueleto de GFAP e um aumento no imunoconteúdo da GFAP na fração citoesquelética. Também verificamos que o CIC, o CMV e o CIV, em concentrações encontradas em pacientes portadores de DXB, levaram a uma reorganização dos filamentos de GFAP e do citoesqueleto de actina de astrócitos em cultura, causando uma importante alteração na morfologia destas células. As alterações do citoesqueleto levaram a morte celular progressiva quando os astrócitos foram expostos por várias horas aos metabólitos. Demonstramos também que os efeitos dos CACR sobre a morfologia dos astrócitos foram desencadeados por mecanismos de membrana que diminuíram a atividade da Rho GTPase. Esse mecanismo foi evidenciado utilizando-se ácido lisofosfatídico (LPA), um ativador específico da RhoA, o qual preveniu os efeitos causados pelos CACR em culturas de astrócitos. É importante salientar que as alterações morfológicas e a morte celular induzida pelos CACR em culturas de astrócitos foram totalmente evitadas com a suplementação de creatina às culturas. Também verificamos que a atividade da creatina quinase foi inibida pelos metabólitos, indicando que a homeostase energética provavelmente estaria envolvida nos efeitos causados pelos CACR. XI Sabe-se que as alterações do citoesqueleto estão relacionadas com inúmeras doenças neurodegenerativas. Portanto, é provável que as alterações causadas pelos CACR nos mecanismos de membrana que regulam níveis de segundos mensageiros intracelulares e cascatas de sinalização celular, na perda do equilíbrio fisiológico do sistema fosforilante associado ao citoesqueleto e conseqüentemente na sua reorganização, possam ter importantes conseqüências para a função neural. Com base nos presentes resultados demonstramos que os CACR que se acumulam na DXB levam à desorganização do citoesqueleto em um modelo experimental podendo ser uma contribuição importante para o estudo da patogênese do sistema nervoso central característica dos pacientes portadores de DXB.

Bothrops jararacussu myotoxin I (BthTx-I; Lys 49) and II (BthTX-II; Asp 49) were purified by ion-exchange chromatography and reverse phase HPLC. In this work we used the isolated perfused rat kidney method to evaluate the renal effects of B. jararacussu myotoxins I (Lys49 PLA(2)) and II (Asp49 PLA(2)) and their possible blockage by indomethacin. BthTX-1 (5 mu g/ml) and BthTX-II (5 mu g/ml) increased perfusion pressure (PP; ct(120) = 110.28+/-3.70 mmHg; BthTX I = 171.28+/-6.30* mmHg; BthTX II = 175.50+/-7.20* mmHg), renal vascular resistance (RVR; ct(120) = 5.49+/-0.54 mmHg/ml.g(-1) min(-1); BthTX I = 8.62+/-0.37* mmHg/ml g(-1) min(-1); BthTX II=8.9+/-0.36* mmHg/ml g(-1) min(-1)), urinary flow (UF; ct(120)= 0.14+/-0.01 ml g(-1) min(-1); BthTX I=0.32+/-0.05* ml g(-1) min(-1); BthTX II=0.37+/-0.01* ml g(-1) min(-1)) and glomerular filtration rate (GFR; ct(120)=0.72+/-0.10 ml g(-1) min(-1); BthTX I=0.85+/-0.13* ml g(-1) min(-1); BthTX II=1.22+/-0.28* ml g(-1) min(-1)). In contrast decreased the percent of sodium tubular transport (%TNa+; ct(120)=79,76+/-0.56; BthTX I=62.23+/-4.12*; BthTX II=70.96+/-2.93*) and percent of potassium tubular transport (%TK+;ct(120)=66.80+/-3.69; BthTX I=55.76+/-5.57*; BthTX II=50.86+/-6.16*). Indomethacin antagonized the vascular, glomerular and tubular effects promoted by BthTX I and it's partially blocked the effects of BthTX II. In this work also evaluated the antibacterial effects of BthTx-I and BthTx-II against Xanthomonas axonopodis. pv. passiflorae (Gram-negative bacteria) and we observed that both PLA2 showed antibacterial activity. Also we observed that proteins Also we observed that proteins chemically modified with 4-bromophenacyl bromide (rho-BPB) decrease significantly the antibacterial effect of both PLA(2). In conclusion, BthTx I and BthTX II caused renal alteration and presented activity antimicrobial. The indomethacin was able to antagonize totally the renal effects induced by BthTx I and partially the effects promoted by BthTx II, suggesting involvement of inflammatory mediators in the renal effects caused by myotoxins. In the other hand, other effects could be independently of the enzymatic activity of the BthTX II and the C-terminal domain could be involved in both effects promoted for PLA(2). In the present article we report on the biological characterization and amino acid sequence of a new basic Phospholipases A(2) (PLA(2)) isolated from the Crotalus durissus collilineatus venom (Cdcolli F6), which showed the presence of 122 amino acid residues with a pI value of 8.3, molecular mass of 14 kDa and revealed an amino acid sequence identity of 80% with crotalic PLA(2)s such as Mojave B, Cdt F15, and CROATOX. This homology, however, dropped to 50% if compared to other sources of PLA(2)s such as from the Bothrops snake venom. Also, this PLA(2) induced myonecrosis, although this effect was lower than that of BthTx-I or whole crotoxin and it was able to induce a strong blockage effect on the chick biventer neuromuscular preparation, independently of the presence of the acid subunid (crotapotin). The neurotoxic effect was strongly reduced by pre-incubation with heparin or with anhydrous acetic acid and rho-BPB showed a similar reduction. The rho-BPB did not reduce significantly the myotoxic activity induced by the PLA(2), but the anhydrous acetic acid treatment and the pre-incu-bation of PLA(2) with heparin reduced significantly its effects. This protein showed a strong antimicrobial activity against Xanthomonas axonopodis passiflorae (Gram-negative), which was drastically reduced by incubation of this PLA(2) with rho-BPB, but this effect was marginally reduced after treatment with anhydrous acetic acid. Our findings here allow to speculate that basic amino acid residues on the C-terminal and molecular regions near catalytic site regions such as Calcium binding loop or rho-wing region may be involved in the binding of this PLA(2) to the molecular receptor to induce the neurotoxic effect. The bactericidal effect, however, was completely dependent on the enzymatic activity of this protein.

OBJETIVO: Estudar a confiabilidade, da versão em português, do questionário para o diagnóstico psicológico e psicossocial dos indivíduos com desordens temporomandibulares (RDC/TMD). MÉTODOS: Foram entrevistados 109 indivíduos, de ambos sexos, que demandaram atendimento junto à Clínica de Fisioterapia do Centro Universitário de Araraquara, de janeiro a julho de 2006. Os questionários foram aplicados por um único examinador. Após duas semanas, o mesmo foi reaplicado em 36 indivíduos. Para avaliação da consistência interna do método, utilizou-se o Coeficiente Alfa de Cronbach; para análise da reprodutibilidade intra-examinador, o Coeficiente de Correlação Intraclasse (ro) e a estatística Kappa (kapa), respectivamente às variáveis de natureza quantitativa e qualitativa. RESULTADOS: A consistência interna para as dimensões intensidade da dor crônica e incapacidade; limitação da função mandibular; sintomas físicos não-específicos, incluindo os itens de dor; sintomas físicos não-específicos, excluindo os itens de dor e depressão foi de 0,8479, 0,8971, 0,8673, 0,8080 e 0,9270 respectivamente, atestando ao método excelente validade interna. Obteve-se excelente concordância intra-examinador para as questões referentes ao tempo de presença da dor e sua gradação, e boa para a questão referente à dor presente. Os menores valores de kapa relacionaram-se aos itens de sintomas físicos e depressão. A percepção de estalos ou rangidos pelos indivíduos apresentou concordância regular bem como a questão referente à procura de profissional para tratamento da dor. As demais questões apresentaram reprodutibilidade boa e ótima, sendo que a maioria dessas apresentou nível máximo de concordância. CONCLUSÃO: A versão adaptada para o português mostrou-se confiável para detecção das alterações psicológicas e psicossociais associadas às desordens temporomandibulares.

A contaminated site from a downstream municipal solid waste disposal site in Brazil was investigated by using a 3D resistivity and induced polarization (IP) imaging technique. This investigation purpose was to detect and delineate contamination plume produced by wastes. The area was selected based on previous geophysical investigations, and chemical analyses carried out in the site, indicating the presence of a contamination plume in the area. Resistivity model has successfully imaged waste presence (rho < 20 Omega m), water table depth, and groundwater flow direction. A conductive anomaly (rho < 20 Omega m) outside wastes placement was interpreted as a contamination plume. Chargeability model was also able to imaging waste presence (m > 31 mV/V), water table depth, and groundwater flow direction. A higher chargeability zone (m > 31 mV/V) outside wastes placement and following conductive anomaly was interpreted as a contamination plume. Normalized chargeability (MN = m/rho) confirmed polarizable zone, which could be an effect of a salinity increase (contamination plume), and the clay presence in the environment.

Diamond-like carbon (DLC) films were grown from radiofrequency plasmas of acetylene-argon mixtures, at different excitation powers, P. The effects of this parameter on the plasma potential, electron density, electron temperature, and plasma activity were investigated using a Langmuir probe. The mean electron temperature increased from about 0.5 to about 7.0 eV while the mean electron density decreased from about 1.2x10(9) to about 0.2x10(9) cm(-3) as P was increased from 25 to 150 W. Both the plasma potential and the plasma activity were found to increase with increasing P. Through actinometric optical emission spectrometry, the relative concentrations of CH, [CH], and H, [H], in the discharge were mapped as a function of the applied power. A rise in [H] and a fall in [CH] with increasing P were observed and are discussed in relation to the plasma characteristics and the subimplantation model. The optical properties of the films were calculated from ultraviolet-visible spectroscopic data; the surface resistivity was measured by the two-point probe method. The optical gap, E(G), and the surface resistivity, rho(s), fall with increasing P. E(G) and rho(s) are in the ranges of about 2.0-1.3 eV and 10(14)-10(16) Omega/square, respectively. The plasma power also influences the film self-bias, V(b), via a linear dependence, and the effect of V(b) on ion bombardment during growth is addressed together with variation in the relative densities of sp(2) and sp(3) bonds in the films as determined by Raman spectroscopy.
