A reação de fixação de complemento é um dos testes usados no diagnóstico confirmatório da brucelose bovina, e para sua realização emprega-se o mesmo antígeno usado na prova de soroaglutinação lenta, porém não foi possível encontrar na literatura estudos sobre a estabilidade desse antígeno para uso na prova de fixação de complemento, de modo a estabelecer um prazo de validade para o mesmo. Por isso, esta investigação teve por objetivo avaliar a estabilidade do antígeno de célula total de Brucella conservado sob refrigeração, para uso na reação de fixação de complemento. Analisaram-se 14 partidas de antígeno, preparado com Brucella abortus amostra 1119/3 e padronizado para uso na prova de soroaglutinação lenta, com tempo de fabricação variando de 9 meses a 23 anos e 11 meses. Testaram-se 167 soros bovinos com títulos variáveis de anticorpos contra Brucella, adotando-se a técnica com incubação a 37ºC nas duas fases da reação e 5 unidades hemolíticas 50% de complemento. Considerou-se como positivo o soro com pelo menos 25% de fixação de complemento na diluição 1:4. Compararam-se os resultados obtidos com as 13 partidas de antígeno com aqueles obtidos com a partida com 9 meses de fabricação, usando o teste de chi2 de McNemar e o coeficiente kappa. A grande maioria dos soros apresentou resultados muito próximos quando testados com as diversas partidas de antígeno, e não se observou relação entre tempo de fabricação do antígeno e diferenças nos resultados obtidos.

The aim of this work is the numerical simulation of the mechanical performance of concrete affected by Alkali-Aggregate Reaction or RAA, reported by Stanton in 1940. The RAA has aroused attention in the context of Civil Engineering from the early 80, when they were reported consequences of his swelling effect in concrete structures, including cracking, failure and loss of serviceability. Despite the availability of experimental results the problem formulation still lacks refinement so that your solution remains doubtful. The numerical simulation is important resource for the assessment of damages in structures caused by the reaction, and their recoveries The tasks of support of this work were performed by means of the finite element approach, about orthotropic non-linear formulation, and, thermodynamic model of deformation by RAA. The results obtained revealed that the swelling effect of RAA induced decline of the mechanical performance of concrete by decreasing the margin of safety prior to the material failure. They showed that the temperature influences, exclusively, the kinetics of the reaction, so that the failure was the more precocious the higher the temperature of the solid mass of concrete

In this work, we propose methodologies and computer tools to insert robots in cultural environments. The basic idea is to have a robot in a real context (a cultural space) that can represent an user connected to the system through Internet (visitor avatar in the real space) and that the robot also have its representation in a Mixed Reality space (robot avatar in the virtual space). In this way, robot and avatar are not simply real and virtual objects. They play a more important role in the scenery, interfering in the process and taking decisions. In order to have this service running, we developed a module composed by a robot, communication tools and ways to provide integration of these with the virtual environment. As welI we implemented a set of behaviors with the purpose of controlling the robot in the real space. We studied available software and hardware tools for the robotics platform used in the experiments, as welI we developed test routines to determine their potentialities. Finally, we studied the behavior-based control model, we planned and implemented alI the necessary behaviors for the robot integration to the real and virtual cultural spaces. Several experiments were conducted, in order to validate the developed methodologies and tools

Avaliou-se a ocorrência da murcha-de-curtobacterium em lavouras de feijoeiro comum em algumas localidades do Estado de Santa Catarina, nas safras 2002/03 e 2003/04, e o comportamento dos genótipos BRS Valente, Carioca, CHC 97-29, CHP 97-26, CNPF 8104, Diamante Negro, Empasc 201 - Chapecó, IAPAR 44, IPR Graúna, IPR Juriti, IPR Uirapuru, LP 9728, Pérola, SCS 202-Guará, Sel. CP 9310635, TPS Bionobre, TPS Bonito, TPS Magnífico, TPS Nobre, TPS Soberano e Xamego perante Curtobacterium flaccumfaciens pv. flaccumfaciens (Cff), em condições de casa-de-vegetação. As cultivares IAC Carioca Akytã, IAC Carioca Aruã e IAC Carioca Pyatã foram empregadas como padrões de resistência a Cff. As avaliações dos sintomas ocorreram aos 5, 10, 15, 20 e 25 dias após a inoculação (DAI) e, posteriormente, foi estimada a área abaixo da curva de progresso da murcha-de-curtobacterium (AACPMC), em cada genótipo. A doença esteve presente nos municípios de Campos Novos, Faxinal dos Guedes, Guatambu, Ipuaçu, Ponte Serrada e Tigrinhos e que, aos 10 DAI, as cultivares SCS 202 - Guará e IPR Juriti mostraram baixa severidade. Porém, aos 25 DAI, somente as cultivares padrões foram resistentes e apresentaram menor AACPMC.

Conduziu-se simultaneamente dois experimentos, em casa de vegetação, com o objetivo de avaliar a resistência de híbridos de brócolis 'tipo cabeça única' (AF 649, Titleist, Centenário, Green Power, BR068, Magestic Crown, Marathon, Laguna, Legacy, Green Parasol, Packman e Mônaco) à podridão negra, causada por Xanthomonas campestris pv. campestris. Foram utilizados os métodos de inoculação no ápice das folhas por cortes com tesoura embebida na suspensão bacteriana e por ferimento provocado no caule com palito de dente umedecido na suspensão bacteriana. A inoculação foi realizada aos 25 dias após transplante (6 a 8 folhas definitivas). Avaliou-se no experimento de inoculação com tesoura, as áreas abaixo da curva de progresso da doença nas folhas inoculadas. No experimento de inoculação por palito de dente, avaliou-se a proporção de altura necrosada do caule, aos 26 dias após inoculação. Verificou-se que, em ambos os experimentos, o híbrido BRO68 apresentou-se mais suscetível à podridão negra e os híbridos Marathon, Legacy e Green Power foram os que apresentaram os maiores níveis de resistência à podridão negra.

O objetivo do presente trabalho foi verificar a resistência ao nematóide Meloidogyne mayaguensis em oito porta-enxertos de tomateiro considerados resistentes à Meloidogyne incognita, M. javanica e M. arenaria, comercializados no Brasil. Os porta-enxertos testados foram: 'Guardião', 'Helper-M', 'Anchor-T', 'Dr. K', 'Kagemuscha', 'TMA 809', 'Magnet' e 'He-Man'. O experimento constou de 9 tratamentos (8 porta-enxertos e a cultivar Rutgers utilizada como padrão de suscetibilidade), com 6 repetições, sendo cada parcela constituída por 1 planta por vaso, mantidas em casa de vegetação. As plantas foram inoculadas com 5.000 ovos e eventuais juvenis infectantes de M. mayaguensis. O experimento seguiu o delineamento inteiramente casualizado. Aos 60 dias da inoculação procederam-se as avaliações, quando foram avaliados os índices de galhas e massas de ovos, número de nematóides no solo e na raiz, peso do sistema radicular e o fator de reprodução. Todos os porta-enxertos estudados demonstraram-se suscetíveis a M. mayaguensis.

The nanostructures materials are characterized to have particle size smaller than 100 nm and could reach 1 nm. Due to the extremely reduced dimensions of the grains, the properties of these materials are significantly modified relatively when compared with the conventional materials. In the present work was accomplished a study and characterization of the molybdenum carbide, seeking obtain it with particles size in the nanometers order and evaluate its potential as catalyst in the reaction of partial methane oxidation. The method used for obtaining the molybdenum carbide was starting from the precursor ammonium heptamolybdate of that was developed in split into two oven, in reactor of fixed bed, with at a heating rate of 5ºC/min, in a flow of methane and hydrogen whose flow was of 15L/h with 5% of methane for all of the samples. The studied temperatures were 350, 500, 600, 650, 660, 675 and 700ºC and were conducted for 0, 60, 120 and 180 minutes, and the percent amount and the crystallite size of the intermediate phases were determined by the Rietveld refinement method. The carbide obtained at 660ºC for 3 hours of reaction showed the best results, 24 nm. Certain the best synthesis condition, a passivating study was accomplished, in these conditions, to verify the stability of the carbide when exposed to the air. The molybdenum carbide was characterized by SEM, TEM, elemental analysis, ICP-AES, TG in atmosphere of hydrogen and TPR. Through the elemental analysis and ICP-AES the presence carbon load was verified. TG in atmosphere of hydrogen proved that is necessary the passivating of the molybdenum carbide, because occur oxidation in room temperature. The catalytic test was accomplished in the plant of Fischer-Tropsch of CTGAS, that is composed of a reactor of fixed bed. Already the catalytic test showed that the carbide presents activity for partial oxidation, but the operational conditions should be adjusted to improve the conversion

Among the heterogeneous catalysts materials made from niobium show up as an alternative to meet the demand of catalysts for biodiesel production. This study aims to evaluate the potential of a heterogeneous catalyst derived from a complex of niobium in the reaction of methyl esterification of oleic acid. The catalyst was synthesized after calcination at different temperatures of a niobium complex ((NH4)3[NbO(C2O4)3].H2O) generating a niobium oxide nanostructure with a different commercial niobium oxide used to synthesize the complex. The commercial niobium oxide, the complex niobium and niobium catalyst were characterized by thermogravimetry (TG and DTA), surface area analysis (BET), scanning electron microscopy (SEM) and X-ray diffraction (XRD), showing the catalyst has researched morphological and crystallographic indicating a catalytic potential higher than that of commercial niobium oxide characteristics. Factorial with central composite design point, with three factors (calcination temperature, molar ratio of alcohol/oleic acid and mass percentage of catalyst) was performed. Noting that the optimal experimental point was given by the complex calcination temperature of 600°C, a molar ratio alcohol/oleic acid of 3.007/1 and the catalyst mass percentage of 7.998%, with a conversion of 22.44% oleic acid in methyl oleate to 60 min of reaction. We performed a composite linear and quadratic regression to determine an optimal statistical point of the reaction, the temperature of calcination of the complex at 450°C, the molar ratio of alcohol/oleic acid 3.3408/1 and mass percentage of catalyst of 7.6833% . Kinetic modeling to estimate parameters for heterogeneous catalysis it set well the experimental results with a final conversion of 85.01% with 42.38% of catalyst and without catalyst at 240 min reaction was performed. Allowing to evaluate the catalyst catalytic studied has the potential to be used in biodiesel production

A incontinência fecal, também conhecida como encoprese, é um transtorno de evacuação que acarreta prejuízos ao desenvolvimento psicossocial e orgânico da criança e do adolescente, e que demanda atenção e cuidado de pais e profissionais de saúde. No amplo contexto de tratamento da encoprese, a psicoterapia constitui importante recurso, sendo a terapia comportamental apontada como uma das modalidades mais promissoras e eficazes para o tratamento dessa dificuldade de eliminação. Este artigo apresenta o estudo dos efeitos do manejo comportamental de quadro de incontinência fecal em um adolescente de 14 anos, atendido em clínica-escola de Psicologia do interior do Estado de São Paulo durante 14 meses. A partir do referencial teórico da análise do comportamento, foi desenvolvido, em contexto psicoterápico, um conjunto de estratégias comportamentais com o cliente, bem como orientações aos pais, visando à gradativa extinção encoprética. No decorrer desse processo, o cliente apresentou significativas aquisições comportamentais de uso regular do banheiro e adequado controle esfincteriano, monitoradas semanalmente, que possibilitaram a plena extinção das ocorrências de sujidade, sendo avaliado o efeito em follow-up realizado três meses após o encerramento dessa intervenção.

Autism comprises a heterogeneous group of neurodevelopmental disorders that affects the brain maturation and produces sensorial, motor, language and social interaction deficits in early childhood. Several studies have shown a major involvement of genetic factors leading to a predisposition to autism, which are possibly affected by environmental modulators during embryonic and post-natal life. Recent studies in animal models indicate that alterations in epigenetic control during development can generate neuronal maturation disturbances and produce a hyper-excitable circuit, resulting in typical symptoms of autism. In the animal model of autism induced by valproic acid (VPA) during rat pregnancy, behavioral, electrophysiological and cellular alterations have been reported which can also be observed in patients with autism. However, only a few studies have correlated behavioral alterations with the supposed neuronal hyper-excitability in this model. The aim of this project was to generate an animal model of autism by pre-natal exposure to VPA and evaluate the early post-natal development and pre-puberal (PND30) behavior in the offspring. Furthermore, we quantified the parvalbumin-positive neuronal distribution in the medial prefrontal cortex and Purkinje cells in the cerebellum of VPA animals. Our results show that VPA treatment induced developmental alterations, which were observed in behavioral changes as compared to vehicle-treated controls. VPA animals showed clear behavioral abnormalities such as hyperlocomotion, prolonged stereotipies and reduced social interaction with an unfamiliar mate. Cellular quantification revealed a decrease in the number of parvalbumin-positive interneurons in the anterior cingulate cortex and in the prelimbic cortex of the mPFC, suggesting an excitatory/inhibitory unbalance in this animal model of autism. Moreover, we also observed that the neuronal reduction occurred mainly in the cortical layers II/III and V/VI. We did not detect any change in the density of Purkinje neurons in the Crus I region of the cerebellar cortex. Together, our results strengthens the face validity of the VPA model in rats and shed light on specific changes in the inhibitory circuitry of the prefrontal cortex in this autism model. Further studies should address the challenges to clarify particular electrophysiological correlates of the cellular alterations in order to better understand the behavioral dysfunctions

The use of non-human primates in scientific research has contributed significantly to the biomedical area and, in the case of Callithrix jacchus, has provided important evidence on physiological mechanisms that help explain its biology, making the species a valuable experimental model in different pathologies. However, raising non-human primates in captivity for long periods of time is accompanied by behavioral disorders and chronic diseases, as well as progressive weight loss in most of the animals. The Primatology Center of the Universidade Federal do Rio Grande do Norte (UFRN) has housed a colony of C. jacchus for nearly 30 years and during this period these animals have been weighed systematically to detect possible alterations in their clinical conditions. This procedure has generated a volume of data on the weight of animals at different age ranges. These data are of great importance in the study of this variable from different perspectives. Accordingly, this paper presents three studies using weight data collected over 15 years (1985-2000) as a way of verifying the health status and development of the animals. The first study produced the first article, which describes the histopathological findings of animals with probable diagnosis of permanent wasting marmoset syndrome (WMS). All the animals were carriers of trematode parasites (Platynosomum spp) and had obstruction in the hepatobiliary system; it is suggested that this agent is one of the etiological factors of the syndrome. In the second article, the analysis focused on comparing environmental profile and cortisol levels between the animals with normal weight curve evolution and those with WMS. We observed a marked decrease in locomotion, increased use of lower cage extracts and hypocortisolemia. The latter is likely associated to an adaptation of the mechanisms that make up the hypothalamus-hypophysis-adrenal axis, as observed in other mammals under conditions of chronic malnutrition. Finally, in the third study, the animals with weight alterations were excluded from the sample and, using computational tools (K-means and SOM) in a non-supervised way, we suggest found new ontogenetic development classes for C. jacchus. These were redimensioned from five to eight classes: infant I, infant II, infant III, juvenile I, juvenile II, sub-adult, young adult and elderly adult, in order to provide a more suitable classification for more detailed studies that require better control over the animal development

Goat breeding in the state of Rio Grande do Norte, Brazil has promising economic possibilities, with the proper handling of the natural resources. The introduction of specialized animals has been one of the ways used to improve herd genetics and increase productivity. However, climate has been one of the regional factors that most interferes with the adaptation of the new genetic prevalence resulting from the introduction of exotic breeds, because in their country of origin, the air temperature during most of the year is lower than the animals body temperature. With this in mind, the aim of this study was to characterize behavioral, physiological and morphological profiles and milk production of female Saanen goats belonging to different genetic groups raised in the semi-arid region of Rio Grande do Norte in Northeast Brazil. The study was conducted in the city of Lages (5° 42 00 S and 36° 14 41 W). We used 25 lactating female Saanen goats, distributed into 3 genetic groups: 5 purebred animals, 11 three-quarter bred and 9 half-bred. Behavioral observations were made over three consecutive days in the months of August and September, between 09:00 and 11:30h, when the animals were grazing. Physiological and meteorological data were recorded in the last three days of June, July, August and September at 05:00h and at 16:00h. In the semi-intensive breeding system, the animals from different genetic groups were similar in both field behavior and physiological response patterns. Although the purebred goats had longer hair, they did not show symptoms of thermal discomfort. Their white hair helped to reflect the short wavelength rays and thus eliminate those at the longer wave lengths. We concluded that the animals raised in the semi-intensive milk production system in this study seem to have adapted to the climatic conditions of the semi-arid region of Rio Grande do Norte, Brazil

Regarding the growing number of human beings with physical and mental pathologies associated to different stressor agents, attempts are being made to validate animal models with a close phylogenetic resemblance to man, to study stress response. Callithrix jacchus has been widely used in biomedical research, including on stress, but there is scarce information in the literature about how individual and social factors modulate stressor response in this species. This study uses 4 approaches to investigate the response of male and female adult C. jacchus, under situations of stress, and in the first we show evidence of the importance of this animal as an experimental model in research involving the hypothalamus-pituitary-adrenal axis. And we investigate if sex and baseline cortisol levels modulate the behavioral and hormonal response to separation. In two additional approaches investigate if type of social support (co-specific parent or non-parent) and social rank interfere in behavioral and hormonal when the animal are exposure to a new environment, paired with a co-specific (F2), exposure of the animal to a new environment, isolated (F3) or during reunion (F4). Finally, we also investigated the androgen levels in the males, with a focus on the challenge hypothesis, referring to environmental responsiveness and male-male exposure to relatives and non-relatives of C. jacchus. It was observed that: (1) the baseline cortisol of the animal is predictive of cortisol reactivity at separation; (2) males and females do not show dimorphism in the response of cortisol to stressors, although the females have higher baseline levels of this hormone and exhibit higher frequencies of anxiety-related behaviors; (3) only social support provided by relatives proved to be effective in buffering the cortisol response. In behavioral terms this response was dimorphic, showing that only the male dyads displayed an attenuated response to stress; (4) the males showed differences in cortisol levels as a function of social rank and study phases, whereas in the females no such alterations were observed. The males with indefinite dominance hierarchy (IDH) had reduced cortisol in F2 and F4, while the IDH females showed an increase in F3 and F4; (5) the males of relative and non-relative dyads did not exhibit variations in androgen levels as a function of a new environment. These results, taken together, (a) corroborate the use of C. jacchus as a good animal model for stress-related studies, given that they exhibit similar behavioral and physiological alterations to those of human beings in response to stressor agents; (b) point to the importance of considering individual and social modulating factors during experiments with stressors; (c) provide more reliable comparison parameters in studies where these primates are used as animal models, and (d) show that androgens vary as a function of genetic proximity (relative or non-relative) when the animals are faced with physical and social environmental challenges, thus providing important information for studying the challenge hypothesis in this species

Parental care in mammals is influenced by somatosensory stimuli from infants, such as vocalization and sight and by changes in the hormone levels of caretakers. The aim of this study was to evaluate the behavioral and hormonal responses of twelve non reproductive adult male common marmosets (Callithrix jacchus) to infant cues, vocalization recordings, sight and physical contact with newborn. Six out of twelve males had previous experience in caretaking. In article 1, adult males were exposed to newborn vocalizations for 10 minutes. On control condition no sound was presented. In article 2, males were tested on two conditions: a) Control: an empty acrylic transparent box (test box) was placed in male s cage for 15 minutes, and b) Experimental: males were exposed to newborns into a closed text box for 15 minutes. The cage was kept closed to prevented from tactile, smell and acoustic stimulation by the infant on common marmoset males. In article 3, males were exposed to an open or closed text box, which allowed or not their access to and social interaction with the infants. After each observation sessions, blood samples were collected to evaluate the cortisol levels of males. In all studies, behavioral response of adult males was significantly modified by newborns sight, vocalization and physical contact. Males approached and spent more time near the sound source and showed an increase in locomotion during sound exposure. Furthermore, males approached, smelled and spent more time near the test box when the newborn was inside it. There was no difference in behavioral pattern between experienced and non-experienced males in articles 1 and 2. In article 3, behavioral pattern of males was influence by previous caretaking experience. Experienced males recovered quicker and carried the infants more than the inexperienced ones. However, inexperienced males showed a decrease in recovery latency and an increase in carrying time after successive exposure to infants. Cortisol levels changed after exposure to infant s vocalization, especially for experienced adult males. Male hormonal profile was not affected by the sight of infants neither by their previous experienced in caretaking. The occurrence of social interaction between the caretaker and infant did not modify the hormonal profile of common marmoset males; however, as much as experienced males carried the infants their cortisol levels decreased. Thus, members of a social group or potential caretakers common marmosets exposed to sensory cues from dependent infant such as vocalization, sight, smell and physical contact, changed their behavioral and hormonal responses that are physiological modulators of parental behavior in common marmoset
