Objective The protein Hwp1, expressed on the pathogenic phase of Candida albicans, presents sequence analogy with the gluten protein gliadin and is also a substrate for transglutaminase. This had led to the suggestion that C. albicans infection (CI) may be a triggering factor for Celiac disease (CeD) onset. We investigated cross-immune reactivity between CeD and CI. Methods Serum IgG levels against recombinant Hwp1 and serological markers of CeD were measured in 87 CeD patients, 41 CI patients, and 98 healthy controls (HC). IgA and IgG were also measured in 20 individuals from each of these groups using microchips sensitized with 38 peptides designed from the N-terminal of Hwp1. Results CI and CeD patients had higher levels of anti-Hwp1 (p= 0.0005 and p= 0.004) and anti-gliadin (p= 0.002 and p= 0.0009) antibodies than HC but there was no significant difference between CeD and CI patients. CeD and CI patients had higher levels of anti-transglutaminase IgA than HC (p= 0.0001 and p= 0.0039). During CI, the increase in anti-Hwp1 paralleled the increase in anti-gliadin antibodies. Microchip analysis showed that CeD patients were more reactive against some Hwp1 peptides than CI patients, and that some deamidated peptides were more reactive than their native analogs. Binding of IgG from CeD patients to Hwp1 peptides was inhibited by gamma III gliadin peptides. Conclusions Humoral cross-reactivity between Hwp1 and gliadin was observed during CeD and CI. Increased reactivity to Hwp1 deamidated peptide suggests that transglutaminase is involved in this interplay. These results support the hypothesis that CI may trigger CeD onset in genetically-susceptible individuals.

The recovery of benthic communities inside the western Gulf of Maine fishing closure area was evaluated by comparing invertebrate assemblages at sites inside and outside of the closure four to six years after the closure was established. The major restriction imposed by the closure was a year-round prohibition of bottom gillnets and otter trawls. A total of 163 seafloor sites (~half inside and half outside the closure) within a 515-km2 study area were sampled with some combination of Shipek grab, Wildco box corer, or underwater video. Bottom types ranged from mud (silt and clay) to boulders, and the effects of the closure on univariate measures (total density, biomass, taxonomic richness) of benthos varied widely among sediment types. For sites with predominantly mud sediments, there were mixed effects on inside and outside infauna and no effect on epifauna. For sites with mainly sand sediments, there were higher density, biomass, and taxonomic richness for infauna inside the closure, but no significant effects on epifauna. For sites dominated by gravel (which included boulders in some areas), there were no effects on infauna but strong effects on epifaunal density and taxonomic richness. For fishing gear, the data indicated that infauna recovered in sand from the impacts of otter trawls operated inside the closure but that they did not recover in mud, and that epifauna recovered on gravel bottoms from the impact of gillnets used inside the closure. The magnitudes of impact and recovery, however, cannot be inferred directly from our data because of a confounding factor of different fishing intensities outside the closure for a direct comparison of preclosure and postclosure data. The overall negative impact of trawls is likely underestimated by our data, whereas the negative impact of gillnets is likely overestimated.

Commercial catch and effort data were fit to the Leslie model to estimate preexploitation abundance and the catchability coefficient of slipper lobster, Scyllarides squammosus, in the Northwestern Hawaiian Islands (NWHI). A single vessel fished for 34 consecutive days in the vicinity of Laysan Island and caught 126,127 total slipper lobster in 36,170 trap hauls. Adjusted catch of legal slipper lobster dropped from a high of 3.70 to 1.16 lobster per trap haul. Preexploitation abundance at Laysan Island was an estimated 204,000 legal slipper lobster, which was extrapolated to yield an estimate of 1.2 X 106 to 3.8 X 106 lobster for the entire NWHI slipper lobster fishery.

The primary objective of this study was to predict the distribution of mesophotic hard corals in the Au‘au Channel in the Main Hawaiian Islands (MHI). Mesophotic hard corals are light-dependent corals adapted to the low light conditions at approximately 30 to 150 m in depth. Several physical factors potentially influence their spatial distribution, including aragonite saturation, alkalinity, pH, currents, water temperature, hard substrate availability and the availability of light at depth. Mesophotic corals and mesophotic coral ecosystems (MCEs) have increasingly been the subject of scientific study because they are being threatened by a growing number of anthropogenic stressors. They are the focus of this spatial modeling effort because the Hawaiian Islands Humpback Whale National Marine Sanctuary (HIHWNMS) is exploring the expansion of its scope—beyond the protection of the North Pacific Humpback Whale (Megaptera novaeangliae)—to include the conservation and management of these ecosystem components. The present study helps to address this need by examining the distribution of mesophotic corals in the Au‘au Channel region. This area is located between the islands of Maui, Lanai, Molokai and Kahoolawe, and includes parts of the Kealaikahiki, Alalākeiki and Kalohi Channels. It is unique, not only in terms of its geology, but also in terms of its physical oceanography and local weather patterns. Several physical conditions make it an ideal place for mesophotic hard corals, including consistently good water quality and clarity because it is flushed by tidal currents semi-diurnally; it has low amounts of rainfall and sediment run-off from the nearby land; and it is largely protected from seasonally strong wind and wave energy. Combined, these oceanographic and weather conditions create patches of comparatively warm, calm, clear waters that remain relatively stable through time. Freely available Maximum Entropy modeling software (MaxEnt 3.3.3e) was used to create four separate maps of predicted habitat suitability for: (1) all mesophotic hard corals combined, (2) Leptoseris, (3) Montipora and (4) Porites genera. MaxEnt works by analyzing the distribution of environmental variables where species are present, so it can find other areas that meet all of the same environmental constraints. Several steps (Figure 0.1) were required to produce and validate four ensemble predictive models (i.e., models with 10 replicates each). Approximately 2,000 georeferenced records containing information about mesophotic coral occurrence and 34 environmental predictors describing the seafloor’s depth, vertical structure, available light, surface temperature, currents and distance from shoreline at three spatial scales were used to train MaxEnt. Fifty percent of the 1,989 records were randomly chosen and set aside to assess each model replicate’s performance using Receiver Operating Characteristic (ROC), Area Under the Curve (AUC) values. An additional 1,646 records were also randomly chosen and set aside to independently assess the predictive accuracy of the four ensemble models. Suitability thresholds for these models (denoting where corals were predicted to be present/absent) were chosen by finding where the maximum number of correctly predicted presence and absence records intersected on each ROC curve. Permutation importance and jackknife analysis were used to quantify the contribution of each environmental variable to the four ensemble models.

EXTRACT (SEE PDF FOR FULL ABSTRACT): Since 1986, the U.S. Geological Survey and National Weather Service have operated a warning system for debris flows triggered by severe rainstorms in the San Francisco Bay region. The NWS tracks storm systems as they approach the region, forecasts precipitation, and observes rainfall with a network of radiotelemetered rain gauges (ALERT). The USGS also monitors ALERT data and compares the observed and forecast rainfall to thresholds for debris-flow initiation. Both groups jointly assess debris-flow hazards and issue public advisories when rainfall conditions reach or approach critical levels.

EXTRACT (SEE PDF FOR FULL ABSTRACT): As part of a study of climatic influences on landslide initiation, a statistical analysis of long-term (>40 years) records of daily rainfall from 24 Pacific coastal stations, from San Diego to Cape Flattery, disclosed an unexpected result - the square root of the daily rainfall closely approximates a normal distribution function. ... This paper illustrates the use of the square-root-normal distribution to analyze variations in precipitation along the mainland United States Pacific Coast with examples of orographic enhancement, rain shadows, and increase in precipitation frequency with geographic latitude.

Biochemical techniques designed to compare species on the basis of protein differences were started by NUTTALL (1904) who used immunological methods to compare the serum of humans with that of other primates. Since then more refined techniques have led to better results at the protein level in taxonomy, The analyses of proteins are considered to be the simplest indirect approach to understanding the structure and function of the genetic material, deoxyribonucleic acid (DNA). Interest in these analyses arises because of the close relationship between protein structure and gene structure. Thus by comparing the properties of homologous proteins from different taxa one is in essence comparins their genes (GORMAN er al., 1971). It is now an established fact that genetic information coded in molecules of DNA is translated through a series of reactions in the structure of proteins which form the principal morphological units of the animal body at the molecular level of organization (SIBLEY, 1952). A convenient method of comparing molecular differences between species is to measure the electrophoretic mobility of proteins in a starch gel medium (ASPINWALL and TSUYUKI, 1968) or acrylamide gel (RAYMOND and WEINTRAUB, 1959; BOUCK and BALL, 1968). Proteins with enzymatic properties can be compared on the basis of catalytic activity in the presence or absence of inhibitors (KAPLAN et al., 1959); BAILEY et al., t 1970). A combination of gel electrophoresis and histochemical enzyme detection techniques (HUNTER and MARKERT, 1957) makes it possible to combine electrophoretic mobility anti catalytic activity comparison, Enzyme patterns exhibited in starch gel or acrylamide gel have been used to classify different species. BOUCK and BALL (1968)working with lactate dehydrogenase in species of Trout found that each Trout species had LDH pattern characterbtic of that species. ASPINIWALL and TSUYUKI (1968) used muscle protein electrophoretic patterns to identify hybrid fishes. TSUYUKI and ROBERTS (1963) and TSUYUKI et al. (1964-65) found that myogen protein patterns in fishes were species specific. The myogen patterns within one family were remarkably parallel with the existing morphometric classification and these patterns constituted a single criterion by which the fishes could be identified. The fish used in these investigations were collected from shallow waters (10 metres) of Lake Victoria in two areas, Jinja and Kisumu, using gillnets and beach-seines. The study included ten specimens of each of the following specIes: (l) Haplochromis michaeli (2) Haploehromis obems (3) Astatoreochromis ulluaudi (4) Tilapia zillii and (5) Tilapia nilotica.

In most parts of China, mosquitoes have been subjected to organophosphate (OP) insecticide treatments since the mid-1960s, and resistance gene monitoring in the Culex pipiens complex (Diptera: Culicidae) started in only a few locations from the end of the 1980s. Many resistant alleles at the Ester locus have been found in field populations, including those commonly found around the world (Ester(B1) and Ester(2)), and those endemic to China (Ester(B6), Ester(B7), Ester(8), and Ester(9)). This situation is atypical, and may represent a complex situation for the evolution of insecticide resistance genes in China. To increase our understanding of the Chinese situation and our ability to manage resistance in the C. pipiens complex, a large study was performed. Twenty field populations were sampled from Beijing to Guangzhou. Bioassays with five insecticides (dichlorvos, parathion, chlorpyrifos, 2-sec-butylphenyl methyl carbamate, and propoxur) disclosed resistance levels variable according to the geographic origin, and up to 85-fold for dichlorvos. Six overproduced esterases were identified, including two that have not been previously described. Most of them were found in all samples, although at variable frequencies, suggesting variable selection or a transient situation, e.g., each one was recently restricted to a particular geographic area. The results are discussed in the context of recent alterations to insecticide campaigns, and of the evolution of resistance genes in Chinese C. pipiens populations.
