Extracellular signal-regulated kinases 1/2 (ERK1/2) and their substrates, p90 ribosomal S6 kinases (RSKs), phosphorylate different transcription factors, contributing differentially to transcriptomic profiles. In cardiomyocytes, ERK1/2 are required for >70% of the transcriptomic response to endothelin-1. Here, we investigated the role of RSKs in the transcriptomic responses to Gq protein-coupled receptor agonists, endothelin-1, phenylephrine (generic α1-adrenergic receptor agonist) and A61603 (α1A-adrenergic receptor selective). Phospho-ERK1/2 and phospho-RSKs appeared in cardiomyocyte nuclei within 2-3 min of stimulation (endothelin-1>a61603≈phenylephrine). All agonists increased nuclear RSK2, but only endothelin-1 increased nuclear RSK1 content. PD184352 (inhibits ERK1/2 activation) and BI-D1870 (inhibits RSKs) were used to dissect the contribution of RSKs to the endothelin-1-responsive transcriptome. Of 213 RNAs upregulated at 1 h, 51% required RSKs for upregulation whereas 29% required ERK1/2 but not RSKs. The transcriptomic response to phenylephrine overlapped with, but was not identical to, endothelin-1. As with endothelin-1, PD184352 inhibited upregulation of most phenylephrine-responsive transcripts, but the greater variation in effects of BI-D1870 suggests that differential RSK signalling influences global gene expression. A61603 induced similar changes in RNA expression in cardiomyocytes as phenylephrine, indicating that the signal was mediated largely through α1A-adrenergic receptors. A61603 also increased expression of immediate early genes in perfused adult rat hearts and, as in cardiomyocytes, upregulation of the majority of genes was inhibited by PD184352. PD184352 or BI-D1870 prevented the increased surface area induced by endothelin-1 in cardiomyocytes. Thus, RSKs play a significant role in regulating cardiomyocyte gene expression and hypertrophy in response to Gq protein-coupled receptor stimulation.

AIMS: The objective of the present investigation was to examine the relationship of three polymorphisms, Thr394Thr, Gly482Ser and +A2962G, of the peroxisome proliferator activated receptor-gamma co-activator-1 alpha (PGC-1alpha) gene with Type 2 diabetes in Asian Indians. METHODS: The study group comprised 515 Type 2 diabetic and 882 normal glucose tolerant subjects chosen from the Chennai Urban Rural Epidemiology Study, an ongoing population-based study in southern India. The three polymorphisms were genotyped using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Haplotype frequencies were estimated using an expectation-maximization (EM) algorithm. Linkage disequilibrium was estimated from the estimates of haplotypic frequencies. RESULTS: The three polymorphisms studied were not in linkage disequilibrium. With respect to the Thr394Thr polymorphism, 20% of the Type 2 diabetic patients (103/515) had the GA genotype compared with 12% of the normal glucose tolerance (NGT) subjects (108/882) (P = 0.0004). The frequency of the A allele was also higher in Type 2 diabetic subjects (0.11) compared with NGT subjects (0.07) (P = 0.002). Regression analysis revealed the odds ratio for Type 2 diabetes for the susceptible genotype (XA) to be 1.683 (95% confidence intervals: 1.264-2.241, P = 0.0004). Age adjusted glycated haemoglobin (P = 0.003), serum cholesterol (P = 0.001) and low-density lipoprotein (LDL) cholesterol (P = 0.001) levels and systolic blood pressure (P = 0.001) were higher in the NGT subjects with the XA genotype compared with GG genotype. There were no differences in genotype or allelic distribution between the Type 2 diabetic and NGT subjects with respect to the Gly482Ser and +A2962G polymorphisms. CONCLUSIONS: The A allele of Thr394Thr (G --> A) polymorphism of the PGC-1 gene is associated with Type 2 diabetes in Asian Indian subjects and the XA genotype confers 1.6 times higher risk for Type 2 diabetes compared with the GG genotype in this population.

Background Autism Spectrum Conditions (ASC) are a group of neurodevelopmental conditions characterized by impairments in communication and social interaction, alongside unusually repetitive behaviors and narrow interests. ASC are highly heritable and have complex patterns of inheritance where multiple genes are involved, alongside environmental and epigenetic factors. Asperger Syndrome (AS) is a subgroup of these conditions, where there is no history of language or cognitive delay. Animal models suggest a role for oxytocin (OXT) and oxytocin receptor (OXTR) genes in social-emotional behaviors, and several studies indicate that the oxytocin/oxytocin receptor system is altered in individuals with ASC. Previous studies have reported associations between genetic variations in the OXTR gene and ASC. Methods The present study tested for an association between nine single nucleotide polymorphisms (SNPs) in the OXTR gene and AS in 530 individuals of Caucasian origin, using SNP association test and haplotype analysis. Results There was a significant association between rs2268493 in OXTR and AS. Multiple haplotypes that include this SNP (rs2268493-rs2254298, rs2268490-rs2268493-rs2254298, rs2268493-rs2254298-rs53576, rs237885-rs2268490-rs2268493-rs2254298, rs2268490-rs2268493-rs2254298-rs53576) were also associated with AS. rs2268493 has been previously associated with ASC and putatively alters several transcription factor-binding sites and regulates chromatin states, either directly or through other variants in linkage disequilibrium (LD). Conclusions This study reports a significant association of the sequence variant rs2268493 in the OXTR gene and associated haplotypes with AS.

Nuclear receptors are ligand-activated transcription factors, which have the potential to integrate internal metabolic events in an organism, with consequences for control of behaviour. Previous studies from this laboratory have shown that thyroid hormone receptor (TR) isoforms can inhibit oestrogen receptor (ER)alpha-mediated induction of preproenkephalin (PPE) gene expression in the hypothalamus. Also, thyroid hormone administration inhibits lordosis, a behaviour facilitated by PPE expression. We have examined the effect of multiple ligand-binding TR isoforms on the ER-mediated induction of the PPE gene in transient transfection assays in CV-1 cells. On a natural PPE gene promoter fragment containing two putative oestrogen response elements (EREs), both ER alpha and beta isoforms mediate a four to five-fold induction by oestrogen. Cotransfection of TR alpha 1 along with ER alpha inhibited the ER alpha transactivation of PPE by approximately 50%. However, cotransfection with either TR beta 1 or TR beta 2 expression plasmids produced no effect on the ER alpha or ER beta mediated induction of PPE. Therefore, under these experimental conditions, interactions with a single ER isoform are specific to an individual TR isoform. Transfection with a TR alpha 1 DNA-binding mutant could also inhibit ER alpha transactivation, suggesting that competition for binding on the ERE may not be the exclusive mechanism for inhibition. Data with the coactivator, SRC-1, suggested that coactivator squelching may participate in the inhibition. In dramatic contrast, when ER beta is cotransfected, TR alpha 1 stimulated ER beta-mediated transactivation of PPE by approximately eight-fold over control levels. This is the first study revealing specific interactions among nuclear receptor isoforms on a neuroendocrine promoter. These data also suggest that the combinatorics of ER and TR isoforms allow multiple forms of flexible gene regulations in the service of neuroendocrine integration.

Oestrogens are critical for the display of lordosis behaviour and, in recent years, have also been shown to be involved in synaptic plasticity. In the brain, the regulation of ionotropic glutamate receptors has consequences for excitatory neurotransmission. Oestrogen regulation of the N-methyl-d-aspartate receptor subunit 2D (NR2D) has generated considerable interest as a possible molecular mechanism by which synaptic plasticity can be modulated. Since more than one isoform of the oestrogen receptor (ER) exists in mammals, it is possible that oestrogen regulation via the ERalpha and ERbeta isoforms on the NR2D oestrogen response element (ERE) is not equivalent. In the kidney fibroblast (CV1) cell line, we show that in response to 17beta-oestradiol, only ERalpha, not ERbeta, could upregulate transcription from the ERE which is in the 3' untranslated region of the NR2D gene. When this ERE is in the 5' position, neither ERalpha nor ERbeta showed transactivation capacity. Thyroid hormone receptor (TR) modulation of ER mediated induction has been shown for other ER target genes, such as the preproenkephalin and oxytocin receptor genes. Since the various TR isoforms exhibit distinct roles, we hypothesized that TR modulation of ER induction may also be isoform specific. This is indeed the case. The TRalpha1 isoform stimulated ERalpha mediated induction from the 3'-ERE whereas the TRbeta1 isoform inhibited this induction. This study shows that isoforms of both the ER and TR have different transactivation properties. Such flexible regulation and crosstalk by nuclear receptor isoforms leads to different transcriptional outcomes and the combinatorial logic may aid neuroendocrine integration.

Estrogen is an important steroid hormone that mediates most of its effects on regulation of gene expression by binding to intracellular receptors. The consensus estrogen response element (ERE) is a 13 bp palindromic inverted repeat with a three nucleotide spacer. However, several reports suggest that many estrogen target genes are regulated by diverse elements, such as imperfect EREs and ERE half sites (ERE 1/2), which are either the proximal or the distal half of the palindrome. To gain more insight into ERE half site-mediated gene regulation, we used a region from the estrogen-regulated chicken riboflavin carrier protein (RCP) gene promoter that contains ERE half sites. Using moxestrol, an analogue of estrogen and transient transfection of deletion and mutation containing RCP promoter/reporter constructs in chicken hepatoma (LMH2A) cells, we identified an estrogen response unit (ERU) composed of two consensus ERE 1/2 sites and one non-consensus ERE 1/2 site. Mutation of any of these sites within this ERU abolishes moxestrol response. Further, the ERU is able to confer moxestrol responsiveness to a heterologous promoter. Interestingly, RCP promoter is regulated by moxestrol in estrogen responsive human MCF-7 cells, but not in other cell lines such as NIH3T3 and HepG2 despite estrogen receptor-alpha (ER-�) co transfection. Electrophoretic mobility shift assays (EMSAs) with promoter regions encompassing the half sites and nuclear extracts from LMH2A cells show the presence of a moxestrol-induced complex that is abolished by a polyclonal anti-ER� antibody. Surprisingly, estrogen receptor cannot bind to these promoter elements in isolation. Thus, there appears to be a definite requirement for some other factor(s) in addition to estrogen receptor, for the generation of a suitable response of this promoter to estrogen. Our studies therefore suggest a novel mechanism of gene regulation by estrogen, involving ERE half sites without direct binding of ER to the cognate elements.

This work examined how the conceptus modulates endometrial tissue remodeling and vascular development prior to implantation in mares. A macroscopic uterine examination was completed at day 21 of pregnancy. In situ morphology revealed that the endometrium involved in encroachment is restricted to the dorsal endometrium immediately overlying the yolk sac. The amount of stromal area occupied by blood vessels and the number of endometrial glands were increased during early pregnancy. Endometrial histomorphometry as well as the endometrial mRNA abundance and immunolocalization of VEGF, VEGFR1, VEGFR2, and Ki-67 was completed at days 14 and 21 of pregnancy, at day 10 of the estrous cycle, and during estrus. No obvious differences in VEGF and VEGFR1 protein localization were detected between pregnant and cycling mares but differential staining pattern for VEGFR2 and Ki-67 was observed. VEGFR2 localized to luminal and glandular epithelium of pregnant mares, while luminal epithelium was negative in cycling mares. Ki-67 staining was weak during the luteal phase but exhibited prominent luminal epithelium staining during estrus. In pregnant mares, all endometrial layers were Ki-67 positive. Quantitative RT-PCR revealed a greater abundance of VEGF mRNA during pregnancy. VEGFR2 transcript abundance was greatest in pregnant mares on day 21. This study supports the concept that the conceptus plays an active role in directing vasculogenesis within the uterus and thereby establishing hemotrophic nutrition that supports pregnancy after implantation. As origens das populações e o processo de povoamento das Américas são questões bastante controversas e, por isso, têm sido amplamente estudadas. O gene LDLR (low density lipoprotein receptor), que está localizado no braço curto do cromossomo 19 (p13.1-p13.3) e possui 18 éxons, bem como uma porção 3’UTR onde ocorrem dois elementos Alu completos (chamados de U e D) e um parcial, foi escolhido para esclarecer esses e outros problemas de evolução e variação genética humana. O objetivo geral deste trabalho foi verificar o que esta região hipervariável, especificamente na sua porção U, nos indicaria sobre a história evolutiva das populações ameríndias. Para este trabalho foram analisadas amostras de DNA de indivíduos nativos da Mongólia (n=24), da Sibéria (n=26), das Américas do Norte (n=11), Central (n=26) e do Sul (n=16), totalizando 14 populações e 103 indivíduos. Essas amostras foram amplificadas pela técnica de PCR (polymerase chain reaction), utilizando-se primers específicos para o segmento hipervariável U e, posteriormente, foram elas seqüenciadas automaticamente. Quatorze sítios polimórficos foram encontrados, classificáveis em sete haplótipos, sendo que o sítio 3809 apresentou uma transversão de C para G não descrita na literatura. A estimativa geral de diversidade nucleotídica (π) foi de 0.62%, considerada alta para um marcador autossômico. Verificou-se uma certa uniformidade haplotípica entre a Ásia e a América, mas com a diferenciação maior ocorrendo entre a Mongólia e a América+Sibéria. Não foi detectada diferenciação significativa entre nativos sul e centro-americanos. De um modo geral, o estudo desse marcador confirmou uma provável origem asiática para as populações ameríndias e apoiou a hipótese de uma única onda de migração no processo de povoamento pré-histórico das Américas.

Introdução: Os tumores do Sistema Nervoso Central (SNC) constituem um grupo heterogêneo de neoplasias e representam o tumor sólido mais comum na infância e adolescência (DEANGELIS 2001). A tumorigênese é um processo de múltiplas etapas envolvendo alterações genéticas que levam a transformação progressiva de células normais para a formação de derivados altamente malignos (HANAHAN 2000). O receptor do fator de crescimento epidermal (EGFR) é um receptor transmembrana que atua num sistema de sinalização fundamental para a fisiologia normal da célula e na manutenção do estado tumorigênico (JORISSEM 2003). O EGFR e seus ligantes estão envolvidos em mais de 70% de todas as neoplasias (YARDEN 2001). Em vários tipos de tumores, incluindo os tumores cerebrais, o EGFR é expresso aproximadamente 100 vezes mais do que o número normal de receptores encontrados na superfície de células normais. A hiperexpressão do EGFR e do ErbB2, tem sido associada com neoplasias que apresentam um comportamento clínico mais agressivo (ALROY 1997). Objetivos: Analisar a expressão do gene do EGFR em tumores primários do SNC. Material e Método: Análise de 18 amostras de tumores primários do SNC em crianças de 01 a 14 anos de idade utilizando uma técnica semi-quantitativa de RT-PCR. A coleta das amostras foi feita no período de 2002 a 2004. Resultados: A amostra foi composta por: um astrocitoma grau I, três astrocitomas grau III, três astrocitomas grau IV, três carcinomas de plexo coróide, 1 craniofaringeoma, três ependimomas, um tumor neuroectodérmico primitivo e três meduloblastomas. 33% da amostra apresentou hiperexpressão de EGFR. Ao analisar-se os resultados de expressão do EGFR nas amostras de astrocitomas verificou-se um aumento na expressão do EGFR para os tumores de grau IV, com exceção de uma amostra que apresentou um índice baixo de 0.13 RNAm. Para as demais amostras de astrocitoma grau 3 a expressão do EGFR foi baixa e os pacientes apresentaram sobrevida com doença, uma vez que não foi possível realizar ressecção cirúrgica total. Conclusão:Ainda que este estudo forneça dados de uma população bastante heterogênea é possível evidenciar diferenças nítidas de expressão do EGFR entre as diversas histologias que compreendem os tumores pediátrico do SNC. Tais resultados sugerem um importante papel do EGFR nos tumores cerebrais da infância e encorajam a continuidade de investigações que elucidem a real aplicabilidade deste fator de crescimento como marcador prognóstico e alvo terapêutico.

A esquizofrenia é uma doença psiquiátrica grave que afeta aproximadamente 1% da população mundial em que fatores genéticos exercem importante papel na etiopatogenia. Nosso grupo sugeriu a participação do sistema adenosinérgico na fisiopatologia desta doença. Neste estudo genético de associação, uma amostra de 88 pacientes com esquizofrenia foi comparada a um grupo de 100 controles sem histórico pessoal e familiar de doença psiquiátrica, em relação aos polimorfismos 2592C>Tins e 263C>T do gene que codifica o receptor A2A de adenosina. O grupo total de pacientes foi dividido nos seguintes subgrupos: pacientes com predomínio de sintomas positivos, pacientes com sintomas negativos, pacientes com sintomas desorganizados, pacientes com sintomas negativos e desorganizados e pacientes com manifestações mais e menos graves da doença, conforme parâmetro farmacológico. Os grupos de pacientes foram comparados entre si e ao grupo controle em relação aos polimorfismos em estudo. Nossos resultados indicam uma associação do genótipo 2592C/C com a doença, em que variações geneticamente determinadas do nível de expressão dos receptores A2A de adenosina desempenhariam papel no seu desenvolvimento. Particularmente, a sintomatologia negativa e desorganizada, relacionada à proposta da esquizotaxia, esteve associada ao genótipo 2592C/C. Nosso estudo reforça as evidências da participação do sistema adenosinérgico na fisiopatologia da esquizofrenia, mais marcadamente na esquizotaxia, fornecendo evidências genéticas preliminares para esta teoria.
