Data are presented on concentration and composition of aliphatic and polycyclic aromatic hydrocarbons (HC) in water, suspended matter (collected with a Juday net and by a separator), and in bottom sediments of the White Sea. It was found that during the last years the level of aliphatic HC concentrations in waters of the White Sea (aver. 18 µg/l) practically did not change and was comparable with average concentrations in shelf areas of the World Ocean. In water and bottom sediments distribution of HC is determined by discharge of river marginal filters. Here sedimentation of the bulk of anthropogenic HC occurs. That is why a grain-size controlling factor is not active in the zone of the river depocenter (in particular, of the North Dvina River). The same reasons most probably may explain differences in degree of geochemical relationships between contents of TOC and HC in suspended matter and bottom sediments. After passing through marginal filters natural HC are dominant in all migration forms.

Materials from different spheres of the Earth are ultimately delivered to bottom sediments, which serve as a natural recorder of the functioning of other spheres and originate as a result of the accumulation of their substances. Sedimentary material and species of river-transported elements are subjected to dramatic reworking in marginal filters, where river and sea waters are mixed. These processes are most important for the Caspian Sea, where runoffs of rivers (especially the Volga River) and the intense development and transportation of hydrocarbon fuel by tankers and pipelines (related to the coastal petroleum industry in the Sumgait and Baku ports, Apsheron Peninsula) are potential sources of hydrocarbon pollution. Previously obtained data showed that the total content of hydrocarbon fraction (i.e., the sum of aliphatic hydrocarbons (AHC) and polycyclic aromatic hydrocarbons (PAH)) in bottom sediments varied within 29-1820 µg/g. The content of petroleum hydrocarbons in the northeastern Caspian region varied from 0.052 to 34.09 µg/g with the maximum content in the Tengiz field. The content of six polyarenes in the Volga delta sediments was no more than 40 ng/g. To determine the recent HC pollution of bottom sediments and trends in the functioning of the Volga marginal filter, in summer of 2003 and 2004 we analyzed bottom sediments (58 samples) in the river waterway; Kirovsk channel; Bakhtemir and Ikryanoe branches; tributaries of the Kizan, Chagan, and other rivers; and the Caspian seashore.

Abundances and compositions of aliphatic hydrocarbons (AHC) and polyaromatic hydrocarbons (PAH) were investigated in water and bottom sediments of the southwestern Amur Bay, Sea of Japan. Water contained 0-129 ?g/l AHC (average 42.2 ?g/l) and 5-85 ng/l PAH (average 18 ng/l). Bottom sediments contained 168-2098 ?g/g AHC and 7.2-1100 ng/g dry mass PAH. It was shown that input of anthropogenic hydrocarbons is better recorded by molecular markers than by distribution of AHC and PAH concentrations. Discovery of elevated hydrocarbon concentrations in the bottom water layer suggests that bottom sediments induced secondary contamination of the water body.

The Canadian economy is largely dependent on the distribution of large volumes of oil to domestic and international markets by a long network of pipelines. Unfortunately, accidents occur, and oil can leak or spill from these pipelines before it reaches its destination. Of particular concern are the long-term consequences of oil spills in freshwater, which include sinking of oil in water and the contamination of sensitive areas, such as where fish (e.g., salmon) deposit their eggs in gravel-dominated river sediments. There is a knowledge gap regarding the fate and behaviour of oil in river sediment. To this end, this study aimed at finding the potential for diluted bitumen (dilbit) oil to become trapped in gravel and to transfer hydrocarbons into water by dissolution, which are harmful to aquatic life. Two sets of laboratory experiments were conducted to simulate conditions of an oil spill on an exposed bank or in shallow water. In the first set, by conducting capillary pressure-saturation (Pc-Sw) experiments it was found that dilbit can enter gravel pores without much resistance and approximately 14% of the pore volume can be occupied by discontinuous single or multipore blobs of dilbit following imbibition of water. Air-water Pc-Sw experiments done in laboratory 1-D columns required gravity correction and could be successfully scaled to predict dilbit-water Pc-Sw curves, except for the trapped amount of dilbit. Trapped dilbit constituents can be dissolved into the water flowing through gravel pores (hyporheic flow) at different velocities. In the second set, dissolution experiments suggested that for the duration of the test, hydrocarbons that cause acute toxicity dissolve rapidly, likely resulting in a decrease in their effective solubility. However, dilbit saturation changed only <2% within that time. Chronically toxic PAH compounds were also detected in the effluent water. The total concentration of all detected PAHs and alkylPAHs exceeded the threshold literature value to protect early-life stage fish. Observations of decreased concentrations with increased aqueous velocities as well as less than equilibrium concentrations indicated that the mass transfer was rate-limited. A correlation was developed for the mass transfer rate coefficient to understand the mass transfer behaviour beyond the conditions used in the experiments, which had a Reynolds number exponent similar to the studies of NAPL dissolution in groundwater.

Vascular smooth muscle cell (VSMC) behaviour and phenotypic modulation is critical to vessel repair following damage, and the progression of various cardiovascular diseases. The second messenger cyclic adenosine monophosphosphate (cAMP) plays a key role in VSMC function under the synthetic/activated phenotype, which is typically associated with unhealthy cell behaviour. Consequently, cAMP signaling is often targeted in attempts to impact several pathological diseases, including atherosclerosis, restenosis, and pulmonary arterial hypertension (PAH). The cyclic nucleotide phosphodiesterases (PDEs) catalyze hydrolysis of cAMP to an inactive form, and therefore directly regulate cAMP signaling. The PDE4D family dominates in synthetic VSMCs, and there is considerable interest in determining how distinct PDE4D isoforms affect cell function. Specifically, we are interested in the potential link between short isoforms of PDE4D and VSMC desensitization to pharmacological agents that impact cardiovascular disease via cAMP signaling. This study extends on previous work that assessed the expression of PDE4D splice variants in rat aortic VSMCs following prolonged challenge with cAMP-elevating agents. It was determined that PDE4D1 and PDE4D2 were uniquely expressed in synthetic VSMCs incubated with these agents, and that this upregulation impacted PDE activity and cAMP accumulation in these cells. Here, we report that PDE4D1 and PDE4D2 are markedly upregulated in synthetic human aortic smooth muscle cells (HASMCs) following prolonged challenge with cAMP-elevating agents. Using a combination of RNAi-based and pharmacological approaches, we establish that this upregulation is reflected in levels of cAMP PDE activity, and restricted to the cytosolic sub-cellular compartment. Our results suggest a role for localized PDE4D1 and PDE4D2 activity in regulating cAMP-mediated desensitization in HASMCs, and highlight their therapeutic potential in treating various cardiovascular diseases.

Los desahucios en España se han convertido en los últimos años en un fenómeno social emergente, de interés tanto para la investigación como para la intervención de los profesionales de la acción social. Aun así, no existen estudios suficientes relacionados con esta situación adversa, y menos aún centrados en las respuestas resilientes que las personas son capaces de ofrecer ante ella. Con el objetivo de identificar los factores internos y externos que se presentan en la experiencia de las familias que viven procesos de desahucios y que les están permitiendo desarrollar estrategias resilientes ante dicha adversidad, se lleva a cabo la presente investigación en la que participan 20 sujetos, pertenecientes a familias que acuden a la Plataforma de Afectados por las Hipotecas (PAH) de Málaga. A los participantes se les realiza una entrevista semi-estructurada y a través de ellas se han podido definir las características socio-familiares de las personas afectadas, las diferentes estrategias de afrontamiento desarrolladas, las principales fuentes de apoyo con las que han contado, así como las preocupaciones que les han acompañado. Los resultados muestran los principales factores internos y externos que están presenten en las estrategias resilientes que han desarrollado las familias afectadas por los desahucios durante el proceso vivido y destacan como fuente de apoyo informacional a la PAH. Se concluye con la necesidad de continuar con esta línea de investigación para tratar de diseñar intervenciones que refuercen y fomenten las estrategias de afrontamiento ante la situación adversa del desahucio.

La actual crisis sistémica ha operado como una estructura de oportunidad sobre la que han cristalizado nuevos discursos, nuevas prácticas y nuevas estructuras políticas. La irrupción del 15M, de la Plataforma de Afectados por la Hipoteca (PAH) o de las llamadas “mareas” han permitido la visibilización de modos de hacer política que marcan una diferencia explícita con las prácticas asociadas al establishment político del llamado “régimen del 78”. Algunas de las prácticas popularizadas por estos movimientos han llegado a ser asumidas por amplias capas de la población. Entre ellas, destaca la asamblea como método legítimo de toma de decisiones. En esta comunicación planteamos un análisis de la asamblea como ritual político, deteniéndonos en la descripción de sus lógicas organizativas, principios de legimitación y características formales. A través de datos etnográficos recabados en la ciudad de Alcalá de Guadaíra (Sevilla) rastreamos la trama específica de discursos, prácticas y relaciones observables en el seno de las asambleas organizadas por el movimiento 15M. Finalmente, y a través de estos datos, reflexionamos sobre el sentido político del ritual en tiempos de crisis.

The source, concentration, and potential impact of sewage discharge and incomplete organic matter (OM) combustion on sedimentary microbial populations were assessed in Dublin Bay, Ireland. Polycyclic aromatic hydrocarbons (PAHs) and faecal steroids were investigated in 30 surface sediment stations in the bay. Phospholipid fatty acid (PLFA) content at each station was used to identify and quantify the broad microbial groups present and the impact of particle size, total organic carbon (%TOC), total hydrogen (%H) and total nitrogen (%N) was also considered. Faecal sterols were found to be highest in areas with historical point sources of sewage discharge. PAH distribution was more strongly associated with areas of deposition containing high %silt and %clay content, suggesting that PAHs are from diffuse sources such as rainwater run-off and atmospheric deposition. The PAHs ranged from 12 to 3072 ng/g, with 10 stations exceeding the suggested effect range low (ERL) for PAHs in marine sediments. PAH isomer pair ratios and sterol ratios were used to determine the source and extent of pollution. PLFAs were not impacted by sediment type or water depth but were strongly correlated to, and influenced by PAH and sewage levels. Certain biomarkers such as 10Me16:0, i17:0 and a17:0 were closely associated with PAH polluted sediments, while 16:1ω9, 16:1ω7c, Cy17:0, 18:1ω6, i16:0 and 15:0 all have strong positive correlations with faecal sterols. Overall, the results show that sedimentary microbial communities are impacted by anthropogenic pollution.

Bacteria that degrade polycyclic aromatic hydrocarbons (PAHs) in the estuarine surface microlayer (SML) of the Ria de Aveiro, Portugal—which is chronically polluted with oil hydrocarbons (OH)—were isolated and characterized; Pseudomonas was dominant among the PAH-degrading bacteria. Screening for PAH dioxygenase genes detected almost identical nahAc genes (encoding the alpha subunits of naphthalene dioxygenase) in 2 phylogenetically distinct isolates: Pseudomonas sp. and an unknown species of the family Enterobacteriaceae; this suggested that horizontal transfer of nah genes might be involved in PAH degradation in the SML. We also investigated the effect of PAH contamination on the spatial variability of the bacterioneuston along a gradient of pollution in the estuarine system of the Ria de Aveiro. Culture-independent techniques—fluorescence in situ hy - bridization (FISH) and denaturing-gradient gel electrophoresis (DGGE)—revealed a similar structure among the bacterioneuston communities along the estuary. In contrast, we detected differences in the relative abundance and diversity of organisms of the Gammaproteobacteria, including those of the genus Pseudomonas (which belongs to the Gammaproteobacteria). This is the first insight into the hydrocarbonoclastic bacterial communities in the SML of an estuarine area polluted with hydrocarbons. Our findings highlight the importance of SML-adapted hydrocarbonoclastic bacterioneuston as a potential source of new PAH-degrading bacteria (including new pseudomonads) with potential use in the bioremediation of hydrocarbon-polluted ecosystems.

L’hypertension artérielle pulmonaire (HTAP) est une maladie caractérisée par l’augmentation progressive des résistances vasculaires pulmonaires causant une augmentation de la pression artérielle pulmonaire qui mène au décès prématuré des patients. Malgré une amélioration rapide ces dernières années des traitements spécifiques, les patients souffrant d’HTAP demeurent dyspnéiques et intolérants à l’effort. L’atteinte vasculaire pulmonaire est actuellement irréversible. Elle est également la source de plusieurs anomalies au niveau des systèmes cardiovasculaires, ventilatoires et musculaires constituant les principaux déterminants physiologiques de la capacité à l’effort des patients. Cette thèse a investigué différentes facettes de la tolérance à l’effort en HTAP : les différents mécanismes ayant un impact sur l’apport musculaire en oxygène, l’altération des voies de signalisation cellulaire impliquées dans l’angiogenèse musculaire et les mécanismes ayant un impact sur la régulation du débit sanguin et l’oxygénation cérébrale en HTAP. Nous avons premièrement documenté une diminution de l’apport en oxygène aux muscles squelettiques à l’effort des patients en relation avec une diminution de la densité capillaire musculaire. Ce défaut d’angiogenèse corrélait d’ailleurs avec la capacité à l’effort des sujets. Par la suite, nous avons étudié les voies de signalisations cellulaires de l’angiogenèse musculaire. Ces résultats ont permis de démontrer une diminution de l’expression de miR-126, unique aux patients HTAP, qui était responsable de la diminution de la densité capillaire et qui contribuait à leur intolérance à l’effort. De plus, il était possible de moduler in vivo l’expression de miR-126. L’expérimentation in vivo, à l’aide d’un modèle murin d’HTAP, a permis de rétablir l’expression de miR-126, d’augmenter la microcirculation musculaire et d’améliorer la tolérance à l’effort des animaux, ce qui met en lumière le potentiel thérapeutique de l’angiogenèse musculaire pour améliorer la capacité à l’effort en HTAP. Notre dernier projet a démontré que les patients HTAP présentaient une diminution de débit sanguin cérébral. Ce projet a également démontré que les changements de pression artérielle sont moins bien amortis par les vaisseaux cérébraux des patients et que leurs vaisseaux cérébraux étaient moins réactifs aux changements de CO2. Les patients présentaient aussi une augmentation de la sensibilité des chémorécepteurs centraux qui contribuait à augmenter leur ventilation au repos, mais aussi à l’exercice. Finalement, à l’effort, nous avons démontré que le débit sanguin cérébral des patients HTAP était principalement influencé par la pression artérielle alors que chez les sujets sains, le débit sanguin cérébral était influencé principalement par la PETCO2. Nous avons également démontré que les patients HTAP présentaient une diminution progressive de leur oxygénation cérébrale, qui corrélait avec leur capacité à l’effort. Les résultats obtenus au cours de ce doctorat démontrent bien que la capacité à l’effort en HTAP est aussi déterminée par plusieurs anomalies physiopathologiques périphériques.

Contaminantes orgânicos, como os hidrocarbonetos policíclicos aromáticos (HPAs), podem atingir corpos da água e possuem potencial para causar efeitos tóxicos em organismos. A exposição aos HPAs causa indução nos níveis de citocromo P450 1A (CYP1A) em peixes, e portanto, é utilizado como um biomarcador de contaminação ambiental. O guarú Phalloceros caudimaculatus ocorre naturalmente em ambientes aquáticos dulcícolas e mixohalinos na América do Sul. O presente estudo identificou a sequência nucleotídica do transcrito CYP1A de P. caudimaculatus, que codifica uma proteína com 521 aminoácidos, e que apresenta 91% e 70% de identidade com CYP1A de killifish e paulistinha, respectivamente. A partir desta sequência foi possível realizar a avaliação dos níveis de mRNA de CYP1A deste peixe por RTq-PCR. Foi realizada uma caracterização de sua indução órgão- e tempo-dependente frente a exposição ao HPA beta-naftoflavona (BNF) e ao elutriato preparado a partir de sedimento de dois corpos da água possivelmente contaminados com HPAs. Foi constatado um aumento significativo nos níveis de mRNA de CYP1A em fígado, brânquia, intestino, cérebro, nadadeira anal de macho adultos e em alevinos na primeira hora de exposição a 1 µM de BNF, em relação ao grupo controle. O rim e as nadadeiras caudal e dorsal apresentaram indução de CYP1A após duas horas de exposição ao BNF. As maiores induções nos peixes dos grupos expostos ao BNF em relação ao controle foram de 176 no rim e 122 vezes no cérebro, observadas respectivamente após 8 e 48 horas de exposição. Os níveis de mRNA de CYP1A nos órgãos e tecidos de alevino, mantiveramse induzidos pela exposição ao BNF até o final das 96 horas de exposição. A exposição dos peixes ao elutriato produzido a partir dos sedimentos coletados em dois locais potencialmente contaminados causou indução do CYP1A no fígado de 22 e 122 vezes em relação ao controle. Os resultados demonstram que a indução de CYP1A em Phalloceros caudimaculatus ocorre em tempos curtos de exposição, além da variação de acordo com o tempo de exposição e com o órgão analisado. Além disso, foi demonstrado que tecidos externos também podem ser utilizados para tais análises e que o elutriato feito a partir de sedimento de locais que recebem descargas de contaminantes podem causar indução de CYP1A nos organismos.

The explosion of the Deepwater Horizon (DWH) oil platform resulted in large amounts of crude oil and dispersant Corexit 9500A® released into the Gulf of Mexico and coincided with the spawning season of the oyster, Crassostrea virginica. The effects of exposing gametes and embryos of C. virginica to dispersant alone (Corexit), mechanically (HEWAF) and chemically dispersed (CEWAF) DWH oil were evaluated. Fertilization success and the morphological development, growth, and survival of larvae were assessed. Gamete exposure reduced fertilization (HEWAF: EC201 h = 1650 μg tPAH50 L− 1; CEWAF: EC201 h = 19.4 μg tPAH50 L− 1; Corexit: EC201 h = 6.9 mg L− 1). CEWAF and Corexit showed a similar toxicity on early life stages at equivalent nominal concentrations. Oysters exposed from gametes to CEWAF and Corexit experienced more deleterious effects than oysters exposed from embryos. Results suggest the presence of oil and dispersant during oyster spawning season may interfere with larval development and subsequent recruitment.

This thesis presents an analysis of the largest catalog to date of infrared spectra of massive young stellar objects in the Large Magellanic Cloud. Evidenced by their very different spectral features, the luminous objects span a range of evolutionary states from those most embedded in their natal molecular material to those that have dissipated and ionized their surroundings to form compact HII regions and photodissociation regions. We quantify the contributions of the various spectral features using the statistical method of principal component analysis. Using this analysis, we classify the YSO spectra into several distinct groups based upon their dominant spectral features: silicate absorption (S Group), silicate absorption and fine-structure line emission (SE), polycyclic aromatic hydrocarbon (PAH) emission (P Group), PAH and fine-structure line emission (PE), and only fine-structure line emission (E). Based upon the relative numbers of sources in each category, we are able to estimate the amount of time massive YSOs spend in each evolutionary stage. We find that approximately 50% of the sources have ionic fine-structure lines, indicating that a compact HII region forms about half-way through the YSO lifetime probed in our study. Of the 277 YSOs we collected spectra for, 41 have ice absorption features, indicating they are surrounded by cold ice-bearing dust particles. We have decomposed the shape of the ice features to probe the composition and thermal history of the ice. We find that most the CO2 ice is embedded a polar ice matrix that has been thermally processed by the embedded YSO. The amount of thermal processing may be correlated with the luminosity of the YSO. Using the Australia Telescope Compact Array, we imaged the dense gas around a subsample of our sources in the HII complexes N44, N105, N113, and N159 using HCO+ and HCN as dense gas tracers. We find that the molecular material in star forming environments is highly clumpy, with clumps that range from subparsec to ~2 parsecs in size and with masses between 10^2 to 10^4 solar masses. We find that there are varying levels of star formation in the clumps, with the lower-mass clumps tending to be without massive YSOs. These YSO-less clumps could either represent an earlier stage of clump to the more massive YSO-bearing ones or clumps that will never form a massive star. Clumps with massive YSOs at their centers have masses larger than those with massive YSOs at their edges, and we suggest that the difference is evolutionary: edge YSO clumps are more advanced than those with YSOs at their centers. Clumps with YSOs at their edges may have had a significant fraction of their mass disrupted or destroyed by the forming massive star. We find that the strength of the silicate absorption seen in YSO IR spectra feature is well-correlated with the on-source HCO+ and HCN flux densities, such that the strength of the feature is indicative of the embeddedness of the YSO. We estimate that ~40% of the entire spectral sample has strong silicate absorption features, implying that the YSOs are embedded in circumstellar material for about 40% of the time probed in our study.

Pulmonary hypertension (PH) is a rare but serious condition that causes progressive right ventricular (RV) failure and death. PH may be idiopathic, associated with underlying connective-tissue disease or hypoxic lung disease, and is also increasingly being observed in the setting of heart failure with preserved ejection fraction (HFpEF). The management of PH has been revolutionised by the recent development of new disease-targeted therapies which are beneficial in pulmonary arterial hypertension (PAH), but can be potentially harmful in PH due to left heart disease, so accurate diagnosis and classification of patients is essential. These PAH therapies improve exercise capacity and pulmonary haemodynamics, but their overall effect on the right ventricle remains unclear. Current practice in the UK is to assess treatment response with 6-minute walk test and NYHA functional class, neither of which truly reflects RV function. Cardiac magnetic resonance (CMR) imaging has been established as the gold standard for the evaluation of right ventricular structure and function, but it also allows a non-invasive and accurate study of the left heart. The aims of this thesis were to investigate the use of CMR in the diagnosis of PH, in the assessment of treatment response, and in predicting survival in idiopathic and connective-tissue disease associated PAH. In Chapter 3, a left atrial volume (LAV) threshold of 43 ml/m2 measured with CMR was able to distinguish idiopathic PAH from PH due to HFpEF (sensitivity 97%, specificity 100%). In Chapter 4, disease-targeted PAH therapy resulted in significant improvements in RV and left ventricular ejection fraction (p<0.001 and p=0.0007, respectively), RV stroke volume index (p<0.0001), and left ventricular end-diastolic volume index (p=0.0015). These corresponded to observed improvements in functional class and exercise capacity, although correlation coefficients between Δ 6MWD and Δ RVEF or Δ LVEDV were low. Finally, in Chapter 5, one-year and three-year survival was worse in CTD-PAH (75% and 53%) than in IPAH (83% and 74%), despite similar baseline clinical characteristics, lung function, pulmonary haemodynamics and treatment. Baseline right ventricular stroke volume index was an independent predictor of survival in both conditions. The presence of LV systolic dysfunction was of prognostic significance in CTD-PAH but not IPAH, and a higher LAV was observed in CTD-PAH suggesting a potential contribution from LV diastolic dysfunction in this group.

The concentrations of six urinary monohydroxyl metabolites (OH-PAHs) of polycyclic aromatic hydrocarbons, namely 1-hydroxynaphthalene, 1-hydroxyacenaphthene, 2-hydroxyfluorene, 1-hydroxyphenanthrene, 1-hydroxypyrene (1OHPy), and 3-hydroxybenzo[a]pyrene, were assessed in the post-shift urine of wildland firefighters involved in fire combat activities at six Portuguese fire corporations, and compared with those of non-exposed subjects. Overall, median levels of urinary individual and total OH-PAHs (ΣOH-PAHs) suggest an increased exposure to polycyclic aromatic hydrocarbons during firefighting activities with ΣOH-PAH levels in exposed firefighters 1.7-35 times higher than in non-exposed ones. Urinary 1-hydroxynaphthalene and/or 1-hydroxyacenapthene were the predominant compounds, representing 63-98% of ΣOH-PAHs, followed by 2-hydroxyfluorene (1-17%), 1-hydroxyphenanthrene (1-13%), and 1OHPy (0.3-10%). A similar profile was observed when gender discrimination was considered. Participation in fire combat activities promoted an increase of the distribution percentage of 1-hydroxynaphthalene and 1-hydroxyacenaphthene, while contributions of 1-hydroxyphenanthrene and 1OHPy decreased. The detected urinary 1OHPy concentrations (1.73×10(-2) to 0.152μmol/mol creatinine in exposed subjects versus 1.21×10(-2) to 5.44×10(-2)μmol/mol creatinine in non-exposed individuals) were lower than the benchmark level (0.5μmol/mol creatinine) proposed by the American Conference of Governmental Industrial Hygienists. This compound, considered the biomarker of exposure to PAHs, was the less abundant one from the six analyzed biomarkers. Thus the inclusion of other metabolites, in addition to 1OHPy, in future studies is suggested to better estimate firefighters' occupational exposure to PAHs. Moreover, strong to moderate Spearman correlations were observed between individual compounds and ΣOH-PAHs corroborating the prevalence of an emission source.
