Sendo a malária uma doença infecciosa que põe em risco uma elevada percentagem da população mundial, a presente dissertação apresenta o desenvolvimento de um teste de diagnóstico rápido (RDT) para a detecção da infecção. Para a detecção dos antigénios de Plasmodium falciparum (Pf), parasita da malária, foram estudadas nanopartículas de ouro (AuNPs) com dois diâmetros médios diferentes, 17nm e 48nm, funcionalizadas e conjugadas com anticorpos específicos, formando os bionanoconjugados. Este trabalho abrange dois focos de investigação, interligados, de forma a atingir-se o objectivo prosposto: i) Caracterização físico-quimica das AuNPs e dos bionanoconjugados em solução, usando a espectrocopia de UV-Visível, variando a força iónica e o pH do meio. A compreensão do comportamento e estabilidade das partículas consoante o seu diâmetro e revestimento tem em vista o melhoramento do RDT, tanto em sensibilidade como em custo; ii) Desenvolvimento do RDT usando um método que envolve uma ligação competitiva entre o antigénio e o anticorpo específico imobilizado à superfície das AuNPs. Os antigénios correspondem a biomarcadores da presença de Pf: Heat Shock Protein 70 (PfHsp70) e Histidine Rich Protein II (PfHRPII). Como superfícies para o desenvolvimento do RDT foram usados a nitrocelulose e o papel de filtro. Quanto maior é o diâmetro das AuNPs, maior é a estabilidade apresentada em solução quando aumentada a força iónica. Quando é variado o valor de pH do meio, as partículas adoptam um comportamento dependente do seu revestimento, independente do diâmetro. Os bionanoconjugados formados com AuNPs de ambos os diâmetros revelaram-se muito estáveis em solução, numa gama de forças iónicas de 0 a 0,5M de cloreto de sódio, e de pH de 2 a 7. Uma vez que a proteína Hsp70 é produzida constitutivamente em diversas células humanas, é necessário trabalhar com proteínas específicas da infecção, que nesta dissertação foi a PfHRPII. Foram efectuados testes no suporte de nitrocelulose que provaram o reconhecimento da PfHRPII pelos bionanoconjugados AuNPs-MUA-anti-PfHRPII. Estes ensaios necessitam ainda de algumas optimizações. Ensaios de Western-Blot permitiram a identificação da presença da PfHRPII em culturas infectadas, bem como a confirmação da sua ligação ao anticorpo específico, anti-PfHRPII. Um novo método de revelação é introduzido nesta técnica, efectuado através do uso dos bionanoconjugados. Este método mostrou-se promissor na medida em que é rápido, específico e de baixo custo. O principal contributo do trabalho consistiu no desenvolvimento, com sucesso, de um RDT para diagnóstico de malária, usando o antigénio PfHsp70, tendo como base a detecção colorimétrica com AuNPs de 17nm de diâmetro. Usando uma tira de nitrocelulose foi estabelecido um limite de detecção correspondente a 1600parasitas/μL, valor este considerado válido pela OMS para um correcto diagnóstico de malária. O limite de detecção obtido para as AuNPs de 48nm foi superior, não tendo trazido melhorias para o teste desenvolvido com AuNPs de menor diâmetro.

Grapevine (Vitis vinifera) is one of most agro-economically important fruit crops worldwide, with a special relevance in Portugal where over 300 varieties are used for wine production. Due to global warming, temperature stress is currently a serious issue affecting crop production especially in temperate climates. Mobile genetic elements such as retrotransposons have been shown to be involved in environmental stress induced genetic and epigenetic modifications. In this study, sequences related to Grapevine Retrotransposon 1 (Gret1) were utilized to determine heat induced genomic and transcriptomic modifications in Touriga Nacional, a traditional Portuguese grapevine variety. For this purpose, growing canes were treated to 42 oC for four hours and leaf genomic DNA and RNA was utilized for various techniques to observe possible genomic alterations and variation in transcription levels of coding and non-coding sequences between non-treated plants and treated plants immediately after heat stress (HS-0 h) or after a 24 hour recovery period (HS-24 h). Heat stress was found to induce a significant decrease in Gret1 related sequences in HS-24 h leaves, indicating an effect of heat stress on genomic structure. In order to identify putative heat induced DNA modifications, genome wide approaches such as Amplified Fragment Length Polymorphism were utilized. This resulted in the identification of a polymorphic DNA fragment in HS-0 h and HS-24 h leaves whose sequence mapped to a genomic region flanking a house keeping gene (NADH) that is represented in multiple copies in the Vitis vinifera genome. Heat stress was also found to affect the transcript levels of various non-coding and gene coding sequences. Accordingly, quantitative real time PCR results established that Gret1 related sequences are up regulated immediately after heat stress whereas the level of transcript of genes involved in identification and repair of double strand breaks are significantly down regulated in HS-0 h plants. Taken together, the results of this work demonstrated heat stress affects both genomic integrity and transcription levels.

INTRODUCTION: For a long time, the importance of Chagas disease in Mexico, where many regarded it as an exotic malady, was questioned. Considering the great genetic diversity among isolates of Trypanosoma cruzi, the importance of this biological characterization, and the paucity of information on the clinical and biological aspects of Chagas disease in Mexico, this study aimed to identify the molecular and biological characterization of Trypanosoma cruzi isolates from different endemic areas of this country, especially of the State of Jalisco. METHODS: Eight Mexican Trypanosoma cruzi strains were biologically and genetically characterized (PCR specific for Trypanosoma cruzi, multiplex-PCR, amplification of space no transcript of the genes of the mini-exon, amplification of polymorphic regions of the mini-exon, classification by amplification of intergenic regions of the spliced leader genes, RAPD - (random amplified polymorphic DNA). RESULTS: Two profiles of parasitaemia were observed, patent (peak parasitaemia of 4.6×10(6) to 10(7) parasites/mL) and subpatent. In addition, all isolates were able to infect 100% of the animals. The isolates mainly displayed tropism for striated (cardiac and skeletal) muscle. PCR amplification of the mini-exon gene classified the eight strains as TcI. The RAPD technique revealed intraspecies variation among isolates, distinguishing strains isolated from humans and triatomines and according to geographic origin. CONCLUSIONS: The Mexican T. cruzi strains are myotrophic and belong to group TcI.

A presente dissertação de mestrado visa reflectir acerca da construção e exposição da imagem feminina constante em três produtos de ficção, transmitidos pela televisão portuguesa desde a entrada no novo século. Considerando a televisão um dos meios de comunicação social mais dominantes da sociedade atual, a telenovela destaca-se da programação desde 1977, com a chegada de Gabriela, tornando-se, a partir de então, um dos produtos líderes de audiências, sendo portanto dos formatos mais consumidos pelos espetadores desse meio. Atua, na maior parte das vezes, como um veículo de representação social, uma forma particular de ver o mundo e um modo válido de denúncia das problemáticas sociais. A partir da transcrição de dois pequenos fragmentos de cada uma das três obras ficcionais selecionadas, pretende-se analisar e descrever os fenómenos e processos discursivos utilizados na representação das três personagens femininas, observando comportamentos e examinando relações sociais que possam conduzir à denúncia de situações de discriminação de género.

DNA microarrays are one of the most used technologies for gene expression measurement. However, there are several distinct microarray platforms, from different manufacturers, each with its own measurement protocol, resulting in data that can hardly be compared or directly integrated. Data integration from multiple sources aims to improve the assertiveness of statistical tests, reducing the data dimensionality problem. The integration of heterogeneous DNA microarray platforms comprehends a set of tasks that range from the re-annotation of the features used on gene expression, to data normalization and batch effect elimination. In this work, a complete methodology for gene expression data integration and application is proposed, which comprehends a transcript-based re-annotation process and several methods for batch effect attenuation. The integrated data will be used to select the best feature set and learning algorithm for a brain tumor classification case study. The integration will consider data from heterogeneous Agilent and Affymetrix platforms, collected from public gene expression databases, such as The Cancer Genome Atlas and Gene Expression Omnibus.

The present work evaluated several aspects of the generalized stress response [endocrine (cortisol), metabolic (glucose), hematologic (hematocrit and hemoglobin) and cellular (HSP70)] in the Amazonian warm-water fish matrinxã (Brycon amazonicus ) subjected to an acute cold shock. This species farming has been done in South America, and growth and feed conversion rates have been interesting. However, in subtropical areas of Brazil, where the water temperature can rapidly change, high rates of matrinxã mortality have been associated with abrupt decrease in the water temperature. Thus, we subjected matrinxã to a sudden cold shock by transferring the fish directly to tanks in which the water temperature was 10ºC below the initial conditions (cold shock from 28ºC to 18ºC). After 1h the fish were returned to the original tanks (28ºC). The handling associated with tank transfer was also imposed on control groups (not exposed to cold shock). While exposure to cold shock did not alter the measured physiological conditions within 1h, fish returned to the ambient condition (water at 28º C) significantly increased plasma cortisol and glucose levels. Exposure to cold shock and return to the warm water did not affect HSP70 levels. The increased plasma cortisol and glucose levels after returning the fish to warm water suggest that matrinxã requires cortisol and glucose for adaptation to increased temperature.

Prostate cancer (PCa) is one of the most incident cancers worldwide but clinical and pathological parameters have limited ability to discriminate between clinically significant and indolent PCa. Altered expression of histone methyltransferases and histone methylation patterns are involved in prostate carcinogenesis. SMYD3 transcript levels have prognostic value and discriminate among PCa with different clinical aggressiveness, so we decided to investigate its putative oncogenic role on PCa.We silenced SMYD3 and assess its impact through in vitro (cell viability, cell cycle, apoptosis, migration, invasion assays) and in vivo (tumor formation, angiogenesis). We evaluated SET domain's impact in PCa cells' phenotype. Histone marks deposition on SMYD3 putative target genes was assessed by ChIP analysis.Knockdown of SMYD3 attenuated malignant phenotype of LNCaP and PC3 cell lines. Deletions affecting the SET domain showed phenotypic impact similar to SMYD3 silencing, suggesting that tumorigenic effect is mediated through its histone methyltransferase activity. Moreover, CCND2 was identified as a putative target gene for SMYD3 transcriptional regulation, through trimethylation of H4K20.Our results support a proto-oncogenic role for SMYD3 in prostate carcinogenesis, mainly due to its methyltransferase enzymatic activity. Thus, SMYD3 overexpression is a potential biomarker for clinically aggressive disease and an attractive therapeutic target in PCa.

Renal cell tumors (RCTs) are the most lethal of the common urological cancers. The widespread use of imaging entailed an increased detection of small renal masses, emphasizing the need for accurate distinction between benign and malignant RCTs, which is critical for adequate therapeutic management. Histone methylation has been implicated in renal tumorigenesis, but its potential clinical value as RCT biomarker remains mostly unexplored. Hence, the main goal of this study was to identify differentially expressed histone methyltransferases (HMTs) and histone demethylases (HDMs) that might prove useful for RCT diagnosis and prognostication, emphasizing the discrimination between oncocytoma (a benign tumor) and renal cell carcinoma (RCC), especially the chromophobe subtype (chRCC). We found that the expression levels of three genes-SMYD2, SETD3, and NO66-was significantly altered in a set of RCTs, which was further validated in a large independent cohort. Higher expression levels were found in RCTs compared to normal renal tissues (RNTs) and in chRCCs comparatively to oncocytomas. SMYD2 and SETD3 mRNA levels correlated with protein expression assessed by immunohistochemistry. SMYD2 transcript levels discriminated RCTs from RNT, with 82.1% sensitivity and 100% specificity (AUC=0.959), and distinguished chRCCs from oncocytomas, with 71.0% sensitivity and 73.3% specificity (AUC: 0.784). Low expression levels of SMYD2, SETD3, and NO66 were significantly associated with shorter disease-specific and disease-free survival, especially in patients with non-organ confined tumors. We conclude that expression of selected HMTs and HDMs might constitute novel biomarkers to assist in RCT diagnosis and assessment of tumor aggressiveness.

Tau-mediated neurodegeneration is a central event in Alzheimer's disease (AD) and other tauopathies. Consistent with suggestions that lifetime stress may be a clinically-relevant precipitant of AD pathology, we previously showed that stress triggers tau hyperphosphorylation and accumulation; however, little is known about the etiopathogenic interaction of chronic stress with other AD risk factors, such as sex and aging. This study focused on how these various factors converge on the cellular mechanisms underlying tau aggregation in the hippocampus of chronically stressed male and female (middle-aged and old) mice expressing the most commonly found disease-associated Tau mutation in humans, P301L-Tau. We report that environmental stress triggers memory impairments in female, but not male, P301L-Tau transgenic mice. Furthermore, stress elevates levels of caspase-3-truncated tau and insoluble tau aggregates exclusively in the female hippocampus while it also alters the expression of the molecular chaperones Hsp90, Hsp70, and Hsp105, thus favoring accumulation of tau aggregates. Our findings provide new insights into the molecular mechanisms through which clinically-relevant precipitating factors contribute to the pathophysiology of AD. Our data point to the exquisite sensitivity of the female hippocampus to stress-triggered tau pathology.
