Uma das prioridades do Programa de Melhoramento Genético de Macieira da Epagri é a obtenção de cultivares resistentes à mancha foliar de glomerella - MFG, doença essa causada principalmente pela espécie Colletotrichum gloeosporioides, que tem causado elevadas perdas na produção. Os resultados obtidos neste estudo têm, dentre outras, a finalidade de subsidiar os programas de melhoramento genético da macieira na seleção de parentais, objetivando a incorporação de resistência genética à MFG nas futuras novas cultivares. Para tanto, o primeiro passo é a identificação de germoplasma portador dos genes de resistência. No presente trabalho, foram utilizados 3 isolados de C. gloeosporioides provenientes de diferentes regiões produtoras de maçã. Estes isolados foram inoculados em 245 acessos pertencentes ao Banco de Germoplasma de Macieira - BGM, da Epagri / Estação Experimental de Caçador - EECd, SC. Para a inoculação em tecido foliar, a concentração de conídios foi ajustada para 10³ esporos/mL. Após 72h de incubação, sob temperatura de 25ºC, foi realizada a avaliação da incidência para presença ou ausência de sintomas e para a severidade da doença. O delineamento experimental foi o inteiramente casualizado, com três repetições. As diferenças de severidade entre os acessos foram testadas através da Anova não paramétrica Kruskal Wallis. Entre os acessos testados, 187 (76,3%) manifestaram resistência e 58 (23,7%) suscetibilidade à MFG. Entre as cultivares suscetíveis, não houve diferença no grau de severidade de ataque da MFG.

O rambotã (Nephelium lappaceum L.), fruta originária da Ásia, é cultivado comercialmente no Brasil, nos Estados da Pará e Bahia, mas devido à propagação sexuada, apresenta grande variabilidade para características morfológicas e químicas dos frutos. Visando a identificar potenciais matrizes, foram avaliados, em Ituberá-BA, frutos de 105 genótipos de rambotã com base na coloração da casca, suculência e teor de sólidos solúveis. Nesse grupo de plantas, 80 genótipos (76,2%) apresentaram frutos com teor de sólidos solúveis igual ou superior a 16 ºBrix e tiveram uma amostra coletada para análise em laboratório. Após a pesagem, foram identificados 20 genótipos cujos frutos apresentaram peso médio acima de 30 g e foram submetidos às análises físico-químicas. Os genótipos avaliados apresentaram frutos com peso médio de 33,2 g (30,2 a 39,4 g), rendimento do arilo de 42,3% (35,1 a 50,2%), sólidos solúveis de 17,6 ºBrix (15,8 a 19,7 ºBrix) e acidez titulável 0,44% (0,19 a 0,86%). A amostra de rambuteiras avaliadas apresentou grande variabilidade fenotípica e permitiu a identificação de 20 19 ou 19,05% do total de genótipos avaliados, cujas características dos frutos atendem aos padrões estabelecidos em outros países (peso acima de 30 g e sólidos solúveis acima de 16 ºBrix).

Four formulations of mixed açaí (Euterpe oleracea) (A) and cocoa´s honey (Theobroma cacao) (CH) jellies were prepared according to the following proportions: T1 (40% A:60% CH), T2 (50% A:50% CH), T3 (60% A: 40% CH) and T4 (100% A - control). All formulations were prepared using a rate 60:40 (w/w) of sucrose and pulp, plus 0.5% pectin and the products reached to average of 65% soluble solids content. The jellies were analyzed by chemical and physicochemical (titratable acidity, pH, soluble solid content, dry matter, total protein, lipids, vitamin C and calories) and sensory characteristics; also were evaluated levels of P, K, Ca, Mg, Fe, Zn, Cu and Mn. It was used a hedonic scale of 7 points to evaluate the attributes: overall impression, spreadability, brightness, flavor, texture and color, and also was verified the purchase intention score. The titratable acidity and pH ranged from 0.46 to 0.64% and 3.35 to 3.64, respectively, that are within the range found at most fruit jellies. The soluble solids content ranged between 65.2 and 65.5 ºBrix. The sensory acceptance results showed that all treatments (T1, T2, T3 and T4) presented means of sensory attributes above 4, demonstrating good acceptance of the product, but the treatment T1 presented the higher scores for the evaluated attributes. Cocoa´s honey added a positive influence on the attributes of color, texture and spreadability.

O objetivo deste trabalho foi avaliar o efeito da inoculação de fungos micorrízicos arbusculares (Glomus clarum Nicolson e Schenck, Glomus etunicatum Becker e Gerd Gigaspora margarita Becker e Hall e Scutellospora heterogama Nicolson e Gerd) em mudas micropropagadas de mirtileiro da cultivar Woodard. O delineamento experimental utilizado foi o de blocos casualizados, com cinco tratamentos, quatro repetições por tratamento e seis plantas por parcela. As plantas inoculadas com G. margarita apresentaram maior altura e biomassa seca das raízes. Os tratamentos com S. heterogama e G. margarita apresentaram os maiores resultados para a biomassa verde das raízes. O desempenho foi relacionado com as taxas de colonização das raízes que, nas plantas inoculadas com G. margarita e S. heterogama, foram de 48,2 e 45,1%, respectivamente. Para os teores nutricionais, as plantas inoculadas com S. heterogama, G. etunicatum, G. clarum e G. margarita proporcionaram maiores teores de N e P na parte aérea das plantas.

Homozygosity has long been associated with rare, often devastating, Mendelian disorders, and Darwin was one of the first to recognize that inbreeding reduces evolutionary fitness. However, the effect of the more distant parental relatedness that is common in modern human populations is less well understood. Genomic data now allow us to investigate the effects of homozygosity on traits of public health importance by observing contiguous homozygous segments (runs of homozygosity), which are inferred to be homozygous along their complete length. Given the low levels of genome-wide homozygosity prevalent in most human populations, information is required on very large numbers of people to provide sufficient power. Here we use runs of homozygosity to study 16 health-related quantitative traits in 354,224 individuals from 102 cohorts, and find statistically significant associations between summed runs of homozygosity and four complex traits: height, forced expiratory lung volume in one second, general cognitive ability and educational attainment (P < 1 × 10(-300), 2.1 × 10(-6), 2.5 × 10(-10) and 1.8 × 10(-10), respectively). In each case, increased homozygosity was associated with decreased trait value, equivalent to the offspring of first cousins being 1.2 cm shorter and having 10 months' less education. Similar effect sizes were found across four continental groups and populations with different degrees of genome-wide homozygosity, providing evidence that homozygosity, rather than confounding, directly contributes to phenotypic variance. Contrary to earlier reports in substantially smaller samples, no evidence was seen of an influence of genome-wide homozygosity on blood pressure and low density lipoprotein cholesterol, or ten other cardio-metabolic traits. Since directional dominance is predicted for traits under directional evolutionary selection, this study provides evidence that increased stature and cognitive function have been positively selected in human evolution, whereas many important risk factors for late-onset complex diseases may not have been.

Cet article montre, en premier lieu, que le degré d'investissement politico-moral des policières et des policiers dans leur métier est fortement variable. Si une partie des individus nouvellement entrés dans la police expriment un sentiment de distance sociale doublé d'une distinction morale par rapport à la population, d'autres ne s'investissement pas dans leur métier comme dans une « croisade morale » (Becker, 1985 [1963]), et estiment au contraire que les justiciables appartiennent à la même « communauté morale » (Fassin, 2011, p. 313) qu'eux-mêmes. La perspective adoptée prolonge ainsi les critiques des approches classiques de la « culture policière », qui ont démontré leur caractère trop uniformisant et leurs biais mécaniste. Parmi les études de sociologie de la police n'ayant pas adopté une telle approche, celle de W. Ker Muir (1977), pourtant ancienne, a montré que tous les policiers ne vivaient pas leur appartenance au groupe professionnel sur le mode d'une distinction morale d'avec les justiciables. Aucune autre étude n'a pourtant traité des variations dans l'investissement politico moral des policières et des policiers, et n'a donc tenté d'en fournir des explications. Le second axe d'analyse vise ainsi à combler ce déficit explicatif, en montrant que l'investissement politico-moral dans le métier dépend largement de la trajectoire antérieure des individus recrutés dans la police, en particulier des expériences de pertes de statut qu'ils ont pu subir, ainsi que du type de stratégies d'ascension sociale ou de récupération de ce statut qu'ils mettent en place.

BACKGROUND: According to the gateway hypothesis, tobacco use is a gateway of cannabis use. However, there is increasing evidence that cannabis use also predicts the progression of tobacco use (reverse gateway hypothesis). Unfortunately, the importance of cannabis use compared to other predictors of tobacco use is less clear. The aim of this study was to examine which variables, in addition to cannabis use, best predict the onset of daily cigarette smoking in young men. METHODS: A total of 5,590 young Swiss men (mean age = 19.4 years, SD = 1.2) provided data on their substance use, socio-demographic background, religion, health, social context, and personality at baseline and after 18 months. We modelled the predictors of progression to daily cigarette smoking using logistic regression analyses (n = 4,230). RESULTS: In the multivariate overall model, use of cannabis remained among the strongest predictors for the onset of daily cigarette use. Daily cigarette use was also predicted by a lifetime use of at least 50 cigarettes, occasional cigarette use, educational level, religious affiliation, parental situation, peers with psychiatric problems, and sociability. CONCLUSIONS: Our results highlight the relevance of cannabis use compared to other potential predictors of the progression of tobacco use and thereby support the reverse gateway hypothesis.

Waddlia chondrophila is a obligate intracellular bacterium belonging to the Chlamydiales order, a clade that also includes the well-known classical Chlamydia responsible for a number of severe human and animal diseases. Waddlia is an emerging pathogen associated with adverse pregnancy outcomes in humans and abortion in ruminants. Adhesion to the host cell is an essential prerequisite for survival of every strict intracellular bacteria and, in classical Chlamydia, this step is partially mediated by polymorphic outer membrane proteins (Pmps), a family of highly diverse autotransporters that represent about 15% of the bacterial coding capacity. Waddlia chondrophila genome however only encodes one putative Pmp-like protein. Using a proteomic approach, we identified several bacterial proteins potentially implicated in the adhesion process and we characterized their expression during the replication cycle of the bacteria. In addition, we demonstrated that the Waddlia Pmp-like autotransporter as well as OmpA2 and OmpA3, two members of the extended Waddlia OmpA protein family, exhibit adhesive properties on epithelial cells. We hypothesize that the large diversity of the OmpA protein family is linked to the wide host range of these bacteria that are able to enter and multiply in various host cells ranging from protozoa to mammalian and fish cells.

The recent trend for journals to require open access to primary data included in publications has been embraced by many biologists, but has caused apprehension amongst researchers engaged in long-term ecological and evolutionary studies. A worldwide survey of 73 principal investigators (Pls) with long-term studies revealed positive attitudes towards sharing data with the agreement or involvement of the PI, and 93% of PIs have historically shared data. Only 8% were in favor of uncontrolled, open access to primary data while 63% expressed serious concern. We present here their viewpoint on an issue that can have non-trivial scientific consequences. We discuss potential costs of public data archiving and provide possible solutions to meet the needs of journals and researchers.

Temporal lobe epilepsy (TLE) is a common epilepsy syndrome with a complex etiology. Despite evidence for the participation of genetic factors, the genetic basis of TLE remains largely unknown. A role for the galanin neuropeptide in the regulation of epileptic seizures has been established in animal models more than two decades ago. However, until now there was no report of pathogenic mutations in GAL, the galanin-encoding gene, and therefore its role in human epilepsy was not established. Here, we studied a family with a pair of monozygotic twins affected by TLE and two unaffected siblings born to healthy parents. Exome sequencing revealed that both twins carried a novel de novo mutation (p.A39E) in the GAL gene. Functional analysis revealed that the p.A39E mutant showed antagonistic activity against galanin receptor 1 (GalR1)-mediated response, and decreased binding affinity and reduced agonist properties for GalR2. These findings suggest that the p.A39E mutant could impair galanin signaling in the hippocampus, leading to increased glutamatergic excitation and ultimately to TLE. In a cohort of 582 cases, we did not observe any pathogenic mutations indicating that mutations in GAL are a rare cause of TLE. The identification of a novel de novo mutation in a biologically-relevant candidate gene, coupled with functional evidence that the mutant protein disrupts galanin signaling, strongly supports GAL as the causal gene for the TLE in this family. Given the availability of galanin agonists which inhibit seizures, our findings could potentially have direct implications for the development of anti-epileptic treatment.

Human biomonitoring (HBM) is an effective tool for assessing actual exposure to chemicals that takes into account all routes of intake. Although hair analysis is considered to be an optimal biomarker for assessing mercury exposure, the lack of harmonization as regards sampling and analytical procedures has often limited the comparison of data at national and international level. The European-funded projects COPHES and DEMOCOPHES developed and tested a harmonized European approach to Human Biomonitoring in response to the European Environment and Health Action Plan. Herein we describe the quality assurance program (QAP) for assessing mercury levels in hair samples from more than 1800 mother-child pairs recruited in 17 European countries. To ensure the comparability of the results, standard operating procedures (SOPs) for sampling and for mercury analysis were drafted and distributed to participating laboratories. Training sessions were organized for field workers and four external quality-assessment exercises (ICI/EQUAS), followed by the corresponding web conferences, were organized between March 2011 and February 2012. ICI/EQUAS used native hair samples at two mercury concentration ranges (0.20-0.71 and 0.80-1.63) per exercise. The results revealed relative standard deviations of 7.87-13.55% and 4.04-11.31% for the low and high mercury concentration ranges, respectively. A total of 16 out of 18 participating laboratories the QAP requirements and were allowed to analyze samples from the DEMOCOPHES pilot study. Web conferences after each ICI/EQUAS revealed this to be a new and effective tool for improving analytical performance and increasing capacity building. The procedure developed and tested in COPHES/DEMOCOPHES would be optimal for application on a global scale as regards implementation of the Minamata Convention on Mercury.

Selon le modèle de demandes-contrôle (Karasek, 1979), les conditions de travail les plus néfastes sont caractérisées par des exigences mentales élevées et par une latitude décisionnelle faible. Peu d'études se sont penchées sur le rôle des ressources individuelles en lien avec ce modèle. Pour cette raison, notre étude examine les effets médiateurs des traits de la personnalité et des valeurs culturelles sur la relation entre les conditions psychosociales du travail et le bien-être. En comparant deux échantillons de travailleurs, en Suisse (n = 585) et en Afrique du Sud (n = 818), nos résultats indiquent que les mêmes ressources et vulnérabilités individuelles n'interagissent pas de la même manière avec l'environnement de travail pour les travailleurs suisse que ceux d'Afrique du Sud. Ces résultats peuvent contribuer à l'amélioration des compétences culturelles dans les organisations où la gestion de la diversité culturelle des travailleurs devient une réalité complexe.

Le syndrome douloureux fémoro-patellaire (SDFP) est l'une des causes les plus fréquentes de douleur antérieure du genou chez l'adolescent et l'adulte. De par son étiologie complexe, multifactorielle et encore mal comprise, sa prise en charge est un important challenge pour le praticien. Le diagnostic se fait principalement sur l'anamnèse et l'examen clinique du genou mais aussi de l'ensemble du membre inférieur, pouvant parfois nécessiter la réalisation d'une imagerie. Le traitement est dans la grande majorité des cas conservateur, principalement axé sur la rééducation avec de la physiothérapie ciblée et personnalisée. Le traitement chirurgical est réservé aux cas présentant une anomalie structurelle causale. Patellofemoral pain syndrome (PFPS) is one of the most frequent cause of anterior knee pain in adolescents and adults. Due to its complex etiology, which is multifactorial and still poorly understood, its management is a major challenge for the practitioner. The diagnosis is made primarily on the history and clinical examination of the knee, but also of the entire lower limb, which may sometimes require the completion of imaging. The treatment is mostly conservative, focussing on rehabilitation with targeted and personalized therapy. Surgical treatment is reserved for cases with a causal structural lesion.

The occurrence of cognitive disturbances upon CNS inflammation or infection has been correlated with increased levels of the cytokine tumor necrosis factor-α (TNFα). To date, however, no specific mechanism via which this cytokine could alter cognitive circuits has been demonstrated. Here, we show that local increase of TNFα in the hippocampal dentate gyrus activates astrocyte TNF receptor type 1 (TNFR1), which in turn triggers an astrocyte-neuron signaling cascade that results in persistent functional modification of hippocampal excitatory synapses. Astrocytic TNFR1 signaling is necessary for the hippocampal synaptic alteration and contextual learning-memory impairment observed in experimental autoimmune encephalitis (EAE), an animal model of multiple sclerosis (MS). This process may contribute to the pathogenesis of cognitive disturbances in MS, as well as in other CNS conditions accompanied by inflammatory states or infections.
