This research is concerned with assessing from a national perspective the role, work and historical impact of the Irish Red Cross Society (IRCS) between 1939 and 1971. During this period the IRCS discharged three primary functions: it provided first aid services both in war-time and peace-time; it pioneered public health and social care services; and acted as the State’s main agency for international humanitarian relief measures. Although primarily a national organisational history of the Society, it is not a history in isolation. A broader perspective demonstrates that the work undertaken by the IRCS has relevance to the medical, social, religious, cultural, political and diplomatic history of twentieth century Ireland. This study assesses the impact of a number of significant public health and social care initiatives which the IRCS implemented and developed since its inception and how most of these were subsequently developed independently by the State. During the early 1940s, the Society’s formation of a national blood transfusion service ultimately laid the foundations for the establishment of a national blood transfusion service. The Society’s steering of a national anti-tuberculosis campaign in the 1940s brought the issue of the eradication of TB to the fore and helped to change public attitudes towards the disease. The concept of caring for the needs of the elderly in Ireland was largely unknown until the IRCS began addressing the issue in the 1950s and, for more than two decades, was effectively the only organisation in the State that campaigned and introduced innovative services for the aged. The IRCS made a significant impact in terms of its commitment to the needs of refugees and the provision of international humanitarian relief from Ireland. The Society’s donation in 1945 of a fully equipped hospital to the population of Saint-Lo in France, its war-time overseas relief efforts and its post-war work for child refugees earned Ireland significant international recognition and prestige and, more importantly, justified Ireland’s war-time policy of neutrality. With Ireland’s admission to the UN, the government became more dependent on the IRCS to consolidate that position.

This text presents an analysis of aggregated membership’s dynamics for Spanish trade unions, using ECVT data, as well as union memberships’ trajectories, or members’ decisions about joining the organization, permanency and responsibilities, and subsequent attrition. For the analysis of trajectories we make use of information of the records of actual memberships and the record of quitting of CCOO, and of a survey-questionnaire to a sample of leavers of the same union. This study allows us to confirm a linkage between the decision and motivations to become union member, to participate in union activities, the time of permanency, and the motives to quit the organization. We also identify five types of union members’ trajectories, indicating that, far from views that assert a monolithic structure, unions are complex organizations.

A novel lysozyme exhibiting antifungal activity and with a molecular mass of 14.4 kDa in SDS–polyacrylamide gel electrophoresis was isolated from mung bean (Phaseolus mungo) seeds using a procedure that involved aqueous extraction, ammonium sulfate precipitation, ion exchange chromatography on CM-Sephadex, and high-performance liquid chromatography on POROS HS-20. Its N-terminal sequence was very different from that of hen egg white lysozyme. Its pI was estimated to be above 9.7. The specific activity of the lysozyme was 355 U/mg at pH 5.5 and 30 °C. The lysozyme exhibited a pH optimum at pH 5.5 and a temperature optimum at 55 °C. It is reported herein, for the first time, that a novel plant lysozyme exerted an antifungal action toward Fusarium oxysporum, Fusarium solani, Pythium aphanidermatum, Sclerotium rolfsii, and Botrytis cinerea, in addition to an antibacterial action against Staphylococcus aureus.

Carbon stable-isotope analysis showed that individual brown trout Salmo trutta in Loch Lomond adopted strategies intermediate to that of freshwater residency or anadromy, suggesting either repeated movement between freshwater and marine environments, or estuarine residency. Carbon stable-isotope (delta C-13) values from Loch Lomond brown trout muscle tissue ranged from those indicative of assimilation of purely freshwater-derived carbon to those reflecting significant utilization of marine-derived carbon. A single isotope, two-source mixing model indicated that, on average, marine C made a 33% contribution to the muscle tissue C of Loch Lomond brown trout. Nitrogen stable isotope, delta N-15, but not delta C-13 was correlated with fork length suggesting that larger fish were feeding at a higher trophic level but that marine feeding was not indicated by larger body size. A key for three putative species apparently found in three geographic areas, i.e. Coregonus clupeoides (in Scotland), Coregonus stigmaticus (in England), and Coregonus pennantii (in Wales) given in a recent review was tested quantitatively using 544 individuals from nine populations. The classification success of the key was very low (27%). It was concluded that there is currently no robust evidence for the recognition of the three putative species. Furthermore, the use of phenotypic characters alone to distinguish putative species in postglacial fish species such as those of the genus Coregonus that show homoplasy in many of these traits is questioned. In the absence of further evidence, it was concluded that a single highly variable species best describes the pattern of phenotypic variation in these U.K. populations. On this basis it is argued that taxonomic subdivision of U.K. European coregonids is inappropriate and that Coregonus lavaretus should prevail as the species name applicable to all populations.

Differences in stable-isotope values, morphology and ecology in whitefish Coregonus lavaretus were investigated between the three basins of Loch Lomond. The results are discussed with reference to a genetic investigation to elucidate any substructuring or spawning site fidelity. Foraging fidelity between basins of Loch Lomond was indicated by delta 13C and delta 15N values of C. lavaretus muscle tissue. There was, however, no evidence of the existence of sympatric morphs in the C. lavaretus population. A previous report of two C. lavaretus 'species' in Loch Lomond probably reflects natural variation between individuals within a single mixed population.

Recovery of cellulose fibres from paper mill effluent has been studied using common polysaccharides or biopolymers such as Guar gum, Xanthan gum and Locust bean gum as flocculent. Guar gum is commonly used in sizing paper and routinely used in paper making. The results have been compared with the performance of alum, which is a common coagulant and a key ingredient of the paper industry. Guar gum recovered about 3.86 mg/L of fibre and was most effective among the biopolymers. Settling velocity distribution curves demonstrated that Guar gum was able to settle the fibres faster than the other biopolymers; however, alum displayed the highest particle removal rate than all the biopolymers at any of the settling velocities. Alum, Guar gum, Xanthan gum and Locust bean gum removed 97.46%, 94.68%, 92.39% and 92.46% turbidity of raw effluent at a settling velocity of 0.5 cm/min, respectively. The conditions for obtaining the lowest sludge volume index such as pH, dose and mixing speed were optimised for guar gum which was the most effective among the biopolymers. Response surface methodology was used to design all experiments, and an optimum operational setting was proposed. The test results indicate similar performance of alum and Guar gum in terms of floc settling velocities and sludge volume index. Since Guar gum is a plant derived natural substance, it is environmentally benign and offers a green treatment option to the paper mills for pulp recycling.

The breast cancer susceptibility gene BRCA1 encodes a protein implicated in the cellular response to DNA damage, with postulated roles in homologous recombination as well as transcriptional regulation. To identify downstream target genes, we established cell lines with tightly regulated inducible expression of BRCA1. High-density oligonucleotide arrays were used to analyze gene expression profiles at various times following BRCA1 induction. A major BRCA1 target is the DNA damage-responsive gene GADD45. Induction of BRCA1 triggers apoptosis through activation of c-Jun N-terminal kinase/stress-activated protein kinase (JNK/SAPK), a signaling pathway potentially linked to GADD45 gene family members. The p53-independent induction of GADD45 by BRCA1 and its activation of JNK/SAPK suggest a pathway for BRCA1-induced apoptosis.

The Wilms tumor suppressor WT1 encodes a zinc finger transcription factor that is expressed in glomerular podocytes during a narrow window in kidney development. By immunoprecipitation and protein microsequencing analysis, we have identified a major cellular protein associated with endogenous WT1 to be the inducible chaperone Hsp70. WT1 and Hsp70 are physically associated in embryonic rat kidney cells, in primary Wilms tumor specimens and in cultured cells with inducible expression of WT1. Colocalization of WT1 and Hsp70 is evident within podocytes of the developing kidney, and Hsp70 is recruited to the characteristic subnuclear clusters that contain WT1. The amino-terminal transactivation domain of WT1 is required for binding to Hsp70, and expression of that domain itself is sufficient to induce expression of Hsp70 through the heat shock element (HSE). Substitution of a heterologous Hsp70-binding domain derived from human DNAJ is sufficient to restore the functional properties of a WT1 protein with an amino-terminal deletion, an effect that is abrogated by a point mutation in DNAJ that reduces binding to Hsp70. These observations indicate that Hsp70 is an important cofactor for the function of WT1, and suggest a potential role for this chaperone during kidney differentiation.

Ataxia telangiectasia (AT) is a recessive syndrome, including cerebellar degeneration, immunologic defects and cancer predisposition, attributed to mutations in the recently isolated ATM (ataxia telangiectasia, mutated) gene. AT is diagnosed in 1/40,000 to 1/100,000 live births, with carriers calculated to comprise approximately 1% of the population. Studies of AT families have suggested that female relatives presumed to be carriers have a 5 to 8-fold increased risk for developing breast cancer, raising the possibility that germline ATM mutations may account for approximately 5% of all breast cancer cases. The increased risk for breast cancer reported for AT family members has been most evident among younger women, leading to an age-specific relative risk model predicting that 8% of breast cancer in women under age 40 arises in AT carriers, compared with 2% of cases between 40-59 years. To test this hypothesis, we undertook a germ-line mutational analysis of the ATM gene in a population of women with early onset of breast cancer, using a protein truncation (PTT) assay to detect chain-terminating mutations, which account for 90% of mutations identified in children with AT. We detected a heterozygous ATM mutation in 2/202 (1%) controls, consistent with the frequency of AT carriers predicted from epidemiologic studies. ATM mutations were present in only 2/401 (0.5%) women with early onset of breast cancer (P = 0.6). We conclude that heterozygous ATM mutations do not confer genetic predisposition to early onset of breast cancer.

O presente trabalho apresenta novas metodologias desenvolvidas para a análise das propriedades magnéticas e magnetocalóricas de materiais, sustentadas em considerações teóricas a partir de modelos, nomeadamente a teoria de transições de fase de Landau, o modelo de campo médio molecular e a teoria de fenómeno crítico. São propostos novos métodos de escala, permitindo a interpretação de dados de magnetização de materiais numa perspectiva de campo médio molecular ou teoria de fenómeno crítico. É apresentado um método de estimar a magnetização espontânea de um material ferromagnético a partir de relações entropia/magnetização estabelecidas pelo modelo de campo médio molecular. A termodinâmica das transições de fase magnéticas de primeira ordem é estudada usando a teoria de Landau e de campo médio molecular (modelo de Bean-Rodbell), avaliando os efeitos de fenómenos fora de equilíbrio e de condições de mistura de fase em estimativas do efeito magnetocalórico a partir de medidas magnéticas. Efeitos de desordem, interpretados como uma distribuição na interacção magnética entre iões, estabelecem os efeitos de distribuições químicas/estruturais nas propriedades magnéticas e magnetocalóricas de materiais com transições de fase de segunda e de primeira ordem. O uso das metodologias apresentadas na interpretação das propriedades magnéticas de variados materiais ferromagnéticos permitiu obter: 1) uma análise quantitativa da variação de spin por ião Gadolínio devido à transição estrutural do composto Gd5Si2Ge2, 2) a descrição da configuração de cluster magnético de iões Mn na fase ferromagnética em manganites da família La-Sr e La-Ca, 3) a determinação dos expoentes críticos β e δ do Níquel por métodos de escala, 4) a descrição do efeito da pressão nas propriedades magnéticas e magnetocalóricas do composto LaFe11.5Si1.5 através do modelo de Bean-Rodbell, 5) uma estimativa da desordem em manganites ferromagnéticas com transições de segunda e primeira ordem, 6) uma descrição de campo médio das propriedades magnéticas da liga Fe23Cu77, 7) o estudo de efeitos de separação de fase na família de compostos La0.70-xErxSr0.30MnO3 e 8) a determinação realista da variação de entropia magnética na família de compostos de efeito magnetocalórico colossal Mn1-x-yFexCryAs.

O uso de polímeros naturais no âmbito da preparação de nanocompósitos não tem sido tão amplamente estudado quando comparado com os polímeros sintéticos. Assim, esta tese tem como objectivo estudar metodologias para a preparação de novos materiais nanocompósitos sob a forma de dispersões e filmes utilizando polissacarídeos como matriz. A tese está dividida em cinco capítulos sendo o último capítulo dedicado às conclusões gerais e a sugestões para trabalhos futuros. Inicialmente é apresentada uma breve revisão bibliográfica sobre os principais temas colocando esta tese em contexto. Considerações sobre o uso de polímeros naturais e a sua combinação com a utilização de nanopartículas inorgânicas para a fabricação de novos bionanocomposites são descritas e os objectivos e outline da tese são também apresentados. No segundo capítulo, a preparação de partículas de sílica puras ou modificadas bem como a sua caracterização por FTIR, SEM, TEM, TGA, DLS (tamanho e potencial zeta) e medições de ângulo de contacto são discutidas. De modo a melhorar a compatibilidade da sílica com os polissacarídeos, as partículas SiO2 foram modificados com dois compostos do tipo organosilano: 3- metacril-oxipropil-trimetoxissilano (MPS) e 3-aminopropil-trimetoxissilano (APS). As partículas SiO2@MPS foram posteriormente encapsuladas com de poli(metacrilato de glicidilo) utilizando a técnica de polimerização em emulsão. A utilização dos nanocompósitos resultantes na preparação de dispersões de bionanocompósitos não foi bem sucedida e por esse motivo não os estudos não foram prosseguidos. O uso de SiO2@APS na preparação de dispersões bionanocomposite foi eficiente. No terceiro capítulo é apresentada uma revisão sobre dispersões bionanocompósitas e respectiva caracterização destacando aspectos fundamentais sobre reologia e microestrutura. Em seguida, é discutido o estudo sistemático realizado sobre o comportamento reológico de dispersões de SiO2 utilizando três polissacarídeos distintos no que concerne a carga e as características gelificantes: a goma de alfarroba (não iónica), o quitosano (catiónico) e a goma xantana (aniónica) cujas propriedades reológicas são amplamente conhecidas. Os estudos reológicos realizados sob diferentes condições demonstraram que a formação de géis frágeis e/ou bem estruturados depende do tamanho SiO2, da concentração, do pH e da força iónica. Estes estudos foram confirmados por análises microestruturais usando a microscopia electrónica a baixas temperaturas (Cryo-SEM). No quarto capítulo, são apresentados os estudos relativos à preparação e caracterização de filmes bionanocompósitos utilizando quitosano como matriz. Primeiramente é apresentada uma revisão sobre filmes de bionanocompósitos e os aspectos fundamentais das técnicas de caracterização utilizadas. A escolha do plasticizante e da sua concentração são discutidas com base nas propriedades de filmes de quitosano preparados. Em seguida, o efeito da concentração de sílica e dos métodos utilizados para a dispersar na matriz de polissacarídeo, bem como o efeito da modificação da superfície da sílica é avaliado. As características da superfície e as propriedades de barreira, mecânicas e térmicas são discutidas para cada conjunto de filmes preparados antes e após a sua neutralização. Os resultados obtidos mostraram que a dispersão das cargas no plasticizante e posterior adição à matriz polissacarídica resultaram apenas em pequenas melhorias já que o problema da agregação de sílica não foi ultrapassado. Por esse motivo foram preparados filmes com SiO2@APS os quais apresentaram propriedades melhores apesar da agregação das partículas não ter sido completamente impedida. Tal pode estar relacionado com o processo de secagem dos filmes. Finalmente, no capítulo 5, são apresentadas as principais conclusões obtidas e algumas sugestões para trabalho futuro.
