Outer membrane vesicles (OMVs) are ubiquitously secreted from the outer membrane (OM) of Gram-negative bacteria. These heterogeneous structures are composed of OM filled with periplasmic content from the site of budding. By analyzing mutants that have vesicle production phenotypes, we can gain insight into the mechanism of OMV budding in wild-type cells, which has thus far remained elusive. In this study, we present data demonstrating that the hypervesiculation phenotype of the nlpI deletion mutant of Escherichia coli correlates with changes in peptidoglycan (PG) dynamics. Our data indicate that in stationary phase cultures the nlpI mutant exhibits increased PG synthesis that is dependent on spr, consistent with a model in which NlpI controls the activity of the PG endopeptidase Spr. In log phase, the nlpI mutation was suppressed by a dacB mutation, suggesting that NlpI regulates penicillin-binding protein 4 (PBP4) during exponential growth. The data support a model in which NlpI negatively regulates PBP4 activity during log phase, and Spr activity during stationary phase, and that in the absence of NlpI, the cell survives by increasing PG synthesis. Further, the nlpI mutant exhibited a significant decrease in covalent outer membrane (OM-PG) envelope stabilizing cross-links, consistent with its high level of OMV production. Based on these results, we propose that one mechanism wild-type Gram-negative bacteria can use to modulate vesiculation is by altering PG-OM cross-linking via localized modulation of PG degradation and synthesis.

This study is part of the area of research in Psychology of Mathematics Education that investigates, among other things, knowledge relating to the formation of mathematical concepts. The objective was to investigate the conceptual knowledge of polygons of 76 high school students in terms of defining attributes and examples and non-examples. The instruments for collecting data was a test with two questions about polygons, defining attributes of a test and a test of examples and non-examples, based on the theory of Klausmeier and Goodwin (1977) on formation of concepts. The results showed that participants of the survey had difficulties in identifying defining attributes of polygons and non-discriminating examples of examples, showing the formation of this concept to the level of identity.

En éste trabajo se reportan resultados de la investigación que referencia el título. El proyecto se desarrolló en estudiantes de noveno grado, de educación básica, a través de situaciones problema del contexto sociocultural y de las ciencias, bajo un diseño cualitativo y en las tres fases ; diseño y aplicación de una prueba diagnóstica, para reconocimiento de posibles dificultades de los estudiantes, intervención en el aula, para superación de las dificultades detectadas, y una prueba de contraste, para valorar el logro de las estrategias aplicadas y obtener información para mejoramiento del aprendizaje de los estudiantes. Los resultados muestran avances significativos de los estudiantes en cuanto a la comprensión de los conceptos, procedimientos y aplicaciones del pensamiento métrico.

The performance of loadsharing algorithms for heterogeneous distributed systems is investigated by simulation. The systems considered are networks of workstations (nodes) which differ in processing power. Two parameters are proposed for characterising system heterogeneity, namely the variance and skew of the distribution of processing power among the network nodes. A variety of networks are investigated, with the same number of nodes and total processing power, but with the processing power distributed differently among the nodes. Two loadsharing algorithms are evaluated, at overall system loadings of 50% and 90%, using job response time as the performance metric. Comparison is made with the ideal situation of ‘perfect sharing’, where it is assumed that the communication delays are zero and that complete knowledge is available about job lengths and the loading at the different nodes, so that an arriving job can be sent to the node where it will be completed in the shortest time. The algorithms studied are based on those already in use for homogeneous networks, but were adapted to take account of system heterogeneity. Both algorithms take into account the differences in the processing powers of the nodes in their location policies, but differ in the extent to which they ‘discriminate’ against the slower nodes. It is seen that the relative performance of the two is strongly influenced by the system utilisation and the distribution of processing power among the nodes.

Over recent years there has been an increase in the use of generic Computational Fluid Dynamics (CFD) software packages spread across various application fields. This has created the need for the integration of expertise into CFD software. Expertise can be integrated into CFD software in the form of an Intelligent Knowledge-Based System (IKBS). The advantages of integrating intelligence into generic engineering software are discussed with a special view to software engineering considerations. The software modelling cycle of a typical engineering problem is identified and the respective expertise and user control needed for each modelling phase is shown. The requirements of an IKBS for CFD software are discussed and compared to current practice. The blackboard software architecture is presented. This is shown to be appropriate for the integration of an IKBS into an engineering software package. This is demonstrated through the presentation of the prototype CFD software package FLOWES.

This paper describes the introduction of chained signage systems into evacuation simulation models. Signage systems are widely used in buildings to provide information for wayfinding, thereby providing exiting information during emergencies and assisting in navigation during normal circulation of pedestrians. Recently a system was developed to introduce simple signs into egress models. The system, known as Visibility Catchment Area or VCA, allowed similated agents to interact with signs which point directly to an exit and signs which are located directly above the exit. However, this approach was not able to represent the more general situation of a sign netwokr within an arbitrarily complex building. In this paper we extend the method to include chained signage systems which provides simulated agents that are unfamiliar with the structure a means by which to navigate to an emergency exit. The model includes the associated navigation behaviours exhibited by occupants that rely on a signage system for navigation including: Searching behaviours, Backtracking behaviours, Lost behaviours and Communication behaviours. The new features are demonstrated through a series of demonstration cases and are shown to produce plausible results.

We consider a problem of scheduling jobs on m parallel machines. The machines are dedicated, i.e., for each job the processing machine is known in advance. We mainly concentrate on the model in which at any time there is one unit of an additional resource. Any job may be assigned the resource and this reduces its processing time. A job that is given the resource uses it at each time of its processing. No two jobs are allowed to use the resource simultaneously. The objective is to minimize the makespan. We prove that the two-machine problem is NP-hard in the ordinary sense, describe a pseudopolynomial dynamic programming algorithm and convert it into an FPTAS. For the problem with an arbitrary number of machines we present an algorithm with a worst-case ratio close to 3/2, and close to 3, if a job can be given several units of the resource. For the problem with a fixed number of machines we give a PTAS. Virtually all algorithms rely on a certain variant of the linear knapsack problem (maximization, minimization, multiple-choice, bicriteria). © 2008 Wiley Periodicals, Inc. Freeze-dried (lyophilised) wafers and solvent cast films from sodium alginate (ALG) and sodium carboxymethylcellulose (CMC) have been developed as potential drug delivery systems for mucosal surfaces including wounds. The wafers (ALG, CMC) and films (CMC) were prepared by freeze-drying and drying in air (solvent evaporation) respectively, aqueous gels of the polymers containing paracetamol as a model drug. Microscopic architecture was examined using scanning electron microscopy, hydration characteristics with confocal laser scanning microscopy and dynamic vapour sorption. Texture analysis was employed to investigate mechanical characteristics of the wafers during compression. Differential scanning calorimetry was used to investigate polymorphic changes of paracetamol occurring during formulation of the wafers and films. The porous freeze-dried wafers exhibited higher drug loading and water absorption capacity than the corresponding solvent evaporated films. Moisture absorption, ease of hydration and mechanical behaviour were affected by the polymer and drug concentration. Two polymorphs of paracetamol were observed in the wafers and films, due to partial conversion of the original monoclinic to the orthorhombic polymorph during the formulation process. The results showed the potential of employing the freeze-dried wafers and solvent evaporated films in diverse mucosal applications due to their ease of hydration and based on different physical mechanical properties exhibited by both type of formulations.

Variations in the concentrations and microheterotrophic degradation rates of selected Polycyclic Aromatic Hydrocarbons (PAH) in the water column of the Tamar Estuary were investigated in relation to the major environmental variables. Concentrations of individual PAH varied typically between i and 50 ng l−1 Based on their observed environmental behaviour the PAH appeared divisible into two groupings: (1) low molecular weight PAH incorporating naphthalene, phenanthrene and anthracence and (a) the larger molecular weight homologues (fluoranthene, pyrene, chrysene, benz(a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)-pyrene). Group 1 PAH showed a complex distribution throughout the estuary with no significant correlations with either salinity or suspended particulates. Based on their relatively low particle affinity and high water solubilities and vapour pressures, volatilization is proposed as an important process in determining their fate. Microheterotrophic turnover times of naphthalene varied between x and 30 days, and were independent of suspended solids with maximum degradation rates located in the central and urban regions of the Estuary. When compared with the flushing times for the Tamar (3–5 days), it is probable that heterotrophic activity is important in the removal of naphthalene (and possibly the other Group 1 PAH) from the estuarine environment. In contrast Group 2 PAH concentrations exhibited highly significant correlations with suspended particulates. Highest concentrations occurred at the turbidity maximum, with a secondary concentration maximum localized to the industrialized portion of the estuary and associated with anthropogenic inputs. Laboratory degradation studies of benzo(a)pyrene in water samples taken from the estuary showed turnover times for the compound of between 2000 and 9000 days. Degradation rates correlated positively with suspended solids. The high particulate affinity and microbial refractivity of Group 2 PAH indicate sediment burial as the principal tate of these PAH in the Tamar Estuary. Estuarine sediments contained typically 50–1500 ng g−1 dry weight of individual PAH which were comparable to the levels of Group 2 PAH associated with the suspended particulates. Highest concentrations occurred at the riverine end of the estuary resulting from unresolved inputs in the catchment. Subsequent dilution by less polluted marine sediments together with slow degradation results in a seaward trend of decreasing concentrations. However, there is a secondary maximum of PAH superimposed on this trend which is associated with urban Plymouth.
