Background: A new intervention aimed at managing patients with medically unexplained symptoms (MUS) based on a specific set of communication techniques was developed, and tested in a cluster randomised clinical trial. Due to the modest results obtained and in order to improve our intervention we need to know the GPs' attitudes towards patients with MUS, their experience, expectations and the utility of the communication techniques we proposed and the feasibility of implementing them. Physicians who took part in 2 different training programs and in a randomised controlled trial (RCT) for patients with MUS were questioned to ascertain the reasons for the doctors' participation in the trial and the attitudes, experiences and expectations of GPs about the intervention. Methods: A qualitative study based on four focus groups with GPs who took part in a RCT. A content analysis was carried out. Results: Following the RCT patients are perceived as true suffering persons, and the relationship with them has improved in GPs of both groups. GPs mostly valued the fact that it is highly structured, that it made possible a more comfortable relationship and that it could be applied to a broad spectrum of patients with psychosocial problems. Nevertheless, all participants consider that change in patients is necessary; GPs in the intervention group remarked that that is extremely difficult to achieve. Conclusion: GPs positively evaluate the communication techniques and the interventions that help in understanding patient suffering, and express the enormous difficulties in handling change in patients. These findings provide information on the direction in which efforts for improving intervention should be directed.

The length-weight relationship and the diets of Clarias lazera were investigated between July 1981 and June 1982. About 450 specimens were examined. The standard lengths of the fish ranged from 8.5 cm to 42.2 cm. Significant differences were found between the standard lengths of the males and females with the latter slightly shorter. Somatic weights varied between 10 g to 502 g. Length-weight regression analysis gave a "b" value of 3.02 for both males and females combined; thus indicating an isometric growth. Analysis of the food in the stomachs showed that the fish is an omnivore although, it fed more on insects and fish than other food items

Samples of C. gariepinus collected from the wild and cultured populations in Plateau and Niger States of Nigeria were analyzed for length-weight relationship and organ indices (Gonadosomatic index (GSI), hepatosomatic index (HSI), renalsomatic index (RSI) and somatic fat deposit index (PDI). High correlation and linear relationship between body length and body weight was observed in all sample population (P<0.05). A significant difference was observed between the GSI of males and females of both wild and cultured population and also between females of the wild and cultured population,(P < 0.05).There was no significant difference in HSI, CSI RSI and PDI of all the sample populations (P < 0.05).The importance of length-weight relationship and organ indices in fish production are discussed

The development of the vulva of the nematode Caenorhabditis elegans is induced by a signal from the anchor cell of the somatic gonad. Activity of the gene lin-3 is required for the Vulval Precursor Cells (VPCs) to assume vulval fates. It is shown here that lin-3 encodes the vulval-inducing signal.
lin-3 was molecularly cloned by transposon-tagging and shown to encode a nematode member ofthe Epidermal Growth Factor (EGF) family. Genetic epistasis experiments indicate that lin-3 acts upstream of let-23, which encodes a homologue of the EGF-Receptor.
lin-3 transgenes that contain multiple copies of wild-type lin-3 genomic DNA clones confer a dominant multivulva phenotype in which up to all six of the VPCs assume vulval fates. The properties of these trans genes suggest that lin-3 can act in the anchor cell to induce vulval fates. Ablation of the gonadal precursors, which prevents the development of the AC, strongly reduces the ability of lin-3 transgenes to stimulate vulval development. A lin-3 recorder transgene that retains the ability to stimulate vulval development is expressed specifically in the anchor cell at the time of vulval induction.
Expression of an obligate secreted form of the EGF domain of Lin-S from a heterologous promoter is sufficient to induce vulval fates in the absence of the normal source of the inductive signal. This result suggests that Lin-S may act as a secreted factor, and that Lin-S may be the sole vulval-inducing signal made by the anchor cell.
lin-3 transgenes can cause adjacent VPCs to assume the 1° vulval fate and thus can override the action of the lateral signal mediated by lin-12 that normally prevents adjacent 1° fates. This indicates that the production of Lin-3 by the anchor cell must be limited to allow the VPCs to assume the proper pattern of fates of so 3° 3° 2° 1° 2° 3°.

In June 1994 and 1995 stations in the North, Irish, Celtic Seas and the Channel were studied for the occurrence of Myxobolus aeglefini in whiting (Merlangius merlangus). The disease was visible externally as either white nodules of a few millimeters diameter in the upper mouth cavity, gill arches and the basis of pelvic fins and in severe cases also on the lower jaws or in the cornea and sclera of the eye. It was verified morphometrically in histological sections of infected eyes by size and shape of spores. Myxobolus aeglefini was present in low prevalences at two North Sea stations and high prevalences of up to 49 % in the Irish Sea (Solway Firth) during both cruises. Whiting between 23 and 55 cm were found to be infected. Neither length-age prevalences nor condition factors and gonado, spleen, liver somatic indices differed in diseased and healthy fishes.

There is little doubt that both mammalian and teleost growth hormones can accelerate growth and increase food conversion efficiency in all commonly-reared species of salmonid fish. In those vertebrates that have been closely studied (predominantly mammals), the pituitary hormone somatotropin (GH or growth hormone) is a prime determinant of somatic growth. The hormone stimulates protein biosynthesis and tissue growth, enhances lipid utilization and lipid release from the adipose tissues (a protein-sparing effect) and suppresses the peripheral utilization of glucose. The present study is a prerequisite for future work on growth hormone physiology in salmonids and should contribute to our understanding of the mechanisms of growth suppression in stressed fish. Plasma growth hormone (GH) levels were measured in rainbow trout using a radioimmunoassay developed against chinook salmon growth hormone.

The role of life-history theory in population and evolutionary analyses is outlined. In both cases general life histories can be analysed, but simpler life histories need fewer parameters for their description. The simplest case, of semelparous (breed-once-then-die) organisms, needs only three parameters: somatic growth rate, mortality rate and fecundity. This case is analysed in detail. If fecundity is fixed, population growth rate can be calculated direct from mortality rate and somatic growth rate, and isoclines on which population growth rate is constant can be drawn in a ”state space” with axes for mortality rate and somatic growth rate. In this space density-dependence is likely to result in a population trajectory from low density, when mortality rate is low and somatic growth rate is high and the population increases (positive population growth rate) to high density, after which the process reverses to return to low density. Possible effects of pollution on this system are discussed. The state-space approach allows direct population analysis of the twin effects of pollution and density on population growth rate. Evolutionary analysis uses related methods to identify likely evolutionary outcomes when an organism's genetic options are subject to trade-offs. The trade-off considered here is between somatic growth rate and mortality rate. Such a trade-off could arise because of an energy allocation trade-off if resources spent on personal defence (reducing mortality rate) are not available for somatic growth rate. The evolutionary implications of pollution acting on such a trade-off are outlined.

The effects of stress on the immune system of various fish species including dab Limanda limanda, flounder Platichthys flesus, sea bass Dicentrarchus labrax and gobies Zosterisessor ophiocephalus, were investigated from laboratory and field experiments, using various assays to measure immunocompetence, correlated with histological and ultrastructural observations. Modulation of the immune system was demonstrated at tissue, cellular and biochemical levels following exposure to various stressors. The spleen somatic index was depressed in dab stressed in the laboratory and gobies collected from polluted sites in the Venice Lagoon. Differential blood cell counts consistently showed an increase in phagocytes and decrease in thrombocytes in fish exposed to various stressors. Phagocytic activity from spleen and kidney adherent cells was stimulated in dab stressed by transportation but depressed in fish exposed to chemical pollutants. Respiratory burst activity in phagocytic cells was also stimulated in stressed dab but depressed in sea bass exposed to cadmium. The results are discussed in relation to current concepts on stress in fish and the regulation of the immune system.

Petiveria alliacea L. pertence à família Phytolaccacceae e é conhecida popularmente como guiné ou amansa-senhor, entre outros nomes. Tem sido muito utilizada na medicina popular como agente terapêutico, devido à diversas propriedades farmacológicas. Estudos fitoquímicos têm contribuído para a descoberta de grande variedade de substâncias biologicamente ativas produzidas em diferentes partes da planta (saponinas, alcalóides, flavonóides, sulfetos, taninos, cumarinas, entre outros). A análise química da raiz tem revelado grande quantidade de derivados sulfurados, principalmente o dibenzil trissulfeto (DTS), com atividade antifúngica, antibacteriana, antioxidante e anticancerígena. Visando avaliar a produção biotecnológica do DTS, o presente trabalho teve como objetivo, otimizar a cultura de novas linhagens de calos, células em suspensão e embriões somáticos, a partir de plantas de P. alliacea L. mantidas in vitro, com o monitoramento da capacidade biossintética das culturas. Os resultados mostraram que a produção de calos friáveis foi possível em explantes foliares inoculados em meio MS suplementado com PIC ou 2,4-D. Além da resposta calogênica, foi observada a produção de estruturas globulares caracterizadas como embriões somáticos. A ocorrência de embriogênese somática direta foi confirmada através da análise histológica do processo regenerativo. A indução de embriões somáticos gerou um processo de embriogênese secundária altamente repetitivo até 150 dias de cultura e conversão a plantas em freqüência de 5%. Em relação à cultura de células em suspensão a partir dos calos friáveis, observou-se uma diminuição do crescimento celular ao longo das subculturas. As culturas em suspensão originadas de tecido embriogênico secundário continuaram o processo repetitivo em meio líquido e apresentaram conversão a plantas em taxas mais baixas que as obtidas em meio sólido. A obtenção de plantas completas a partir dos embriões somáticos demonstrou a possibilidade de utilização desse sistema para a micropropagação dessa espécie. O monitoramento fitoquímico dos sistemas de cultura in vitro e plantas de campo mantidas em casa de vegetação durante 02 anos apresentou diferenças significativas, confirmando que a cultura de tecidos pode alterar as rotas metabólicas. A cromatografia gasosa acoplada à espectrometria de massas realizada com extrato em diclorometano de embriões secos e hexânico de embriões frescos e raízes secas de plantas provenientes de embriões somáticos, demonstrou a presença do DTS, constituindo, portanto, sistemas in vitro importantes para a modulação desta substância.

Neurons obtained directly from human somatic cells hold great promise for disease modeling and drug screening. Available protocols rely on overexpression of transcription factors using integrative vectors and are often slow, complex, and inefficient. We report a fast and efficient approach for generating induced neural cells (iNCs) directly from human hematopoietic cells using Sendai virus. Upon SOX2 and c-MYC expression, CD133-positive cord blood cells rapidly adopt a neuroepithelial morphology and exhibit high expansion capacity. Under defined neurogenic culture conditions, they express mature neuronal markers and fire spontaneous action potentials that can be modulated with neurotransmitters. SOX2 and c-MYC are also sufficient to convert peripheral blood mononuclear cells into iNCs. However, the conversion process is less efficient and resulting iNCs have limited expansion capacity and electrophysiological activity upon differentiation. Our study demonstrates rapid and efficient generation of iNCs from hematopoietic cells while underscoring the impact of target cells on conversion efficiency.

English: Food selection of first-feeding yellowfin tuna larvae was studied in the laboratory during October 1992. The larvae were hatched from eggs obtained by natural spawning of yellowfin adults held in sea pens adjacent to Ishigaki Island, Okinawa Prefecture, Japan. The larvae were fed mixed-prey assemblages consisting of size-graded wild zooplankton and cultured rotifers. Yellowfin larvae were found to be selective feeders during the first four days of feeding. Copepod nauplii dominated the diet numerically, by frequency of occurrence and by weight. The relative importance of juvenile and adult copepods (mostly cyclopoids) in the diet increased over the 4-day period. Rotifers, although they comprised 31 to 40 percent of the available forage, comprised less than 2.1 percent of the diet numerically. Prey selection indices were calculated taking into account the relative abundances of prey, the swimming speeds of yellowfin larvae and their prey, and the microscale influence of turbulence on encounter rates. Yellowfin selected for copepod nauplii and against rotifers, and consumed juvenile and adult copepods in proportion to their abundances. Yellowfin larvae may select copepod nauplii and cyclopoid juveniles and adults based on the size and discontinuous swimming motion of these prey. Rotifers may not have been selected because they were larger or because they exhibit a smooth swimming pattern. The best initial diet for the culture of yellowfin larvae may be copepod nauplii and cyclopoid juveniles and adults, due to the size, swimming motion, and nutritional content of these prey. If rotifers alone are fed to yellowfin larvae, the rotifers should be enriched with a nutritional supplement that is high in unsaturated fatty acids. Mouth size of yellowfin larvae increases rapidly within the first few days of feeding, which minimizes limitations on feeding due to prey size. Although yellowfin larvae initiate feeding on relatively small prey, they rapidly acquire the ability to add relatively large, rare prey items to the diet. This mode of feeding may be adaptive for the development of yellowfin larvae, which have high metabolic rates and live in warm mixed-layer habitats of the tropical and subtropical Pacific. Our analysis also indicates a strong potential for the influence of microscale turbulence on the feeding success of yellowfin larvae. --- Experiments designed to validate the periodicity of otolith increments and to examine growth rates of yellowfin tuna larvae were conducted at the Japan Sea-Farming Association’s (JASFA) Yaeyama Experimental Station, Ishigaki Island, Japan, in September 1992. Larvae were reared from eggs spawned by captive yellowfin enclosed in a sea pen in the bay adjacent to Yaeyama Station. Results indicate that the first increment is deposited within 12 hours of hatching in the otoliths of yellowfin larvae, and subsequent growth increments are formed dailyollowing the first 24 hours after hatching r larvae up to 16 days of age. Somatic and otolith gwth ras were examined and compared for yolksac a first-feeding larvae reared at constant water tempatures of 26�and 29°C. Despite the more rapid develo of larvae reared at 29°C, growth rates were nnificaifferent between the two treatments. Howeve to poor survival after the first four days, it was ssible to examine growth rates beyond the onset of first feeding, when growth differences may become more apparent. Somatic and otolith growth were also examined for larvae reared at ambient bay water temperatures during the first 24 days after hatching. timates of laboratory growth rates were come to previously reported values for laboratory-reared yelllarvae of a similar age range, but were lower than growth rates reported for field-collected larvae. The discrepancy between laboratory and field growth rates may be associated with suboptimal growth conditions in the laboratory. Spanish: Durante octubre de 1992 se estudió en el laboratorio la seleccalimento por larvaún aleta amarillmera alimentación. Las larvas provinieron de huevos obtenidosel desove natural de aletas amarillas adultos mantenidos en corrales marinos adyacentes a la Isla Ishigaki, Prefectura de Okinawa (Japón). Se alimentó a las larvas con presas mixtas de zooplancton silvestre clasificado por tamaño y rotíferos cultivados. Se descubrió que las larvas de aleta amarilla se alimentan de forma selectiva durante los cuatro primeros días de alimentación. Los nauplios de copépodo predominaron en la dieta en número, por frecuencia de ocurrencia y por peso. La importancia relativa de copépodos juveniles y adultos (principalmente ciclopoides) en la dieta aumentó en el transcurso del período de 4 días. Los rotíferos, pese a que formaban del 31 al 40% del alimento disponible, respondieron de menos del 2,1% de la dieta en número. Se calcularon índices de selección de presas tomando en cuenta la abundancia relativa de las presas, la velocidad de natación de las larvas de aleta amarilla y de sus presas, y la influencia a microescala de la turbulencia sobre las tasas de encuentro. Los aletas amarillas seleccionaron a favor de nauplios de copépodo y en contra de los rotíferos, y consumieron copépodos juveniles y adultos en proporción a su abundancia. Es posible que las larvas de aleta amarilla seleccionen nauplios de copépodo y ciclopoides juveniles y adultos con base en el tamaño y movimiento de natación discontinuo de estas presas. Es posible que no se hayan seleccionado los rotíferos a raíz de su mayor tamaño o su patrón continuo de natación. Es posible que la mejor dieta inicial para el cultivo de larvas de aleta amarilla sea nauplios de copépodo y ciclopoides juveniles y adultos, debido al tamaño, movimiento de natación, y contenido nutritivo de estas presas. Si se alimenta a las larvas de aleta amarilla con rotíferos solamente, se debería enriquecerlos con un suplemento nutritivo rico en ácidos grasos no saturados. El tamaño de la boca de las larvas de aleta amarilla aumenta rápidamente en los primeros pocos días de alimentación, reduciendo la limitación de la alimentación debida al tamaño de la presa. Pese a que las larvas de aleta amarilla inician su alimentación con presas relativamente pequeñas, se hacen rápidamente capaces de añadir presas relativamente grandes y poco comunes a la dieta. Este modo de alimentación podría ser adaptivo para el desarrollo de larvas de aleta amarilla, que tienen tasa metabólicas altas y viven en hábitats cálidos en la capa de mezcla en el Pacífico tropical y subtropical. Nuestro análisis indica también que la influencia de turbulencia a microescala es potencialmente importante para el éxito de la alimentación de las larvas de aleta amarilla. --- En septiembre de 1992 se realizaron en la Estación Experimental Yaeyama de la Japan Sea- Farming Association (JASFA) en la Isla Ishigaki (Japón) experimentos diseñados para validar la periodicidad de los incrementos en los otolitos y para examinar las tasas de crecimiento de las larvas de atún aleta amarilla. Se criaron las larvas de huevos puestos por aletas amarillas cautivos en un corral marino en la bahía adyacente a la Estación Yaeyama. Los resultados indican que el primer incremento es depositado menos de 12 horas después de la eclosión en los otolitos de las larvas de aleta amarilla, y que los incrementos de crecimiento subsiguientes son formados a diario a partir de las primeras 24 horas después de la eclosión en larvas de hasta 16 días de edad. Se examinaron y compararon las tasas de crecimiento somático y de los otolitos en larvas en las etapas de saco vitelino y de primera alimentación criadas en aguas de temperatura constante entre 26°C y 29°C. A pesar del desarrollo más rápido de las larvas criadas a 29°C, las tasas de crecimiento no fueron significativamente diferentes entre los dos tratamientos. Debido a la mala supervivencia a partir de los cuatro primeros días, no fue posibación, uando las diferencias en el crecimiento podrían hacerse más aparentes. Se examinó también el crecimiento somático y de los otolitos para larvas criadas en temperaturas de agua ambiental en la bahía durante los 24 días inmediatamente después de la eclosión. Nuestras estimaciones de las tasas de crecimiento en el laboratorio fueron comparables a valores reportados previamente para larvas de aleta amarilla de edades similares criadas en el laboratorio, pero más bajas que las tasas de crecimiento reportadas para larvas capturadas en el mar. We propose an extended form of the von Bertalanffy growth function (VBGF), where the allocation of surplus energy to reproduction is considered. Any function can be used in our model to describe the ratio of energy allocation for reproduction to that for somatic growth. As an example, two models for energy allocation were derived: a step-function and a logistic function. The extended model can jointly describe growth in adult and juvenile stages. The change in growth rate between the two stages can be either gradual or steep; the latter gives a biphasic VBGF. The results of curve fitting indicated that a consideration of reproductive energy is meaningful for model extension. By controlling parameter values, our comprehensive model gives various growth curve shapes ranging from indeterminate to determinate growth. An increase in the number of parameters is unavoidable in practical applications of this new model. Additional information on reproduction will improve the reliability of model estimates.

Juvenile fish in temperate coastal oceans exhibit an annual cycle of feeding, and within this cycle, poor wintertime feeding can reduce body growth, condition, and perhaps survival, especially in food-poor areas. We examined the stomach contents of juvenile walleye pollock (Theragra chalcogramma) to explain previously observed seasonal and regional variation in juvenile body condition. Juvenile walleye pollock (1732 fish, 37–250 mm standard length) of the 2000 year class were collected from three regions in the Gulf of Alaska (Kodiak, Semidi, and Shumagin) representing an area of the continental shelf of ca. 100,000 km2 during four seasons (August 2000 to September 2001). Mean stomach content weight (SCW, 0.72% somatic body weight) decreased with fish body length except from winter to summer 2001. Euphausiids composed 61% of SCW and were the main determinant of seasonal change in the diets of fish in the Kodiak and Semidi regions. Before and during winter, SCW and the euphausiid dietary component were highest in the Kodiak region. Bioenergetics modeling indicated a relatively high growth rate for Kodiak juveniles during winter (0.33 mm standard length/d). After winter, Shumagin juveniles had relatively high SCW and, unlike the Kodiak and Semidi juveniles, exhibited no reduction in the euphausiid dietary component. These patterns explain previous seasonal and regional differences in body condition. We hypothesize that high-quality feeding locations (and perhaps nursery areas) shift seasonally in response to the availability of euphausiid
