Introduction. This study aims to compare the molecular gene expression during ischemia reperfusion injury. Several surgical times were considered: in the beginning of the harvesting (T0), at the end of the cold ischemia period (T1), and after reperfusion (T2) and compared with graft dysfunction after liver transplant (OLT). Methods. We studied 54 patients undergoing OLT. Clinical, laboratory data, and histologic data (Suzuki classification) as well as the Survival Outcomes Following Liver Transplantation (SOFT) score were used and compared with the molecular gene expression of the following genes: Interleukin (IL)-1b, IL-6, tumor necrosis factor-a, perforin, E-selectin (SELE), Fas-ligand, granzyme B, heme oxygenase-1, and nitric oxide synthetase. Results. Fifteen patients presented with graft dysfunction according to SOFT criteria. No relevant data were obtained by comparing the variables graft dysfunction and histologic variables. We observed a statistically significant relation between SELE at T0 (P ¼ .013) and IL-1b at T0 (P ¼ .028) and early graft dysfunction. Conclusions. We conclude that several genetically determined proinflammatory expressions may play a critical role in the development of graft dysfunction after OLT.

No presente estudo camondongos "Swiss" foram submetidos a tratamento com a droga Ro 7-1051 (N-Benzil-2-Nitro-imidazolacetamida) antes de sua inoculação com a cepa Colombiana do T. cruzi e comparados com animais inocuiados com a mesma cepa e tratados em fase inicial de infecção. Com avaliação feita por parasitemia periférica, xenodiagnõstico, sub-inoculação em camondongos recém-nascidos e exame histopatológico foram obtidos melhores resultados com os animais submetidos a tratamento prévio, tendo-se verificado que 55,5% dos animais permaneceram negativados durante o curso da experiência, enquanto os tratados precocemente tiveram um índice de cura de 28,5%.

O trabalho mostra o estudo anatomopatológico de 7 casos de Toxoplasmose do sistema nervoso central, com diagnósticos feitos exclusivamente em autópsias. O trabalho chama atenção para a dificuldade de diagnóstico clínico desta entidade e mostra dois casos de apresentação mais rara com manifestações clínicas e anatomopatológicas que simulam abscessos cerebrais. O comprometimento do plexo coróide, freqüente neste material, fortalece a idéia de que esta estrutura desempenha papel importante na disseminação da doença no sistema nervoso central.

No período de agosto a dezembro de 1993 realizou-se um levantamento de 77 pacientes da casuística de leishmaniose mucosa do programa de Três Braços, registrada no período de 1976 a 1986. Encontrou-se 65 pacientes vivos e 12 falecidos. Os primeiros foram submetidos a exame clínico geral e otorrinolaringológico e à pesquisa de anticorpos por imunofluorescência indireta. Os pacientes com lesões ativas foram submetidos à biópsia da mucosa nasal para isolar o parasita em meio de cultura NNN a partir da inoculação direta do material ou das lesões desenvolvidas no hamster e para estudo histopatológico. Todos os isolados foram caracterizados como Leishmania (Viannia) braziliensis utilizando anticoipos monoclonais. Cinqüenta e seis (86%) pacientes estavam curados e nove (13%) encontravam-se com lesões ativas. Entre os pacientes falecidos, cinco (41%) estavam curados no momento do óbito. O programa teve 79% de pacientes curados ao longo de 17 anos. O tempo médio de seguimento clínico foi de 10 anos (7-17 anos). A doença como causa-mortis foi sugerida em 3 casos.

This study compared the serum levels of IL-6, TNF-alpha and IFN-gamma, in children under 1 year of age with and without dengue. Sera were collected from a total of 41 children living in the Department of Antioquia, Colombia (27 patients with dengue and 14 controls). The results showed higher cytokine levels in children with dengue than without dengue, with statistically significant differences for IL-6 and IFN-gamma. No statistically significant differences were found between clinical forms, although IL-6 and IFN-gamma levels were higher in dengue fever cases than in dengue hemorrhagic fever cases. On the other hand, TNF-alpha levels were higher in dengue hemorrhagic fever than in dengue fever. The levels of IL-6 and TNF-alpha were higher in secondary infection than in primary infection, although IFN-gamma levels were higher in primary infection. These results suggest that IL-6, TNF-alpha and IFN-gamma are involved in dengue infection independently of the clinical form.

Diagnosis of human herpesvirus-7 active infection in transplant patients has proved difficult, because this virus is ubiquitous and can cause persistent infections in the host. The significance of viral DNA detected in leukocytes by PCR is unclear and cross-reaction in serological tests may occur. This study aimed to evaluate nested-PCR to detect human herpesvirus-7 active infection in liver transplant recipients compared to healthy individuals. human herpesvirus-7 nested-PCR was performed on leukocytes and sera of 53 healthy volunteers and sera of 29 liver transplant recipients. In healthy volunteers, human herpesvirus-7 was detected in 28.3% of leukocytes and 0% of serum. human herpesvirus-7 was detected in sera of 48.2% of the liver transplant recipients. Nested-PCR on DNA extracted from leukocytes detected latent infection and the study suggests that nested-PCR performed on serum could be useful to detect human herpesvirus-7 active infection in liver transplant recipients.

Interleukin 8 (CXCL8) is an autocrine chemokine specific for the chemoattraction and activation of granulocytes, NKT cells and T lymphocytes. Patients with tuberculosis and latent Mycobacterium tuberculosis infection were assessed for the spontaneous expression of CXCR1 (CD128) and CXCR2 on lymphocytes and monocytes. Compared with ex vivo profiles, increased spontaneous CXCR2 expression and normal CXCR1 expression were found on lymphocytes in two out of 59 individuals. Monocytes showed normal ex vivo profiles for both receptors. After stimulation with purified protein derivative, the in vitro levels of CXCL8 were below the median levels of all patients with prior tuberculosis. Spontaneous CXCR2 modulation did not cause notable variation in the in vitro levels of CXCL8.

INTRODUCTION: This study evaluated the intracellular profile of interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-10 (IL-10) and interferon-^7; (IFN-^7;) in peripheral blood mononuclear cells (PBMCs) from leprosy patients based on oral infections presence to determine whether these coinfections could be associated with pro-inflammatory activity in leprosy. METHODS: Leprosy patients regardless of clinical form and specific leprosy treatment (n=38) were divided into two groups: Group I - leprosy patients with oral infections (n=19), and Group II - leprosy patients without oral infections (n=19). Non-leprosy patients presenting oral infections were assigned to the control Group (n=10). Intracellular IL-2, IL-4, IL-10 and IFN-^7; production was evaluated by flow cytometry (FACS) before and 7 days after controlling the oral infection in the Group I, before and 7 days after dental prophylaxis in the Group II, and during oral infection process in control Group. RESULTS: Low percentages of CD3+ lymphocytes bearing IL-2, IL-10 and IFN-^7; were observed in the Group I and Group II at baseline and 7 days after therapy or prophylaxis compared to controls. Group I showed reduced percentages of IL-4 at baseline and 7 days after therapy compared to controls, or at baseline of Group II, and the Group II showed reduced percentages of CD3+ cells bearing IL-4 compared to control. An increase of the percentages of CD3+cells bearing IL-4 was observed in the Group I after the oral infections treatment. CONCLUSIONS: The occurrence of oral infections favors the intracellular cytokines expression and, probably, the inflammatory reaction operating as a stimulatory signal triggering the leprosy reactions.

IntroductionPurpureocillium lilacinum is emerging as a causal agent of hyalohyphomycosis that is refractory to antifungal drugs; however, the pathogenic mechanisms underlying P. lilacinum infection are not understood. In this study, we investigated the interaction of P. lilacinum conidia with human macrophages and dendritic cells in vitro.MethodsSpores of a P. lilacinum clinical isolate were obtained by chill-heat shock. Mononuclear cells were isolated from eight healthy individuals. Monocytes were separated by cold aggregation and differentiated into macrophages by incubation for 7 to 10 days at 37°C or into dendritic cells by the addition of the cytokines human granulocyte-macrophage colony stimulating factor and interleukin-4. Conidial suspension was added to the human cells at 1:1, 2:1, and 5:1 (conidia:cells) ratios for 1h, 6h, and 24h, and the infection was evaluated by Giemsa staining and light microscopy.ResultsAfter 1h interaction, P. lilacinum conidia were internalized by human cells and after 6h contact, some conidia became inflated. After 24h interaction, the conidia produced germ tubes and hyphae, leading to the disruption of macrophage and dendritic cell membranes. The infection rate analyzed after 6h incubation of P. lilacinumconidia with cells at 2:1 and 1:1 ratios was 76.5% and 25.5%, respectively, for macrophages and 54.3% and 19.5%, respectively, for cultured dendritic cells.ConclusionsP. lilacinum conidia are capable of infecting and destroying both macrophages and dendritic cells, clearly demonstrating the ability of this pathogenic fungus to invade human phagocytic cells.

O estudo sobre os textos escritos produzidos por alunos da 7ª classe em Angola foi estabelecido por nós com base na hipótese da existência de dificuldades de escrita. Há bastantes dúvidas entre os docentes angolanos sobre a capacidade dos alunos na elaboração de textos espontâneos. Desta forma, com o objectivo de conhecermos as capacidades de produção de textos escritos pelos alunos do ensino geral no Lubango, de elaborarmos uma tipologia de análise dos erros encontrados e de propormos estratégias de superação das dificuldades apresentadas nos textos dos alunos, fomos à Escola do I Ciclo do Ensino Secundário 27 de Março do Lubango – Huíla recolher as composições da Prova Trimestral para verificar as dificuldades de escrita apresentadas nos textos dos alunos. Partimos de 314 textos, dos quais foi extraída a amostra de 10%, correspondendo a 32 textos seleccionados através da tabela de números aleatórios. Os textos foram analisados com base na tipologia de erros de Sousa (1996). Desta forma, foram detectados erros de substituição, de omissão, de divisão/aglutinação, os quais foram muito significativos e ocorreram em 66,856% dos casos no total. Os erros de substituição maiúsculas/minúsculas, os de adição, de grafias homófonas constituem 30.856% dos casos. Confirmam-se as dificuldades apresentadas pelos alunos que devem merecer tratamento através das propostas de remediação apresentadas. As dificuldades encontradas nos textos destes alunos devem servir de base para a planificação da disciplina de Língua Portuguesa no Ensino Primário de forma a dotar o aluno de competências para a melhoria da comunicação escrita na sala de aula.

No que diz respeito à sua preservação e apresentação grande parte da arte contemporânea exige respostas eficazes e pragmáticas do ponto de vista da museologia. Esta situação torna-se mais evidente em obras de instalação e media art, que recorrem a múltiplos objectos, mantêm uma relação com o espaço e são alvo de fenómenos de obsolescência tecnológica, reclamando assim, estratégias que passam pela produção activa de registos documentais, a fim de garantir o seu legado a gerações futuras. A partir da obra de Alberto Carneiro "Árvore jogo / lúdico em 7 imagens espelhadas", que foi pela primeira vez materializada na Galeria Quadrum, em Lisboa, em 1975, e destruída no ano seguinte pelo autor, após a Bienal de Veneza, discutimos as estratégias documentais mais adequadas. A metodologia assenta num trabalho de base antropológica, através de entrevistas com o artista, que permitam identificar todos os aspectos relacionados com o processo criativo e criar um manual de instalação, que se pretende de fácil acesso a investigadores, conservadores e até ao público em geral. Para este efeito iniciou-se um trabalho de colaboração com o Departamento de Informática da FCT-UNL com vista à construção de uma ferramenta multimédia que permita reunir num só local toda a informação relativa à obra.

PURPOSE: To test an experimental model of chemical mammary carcinogenesis induction in rats. METHODS: Twenty young virgin Sprague-Dawley female rats, aged 47 days, received 20 mg of 7,12-dimethylbenz(a)anthracene (DMBA) intragastrically by gavage. Afterwards, at 8 and 13 weeks, their mammary glands were examined. At the end of the experiment, the animals were sacrificed, and the mammary tumors were measured and weighed. Tumor fragments were analyzed using light microscopy. RESULTS: Eight weeks after DMBA injection, 16 rats presented at least 1 breast tumor (80%). After 13 weeks, all of them (100%) developed breast carcinomas that were confirmed by histopathological analysis. CONCLUSION: This experimental animal model of chemical mammary induced carcinogenesis is feasible and can be used in further experiments on the role of tumorigenic biomodulator substances.
