Previously, we reported on the discovery and characterization of a mammalian chromatin-associated protein, CHD1 (chromo-ATPase/helicase-DNA-binding domain), with features that led us to suspect that it might have an important role in the modification of chromatin structure. We now report on the characterization of the Drosophila melanogaster CHD1 homologue (dCHD1) and its localization on polytene chromosomes. A set of overlapping cDNAs encodes an 1883-aa open reading frame that is 50% identical and 68% similar to the mouse CHD1 sequence, including conservation of the three signature domains for which the protein was named. When the chromo and ATPase/helicase domain sequences in various CHD1 homologues were compared with the corresponding sequences in other proteins, certain distinctive features of the CHD1 chromo and ATPase/helicase domains were revealed. The dCHD1 gene was mapped to position 23C-24A on chromosome 2L. Western blot analyses with antibodies raised against a dCHD1 fusion protein specifically recognized an approximately 210-kDa protein in nuclear extracts from Drosophila embryos and cultured cells. Most interestingly, these antibodies revealed that dCHD1 localizes to sites of extended chromatin (interbands) and regions associated with high transcriptional activity (puffs) on polytene chromosomes from salivary glands of third instar larvae. These observations strongly support the idea that CHD1 functions to alter chromatin structure in a way that facilitates gene expression.

The regulatory protein calmodulin is a major mediator of calcium-induced changes in cellular activity. To analyze the roles of calmodulin in an intact animal, we have generated a calmodulin null mutation in Drosophila melanogaster. Maternal calmodulin supports calmodulin null individuals throughout embryogenesis, but they die within 2 days of hatching as first instar larvae. We have detected two pronounced behavioral abnormalities specific to the loss of calmodulin in these larvae. Swinging of the head and anterior body, which occurs in the presence of food, is three times more frequent in the null animals. More strikingly, most locomotion in calmodulin null larvae is spontaneous backward movement. This is in marked contrast to the wild-type situation where backward locomotion is seen only as a stimulus-elicited avoidance response. Our finding of spontaneous avoidance behavior has striking similarities to the enhanced avoidance responses produced by some calmodulin mutations in Paramecium. Thus our results suggest evolutionary conservation of a role for calmodulin in membrane excitability and linked behavioral responses.

In a search for retinoid X receptor-like molecules in Drosophila, we have identified an additional member of the nuclear receptor superfamily, XR78E/F. In the DNA-binding domain, XR78E/F is closely related to the mammalian receptor TR2, as well as to the nuclear receptors Coup-TF and Seven-up. We demonstrate that XR78E/F binds as a homodimer to direct repeats of the sequence AGGTCA. In transient transfection assays, XR78E/F represses ecdysone signaling in a DNA-binding-dependent fashion. XR78E/F has its highest expression in third-instar larvae and prepupae. These experiments suggest that XR78E/F may play a regulatory role in the transcriptional cascade triggered by the hormone ecdysone in Drosophila.

We have designed a rapid cloning and screening strategy to identify new members of the nuclear hormone receptor superfamily that are expressed during the onset of Drosophila metamorphosis. Using this approach, we isolated three Drosophila genes, designated DHR38, DHR78, and DHR96. All three genes are expressed throughout third-instar larval and prepupal development. DHR38 is the Drosophila homolog of NGFI-B and binds specifically to an NGFI-B response element. DHR78 and DHR96 are orphan receptor genes. DHR78 is induced by 20-hydroxyecdysone (20E) in cultured larval organs, and its encoded protein binds to two AGGTCA half-sites arranged as either direct or palindromic repeats. DHR96 is also 20E-inducible, and its encoded protein binds selectively to the hsp27 20E response element. The 20E receptor can bind to each of the sequences recognized by DHR78 and DHR96, indicating that these proteins may compete with the receptor for binding to a common set of target sequences.

Anopheles é o gênero da família Culicidae mais estudado devido sua importância médica. Atualmente o gênero Anopheles compreende 472 espécies válidas que estão divididas em sete subgêneros. Os principais vetores de plasmódio da Malária no Brasil pertencem ao subgênero Nyssorhynchus, que inclui 39 espécies oficialmente reconhecidas e um número crescente de complexos de espécies crípticas que estão distribuídas em três Seções: Myzorhynchella, Albimanus e Argyritarsis. Atualmente a Seção Myzorhynchella é formada por seis espécies: An. lutzii, An. parvus, An. nigritarsis, An. guarani, An. antunesi e An. pristinus. Para o desenvolvimento da análise morfológica, observou-se material-tipo depositado em diferentes coleções, espécimes depositados na coleção entomológica da FSP/USP, além de outros obtidos em coletas realizadas durante o presente estudo em diferentes localidades do Brasil. As análises moleculares foram desenvolvidas a partir de espécimes obtidos nas coletas. Revisão taxonômica da Seção Myzorhynchella é apresentada, incluindo-se descrições de quatro novas espécies e redescrições das demais, informações sobre bionomia, importância médica, caracterização molecular, distribuição geográfica, estado de preservação do material-tipo, além de chaves de identificação de adultos, larva de quarto estádio e genitália masculina. Os resultados das análises filogenéticas utilizando sequências de ITS2, COI e Catalase indicam a existência de pelo menos doze espécies dentro da Seção Myzorhynchella, os espécimes que vêm sendo identificados como An. antunesi constitui um complexo formado por possíveis cinco espécies e aqueles de An. parvus e An. pristinus também podem representar complexos de espécies. As sequências de ITS2 podem ser utilizadas como marcador diagnóstico para espécies da Seção Myzorhynchella. Contudo, o estudo ainda demonstra que pouco se conhece sobre a diversidade de espécies de Anopheles que ocorrem em ambientes onde a malária ocorre em baixa endemicidade. Pelo número de espécies novas encontradas e pela escassez de trabalhos com espécies da Seção, fica evidente a necessidade de mais estudos.

Esta tese é dividida em duas partes: traçar a filogenia de Sericothripinae Karny, 1921 e realizar uma revisão taxonômica das espécies neotropicais. Para a análise filogenética, estados de caracteres morfológicos foram examinados entre Sericothripinae e Thysanoptera-Terebrantia relacionados, e as relações genéricas e supragenéricas foram exploradas. O monofiletismo de Sericothripinae foi recuperado, mas o formato do metasterno, usado na classificação dos gêneros, provavelmente não reflete a filogenia. De acordo com estudos moleculares recentes, o grupo genérico Scirtothrips em conjunto com o gênero Echinothrips Moulton, 1911 foram recuperados como intimamente relacionados aos Sericothripinae, mas, neste trabalho, Psilothrips Hood, 1927 e Pseudothrips Hinds, 1902 foram recuperados como não relacionados com Sericothripinae. Muitos estados de caracteres morfológicos entre os Sericothripinae são considerados homoplásticos e, na ausência de análises moleculares adequadas, alterações formais de nomenclatura não foram realizadas. Na revisão taxonômica, 14 novas espécies de Sericothripinae da região Neotropical são descritas. Chaves ilustradas foram elaboradas para as fêmeas de sete espécies de Hydatothrips Karny, 1913 e 41 espécies de Neohydatothrips John, 1929, principalmente do Brasil, mas incluindo todas as espécies registradas do sul da fronteira entre o México e os EUA até o extremo sul da América do Sul. Espécies de plantas em que associações-hospedeiras foram registradas são indicadas sempre que possível, comentários são elencados para as poucas espécies de importância econômica e uma chave para imaturos de segundo instar de cinco espécies é proposta. Neohydatothrips burungae (Hood, 1935) stat. rev. e N. aztecus Johansen, 1983 stat. rev. são retiradas de sinonímia com N. signifer (Priesner, 1932), ao passo que Sericothrips denigratus De Santis, 1966 syn. n. é sinonimizada com N. burungae. Hydatothrips williamsi (Hood, 1928) comb. n. é realocado de Neohydatothrips e, com isso, houve um caso de homônimo no gênero. Para resolver esse problema, H. tareei nom. nov. é proposto para H. williamsi Mound e Tree, 2009, da Austrália.

A necessidade por recursos alimentares move os insetos predadores na busca por presas, para garantir a sobrevivência. Existem vários fatores que podem influenciar o comportamento de busca e escolha por presas e, sob determinadas circunstâncias, o inseto pode escolher se alimentar de presas não alvo, como, por exemplo, inimigos naturais e assim prejudicar a população de organismos benéficos. Garantir a estabilidade do agroecossistema é uma forma de manter as populações de pragas em níveis compatíveis com a ação de inimigos naturais e, consequentemente obter o sucesso no controle biológico. Programas bem sucedidos de manejo de pragas requerem previamente a realização de estudos sobre comportamento de insetos. Com o intuito de compreender os fatores que governam o comportamento predatório e as interações ecológicas entre Harmonia axyridis e Chrysoperla externa, considerando a presença de Diaphorina citri como presa, objetivou-se estudar o efeito de interações intraguilda sobre a predação de D. citri, experimentalmente e utilizando modelos estatísticos para compreender a dinâmica de interações tróficas no contexto de potenciais guildas presentes em citros. Três experimentos distintos foram realizados para investigar o comportamento dos insetos predadores. Foram realizados testes com escolha entre larvas dos predadores C. externa e H. axyridis e testes com e sem escolha comparando machos e fêmeas do predador H. axyridis, a fim de investigar padrões de escolha de presas. O segundo experimento foi realizado para investigar o comportamento predatório entre larvas de segundo instar dos predadores sob diferentes densidades de D. citri como presa. As combinações foram: sem escolha de presas intraguilda, combinações com escolha de presas intraguilda e combinações com a presença de dois predadores da mesma espécie para possibilitar o canibalismo. As densidades de D. citri utilizadas foram 5, 10, 15, 30, 60, 80. No último experimento avaliou-se a interação entre larvas de C. externa e H. axyridis, ao longo do desenvolvimento, simulando a existência de apenas uma presa, como fonte de alimento. O desenvolvimento dos predadores foi avaliado e comparado utilizando-se duas presas diferentes, ninfas de D. citri e ovos de Ephestia kuehniella para cada larva de primeiro instar dos predadores C. externa e H. axyridis. As combinações de predadores foram as mesmas citadas no experimento anterior. Observou-se a duração do desenvolvimento larval nas diferentes combinações de predadores e presa, bem como o percentual de predações intraguilda e canibalismos ocorridos. Nem a densidade de presas, nem o tipo de presa ofertada influenciou o comportamento das larvas dos predadores para a realização de predação intraguilda ou canibalismo. O principal fator foi a diferença de idade que reflete no tamanho dos predadores, podendo direcionar a predação intraguilda ou o canibalismo e até mesmo acelerar o ciclo de desenvolvimento do predador intraguilda envolvido. Predadores em estágio inicial de vida buscaram por qualquer tipo de presa para garantir sua sobrevivência, não demonstrando qualquer padrão em suas escolhas. No caso dos adultos de H. axyridis, há diferença de comportamento entre os sexos, dependendo da densidade de presas disponíveis, fêmeas poderão ser mais ágeis na busca e consumo de presas.

A new interaction between insects and carnivorous plants is reported from Brazil. Larvae of the predatory flower fly Toxomerus basalis (Diptera: Syrphidae: Syrphinae) have been found scavenging on the sticky leaves of several carnivorous sundew species (Drosera, Droseraceae) in Minas Gerais and São Paulo states, SE Brazil. This syrphid apparently spends its whole larval stage feeding on prey trapped by Drosera leaves. The nature of this plant-animal relationship is discussed, as well as the Drosera species involved, and locations where T. basalis was observed. 180 years after the discovery of this flower fly species, its biology now has been revealed. This is (1) the first record of kleptoparasitism in the Syrphidae, (2) a new larval feeding mode for this family, and (3) the first report of a dipteran that shows a kleptoparasitic relationship with a carnivorous plant with adhesive flypaper traps. The first descriptions of the third instar larva and puparium of T. basalis based on Scanning Electron Microscope analysis are provided.

A description of Anopheles (Cellia) irenicus Schmidt, sp.n. (formerly A. farauti No. 7) is provided. This species is one of six recorded from the Solomon Islands within the A. punctulatus group, which contains the major vectors of the causative agents of malaria and lymphatic filariasis in the southwest Pacific. Morphological markers are described for adult females, fourth-instar larvae and pupae that identify most specimens of A. irenicus. Keys are presented to distinguish members of the A. punctulatus group in the Solomon Islands.

Properties relevant to the ovipositional activity and lifetime productivity of Coccidoxenoides peregrinus (Timberlake) were assessed in the laboratory, to determine the potential of this species as a biocontrol agent against the citrus mealybug, Planococcus citri (Risso). In general, this species has not performed well in orchards, except for a few localities on different continents. The mode of reproduction of C peregrinus is almost entirely thelytokous, with males produced sporadically and at low frequency. The females have both pro-ovigenic and synovigenic traits, which raises questions of the utility of this distinction. The females have a high reproductive potential with 10-20 eggs per day available within the first two days (after a short (12 h) pre-oviposition period), and 80-150 eggs per day thereafter until death at about eight days. Mean lifetime fecundity was 239.2 +/- 34.3 eggs. C peregrinus oviposits across a range of P. citri instars, but productivity relies predominantly on second instar hosts. Second stage (N2) hosts received most eggs in choice (about 52%) and no-choice (about 50%) tests. Most eggs deposited into N2 hosts (82%) reached adult stage whereas only a few of those deposited into N1 and N3 (about 5% each) developed successfully. The haemolymph of parasitised reproductive mealybugs contained granular structures and no parasitoid eggs were found 24 h after exposure to ovipositing wasps. Also, no wasps emerged from parasitised adult hosts that were kept alive. Parasitoid eggs deposited into adult hosts were presumed encapsulated and destroyed, as control mealybugs (not exposed to female wasps) had no granular structures in their haemolymph. Wasps exposed to an abundance of hosts soon started ovipositing, but only for a relatively short time each day (about 2.5 h out of a 7 h exposure). They stopped ovipositing despite eggs judged to be mature in their ovaries. The reproductive output of C peregrinus is discussed in relation to the ecological factors that could influence this output, and the implications for biocontrol are discussed. (C) 2003 Elsevier Inc. Spiders are among the most abundant predators recorded in grain crops in Australia. They are voracious predators, and combined with their high abundance, may play an important role in the reduction of pest populations. The significance of spider assemblages as biological control agents of key pests such as Helicoverpa spp. in Australian agroecosystems is largely unknown. A thorough inventory was made of the spider fauna inhabiting unsprayed soybean fields at Gatton, south-east Queensland. One-hundred-and-two morphospecies from 28 families were collected using vacuum sampling and pitfall traps across two summer seasons (2000-01, 2001-02). No-choice feeding tests in the laboratory, using eggs and larvae of Helicoverpa armigera (Hubner) as prey, were used to ascertain the predatory potential of each spider group. The field-collected spider assemblage ate on average 2.4 (+/-0.7 standard error) to 5.0 (+/-0.8) eggs per 24 h per spider (10-25% of those available), depending on level of starvation. Clubionidae were the only spiders to readily consume eggs in the laboratory (mean of 18.4 +/- 1.5 eggs per starved spider and 8.2 +/- 3.9 per non-starved spider after 24 h). Starved spiders consumed 9.4 (+/- 0.1) first-instar larvae per 24 h per spider (90% of those available). This information was combined with field observations and literature from Australian and overseas studies to assess the potential of spider groups as predators of Helicoverpa spp. Lycosidae, Clubionidae, Oxyopidae, Salticidae and Thomisidae have the capacity to contribute to control of Helicoverpa spp.

The relative resistance levels of three different populations of mulberry silkworm, Bombyx mori L. to Bacillus thuringiensis (Bt) has been studied. All three populations (two Australian and one Indonesian) were observed for similar characteristics including 3rd instar larval mortality at 24, 48, 72 and 96 h after treatment (HAT), LD50 ratio and probit mortality. Among the Australian two populations, the QuBill (yellow coloured, oval shaped cocoon) population showed higher larval mortality to Bt toxicity compared to the QuBite (white coloured, oval shaped cocoon) population. When all the populations were compared, the Insab (Indonesian population with white Coloured, peanut shaped cocoon) showed lower larval mortality and highest LD50 ratio up to 48 HAT. The Insab population also showed a 24 It longer incubation/latent period prior to the start Of Mortality.

Coccidoxenoides perminutus achieves only low levels of parasitism of its host Planococcus citri in southeast Queensland citrus. Two possible causes were investigated. Adult survival under natural conditions was assessed to determine whether providing adult food sources could enhance survival. Behavioural changes of hosts, induced by C perminutus parasitism, was also investigated to establish if parasitised P. citri move from their feeding site to seek protected shelters some distance away and are thus not accounted for in field assessments of parasitism rates. Unparasitised mealybugs placed in the field for two periods were retrieved before the effects of parasitism were manifested and parasitism rates were still low (0.3% at 5 days and 1.2% at 10 days). Levels of locomotion of P. citri exposed to C perminutus were compared with those of unexposed ones. Parasitised mealybugs, regardless of instar, undergo behavioural changes. In comparison to unparasitised controls, the mealybugs become highly active 7-14 days after exposure to wasps. All parasitised mealybugs undergo physical changes, their body becomes cylindrical, their legs go so rigid that the mealybugs become immobile, and this signifies the typical mummy appearance. All mealybugs that became mummies eventually fell from the host lemon fruit because of impaired locomotion and were caught on sticky traps that had been placed beneath the lemons. Consequently, their final site of mummification was not established. C perminutus adults provided with nectar or honey survived longer (about 5 days) in the field than those without food (about a day). Nectar from two plant species, Alpinia zerumbet and Datura candida, proved to be good sources of food for the adult wasps, and were comparable in quality to honey. The low level of parasitism achieved by C perminutus in southeast Queensland citrus thus appears to be a consequence of the short adult life and the negative effects of a harsh environment. Provision of a suitable food source (e.g., nectar) may well enhance levels of parasitism in the field. (c) 2005 Elsevier Inc. All rights reserved.

This study provides comprehensive documentation of silk production in the pest moth Helicoverpa armigera from gland secretion to extrusion of silk thread. The structure of the silk glands, accessory structures and extrusion apparatus are reported. The general schema of the paired silk glands follows that found for Lepidoptera. Morphology of the duct, silk press, muscle attachments and spigot are presented as a three-dimensional reconstruction and the cuticular crescent-shaped profile of the silk press is demonstrated in both open and closed forms with attendant muscle blocks, allowing advances in our knowledge of how the silk press functions to regulate the extrusion of silk. Growth of the spigot across instars is documented showing a distinctive developmental pattern for this extrusion device. Its shape and structure are related to use and load-bearing activity. The complete and faithful duplication of the genome is essential to ensure normal cell division and organismal development. Eukaryotic DNA replication is initiated at multiple sites termed origins of replication that are activated at different time through S phase. The replication timing program is regulated by the S-phase checkpoint, which signals and repairs replicative stress. Eukaryotic DNA is packaged with histones into chromatin, thus DNA-templated processes including replication are modulated by the local chromatin environment such as post-translational modifications (PTMs) of histones.

One such epigenetic mark, methylation of lysine 20 on histone H4 (H4K20), has been linked to chromatin compaction, transcription, DNA repair and DNA replication. H4K20 can be mono-, di- and tri-methylated. Monomethylation of H4K20 (H4K20me1) is mediated by the cell cycle-regulated histone methyltransferase PR-Set7 and subsequent di-/tri- methylation is catalyzed by Suv4-20. Prior studies have shown that PR-Set7 depletion in mammalian cells results in defective S phase progression and the accumulation of DNA damage, which may be partially attributed to defects in origin selection and activation. Meanwhile, overexpression of mammalian PR-Set7 recruits components of pre-Replication Complex (pre-RC) onto chromatin and licenses replication origins for re-replication. However, these studies were limited to only a handful of mammalian origins, and it remains unclear how PR-Set7 impacts the replication program on a genomic scale. Finally, the methylation substrates of PR-Set7 include both histone (H4K20) and non-histone targets, therefore it is necessary to directly test the role of H4K20 methylation in PR-Set7 regulated phenotypes.

I employed genetic, cytological, and genomic approaches to better understand the role of H4K20 methylation in regulating DNA replication and genome stability in Drosophila melanogaster cells. Depletion of Drosophila PR-Set7 by RNAi in cultured Kc167 cells led to an ATR-dependent cell cycle arrest with near 4N DNA content and the accumulation of DNA damage, indicating a defect in completing S phase. The cells were arrested at the second S phase following PR-Set7 downregulation, suggesting that it was an epigenetic effect that coupled to the dilution of histone modification over multiple cell cycles. To directly test the role of H4K20 methylation in regulating genome integrity, I collaborated with the Duronio Lab and observed spontaneous DNA damage on the imaginal wing discs of third instar mutant larvae that had an alanine substitution on H4K20 (H4K20A) thus unable to be methylated, confirming that H4K20 is a bona fide target of PR-Set7 in maintaining genome integrity.

One possible source of DNA damage due to loss of PR-Set7 is reduced origin activity. I used BrdU-seq to profile the genome-wide origin activation pattern. However, I found that deregulation of H4K20 methylation states by manipulating the H4K20 methyltransferases PR-Set7 and Suv4-20 had no impact on origin activation throughout the genome. I then mapped the genomic distribution of DNA damage upon PR-Set7 depletion. Surprisingly, ChIP-seq of the DNA damage marker γ-H2A.v located the DNA damage to late replicating euchromatic regions of the Drosophila genome, and the strength of γ-H2A.v signal was uniformly distributed and spanned the entire late replication domain, implying stochastic replication fork collapse within late replicating regions. Together these data suggest that PR-Set7-mediated monomethylation of H4K20 is critical for maintaining the genomic integrity of late replicating domains, presumably via stabilization of late replicating forks.

In addition to investigating the function of H4K20me, I also used immunofluorescence to characterize the cell cycle regulated chromatin loading of Mcm2-7 complex, the DNA helicase that licenses replication origins, using H4K20me1 level as a proxy for cell cycle stages. In parallel with chromatin spindown data by Powell et al. (Powell et al. 2015), we showed a continuous loading of Mcm2-7 during G1 and a progressive removal from chromatin through S phase.
