To date there have been few quantitative studies of the distribution of, and relative habitat utilisation by, koalas in the mulgalands of Queensland. To examine these parameters we applied habitat-accessibility and relative habitat-utilisation indices to estimates of faecal pellet density sampled at 149 sites across the region. Modelling the presence of pellets using logistic regression showed that the potential range of accessible habitats and relative habitat use varied greatly across the region, with rainfall being probably the most important determinant of distribution. Within that distribution, landform and rainfall were both important factors affecting habitat preference. Modelling revealed vastly different probabilities of finding a pellet under trees depending on the tree species, canopy size, and location within the region.

This study used faecal pellets to investigate the broadscale distribution and diet of koalas in the mulgalands biogeographic region of south-west Queensland. Koala distribution was determined by conducting faecal pellet searches within a 30-cm radius of the base of eucalypts on 149 belt transects, located using a multi-scaled stratified sampling design. Cuticular analysis of pellets collected from 22 of these sites was conducted to identify the dietary composition of koalas within the region. Our data suggest that koala distribution is concentrated in the northern and more easterly regions of the study area, and appears to be strongly linked with annual rainfall. Over 50% of our koala records were obtained from non-riverine communities, indicating that koalas in the study area are not primarily restricted to riverine communities, as has frequently been suggested. Cuticular analysis indicates that more than 90% of koala diet within the region consists of five eucalypt species. Our data highlights the importance of residual Tertiary landforms to koala conservation in the region.

Predictions of flow patterns in a 600-mm scale model SAG mill made using four classes of discrete element method (DEM) models are compared to experimental photographs. The accuracy of the various models is assessed using quantitative data on shoulder, toe and vortex center positions taken from ensembles of both experimental and simulation results. These detailed comparisons reveal the strengths and weaknesses of the various models for simulating mills and allow the effect of different modelling assumptions to be quantitatively evaluated. In particular, very close agreement is demonstrated between the full 3D model (including the end wall effects) and the experiments. It is also demonstrated that the traditional two-dimensional circular particle DEM model under-predicts the shoulder, toe and vortex center positions and the power draw by around 10 degrees. The effect of particle shape and the dimensionality of the model are also assessed, with particle shape predominantly affecting the shoulder position while the dimensionality of the model affects mainly the toe position. No information is available on the decomposition and nutrient release pattern of Piper aduncum and Imperata cylindrica despite their importance in shifting cultivation systems of Papua New Guinea and other tropical regions. We conducted a litter bag study (24 weeks) on a Typic Eutropepts in the humid lowlands to assess the rate of decomposition of Piper aduncum, Imperata cylindrica and Gliricidia sepium leaves under sweet potato (Ipomoea batatas). Decomposition rates of piper leaf litter were fastest followed closely by gliricidia, and both lost 50% of the leaf biomass within 10 weeks. Imperata leaf litter decomposed much slower and half-life values exceeded the period of observation. The decomposition patterns were best explained by the lignin plus polyphenol over N ratio which was lowest for piper (4.3) and highest for imperata (24.7). Gliricidia leaf litter released 79 kg N ha(-1), whereas 18 kg N ha(-1) was immobilised in the imperata litter. The mineralization of P was similar for the three species, but piper litter released large amounts of K. The decomposition and nutrient release patterns had significant effects on the soil. The soil contained significantly more water in the previous imperata plots at 13 weeks due to the relative slow decomposition of the leaves. Soil N levels were significantly reduced in the previous imperata plots due to immobilisation of N. Levels of exchangeable K were significantly increased in the previous piper plots due to the large addition of K. It can be concluded that piper leaf litter is a significant and easily decomposable source of K which is an important nutrient for sweet potato. Gliricidia leaf litter contained much N, whereas imperata leaf litter releases relatively little nutrients and keeps the soil more moist. Gliricidia fallow is more attractive than an imperata fallow for it improves the soil fertility and produces fuelwood as additional saleable products.

OBJECTIVE - To assess the effect of age on glucose metabolism by examining 1) glucose metabolism in young and middle-aged subjects when total or regional adiposity is taken into account and 2) in vitro glucose transport in adipose tissue explants from young and middle-aged women paired for total and abdominal adiposity. RESEARCH DESIGN AND METHODS - Study 1: body composition, subcutaneous abdominal and visceral adipose tissue areas, and fasting and oral glucose-stimulated glucose and insulin were measured in 84 young and 81 middle-aged men and in 110 young and 91 middle-aged women. Study 2: glucose uptake in subcutaneous abdominal and visceral adipose tissue explants were measured in eight young and eight middle-aged women. RESULTS - Study 1: young and middle-aged men showed similar subcutaneous abdominal tissue area, whereas fat mass and visceral adipose tissue were greater in middle-aged than in young men (P < 0.01). Fat mass and subcutaneous and visceral adipose tissue areas were greater in middle-aged as compared with young women (P < 0.01). Fasting plasma glucose and the glucose response to an oral glucose tolerance test were significantly higher in middle-aged than in young men and women (P < 0.001). Statistical control for visceral adipose tissue area eliminated the difference seen in glucose response in men and women. Study 2: glucose transport in subcutaneous and omental adipose tissue did not differ between young and middle-aged women. CONCLUSIONS - 1) Visceral obesity, more than age per se, correlates with glucose intolerance in middle-aged subjects; 2) aging does not influence in vitro adipose tissue glucose uptake.

Antigen-specific suppression of a previously primed immune response is a major challenge for immunotherapy of autoimmune disease. ReIB activation is required for myeloid DC differentiation. Here, we show that antigen-exposed DCs in which ReIB function is inhibited lack cell surface CD40, prevent priming of immunity, and suppress previously primed immune responses. DCs generated from CD40-deficient mice similarly confer suppression. Regulatory CD4(+) T cells induced by the DCs transfer antigen-specific Infectious tolerance to primed recipients in an interleukin10-dependent fashion. Thus CD40, regulated by ReIB activity, determines the consequences of antigen presentation by myeloid DCs. These observations have significance for autoimmune immunotherapy and suggest a mechanism by which peripheral tolerance might be constitutively maintained by RelB(-) CD40(-) DCs.

CD40 has emerged as a key signaling pathway for the function of B cells, monocytes, and dendritic cells (DC) in the immune system, and plays a major role in inflammatory pathways of nonhemopoletic cells. CD40 is expressed by monocytes and DC and is up-regulated when DC migrate from the periphery to draining lymph nodes (DLN) in response to microbial challenge. CD154 signaling by MHC-restricted, activated CD4* T cells induces differentiation of DC, as defined by an increased surface expression of MHC, costimulatory, and adhesion molecules. Thus, CD40 functions in the adaptive immune response as a trigger for the expression of costimulatory molecules for efficient T-cell activation. CD40 ligation of DC also has the capacity to induce high levels of the cytokine IL-12, which polarizes CD4(+) T cells toward a T helper 1 (Th1) type, enhances proliferation of CD8(+) T cells, and activates NK cells. CD40 may also play an important role in the decision between tolerance and immunity and the generation of regulatory CD4(+) T cells that are thought to maintain peripheral self-tolerance in vivo.

The NFkappaB family of transcription factors is a critical mediator in inflammation. NFkappaB is involved both in the regulation of CD40 expression and in cell signaling after CD40 ligation. This positive feedback loop linking NFkappaB and CD40 plays an important role in the control of the adaptive immune response, with fundamental implications for immunity and tolerance in vivo.

Respiratory syncytial virus (RSV) is a ubiquitous human pathogen and the leading cause of lower respiratory tract infections in infants. Infection of cells and subsequent formation of syncytia occur through membrane fusion mediated by the RSV fusion protein (RSV-F). A novel in vitro assay of recombinant RSV-F function has been devised and used to characterize a number of escape mutants for three known inhibitors of RSV-F that have been isolated. Homology modeling of the RSV-F structure has been carried out on the basis of a chimera derived from the crystal structures of the RSV-F core and a fragment from the orthologous fusion protein from Newcastle disease virus (NDV). The structure correlates well with the appearance of RSV-F in electron micrographs, and the residues identified as contributing to specific binding sites for several monoclonal antibodies are arranged in appropriate solvent-accessible clusters. The positions of the characterized resistance mutants in the model structure identify two promising regions for the design of fusion inhibitors. (C) 2003 Elsevier Science (USA). All rights reserved.

Este trabalho propõe uma metodologia de aprendizagem que permite a um robô aprender uma tarefa adaptando-a e representando-a de acordo com a sua capacidade motora e sensorial. Primeiramente, um mapeamento sensoriomotor é criado e converte informação sensorial em informação motora. Depois, através de imitação, o robô aprende um conjunto de ações elementares formando um vocabulário motor. A imitação é baseada nas representações motoras obtidas com o mapeamento sensoriomotor. O vocabulário motor criado é então utilizado para aprender e realizar tarefas mais sofisticadas, compostas por seqüências ou combinações de ações elementares. Esta metodologia é ilustrada através de uma aplicação de mapeamento e navegação topológica com um robô móvel. O automovimento é utilizado como mapeamento visuomotor, convertendo o fluxo óptico em imagens omnidirecionais em informação motora (translação e rotação), a qual é usada para a criação de um vocabulário motor. A seguir, o vocabulário é utilizado para mapeamento e navegação topológica. Os resultados obtidos são interessantes e a abordagem proposta pode ser estendida a diferentes robôs e aplicações.

O ser humano realiza uma alimentação pouco variada, com grandes teores de açúcar e gorduras saturadas, ao mesmo tempo, está sujeito a profissões cada vez mais competitivas aonde passa longos períodos sentados sem qualquer esforço físico. Estes aspetos levam à acumulação de gorduras em redor de todos os órgãos, que promovem o aparecimento de problemas cardiovascular, que são atualmente, a principal causa de morte no mundo. O tratamento das doenças cardiovasculares é em muitas situações realizado por procedimentos minimamente-invasivos, que são guiados através de imagem médica. Contudo, a utilização de radiação durante a navegação é normalmente requerida o que tem consequências para o paciente e para a equipa médica. Nesta dissertação, focamo-nos nos recentes sistemas de aquisição de imagem sem radiação e no desenvolvimento de sistemas mais inteligentes para facilitar o controlo destes equipamentos durante o procedimento. Assim, pretendemos desenvolver um robô que apoie na aquisição de imagens de ultrassons através de uma sonda transesofágica. O robô desenvolvido possui um conjunto de engrenagens que fazem a transferência de movimento para as rodas dos manípulos da sonda e um encoder magnético que proporciona uma leitura rápida e de alta precisão dos movimentos da sonda. De forma a automaticamente adaptar a posição da sonda na direção do alvo anatómico, um sistema de motion tracking foi acoplado ao robô e ao instrumento cirúrgico utilizado durante o procedimento. Assim, todos os movimentos realizados pelo intervencionista são repetidos pela sonda, permitindo assim adquirir uma imagem de ultrassom sempre centrada no instrumento cirúrgico. Para avaliar a performance do robô foram realizados testes em laboratório. mais concretamente: 1) controlo do robô sem sonda acoplada e 2) controlo do robô com sonda acoplada. Os testes foram realizados com diferentes posições angulares, em todas as gamas de funcionamento do robô, avaliando o erro da posição final em relação posição desejada e o tempo de resposta. Os resultados demonstraram que um erro médio de 2º foi observado para as diferentes situações com um tempo médio de resposta de 300 ms. Os resultados alcançados demonstraram uma boa resposta do sistema, pelo que se espera que sistema desenvolvido venha ser capaz de reduzir o tempo de intervenção, aumentando a qualidade da intervenção e minimizando possíveis erros.
