Data variability analysis has been the focus of a number of studies seeking to capture differences of patterns generated by biological systems. Although several studies related to gait employ the analysis of variability in their observations, we noticed a lack of such information for subjects with unilateral coxarthrosis undergoing total hip arthroplasty (THA). To tackle this deficiency of information, we conducted a study of the gait on a treadmill with10 healthy subjects (30.7 ± 6.75 years old) from G1 and 24 subjects (65 ± 8.5 years old) with unilateral THA from G2. Thus, by means of two inertial measurement units (IMUs) positioned in the pelvis, we have developed a detection method of the step and stride for calculating these intervals and extract the signal characteristics. The variability analysis (coefficient of variation) was performed, taking into consideration the extracted features and the step and stride times. The average and the 95% confidence interval estimate for the average of the step and stride times to each group were in agreement with literature. The mean coefficient of variation for the step and stride times was calculated and compared among groups by the Kruskal-Wallis test with 95% confidence interval. Each component X, Y and Z of the two IMUs (accelerometer, magnetometer and gyroscope) corresponded to a variable. The resultants of each sensor, the linear velocity (accelerometers) and the instantaneous angular displacement (gyroscopes) completed the set of variables. The characteristics were extracted from the signals of these variables to check the variability in the G1 and G2 groups . There were significant differences (p <0.05) between G1 and G2 for the average of the step and stride times. The variability of the step and stride, as well as the variability of all other evaluated characteristics were higher for the group G2 (p <0.05). The method proposed in this study proved to be suitable for the measuring of variability of biomechanical parameters related to the extracted features. All the extracted features categorized the groups. The G2 group showed greater variability, so it is possible that the age and the pathological condition of the hip both contributed to this result.

The main driver for the investigation of fast pyrolysis oil marine fuel blends is EU directive 2012/33/EU which aims to cut the sulphur content of marine fuel and thereby reduce air pollution caused by marine vessels. The aim of this study was to investigate the miscibility of 3- and 4- component blends containing pyrolysis oil, 1-butanol, biodiesel (RME) and/or marine gas oil (MGO). The ideal blend would be a stable homogenous product with a minimum amount of butanol, whilst maximising the amount of pyrolysis oil. A successful blend would have properties suitable for use in marine engines. In order to successfully utilise a marine fuel blend in commercial vessels it should meet minimum specification requirements such as a flash point ≥60°C. Blends of pyrolysis oil, RME, MGO and 1-butanol were evaluated and characterised. The mixed blends were inspected after 48 hours for homogeneity and the results plotted on a tri-plot phase diagram. Homogenous samples were tested for water content, pH, acid number, viscosity and flash point as these give indicate a blend’s suitability for engine testing. The work forms part of the ReShip Project which is funded by Norwegian industry partners and the Research Council of Norway (The ENERGIX programme).

This thesis explores methods for fabrication of nanohole arrays, and their integration into a benchtop system for use as sensors or anti-counterfeit labels. Chapter 1 gives an introduction to plasmonics and more specifically nanohole arrays and how they have potential as label free sensors compared to the current biosensors on the market. Various fabrication methods are explored, including Focused Ion Beam, Electron Beam Lithography, Nanoimprint lithography, Template stripping and Phase Shift Lithography. Focused Ion Beam was chosen to fabricate the nanohole arrays due to its suitability for rapid prototyping and it’s relatively low cost. In chapter 2 the fabrication of nanohole arrays using FIB is described, and the samples characterised. The fabricated nanohole arrays are tested as bulk refractive index sensors, before a bioassay using whole molecule human IgG antibodies and antigen is developed and performed on the senor. In chapter 3 the fabricated sensors are integrated into a custom built system, capable of real time, multiplexed detection of biomolecules. Here, scFv antibodies of two biomolecules relevant to the detection of pancreatic cancer (C1q and C3) are attached to the nanohole arrays, and detection of their complementary proteins is demonstrated both in buffer (10 nM detection of C1q Ag) and human serum. Chapter 4 explores arrays of anisotropic (elliptical) nanoholes and shows how the shape anisotropy induces polarisation sensitive transmission spectra, in both simulations and fabricated arrays. The potential use of such samples as visible and NIR tag for anti-counterfeiting applications is demonstrated. Finally, chapter 5 gives a summary of the work completed and discusses potential future work in this area.

Narrative research has become a very popular field in contemporary social sciences. It promises new fields of inquiry, creative solutions to persistent problems, a way to establish links with other disciplines such as cultural and literary studies, enhanced possibilities of applying research to policy and practice, and a fresh take on the politics of social research (see for instance Andrews et al., 2013 [ 2008]; Andrews et al., 2004 [2000]; Andrews, Squire and Tamboukou,, 2004; Elliott, 2005; Emerson and Frosh, 2004; Freeman, 2009a; Herman, 2009; Hyvarinen et al., 2010; Lieblich et al., 2004; Patterson, 2002; Riessman, 2008; Trahar, 2009; Wells, 2011). This book aims to introduce you, step by step and with contemporary examples, to narrative research in the social sciences. It will give you an overview of a range of narrative methods, and it will situate narrative research in relation to other social science methods. It will show you what narrative research offers, as well as its difficulties. It will do this by drawing on work from a variety of disciplines, in theoretical and applied fields, across diverse topics, from health and the internet, to politics and sexualities, and in a number of different national contexts.

While news stories are an important traditional medium to broadcast and consume news, microblogging has recently emerged as a place where people can dis- cuss, disseminate, collect or report information about news. However, the massive information in the microblogosphere makes it hard for readers to keep up with these real-time updates. This is especially a problem when it comes to breaking news, where people are more eager to know “what is happening”. Therefore, this dis- sertation is intended as an exploratory effort to investigate computational methods to augment human effort when monitoring the development of breaking news on a given topic from a microblog stream by extractively summarizing the updates in a timely manner. More specifically, given an interest in a topic, either entered as a query or presented as an initial news report, a microblog temporal summarization system is proposed to filter microblog posts from a stream with three primary concerns: topical relevance, novelty, and salience. Considering the relatively high arrival rate of microblog streams, a cascade framework consisting of three stages is proposed to progressively reduce quantity of posts. For each step in the cascade, this dissertation studies methods that improve over current baselines. In the relevance filtering stage, query and document expansion techniques are applied to mitigate sparsity and vocabulary mismatch issues. The use of word embedding as a basis for filtering is also explored, using unsupervised and supervised modeling to characterize lexical and semantic similarity. In the novelty filtering stage, several statistical ways of characterizing novelty are investigated and ensemble learning techniques are used to integrate results from these diverse techniques. These results are compared with a baseline clustering approach using both standard and delay-discounted measures. In the salience filtering stage, because of the real-time prediction requirement a method of learning verb phrase usage from past relevant news reports is used in conjunction with some standard measures for characterizing writing quality. Following a Cranfield-like evaluation paradigm, this dissertation includes a se- ries of experiments to evaluate the proposed methods for each step, and for the end- to-end system. New microblog novelty and salience judgments are created, building on existing relevance judgments from the TREC Microblog track. The results point to future research directions at the intersection of social media, computational jour- nalism, information retrieval, automatic summarization, and machine learning.

A indústria de aves brasileira destaca-se economicamente, onde um total de 12,3 milhões de toneladas foi produzido no país em 2013. Esta produção em larga escala gera considerável volume de subprodutos, chegando até 35% da ave viva. Tais resíduos são convertidos, por processos tradicionais, em produtos de baixo valor comercial, como por exemplo, farinhas. O processo de variação de pH constitui um importante processo alternativo de obtenção de proteínas com melhores características funcionais e nutricionais. Estudar as variáveis do processo, efetuando aumento dimensional, é fundamental para aplicação das tecnologias desenvolvidas no laboratório e posterior definição final de processos industriais. A produção de isolados proteicos seria uma tecnologia atraente no aproveitamento de subprodutos da indústria de frango, convertendo-os em uma ótima fonte proteica, agregando valor ao produto obtido. Este trabalho teve por objetivo produzir isolados proteicos em diferentes escalas, utilizando subprodutos não comestíveis da indústria de frango. Foi estudada a solubilização das proteínas da matéria-prima (MP) para definir pHs de solubilização e de precipitação isoelétrica. A curva apontou um pH alcalino de 11,0 para etapa de solubilização e de 5,25 para etapa de precipitação proteica. As proteínas obtidas foram caracterizadas quanto sua composição proximal, índice de acidez (IA), índice de peróxidos (IP) e substâncias reativas ao ácido tiobarbitúrico (TBARS) além de propriedades funcionais de solubilidade, capacidade de retenção de água (CRA) e capacidade de retenção de óleo (CRO); e nutricionais de digestibilidade proteica. Comparativamente foram analisadas farinhas de vísceras comerciais nos mesmos parâmetros. Um aumento de escala do processo foi realizado e avaliado pelas mesmas respostas do produto da escala laboratorial. Foi obtido um teor proteico de 82 e 85% em escala laboratorial e aumento de escala, respectivamente, e também uma redução lipídica de 75%, e de cinzas de 85%, em relação à MP. A composição proximal das farinhas analisadas ficou entre 67-72% para proteína bruta, 17-22% para lipídios e 9-15% para cinzas. O IA, apresentou valores de 2,2 e 3,1 meq/g de isolado e de 1,6 a 2,0 meq/g de farinha. Já para IP, obteve-se valores de 0,003 a 0,005 meq/g de isolado e de 0,002 a 0,049 meq/g de farinha. Os índices de TBARS apontaram valores de 0,081 e 0,214 mg MA/g de isolado e 0,041 a 0,128 mg MA/g de farinha. A solubilidade das proteínas do isolado apontou 84 e 81% em pH 3 e 11 respectivamente e de 5% em pH 5, já para farinhas variaram de 22 a 31% em pH de 3 a 11. A CRA obtida no isolado foi 3,1 a 16,5 g água/g de proteína e de 3,8 a 10,9 g água/g de proteína nas farinhas. A CRO ficou em 4,2 mL de óleo/g de proteína do isolados e 2,6 mL de óleo/g de proteína da farinhas. Os isolados proteicos apresentaram 92 e 95% de digestibilidade das proteínas, em comparação aos 84% das farinhas comerciais. Os índices acumulados e apresentados neste trabalho concluíram que foi possível aumentar a escala do processo de variação de pH, sem perder qualidade nos índices físico-químicos e de digestibilidade proteica.

Neste trabalho é proposto pela primeira vez, o desenvolvimento e validação de um método analítico baseado no emprego da dispersão da matriz em fase sólida (MSPD) modificada, para extração das espécies CH3Hg+ e Hg2+ em amostras de peixe e determinação por cromatografia em fase gasosa acoplada à espectrometria de massas (GC-MS). O método de extração utilizando a MSPD combina o rompimento da estrutura física da amostra, através da maceração e do uso de SiO2 como suporte sólido, com o método da extração ácida, utilizando uma solução de HCl 4,2 mol L-1 e NaCl 0,5 mol L-1. Para otimização da MSPD, foram avaliados parâmetros como massa de amostra, massa de suporte sólido, concentração de HCl, concentração de NaCl, tipo de suporte sólido e o tempo de agitação, com auxílio da metodologia de superfície de resposta. Além disso, a etapa de derivatização e a separação cromatográfica também foram otimizadas na determinação de CH3Hg+ e Hg2+ por GC-MS. O método mostrouse adequado para extração e determinação de espécies de mercúrio através da aplicação em materiais de referência certificados de fígado de peixe (DOLT-3) e músculo de peixe (DORM-2), apresentando boas concordâncias com os valores certificados e desvio padrão relativo inferior a 9,5%. Os limites de detecção foram de 0,06 e 0,12 µg g-1 para CH3Hg+ e Hg2+, respectivamente. Além disso, foi observado um significativo efeito de matriz e, por isso, a calibração foi feita com curvas preparadas com o extrato da MSPD. O método mostrou boa concordância na comparação entre a soma da concentração das espécies e a concentração de mercúrio total determinada por espectrometria de massas com plasma indutivamente acoplado com geração de vapor frio (CVG-ICP-MS), após digestão assistida por micro-ondas (MAD) em peixes do tipo atum (Thunnus thynnus), cação anjo (Squatina squatina) e cação viola (Rhinobatos blochii.).

A importância da encenação de uma transição entre o mundo secular e a vida em clausura, no contexto da arquitectura cartusiana, é amplamente demonstrada através dos percursos de entrada dos conjuntos mais relevantes, bem como em muitos daqueles representados nas gravuras presentes no livro Maisons de l’Ordre des Chartreux – Vues et Notices, uma obra do início do século XX que inclui gravuras da quase totalidade dos 282 mosteiros cartusianos distribuídos por quase toda a Europa. Na maioria das cartuxas podem observarse transições indirectas e prolongadas, capazes de tardar o passo e dar significado à passagem para a clausura – características espaciais que São Bruno, fundador da casa cartusiana, considerava primordiais para a implantação do seu eremitério. A partir de uma cuidada análise dessa transição, nas cartuxas em geral, e da sua evolução morfológica, na cartuxa de Évora em particular – estimulada pela descoberta de dados aqui analisados e interpretados pela primeira vez – apresenta-se, em continuidade com a sua história e tendo como premissa a consolidação do processo de entrada, uma proposta de redefinição da estrutura monacal de Santa Maria Scala Coeli. Esta estrutura originária do século XVI encontra-se incompleta e degradada, em particular na ala sul onde se encontra o seu principal acesso, sendo fundamental o completamento desta área para o cabal reconhecimento da unidade do conjunto e da intenção na qual se traduz a sua génese. De resto, a estratégia de investigação e de projecto descrita nesta dissertação pode ser encarada como um processo plausível de ser utilizado no caso de futuras intervenções noutras estruturas cartusianas, contribuindo assim para a recuperação e preservação do conceito fundador da identidade da arquitectura cartusiana que, no decorrer da expansão da ordem, se foi perdendo; The Architecture of the Carthusian Monastery of Santa Maria Scala Coeli: About the process of entrance. ABSTRACT: The importance of staging a transition between the secular world and life cloistered in the context of Carthusian architecture is amply demonstrated through the input paths of the most important collections, as well as many of those represented in the pictures in the book Maisons de l’Ordre des Chartreux – Vues et Notices, one of the early twentieth century work that includes prints of almost all of the 282 Carthusians monasteries spread across almost all of Europe. In most of Carthusian the transitions that can be observed are indirect and long, able to delay the step and give meaning to the passage to the life cloistered - spatial characteristics that St. Bruno, founder of the Carthusian Order, considered essential for the implementation of his hermitage. From a careful analysis of this transition, looking through the Carthusian in general, and its morphological evolution, in the Charterhouse of Évora in particular, encouraged by the data discovery here analyzed and interpreted for the first time - it is presented in continuity with its history and having premised on the consolidation of the entry process, a proposal for a redefinition of the monastic structure of Santa Maria Scala Coeli. This original structure of the sixteenth century is incomplete and degraded, particularly in the south wing where it is the main access, and where is fundamental the completion of this area to the full recognition of the unity of the whole and to understand the intent in which translates its genesis. Moreover, the research strategy and project described in this paper can be seen as a plausible case to be used in case of future interventions in other cartusianas structures, thus contributing to the recovery and preservation of the founder of the identity concept of the Carthusian architecture that, during the expansion of the order, was missing.

New and promising treatments for coronary heart disease are enabled by vascular scaffolds made of poly(L-lactic acid) (PLLA), as demonstrated by Abbott Vascular’s bioresorbable vascular scaffold. PLLA is a semicrystalline polymer whose degree of crystallinity and crystalline microstructure depend on the thermal and deformation history during processing. In turn, the semicrystalline morphology determines scaffold strength and biodegradation time. However, spatially-resolved information about the resulting material structure (crystallinity and crystal orientation) is needed to interpret in vivo observations.

The first manufacturing step of the scaffold is tube expansion in a process similar to injection blow molding. Spatial uniformity of the tube microstructure is essential for the consistent production and performance of the final scaffold. For implantation into the artery, solid-state deformation below the glass transition temperature is imposed on a laser-cut subassembly to crimp it into a small diameter. Regions of localized strain during crimping are implicated in deployment behavior.

To examine the semicrystalline microstructure development of the scaffold, we employed complementary techniques of scanning electron and polarized light microscopy, wide-angle X-ray scattering, and X-ray microdiffraction. These techniques enabled us to assess the microstructure at the micro and nano length scale. The results show that the expanded tube is very uniform in the azimuthal and axial directions and that radial variations are more pronounced. The crimping step dramatically changes the microstructure of the subassembly by imposing extreme elongation and compression. Spatial information on the degree and direction of chain orientation from X-ray microdiffraction data gives insight into the mechanism by which the PLLA dissipates the stresses during crimping, without fracture. Finally, analysis of the microstructure after deployment shows that it is inherited from the crimping step and contributes to the scaffold’s successful implantation in vivo.

Após todo o processo referente ao Mestrado em Ensino da Educação Física nos Ensinos Básico e Secundário, que tomou parte numa escola secundária no concelho da Amadora, mais especificamente, na Brandoa, Escola Secundária Fernando Namora, no ano letivo de 2011/2012, serve este relatório para dar como finda esta etapa. Este Estágio teve em atenção o Guia de Estágio Pedagógico, do qual constam quatro áreas: 1. Organização e Gestão do Ensino e da Aprendizagem (Área 1); 2. Inovação e Investigação Pedagógica (Área 2); 3. Participação na Escola (Área 3); 4. Relação com a Comunidade (Área 4). Assim, e tendo em conta as competências aglomeradas nestas quatro áreas de referência, foi desenvolvido, ao longo do ano letivo, um projeto com uma turma de 7º ano da já referida escola – Escola Secundária Fernando Namora. Deste relatório constarão as principais análises e reflexões, referentes a todo o trabalho desenvolvido (estratégias, obstáculos, dificuldades, relacionamento, entre outros.), ao longo do ano letivo. Finalmente, apresentarei a reflexão crítica, que pretenderá demonstrar o papel que este Estágio teve na minha formação, tanto pessoal como profissional.

Background: Incidence of jaundice is high in newborn infants. Since well appearing newborns are rapidly and routinely discharged from hospital, performing an inexpensive noninvasive pre-discharge screening test for evaluation of jaundice seems to be necessary. Objectives: This study was conducted to compare the accuracy of cutaneous v/s serum bilirubin measurements in this regard. Patients and Methods: This was a prospective cross sectional study conducted in Mahdieh hospital, Tehran. 613 neonates weighing ≥ 1,800 g with gestational age of ≥ 35 weeks were enrolled. A pre discharge transcutaneous bilirubin test (TcB) was performed in all. Serum samples were taken from neonates with TcB ≥ 5 mg/dL in first and > 8 mg/dL in second 24 hours. Decision for treatment or recheck of bilirubin level after discharge was made based on serum bilirubin results. Results: Based on the study protocol, among 613 studied neonates, 491 (80%) revealed high TcB, of them 240 (49%) cases showed TBC ≥ 5 mg/dL in first and 251 (51 %) in second pre-discharge 24 hours. TcB ranged 3.3 - 17.1, mean TcB in first 24 hours was 6.9 ± 1 .7 (mode 6) and in second 24 hours 9.1 ± 2.1 (mode 10). Of 491 neonates with high TcB, capillary serum sample was taken as the second step and 398 neonates revealed high total serum bilirubin (TsB) with the same protocol for TcB. 108 (27.1%) neonates showed TsB ≥ 5 mg/dL in first and 290 (72.9%) in second 24 hours. According to the study results TcB has a 81% positive predictive value (PPV) in diagnosis of hyperbilirubinemia. Correlation coefficient of TcB and TsB in highest rate is equal to 72% (P value < 0.001). Conclusions: TcB is an inexpensive, noninvasive and precise pre-discharge screening test for evaluation of hyperbilirubinemia, with a high PPV. It is highly recommended to be performed routinely due to high incidence of hyperbilirubinemia in neonates.

Purpose: To develop processes for effective isolation and purification of recombinant human plasminogen activator (rhPA) from transgenic rabbit milk. Methods: Immunoaffinity chromatography was selected and improved by a special polyol-responsive monoclonal antibody (PR-mAb). Alteplase was used as immunogen because of its similarity to rhPA in terms of structure. The PR-mAb was prepared by hybridoma technology and screened by ELISA-elution assay. Screening antibody was performed using rhPA milk in an ELISA-elution assay. The antibody clone C4-PR-mAb was selected for immunoaffinity chromatography. The rhPA was effectively bound to immobilized C4-PR-mAb on the column and was eluted with Tris buffer comprising 0.75 mol/L ammonium sulfate and 40n% propanediol (pH7.9). The rhPA was further purified by passing through Chromdex75 gel filtration column. Results: There were 12 hybridoma strains selected into the polyol-responsive mAbs screen step and three hybridoma strains were superior for producing PR-mAbs (C1, C4, C8). The rhPA can be purified from transgenic rabbit milk and maintained a higher thrombolytic activity in vitro by FAPA. Conclusion: The results demonstrate the suitability of the alternative approach used in this study. Using immunoaffinity chromatography and gel filtration column is feasible and convenient for extracting rhPA from milk, and should be useful for purifying other tPA mutants or other novel recombinant milkderived proteins.

Durante el ciclo de la vida, la figura corporal de la mujer se modifica paulatinamente, pero al llegar la edad mediana, a consecuencia de la pérdida folicular de la función ovárica entre otros factores, se acentúan los cambios fisiológicos que pueden aumentar el riesgo para la salud y el bienestar. La androgenia, en asociación con la obesidad, aumenta el riesgo de padecer diabetes, hipertensión, dislipidemia, arterosclerosis, enfermedades cardiovasculares, colelitiasis, artrosis, artritis, carcinomas femeninos y otras patologías de la edad mediana. El ejercicio físico ha adquirido cada vez mayor importancia dentro de la terapéutica en el climaterio. Se han demostrado los efectos positivos del ejercicio sobre las enfermedades cardiovasculares y el mantenimiento del peso. Por todo ello, el estudio pretende conocer la influencia de un programa de actividad física de cinco meses de duración sobre factores de riesgo cardiovasculares como la hipertensión arterial y la composición corporal en mujeres premenopáusicas. Participaron 20 mujeres premenopáusicas, separadas en dos grupos de mujeres: menores de 35 años (n=10), y mayores de 35 años (n=10). Las valoraciones previas y al finalizar el programa fueron: valoración de la tensión arterial y de la composición corporal. Los resultados mostraron una reducción significativa en mayor medida en la tensión arterial diastólica y en la masa grasa en el grupo de mujeres mayores de 35 años premenopáusicas. Por ello, practicar un programa de ejercicio físico basado en el aeróbic, clases de step y tonificación con mancuernas, mejora factores de riesgo cardiovasculares como la hipertensión arterial y la composición corporal en mujeres adultas mayores de 35 años premenopáusicas.
