The high water content in maca (Lepidium meyenii W.) roots combined with the damage produced during or after harvest makes them vulnerable to attack by enzymes and microorganisms. Although starch degradation has been extensively studied, in maca roots there is a paucity of research regarding the starch reserves. In this paper, parameters of starch degradation are shown to be related to the action of amylolytic enzymes during storage at room temperature. Over the course of three weeks, the starch and protein content, soluble sugar, total amylolytic activity, and alpha- and beta-amylase activity were measured. In addition, the integrity of starch granules was observed by scanning electron microscopy. Despite the evidence of dehydration, there were no significant differences (p <= 0.5) in the total starch content or in the activities of alpha- and beta-amylase. After the third week the roots remained suitable for consumption. The results indicate a postharvest latency that can lead to sprout or to senescence, depending on the environmental conditions. Quality of fresh-cut carambola (Averrhoa carambola L) is related to many chemical and biochemical variables especially those involved with softening and browning, both influenced by storage temperature. To study these effects, a multivariate analysis was used to evaluate slices packaged in vacuum-sealed polyolefin bags, and stored at 2.5 degrees C, 5 degrees C and 10 degrees C, for up to 16 d. The quality of slices at each temperature was correlated with the duration of storage, O(2) and CO(2) concentration in the package, physical chemical constituents, and activity of enzymes involved in softening (PG) and browning (PPO) metabolism. Three quality groups were identified by hierarchical cluster analysis, and the classification of the components within each of these groups was obtained from a principal component analysis (PCA). The characterization of samples by PCA clearly distinguished acceptable and non-acceptable slices. According to PCA, acceptable slices presented higher ascorbic acid content, greater hue angles ((o)h) and final lightness (L-5) in the first principal component (PC1). On the other hand, non-acceptable slices presented higher total pectin content. PPO activity in the PC1. Non-acceptable slices also presented higher soluble pectin content, increased pectin solubilisation and higher CO(2) concentration in the second principal component (PC2) whereas acceptable slices showed lower total sugar content. The hierarchical cluster and PCA analyses were useful for discriminating the quality of slices stored at different temperatures. (C) 2011 Elsevier B.V. All rights reserved.

This work aimed to evaluate the efficiency of fungicides in controlling in vitro and in vivo the causal agents of anthracnose (Colletotrichum gloeosporioides and C. acutatum) and black spot (Guignardia psidii) and evaluate the effect of alternative products to control these diseases. Inhibition of mycelial growth of the pathogens was evaluated for ten fungicides at concentrations of 1, 10 and 100 mg L-1 of active ingredient in potato-dextrose-agar medium. The effectiveness of the fungicides azoxystrobin + difenoconazole, cyproconazole, pyraclostrobin, tebuconazole and tebuconazole + trifloxystrobin in controlling disease incidence and severity of anthracnose, through applications in the field, was measured in fruits collected at three stages of maturation, according to the skin color ( dark green, light green and yellowish green). In postharvest dipping of fruits, the products evaluated were citric acid, peracetic acid, salicylic acid, sodium bicarbonate, chlorine dioxide, Ecolife (R) and chitosan. The fungicides azoxystrobin + difenoconazole, pyraclostrobin, tebuconazole and trifloxystrobin + tebuconazole were highly effective in inhibiting the in vitro mycelial growth of G. psidii and moderately to highly effective in inhibiting C. acutatum and C. gloeosporioides. In field conditions, the fungicide azoxystrobin + difenoconazole was effective in controlling anthracnose and black spot in fruit at three maturity stage ( skin color yellowish green). The alternative products tested were ineffective in the curative control of anthracnose and early blight at postharvest of guava.

The endophytic fungus Epicoccum nigrum was isolated from sugarcane and the bioguided fractionation of the ethyl acetate extract led to the isolation of epicolactone, mellein, and 4,5-dimethylresorcinol. Characterization of epicolactone by MS, NMR and X-ray crystallography revealed a new natural product with an unusual carbon skeleton. The production of this secondary metabolite decreased in mutants of Epicoccum nigrum transformed by Agrobacterium tumefaciens. Additionally, these mutants produced 4-hydroxymellein.

Maintaining the postharvest quality of whole and fresh-cut fruit during storage and distribution is the major challenge facing fruit industry. For this purpose, industry adopt a wide range of technologies to enable extended shelf-life. Many factors can lead to loss of quality in fresh product, hence the common description of these products as ‘perishable’. As a consequence normal factors such as transpiration and respiration lead ultimately to water loss and senescence of the product. Fruits and vegetables are living commodities and their rate of respiration is of key importance to maintenance of quality. It has been commonly observed that the greater the respiration rate of a product, the shorter the shelf-life. The principal problem for fresh-cut fruit industries is the relative shorter shelf-life of minimally processed fruit (MPF) compared to intact product. This fact is strictly connected with the higher ethylene production of fruit tissue stimulated during fresh-cut processing (peeling, cutting, dipping). 1-Methylcyclopropene (1-MCP) is an inhibitor of ethylene action and several researches have shown its effectiveness on the inhibition of ripening and senescence incidence for intact fruit and consequently on their shelf-life extension. More recently 1-MCP treatment has been tested also for shelf-life extension of MPF but discordant results have been obtained. Considering that in some countries 1-MCP is already a commercial product registered for the use on a number of horticultural products, the main aim of this actual study was to enhance our understanding on the effects of 1-MCP treatment on the quality maintenance of whole and fresh-cut climacteric and non-climacteric fruit (apple, kiwifruit and pineapple). Concerning the effects of 1-MCP on whole fruit, was investigated the effects of a semi-commercial postharvest treatment with 1-MCP on the quality of Pink Lady apples as functions of fruit ripening stage, 1-MCP dose, storage time and also in combination with controlled atmospheres storage in order to better understand what is the relationship among these parameters and if is possible to maximize the 1-MCP treatment to meet the market/consumer needs and then in order to put in the market excellent fruit. To achieve this purpose an incomplete three-level three-factor design was adopted. During the storage were monitored several quality parameters: firmness, ripening index, ethylene and carbon dioxide production and were also performed a sensory evaluations after 6 month of storage. In this study the higher retention of firmness (at the end of storage) was achieved by applying the greatest 1-MCP concentration to fruits with the lowest maturity stage. This finding means that in these semi-commercial conditions we may considerate completely blocked the fruit softening. 1-MCP was able to delay also the ethylene and CO2 production and the maturity parameters (soluble solids content and total acidity). Only in some cases 1-MCP generate a synergistic effect with the CA storage. The results of sensory analyses indicated that, the 1-MCP treatment did not affect the sweetness and whole fruit flavour while had a little effect on the decreasing cut fruit flavour. On the contrary the treated apple was more sour, crisp, firm and juicy. The effects of some treatment (dipping and MAP) on the nutrient stability were also investigated showing that in this case study the adopted treatments did not have drastic effects on the antioxidant compounds on the contrary the dipping may enhance the total antioxidant activity by the accumulation of ascorbic acid on the apple cut surface. Results concerning the effects of 1-MCP in combination with MAP on the quality parameters behaviour of the kiwifruit were not always consistent and clear: in terms of colour maintenance, it seemed to have a synergistic effect with N2O MAP; as far as ripening index is concerned, 1-MCP had a preservative effect, but just for sample packed in air.

Blue mould caused by Penicillium expansum Link is one of the most destructive rot of pome fruit in all growing areas (Snowdon, 1990; Jones and Aldwinckle, 1991; Tonini,1996) In the past, Penicillium rot has been controlled by fungicide postharvest treatment mainly by thiabendazole (TBZ) and benomyl (Hardenburg and Spalding, 1972), but their intense use produced the appearance of resistant strains with a great reduction of their activity The aims of the present study were to characterize the isolates of Pencillium sp causing blue mold on pear in Italy by physiological and biochemical parameters. In particular differencing also the behavior of isolates to relationship with sensitivity or resistance to TBZ treatments. We have examined the early stage of infection in relation to enzyme activity, local modulation of pH, production of organic acids, and to secondary metabolism of pathogen. The results described here confirm that the majority of P. expansum isolates from pears packing houses are resistant to TBZ, Among the TBZ-resistant isolates scored in this work, different isolates (RR) showed higher percentage of conidial germination on TBZ-amended medium compared to non amended medium. This may indicate a stimulatory effect of TBZ on conidial germination. Therefore TBZ treatments are not only ineffective for controlling P. expansum, but they may also increase the severity of blue mould on fruits. In the absence of fungicide, isolates showed a significant difference for infection severity, R and RR isolates are characterized by higher pathogenic fitness on fruits, producing larger lesions than S isolates. These data are supported by the study with laboratory-induced resistant isolates, which shows the lack of correlation between TBZ resistance and osmotic sensitivity, and highlights the association between TBZ resistance and infection severity (Baraldi et al 2003). Enzymatic screening gave a positive reaction to esterase, urease, pectinase activity, in addition, the pathogen is able to synthesize a complex enzyme act to degrade the main components of the cell wall especially pectin and cellulose. Isolated sensitive and resistant are characterized by a good activity of pectinase, especially from poligactoronase, which, as already reported by several studies (D'hallewin et al, 2004; Prusky et al, 2004), are the basis of degradative process of cell wall. Also, although the measure was minor also highlighted some activities of cellulase, but even note in the production of this kind of cellulase and hemicellulase P. Expansum were not targeted, studies have found no other source of information in this regard. Twenty isolates of Penicillium expansum, were tested in vitro ad in vivo for acid production ability and pH drop. We have found that modulation of pH and the organic acids extrusion were influence to various parameter: Initial pH: in general, the greatest reduction of pH was observed in isolates grown at pH 7, except for four isolates that maintained the pH of the medium close to 7, the others significantly decreased the pH, ranging from 5.5 to 4.1.. In extreme acid condition (pH 3,0) growth and modulation of pH is most lower respect optimal condition (pH 5,0). Also isolates R and RR have showed a greater adaptation to environmental condition more than isolates S. Time: although the acidification continues for some days, PH modulation is strongest in early hours (48-72 hours)of inoculation process. Time also affects the quality of organic acids, for example in vitro results showed an initial abundant production of succinc acid, followed to important production of galacturoinc acid. Substrates: there are many differences for the type of acids produced in vitro and in vivo. Results showed in vivo an abundant production of galacturonic, malic, and citric acids and some unknown organic acids in smaller concentrations. Secondary metabolite analysis revealed intra-specific differences, and patulin was found in all isolates, but most significant reduction was observed between in vitro and in vivo samples. There was no correlation between the concentration of patulin, and the percentage of infected fruits, but sample with a lower infection severity of rotten area than the others, showed a significantly lower mycotoxin concentration than samples with a higher lesion diameter of rotten area. Beyond of patulin was detected the presence of another secondary metabolite, penitrem A.

The brown rot fungi belong to a group of fungal pathogens that causes considerable damage to cultivated fruits trees, particularly stone fruits and apples in the temperate regions of the World and during the postharvest with an important economic impact. In particular in Italy, it is important to monitor the Monilinia population to control economic losses associated to the peach and nectarine market. This motivates the research steps presented in this dissertation on Monilinia Italian isolates. The Monilinia species collected from stone fruits have been identified using molecular analysis based on specific primers. The relevant role of M. fructicola was confirmed and, for the first time, it was found also on apple fruits. To avoid the development of resistant strains and implement valid treatment strategies, the understanding of the fruit natural resistance during different developmental stages and the assessment of the Monilinia sensitivity/resistance to fungicides are required. The relationship between the inhibition spots and the phenolic compounds in peach fruit peel was highlighted in this research. Three methods were used to assess isolate resistance/sensitivity, the amended medium, the Spiral Gradient Endpoint Method (SGD) and the Alamar Blue method. The PCR was used to find possible mutation points in the b-tubulin gene that is responsible for fungicide resistance. Interestingly, no mutation points were observed in resistant M. laxa isolates, suggesting that the resistance could be stimulated by environmental factors. This lead to the study of the effect of the temperature on the resistance and the preliminary results of in vitro tests showed that maximum inhibition was observed at 30°C.

The postharvest phase has been considered an environment very suitable for successful application of biological control agents (BCAs). However, the tri-interaction between fungal pathogen, host (fruit) and antagonist is influenced by several parameters such as temperature, oxidative stresses, oxygen composition, water activity, etc. that could be determining for the success of biocontrol. Knowledge of the modes of action of BCAs is essential in order to enhance their viability and increase their potentialities in disease control. The thesis focused on the possibility to explain the modes of action of a biological control agent (BCA): Aureobasidium pullulans, in particular the strains L1 and L8, control effective against fruit postharvest fungal pathogen. In particular in this work were studied the different modes of action of BCA, such as: i) the ability to produce volatile organic compounds (VOCs), identified by SPME- gas chromatography-mass spectrometry (GC-MS) and tested by in vitro and in vivo assays against Penicillium spp., Botrytis cinerea, Colletotrichum acutatum; ii) the ability to produce lytic enzymes (exo and endo chitinase and β-1,3-glucanase) tested against Monilinia laxa, causal agent of brown rot of stone fruits. L1 and L8 lytic enzymes were also evaluated through their relative genes by molecular tools; iii) the competition for space and nutrients, such as sugars (sucrose, glucose and fructose) and iron; the latter induced the production of siderophores, molecules with high affinity for iron chelation. A molecular investigation was carried out to better understand the gene regulation strictly correlated to the production of these chelating molucules. The competition for space against M. laxa was verified by electron microscopy techniques; iv) a depth bibliographical analysis on BCAs mechanisms of action and their possible combination with physical and chemical treatments was conducted.

Silvicultural treatments represent disturbances to forest ecosystems often resulting in transient increases in net nitrification and leaching of nitrate and base cations from the soil. Response of soil carbon (C) is more complex, decreasing from enhanced soil respiration and increasing from enhanced postharvest inputs of detritus. Because nitrogen (N) saturation can have similar effects on cation mobility, timber harvesting in N-saturated forests may contribute to a decline in both soil C and base cation fertility, decreasing tree growth. Although studies have addressed effects of either forest harvesting or N saturation separately, few data exist on their combined effects. Our study examined the responses of soil C and N to several commercially used silvicultural treatments within the Fernow Experimental Forest, West Virginia, USA, a site with N-saturated soils. Soil analyses included soil organic matter (SOM), C, N, C/N ratios, pH, and net nitrification. We hypothesized the following gradient of disturbance intensity among silvicultural practices (from most to least intense): even-age with intensive harvesting (EA-I), even-age with extensive harvesting, even-age with commercial harvesting, diameter limit, and single-tree harvesting (ST). We anticipated that effects on soil C and N would be greatest for EA-I and least with ST. Tree species exhibited a response to the gradient of disturbance intensity, with early successional species more predominant in high-intensity treatments and late successional species more predominant in low-intensity treatments. Results for soil variables, however, generally did not support our predictions, with few significant differences among treatments and between treatments and their paired controls for any of the measured soil variables. Multiple regression indicated that the best predictors for net nitrification among samples were SOM (positive relationship) and pH (negative relationship). This finding confirms the challenge of sustainable management of N-saturated forests.

An in situ study was conducted to evaluate the effects of heat treatments on the degradation kinetics and escape protein concentrations of forages (alfalfa and berseem clover). Alfalfa collected at 4 and 7 weeks post-harvest and berseem clover collected at 5 and 7 weeks postharvest were freeze-dried and then heated to 100, 125, and 150o C for 2 hours. Heat treatment effects were determined by placing two bags of sample (for each treatment, maturity, and forage species for a given incubation times) into the rumen of one fistulated steer fed alfalfa hay. Bags were incubated for periods of 0 to 48 hours. Increasing levels of heat treatments of forages increased concentrations of neutral detergent fiber (NDF), acid detergent fiber (ADF), and acid detergent insoluble nitrogen (ADIN) and non-degradable protein (NDP), potentially degradable protein proportion (PDP), and protein escaping rumen degradation (PEP) while decreasing water soluble protein (WSP) and the rates of crude protein (CP), except immature berseem clover and cell wall (CW) degradation. PEP was greater and rate of CP degradation was lower at 100 and 150o C compared to 125o C in immature berseem clover.

The blaESBL and blaAmpC genes in Enterobacteriaceae are spread by plasmid-mediated integrons, insertion sequences, and transposons, some of which are homologous in bacteria from food animals, foods, and humans. These genes have been frequently identified in Escherichia coli and Salmonella from food animals, the most common being blaCTX-M-1, blaCTX-M-14, and blaCMY-2. Identification of risk factors for their occurrence in food animals is complex. In addition to generic antimicrobial use, cephalosporin usage is an important risk factor for selection and spread of these genes. Extensive international trade of animals is a further risk factor. There are no data on the effectiveness of individual control options in reducing public health risks. A highly effective option would be to stop or restrict cephalosporin usage in food animals. Decreasing total antimicrobial use is also of high priority. Implementation of measures to limit strain dissemination (increasing farm biosecurity, controls in animal trade, and other general postharvest controls) are also important.

Se evaluaron dos cultivares de arándano alto por su comportamiento en almacenamiento refrigerado convencional. Frutos completamente azules se seleccionaron, envasaron en "clamshells" y sometieron a almacenamiento refrigerado a 0 °C y 85-90 % HR durante 10, 17, 24 y 31 días. Para cada período de almacenamiento se realizaron muestreos: (1) a salida de frío y (2) después de dos días a temperatura ambiente. Se evaluó la homogeneidad en el empaque, pérdida de peso, firmeza, contenido de sólidos solubles, acidez titulable, pH y relación sólidos solubles:acidez titulable. El peso promedio de los frutos fue de 1.9 g, correspondiéndoles el grado "extralarge". La fruta se mantuvo en buenas condiciones durante 11 días de almacenamiento refrigerado, después perdió calidad como consecuencia de la pérdida de peso. Se encontró que Bluejay superó a Brigitta en firmeza. El contenido de sólidos solubles aumentó proporcionalmente a la duración del período de almacenamiento. En los períodos de 10 y 31 días Brigitta presentó mayor acidez titulable y menor relación sólidos solubles:acidez titulable que Bluejay. En ambos cultivares se observó un aumento significativo del pH al mes de almacenamiento. A partir de la correlación entre las variables evaluadas se infiere que el contenido de sólidos solubles y la firmeza constituyeron buenos índices de la calidad de la fruta en ambas variedades.

Objetivos: reducir pérdidas durante la conservación frigorífica, emplear atmósfera modificada como método suplementario a la refrigeración, alargar el período de aptitud comercial. Metodología: se trabajó con fruta acondicionada a 0±1 °C y 90±5 % HR, según las siguientes variantes: 1. testigo: 20 kg fruta a granel sin seleccionar en caja plástica; 2. granel + film PVC: 10 kg de fruta a granel en bandejas de madera más cartón corrugado recubierta con film de PVC; 3. celpack: bandejas de madera recubiertas de cartón corrugado con dos celpack de 23 frutos cada uno; 4. celpack + atmósfera modificada: ídem anterior pero cada celpack en bolsa de polietileno de baja densidad de 20 μ. A partir de los 30 días de conservación se extrajo semanalmente, durante 9 semanas, una muestra de 46 frutos, de los cuales 23 fueron analizados al momento de ser extraídos y los 23 restantes luego de 48 horas de comercialización simulada (sc). Para la evaluación estadística se aplicó análisis de la varianza con el programa SAS (Statistical Analysis System) y se determinaron las diferencias entre tratamientos con el test de Duncan. Para sabor, en cambio, se aplicó una prueba de homogeneidad de P2. La evaluación de sabor se realizó mediante degustación con panel de 5 catadores entrenados. Resultados: Los frutos tenían las siguientes características al inicio de conservación: calibre 61.4 mm, peso 117.8 g, firmeza de pulpa 3.1 kgf, sabor agridulce, contenido de sólidos solubles 17.5 °Bx, acidez 0.78 g ác. málico%g, % cubrimiento 83.69 %. Luego de la conservación frigorífica (97días): % de color de cobertura 95 %. La firmeza de la pulpa en el tratamiento celpack + bolsa se diferencia con valores más altos, media de 2.8 kgf , el resto con media 2.6 kgf. En sc la firmeza es inferior y esta disminución es menor en celpack + bolsa. Sólidos solubles, media 17.21 °Bx, en sc valores con media de un 0.3 % más. Acidez titulable: disminución progresiva, de 0.68 a 0.47 g%g al fin de conservación. Sabor: a partir de los 59 días aumentan los frutos insípidos y desagradables excepto en celpack + bolsa. Síntomas de deshidratación: a partir de los 79 días la única variante que no presenta síntomas es celpack + bolsa. Conclusiones: El acondicionamiento en celpack redujo la incidencia de ataque por mohos (fue el único tratamiento sin ataque durante 94 días); tampoco presentó sabores desagradables y su limitación en conservación se debió a la deshidratación evidente a partir de 74 días. La fruta embalada en celpack + bolsa tuvo mayores valores de resistencia a la presión y 100 % de frutos sin deshidratación a los 94 días de conservación; a partir de 80 días es evidente el ataque de mohos y frutos con sabores desagradables. Las variantes granel y granel + film presentan deterioro por deshidratación a partir de 74 días. La conservación no debería superar 80 días. Celpack + bolsa muestra mejores resultados, con mayores valores de resistencia a la presión que los otros tratamientos; con respecto al sabor, mantiene una mayor proporción de sabor dulce.

Los estudios realizados en la Argentina sobre poscosecha del género Allium se refieren a A. sativum, no habiéndolos sobre Allium ampeloprasum (ajo elefante), de buenas características organolépticas (sabor más suave) y creciente demanda en los mercados internos y externos. Por tal motivo se decidió investigar la incidencia del método de secado en su pérdida de peso comparando el procedimiento tradicional en caballete con el realizado en horno solar con circulación forzada de aire. En ambos casos se efectuaron 5 repeticiones para control de pérdida de peso. Se sacaron muestras al azar de los distintos niveles de las paseras para establecer la humedad en bulbo entero, bulbo pelado y hojas, efectuando 3 repeticiones para cada determinación y tratamiento. La duración de los tratamientos se determinó en base a la humedad en hoja y bulbo entero, puesto que la variación en bulbo pelado no fue significativa. Los datos fueron evaluados con el análisis de la varianza utilizando un nivel de significancia del 5 % y la prueba de Duncan. En las condiciones del ensayo, el secado en horno solar adelanta 6 días la terminación, con una pérdida de peso del 30,7 %.

En cerezos plantas con excesivo vigor son poco precoces, a menudo poco productivas y de difícil manejo en el cultivo. El exceso de vigor puede ser controlado con el uso de estrategias de riego deficitario controlado (RDC). Para contribuir a la racionalización del uso del recurso hídrico, controlar el crecimiento vegetativo vigoroso y estimular la producción precoz en plantaciones jóvenes de cerezo, se estableció un ensayo de RDC en un monte frutal comercial de la variedad Bing regado por goteo en la localidad de Agua Amarga, Mendoza, Argentina, Se evaluó la respuesta a distintos regímenes de riego poscosecha sobre parámetros de crecimiento vegetativo (crecimiento de brotes y tronco, área y peso seco foliar), reproductivo (densidad de floración, rendimiento y calidad de frutos) y estado nutricional (nutrimentos foliares y reservas de carbohidratos no estructurales). Los tratamientos de riego poscosecha fueron: riego a demanda plena (T1= Etc 100 %) y RDC reponiendo el 75 % (T2= Etc 75 %) y 50 % (T3= Etc 50 %) respecto de T1. Se midió el estado hídrico de la planta a través del potencial agua del tallo a mediodía y del suelo con sonda de capacitancia y gravimetría. En T3 disminuyó la longitud de brotes, número y longitud de entrenudos, número de hojas, área foliar y peso seco foliar, y área de tronco. En T2 disminuyó la longitud de brotes y de entrenudos. En T3 la intensidad del déficit hídrico impuesta aumentó la calidad de los ramilletes y la producción de yemas de flor, flores y frutos en el ciclo vegetativo siguiente. La calidad y madurez de frutos no fue afectada por los tratamientos de RDC, aunque en T3 aumentó levemente la proporción de frutos dobles. Luego del primer año de RDC en las plantas del T3 hubo una disminución significativa, aunque leve, del contenido de Ky P foliares y de almidón en raíces, El potencial hídrico del tallo a mediodía resultó un buen indicador del estado hídrico de las plantas. En cerezos un ajuste preciso del nivel de restricción hidrica poscosecha puede ser una estrategia de manejo para controlar el vigor y estimular la producción precoz, Al mismo tiempo se ahorran importantes cantidades de agua.
