The genome of all organisms is subject to injuries that can be caused by endogenous and environmental factors. If these lesions are not corrected, it can be fixed generating a mutation which can be lethal to the organisms. In order to prevent this, there are different DNA repair mechanisms. These mechanisms are well known in bacteria, yeast, human, but not in plants. Two plant models Oriza sativa and Arabidopsis thaliana had the genome sequenced and due to this some DNA repair genes have been characterized. The aim of this work is to characterized two sugarcane cDNAs that had homology to AP endonuclease: scARP1 and scARP3. In silico has been done with these two sequences and other from plants. It has been observed domain conservation on these sequences, but the cystein at 65 position that is a characteristic from the redox domain in APE1 protein was not so conservated in plants. Phylogenetic relationship showed two branches, one branch with dicots and monocots sequence and the other branch with only monocots sequences. Another approach in order to characterized these two cDNAs was to construct overexpression cassettes (sense and antisense orientation) using the 35S promoter. After that, these cassettes were transferred to the binary vector pPZP211. Furthermore, previously in the laboratory was obtained a plant from nicotiana tabacum containing the overexpression cassette in anti-sense orientation. It has been observed that this plant had a slow development and problems in setting seeds. After some manual crossing, some seeds were obtained (T2) and it was analyzed the T2 segregation. The third approach used in this work was to clone the promoter region from these two cDNAs by PCR walking. The sequences obtained were analyzed using the program PLANTCARE. It was observed in these sequences some motives that may be related to oxidative stress response

Los trabajos de campo son experiencias que docentes y alumnos pueden utilizar como estrategia didáctica para desarrollar destrezas y aumentar la motivación al abordar el tema ciencias. En el presente trabajo se describe una metodología puesta en práctica con alumnos del Profesorado en Ciencias Biológicas, que se propone a los docentes para planificar sus salidas de campo. A partir del trabajo de campo desarrollado se pudo evidenciar la importancia de éste tipo de actividades didácticas extra-áulicas para el desarrollo de actitudes y destrezas, tales como la organización de tareas, colaboración entre pares, redacción de informes científicos, además, sobre la base de hipótesis y objetivos previos, el abordaje interdisciplinario para la observación y análisis del ambiente en estudio.

Colêmbolos são pequenos artrópodes extremamente abundantes na natureza. São semelhantes a insetos em seu padrão de tagmose, ecologicamente muito importantes e podem ser usados como bioindicadores. Mais de 8.300 espécies já foram descritas no mundo, mas se estima que a riqueza real corresponda a um número muito maior. A Mata Atlântica corresponde ao domínio fitogeográfico mais alterado pela ação antrópica no Brasil, restando menos de 8,5% de sua cobertura original. No Rio Grande do Norte a Mata Atlântica cobre apenas 0,27% do território do Estado. Assim, considerando a importância de se conhecer a fauna de Collembola, sobretudo num ambiente ameaçado como a Mata Atlântica, este trabalho objetivou estudar a riqueza de colêmbolos Entomobryoidea e Symphypleona ocorrentes em dois remanescentes de Mata Atlântica do Rio Grande do Norte através de coletas ativas com aspirador entomológico e passivas com pitfall, durante os períodos de chuva e seca. Foram identificadas 22 espécies de colêmbolos, sendo que destas 19 (mais de 85% do total) são novas para a ciência, incluindo um novo gênero da família Sminthuridae. As coletas realizadas no período chuvoso apresentaram praticamente o dobro de espécies em relação às realizadas no período seco. Este fato possivelmente está associado à dependência dos colêmbolos de umidade, assim como estratégias de resistência a período secos. São descritas quatro novas espécies, incluindo o novo gênero (Trogolaphysa sp.nov., Gen.nov. sp.nov., Szeptyckitheca sp.nov. e Sminthurinus sp.nov.). Este expressivo número de espécies novas identificadas, os novos registros para o gêneros e famílias para a Mata Atlântica do Rio Grande do Norte e a descoberta de um novo gênero corroboram o pressuposto que motivou este trabalho: o Rio Grande do Norte apresenta uma fauna de Collembola desconhecida e potencialmente diversificada. Além disso, foram geradas informações importantes sobre a morfologia das espécies descritas, que por sua vez são de grande importância para a sistemática dos Collembola. O novo gênero descrito faz parte de um visível subgrupo dentro da família Sminthuridae (aqueles com o unguis com cavidade) que aparentemente tem potencial para ser reconhecido futuramente em alguma categoria taxonômica supragenérica.

Prospecting pharmacological active polysaccharides from agricultural byproducts, such as corncobs, is an underexplored practice in the scientific community. Thus, this work aims to expand knowledge about pharmacological activities of polysaccharides extracted from corncobs. From corn cob flour a extract was obtained by ultrasound waves in an alkaline medium, and the end of the process the product was termed PECC (polysaccharidic extract from corncobs). This extract was physicochemical characterized and evaluated by in vitro assays as an antioxidant, cytotoxic, anticoagulant and imunomodulator agent. Results indicated significant activity metal chelating by PECC, and the use of PECC in cell culture cells showed no toxic effects to normal cell lines, but toxic action against HeLa tumor cells due promoting cell death by apoptosis. In addition, other pharmacological effects were observed, the PECC decreased nitric oxide (NO) production by activated macrophages, and prolonged blood clotting time through APTT assay. Then methanolic, ethanolic and ketone fractions were obtained from fractionation of PECC polysaccharides. Five methanolic fractions, six ethanolic fractions and two ketones were obtained; and all fractions were evaluated for antioxidant, cytotoxic, anticoagulant, immunomodulatory activities. E1.4 fraction exhibited significant metal chelating effect, a toxic action to induce apoptosis in HeLa cells, decreased NO production by activated macrophages, and extended blood clotting time. These results showed that the PECC pharmacological active polysaccharides would be present in the fraction E1.4. From fractionation of E1.4 polysaccharide six subfractions with different sizes were obtained: <3; 3-10; 10-30; 30-50; 50-100 and >100 KDa. About 80% of E1.4 polysaccharides had lower size to 10 KDa, and all the subfractions showed over 61% sugar in their chemical compositions. These subfractions exhibited different monosaccharide compositions, but xylose was presented in all of them. The subfractions exhibited distinct pharmacological effects in in vitro assays. Smaller subfractions (<30 KDa) had highest metal chelating activity and greater toxic action in tumor cells. The intermediate fractions (between 30-100 KDa) decreased more NO production of activated macrophages, for other side, the larger size (>100 KDa) modulated a greater number of inflammatory cytokines, and the had greatest anticoagulant effect. Therefore, when analyzing all the results together it is evident that the PECC pharmacological polysaccharides are heteroxylans, and were concentrated in E1.4 fraction, and heteroxilanas pharmacological effects depends on their molecular size. Thus, corncobs could be used as source from molecules with biotechnology potential

Agriculture is one of the most discussed topics currently in the conceptual field of sustainability. The debates are increasingly recurrent and put in question the model adopted from post-war, so-called green revolution, for its potential of degradation of natural resources. This type of Agriculture put Brazil at the top of the global agribusiness, where stands out in various sectors such as grain, meat, sugar and horticulture. Discussions are focused on aspects related to the use of agrochemicals, monoculture, conversion of native forest in extensive agricultural areas, among other points taken as deleterious to environmental balance. On the other hand, there is a model, called by family farming, which for many researchers, has attributes closer to the understanding of sustainable agriculture. In the state of Rio Grande do Norte, the agricultural potential lies mainly on horticulture, where stands the agropolo AcuMossoró, as one of the greatest tropical fruit producing regions of Brazil, being melon, the major fruit produced. The cultivation of this vegetable was developed in the region in the late 1980s, from the investment of large agricultural enterprises, whose cultivation techniques were grounded by the green revolution. Currently, the melon cultivation is also developed in agroecosystems whose management is characterized by family participation, including small farmers of rural settlements created by Instituto Nacional de Colonização e Reforma Agrária (INCRA). In view of the inclusion of family farming in a field that recently was dominated by large agribusiness companies, some questions arise about the maintenance of attributes that characterise this type of family agriculture management. This research aimed to assess the sustainability of family agroecosystems in São Romão settlement in Mossoró-RN, cultivated with melon. The study was conducted by the Framework for Evaluation of Natural Resources Management Systems Incorporating Sustainability Indicators (MESMIS), in ten agroecosystems of the mentioned settlement. The data were obtained from semi-structured interviews and field observations, so that the answers, considerations and comments made by settlers, were widely used to cycle through the six steps of the MESMIS evaluation. As a result of the work, were determined seven critical points affecting sustainability, being: water resources, soils, reliance on external inputs, biodiversity, quality of life, family income and community organizing, from which was derived twenty-three indicators that sought to reflect the actual state of sustainability of the agroecosystems

Dengue is a viral disease transmitted by female mosquitoes from genus Aedes, the principal urban vector is Aedes aegypti. Actually dengue has caused, in global scale, substantial morbidity and mortality. Four serotypes (antigenically distinct) are known: DENV-1, DENV-2, DENV-3 and DENV-4. The objective of this study was described the epidemiological profile dengue in the states of Rio Grande do Norte (RN) and Paraíba (PB), 2013. For that, suspected cases of dengue were studied, received for Laboratory of Molecular Biology of infectious disease and cancer (LADIC-UFRN) from different Health Units from RN and PB between January and December of 2013. The viral RNA was obtained from serum samples of patient from health units from RN and PB. It were studied 478 suspected cases of dengue , 252 (52,7%) from Rio Grande do Norte and 226 (47,3%) from Paraíba, showeds a global rate of infection global prevalence of 29,7% (142/478). The co-circulation of three serotypes was observed: DENV-1 (9,8% [14/142]), DENV-2 (3,5% [5/142]) and DENV-4 (86,7% [123/142]). People between 21-30 years old were the most affected by the disease during all the period of the study, representing 63,7% of the cases in both states. The genus most affected was female, representing 63,3% of cases in both states. Pau dos Ferros, Rio Grande do Norte, had the highest circulation of disease, with 8,2% (8/97) of cases. In Paraíba, the city most affected was João Pessoa, with (80% (36/45) of cases. The months with the biggest viral circulation in RN and PB were March and August, respectively. These results are very important to understanding the dengue viral activity in RN and PB, providing data that can guide control actions of this disease in support to local control programs

The use of fossil fuels has been considered one of reason for the increase of pollution in the atmosphere and it may be related to the climate changes. Then, the research of the new sources of fuels will be important. Considering this, the use of biodiesel has been considered not as bad as petrol. The castor bean (Ricinus communis L.) is an important oilseed, which belongs to Euphorbiaceae family, and the oil found in the seed has important characteristics for biodiesel. This plant is considered as “rustic” as it does not need so much water for its development and oil production. Due to this, this plant has been considered to be ideal in semi-arid regions, such as the Northeast of Brazil. The aim of his study is to better understand the responses to abiotic stresses (drought and salinity) from castor bean plants using morphological, physiological and molecular tools. In order to do this, the castor bean plants were subjected to salt stress (50, 100, 150 and 200 mM NaCl) in a controlled environment and drought stress (5, 10, 15 days and 10 days cyclic). After these treatments, these plants were subjected to different analyzes: a) the expansion and retention of water from leaves; b) anatomy using leaves and roots. Based on these results, we found that castor suffered decrease in leaf area with increase drought stress, however restricted water loss, probably by accumulation of compatible solutes in the leaves. The anatomy data showed modifications in the vascular system. These modifications observed suggested that castor bean plant may be resistant to stress as it was verified in 5 days of drought as well as in 100 mM NaCl. In both conditions, these plants were fine. Probably these plants keep some solutes in the cell and then maintain the cell tugor. The data obtained in this study gave a better idea how castor bean plant responds to abiotic stress conditions - drought and salt stress

Chitin is the second most abundant polysaccharide in nature and its derivative chitosan has been widely studied due to its unique chemical and pharmacological properties. However, studies show that when this molecule is used as food, drug, etc. it tends to accumulate in renal tissue and promotes an increase in calcium excretion. Nevertheless, the effect of chitosan on the formation of calcium oxalate (OxCa) crystals has never been evaluated. The formation of kidney stones (urolithiasis) is the disease that most often affects the kidneys and the urinary system. In addition, this is a disease with high prevalence and recurrence. Many molecules with antioxidant activity have been shown to decrease the potential for in vitro OxCa crystals formation. Thus, the aim of this study was to evaluate the effect of low molecular weight chitosan and its derivatives conjugated to gallic acid (AG) as antioxidant and inhibitor of OxCa crystals formation. The physico-chemical analysis confirmed the identity of chitosan. This molecule was subjected to five antioxidant tests and showed an excellent copper chelating activity. However, chitosan did not show other significant antioxidant activity. When chitosan was subjected to in vitro crystal formation tests, it increased the number of OxCa monohydrate crystals, modified the morphology of the crystals, modified the proportions between populations of crystals in solution and increased the zeta potential of these crystals formed. Four molecules of chitosan conjugated with GA were obtained. The physico-chemical analysis confirmed that chitosan and AG were covalently bonded. However, the amount of GA liked to chitosan did not increase even when 10 times more GA was used in experiment. When these derivatives were subjected to antioxidant tests, all chitosan conjugates showed higher antioxidant potential than their precursors. However, they showed different activity between them, which indicating that the position where AG is conjugated is an important factor for chitosan-GA activity. When conjugated chitosans were submitted to in vitro crystal formation tests, a reduction in the crystals number was observed when compared with those formed in the presence of unconjugated chitosan. Chitosan has a strong capacity for inducing OxCa monohydrate crystal formation, as well as modify their morphology and zeta potential. Over all, the process of conjugating AG to chitosan led to an increase in antioxidant potential of this molecule and was also able to decrease its capacity of inducing in vitro crystal formation

The behavioral decisions of animals do not occur randomly, because behaviors are adjusted to ensure the survival and reproduction of the animal. In this research, I examined behavioral decisions in the foraging context of the ant Dinoponera quadriceps with regard to orientation, food avaliation and foraging dynamic to individual level. The study was conducted at the Laboratory of Behavioral Biology at UFRN and in an area of secondary Atlantic Forest in FLONA-ICMBio Nísia Floresta/RN. In all observations and experiments, ants were marked individually with an alphanumeric code label fixed on the thorax. In the first part of the study, I analyzed the orientation cues used by D. quadriceps. The tests were performed in a maze of 17 compartments. Each forager was tested for 10 min in three sessions for six different treatments. The treatments consisted of the presence or absence of odor and superior or frontal visual cues. The workers demonstrated that the presence of odor is indispensable and front visual cues are more effective than superior visual cues. In the second part, I investigated the discrimination of food, considering the parameters, size, weight and volume. In a 'cafeteria' experiment, I offered cylindrical pieces of food (mortadella) in a Petri dish, within an experimental arena 1m². Initially, the pieces were of four different sizes; in a second step, the pieces were of the same size but with different weight; in the last step, the pieces had the same weight but different volumes. The results showed the effect of the size and weight parameters for food choice. In the third part of the study, I evaluated the influence of the activity of active foragers on inactive ones. In this part, the colonies were observed in a natural environment. The observations took place on three consecutive days in 10 episodes, total of 30 days for each colony, 12 hours/day. On the first day, I registered the output and input of workers; on the second day, the most active ants on the first day were taken and given back at the end of the observations; on the third day, the observations were similar to the first day. As a result, the workers of D. quadriceps show autostimulation and they do not show social facilitation and the colony compensates the absence of the most active workers. Based on the stated, I conclude that workers of D. quadriceps use chemical, frontal and superior visual orientation cues during their displacements. They discriminate the chosen food by size and weight. The regulation of activity dynamics of foragers is by autostimulation, an active worker does not influence the activity of an inactive worker, the successful search previous is the stimulus to the successful worker itself to continue foraging activity.
