Globine sind kleine globuläre Proteine mit nahezu ubiquitärem Vorkommen in allen Tiergruppen. Sie weisen eine typische Sandwichstruktur auf, die in der Regel aus acht α-Helices mit einer zentralen prosthetischen Häm-Gruppe besteht und die Proteine zur Bindung gasförmiger Liganden befähigt. Die Funktionen der Globine reichen von O2-Transport und – Speicherung, über eine Beteiligung bei der Entgiftung reaktiver Sauerstoff- und Stickstoffspezies bis hin zu sensorischen physiologischen Aufgaben. Innerhalb der Klasse der Insekten schien das Vorhandensein von Globinen zunächst auf Insekten mit offensichtlich hypoxischen Habitaten beschränkt zu sein. Die Entdeckung des Globins glob1 in Drosophila melanogaster deutete jedoch eine sehr viel weitere Verbreitung der Globine in Insekten an, die sich durch die Identifizierung von Globingenen in einer Vielzahl von normoxisch lebenden Insekten, wie z.B. Apis mellifera oder Aedes aegypti bestätigte. D. melanogaster besitzt drei Globine, glob1, glob2 und glob3. Glob1 ist eng mit anderen intrazellulären Insektenglobinen verwandt, was zu der Annahme führte, dass es sich bei glob1 um das ursprüngliche und bei glob2 und glob3 um abgeleitete D. melanogaster Globine handelt. Glob1 wird in allen Entwicklungsstadien exprimiert, wobei die Hauptexpressionsorte der Fettkörper und das Tracheensystem sind. Die Transkription des glob1 startet von zwei alternativen Promotoren (Promotor I und II), wodurch in Kombination mit alternativem Splicing vier Transkriptvarianten (Isoform A-D) entstehen, deren Translation jedoch in einer Proteinvariante (glob1) resultiert. Hypoxische Bedingungen führen zu einer vermutlich HIF (=‚hypoxia-inducible factor‘) -vermittelten Abnahme der glob1 Genexpression, wohingegen Hyperoxie eine leichte Zunahme der glob1 mRNA Menge bewirkt. Der mithilfe des UAS/Gal4- Systems erzeugte, RNAi-vermittelte glob1 Knockdown führt zu einer schlechteren Überlebensrate adulter Fliegen unter hypoxischen Bedingungen, einer verkürzten Erholungszeit nach hypoxischem Stupor in Weibchen sowie zu einer erhöhten Resistenz gegenüber dem ROS (=‘reactive oxygen species‘) -generierenden Herbizid Paraquat in Larven und adulten Weibchen. Diese Beobachtungen sprechen für eine Funktion des Drosophila glob1 innerhalb der O2-Versorgung. Unter hyperoxischen Bedingungen hingegen wurde kein Unterschied zwischen Fliegen mit wildtypischer und manipulierter glob1-Expression festgestellt, wodurch eine Beteiligung des glob1 bei der Entgiftung reaktiver Sauerstoffspezies als mögliche Funktion vorerst ausscheidet. Bei glob2 und glob3 handelt es sich um duplizierte Gene. Auf phylogenetischen Rekonstruktionen basierend konnte die Entstehung der Globin-Duplikate auf ein Duplikationsereignis vor der Radiation des Subgenus Sophophora vor mindestens 40 Millionen Jahren zurückgeführt werden. Die durchgeführten Analysen zur molekularen Sequenzevolution der Globin-Duplikate deuten darauf hin, dass glob2 und glob3 nach der Duplikation eine Kombination aus Sub- und Neo-Funktionalisierungsprozessen durchlaufen haben. Glob2 und glob3 zeigen eine deckungsgleiche mRNA Expression, die auf die männliche Keimbahn beschränkt ist. Aufgrund des hohen Konservierungsgrads der für die Häm- und O2-Bindung essentiellen Aminosäuren kann von der Funktionalität beider Proteine ausgegangen werden. Die streng auf die männliche Keimbahn begrenzte Expression von glob2 und glob3 deutet auf eine Rolle der Globin-Duplikate innerhalb der Spermatogenese hin, die möglicherweise in einem Schutz der Spermatogenese vor oxidativem Stress besteht. Auch eine Beteiligung beim korrekten Ablauf der Spermien-Individualisierung, beispielsweise durch Regulation von Apoptoseprozessen wäre denkbar.

Die Messung eines möglichen elektrischen Dipolmoments des freien Neutrons erfordert genaustmögliche Kenntnis und Überwachung des magnetischen Feldes im Inneren der n2EDM-Spektrometerkammer. Die freie Spinpräzession von hyperpolarisiertem ³He kann verbunden mit einer Signalauslese mittels optisch gepumpter Cs-Magnetometer dazu genutzt werden, Messempfindlichkeit auf Magnetfeldschwankungen im Bereich weniger Femto-Tesla zu erhalten. Am Institut für Physik der Universität Mainz wurde eine ³He/Cs-Testanlage aufgebaut, um die Möglichkeiten der Signalauslese der ³He-Spinpräzession mittels eines lampengepumpten Cs-Magnetometers zu untersuchen. Darüber hinaus wurde eine ultrakompakte und transportable Polarisationseinheit entwickelt und installiert, welche ermöglicht, eine ³He-Hyperpolarisation von bis zu 55 Prozent zu erreichen. Im Anschluss wird das polarisierte 3He-Gas automatisiert komprimiert und in zwei Magnetometerzellen in Sandwichanordnung innerhalb der n2EDM-Spektrometerkammer gefüllt. In dieser Arbeit werden die Ergebnisse der ersten im Januar 2012 erfolgreich durchgeführten Messungen vorgestellt. Bei diesen Messungen wurde ³He-Gas in der ultrakompakten Polarisationseinheit hyperpolarisiert und über Führungsfelder eines Transfersystems in eine vierlagige Mumetall-Abschirmung transferiert. Im Anschluss konnte im Inneren der magnetischen Abschirmung die freie ³He-Spinpräzession mittels eines lampengepumpten Cs-Magnetometer eindeutig nachgewiesen werden.

Die Transmissionselektronenmikroskopie gepaart mit bioinformatischen Methoden zur digitalen Bildverarbeitung ist ein schneller Weg zur Erstellung dreidimensionaler Rekonstruktionen großer Proteinkomplexe. Durch die Kombination der 3D-Elektronenmikroskopie mit der Röntgenstruktur von Untereinheiten erhält man ein pseudoatomares Modell der Quartärstruktur.rnIn dieser Arbeit wurden auf diese Weise die Quartärstrukturen von drei unterschiedlichen respiratorischen Proteinen analysiert (einem Hämoglobin und zwei Hämocyaninen). Zudem wurden spezielle Software-Tools entwickelt, um vorhandene Softwarepakete besser miteinander kombinieren zu können.rnDie ca. 15Å 3D-Rekonstruktion des Hämoglobins vom Wasserfloh Daphnia pulex klärt die umstrittene Frage, wie viele Untereinheiten die Quartärstruktur aufbauen: Es sind 16 (mit je zwei Häm-Domänen), angeordnet in zwei Schichten als D4-symmetrisches Sandwich. Die ca. 15 Å 3D-Rekonstruktion des 2x6meren Hämocyanins des Flusskrebses Astacus leptodactylus gibt neue Einblicke in die Kontaktstelle zwischen den beiden Hexameren; sie liegt im Bereich der Domäne 3. Bei dem aus 48 Untereinheiten bestehenden Hämocyanin des Pfeilschwanzes Limulus polyphemus wurde eine Auflösung von ca. 7 Å erreicht. Die Homologiemodelle der Untereinheiten wurden flexibel gefittet. An einer der Kontaktstellen zwischen den beiden Halbmolekülen wurden Molekulardynamik (MD)-Simulationen durchgeführt, um mehr über die Art der chemischen Bindung an dieser Kontaktstelle zu erfahren.rnSpeziell für die Kombination von 3D-Elektronenmikroskopie und MD-Simulation wurden verschiedene bioinformatische Werkzeuge und eine leicht zu erweiternde universelle grafische Benutzeroberfläche (GUI) entwickelt.

This thesis investigates metallic nanostructures exhibiting surface plasmon resonance for the amplification of fluorescence signal in sandwich immunoassays. In this approach, an analyte is captured by an antibody immobilized on a plasmonic structure and detected by a subsequently bound fluorophore labeled detection antibody. The highly confined field of surface plasmons originates from collective charge oscillations which are associated with high electromagnetic field enhancements at the metal surface and allow for greatly increased fluorescence signal from the attached fluorophores. This feature allows for improving the signal-to-noise ratio in fluorescence measurements and thus advancing the sensitivity of the sensor platform. In particular, the thesis presents two plasmonic nanostructures that amplify fluorescence signal in devices that rely on epifluorescence geometry, in which the fluorophore absorbs and emits light from the same direction perpendicular to the substrate surface.rnThe first is a crossed relief gold grating that supports propagating surface plasmon polaritons (SPPs) and second, gold nanoparticles embedded in refractive index symmetric environment exhibiting collective localized surface plasmons (cLSPs). Finite-difference time-domain simulations are performed in order to design structures for the optimum amplification of established Cy5 and Alexa Fluor 647 fluorophore labels with the absorption and emission wavelengths in the red region of spectrum. The design takes into account combined effect of surface plasmon-enhanced excitation rate, directional surface plasmon-driven emission and modified quantum yield for characteristic distances in immunoassays. Homebuilt optical instruments are developed for the experimental observation of the surface plasmon mode spectrum, measurements of the angular distribution of surface plasmon-coupled fluorescence light and a setup mimicking commercial fluorescence reading systems in epifluorescence geometry.rnCrossed relief grating structures are prepared by interference lithography and multiple copies are made by UV nanoimprint lithography. The fabricated crossed diffraction gratings were utilized for sandwich immunoassay-based detection of the clinically relevant inflammation marker interleukin 6 (IL-6). The enhancement factor of the crossed grating reached EF=100 when compared to a flat gold substrate. This result is comparable to the highest reported enhancements to date, for fluorophores with relatively high intrinsic quantum yield. The measured enhancement factor excellently agrees with the predictions of the simulations and the mechanisms of the enhancement are explained in detail. Main contributions were the high electric field intensity enhancement (30-fold increase) and the directional fluorescence emission at (4-fold increase) compared to a flat gold substrate.rnCollective localized surface plasmons (cLSPs) hold potential for even stronger fluorescence enhancement of EF=1000, due to higher electric field intensity confinement. cLSPs are established by diffractive coupling of the localized surface plasmon resonance (LSPR) of metallic nanoparticles and result in a narrow resonance. Due to the narrow resonance, it is hard to overlap the cLSPs mode with the absorption and emission bands of the used fluorophore, simultaneously. Therefore, a novel two resonance structure that supports SPP and cLSP modes was proposed. It consists of a 2D array of cylindrical gold nanoparticles above a low refractive index polymer and a silver film. A structure that supports the proposed SPP and cLSP modes was prepared by employing laser interference lithography and the measured mode spectrum was compared to simulation results.rn

Il teorema di Borsuk-Ulam asserisce che, data una funzione continua f da S^n in R^n, esiste una coppia di punti antipodali sulla sfera che vengono mandati da f nello stesso punto. In questa tesi si vede l'equivalenza di sei diverse formulazioni del teorema e se ne dà una dimostrazione nel caso 2-dimensionale, utilizzando spazi di orbite, gruppo fondamentale e rivestimenti. Si studiano alcune sue dirette conseguenze come generalizzazioni di risultati preliminari sulla suddivisione di regioni piane, dandone anche un’interpretazione fisica e si vede come tutto questo si applica al “Ham Sandwich Theorem” in R^3.

BACKGROUND.: Urine is a potentially rich source of biomarkers for monitoring kidney dysfunction. In this study, we have investigated the potential of soluble human leukocyte antigen (sHLA)-DR in the urine for noninvasive monitoring of renal transplant patients. METHODS.: Urinary soluble HLA-DR levels were measured by sandwich enzyme-linked immunosorbent assay in 103 patients with renal diseases or after renal transplantation. sHLA-DR in urine was characterized by Western blotting and mass spectrometry. RESULTS.: Acute graft rejection was associated with a significantly elevated level of urinary sHLA-DR (P<0.0001), compared with recipients with stable graft function or healthy individuals. A receiver operating characteristic curve analysis showed the area under the curve to be 0.88 (P<0.001). At a selected threshold, the sensitivity was 80% and specificity was 98% for detection of acute renal transplant rejection. sHLA-DR was not exosomally associated and was of lower molecular weight compared with the HLA-DR expressed as heterodimer on the plasma membrane of antigen-presenting cells. CONCLUSIONS.: sHLA-DR excreted into urine is a promising indicator of renal transplant rejection.

We reported the first application of in situ shell-isolated nanoparticle enhanced Raman spectroscopy (SHINERS) to an interfacial redox reaction under electrochemical conditions. We construct gap-mode sandwich structures composed of a thiol-terminated HS-6V6H viologen adlayer immobilized on a single crystal Au(111)-(1x1) electrode and covered by Au(60 nm)@SlO(2) core shell nanoparticles acting as plasmonic antennas. We observed high-quality, potential-dependent Raman spectra of the three viologen species V(2+),V(+center dot) and V(0) on a well-defined Au(111) substrate surface and could map their potential-dependent evolution. Comparison with experiments on powder samples revealed an enhancement factor of the nonresonant Raman modes of similar to 3 x 10(5), and up to 9 x 10(7) for the resonance modes. The study illustrates the unique capability of SHINERS and its potential in the entire field of electrochemical surface science to explore structures and reaction pathways on well-defined substrate surfaces, such as single crystals, for molecular, (electro-)- catalytic, bioelectrochemical systems up to fundamental double layer studies at electrified solid/liquid interfaces.

Marginal generalized linear models can be used for clustered and longitudinal data by fitting a model as if the data were independent and using an empirical estimator of parameter standard errors. We extend this approach to data where the number of observations correlated with a given one grows with sample size and show that parameter estimates are consistent and asymptotically Normal with a slower convergence rate than for independent data, and that an information sandwich variance estimator is consistent. We present two problems that motivated this work, the modelling of patterns of HIV genetic variation and the behavior of clustered data estimators when clusters are large.

BACKGROUND: In contrast to RIA, recently available ELISAs provide the potential for fully automated analysis of adiponectin. To date, studies reporting on the diagnostic characteristics of ELISAs and investigating on the relationship between ELISA- and RIA-based methods are rare. METHODS: Thus, we established and evaluated a fully automated platform (BEP 2000; Dade-Behring, Switzerland) for determination of adiponectin levels in serum by two different ELISA methods (competitive human adiponectin ELISA; high sensitivity human adiponectin sandwich ELISA; both Biovendor, Czech Republic). Further, as a reference method, we also employed a human adiponectin RIA (Linco Research, USA). Samples from 150 patients routinely presenting to our cardiology unit were tested. RESULTS: ELISA measurements could be accomplished in less than 3 h, measurement of RIA had a duration of 24 h. The ELISAs were evaluated for precision, analytical sensitivity and specificity, linearity on dilution and spiking recovery. In the investigated patients, type 2 diabetes, higher age and male gender were significantly associated with lower serum adiponectin concentrations. Correlations between the ELISA methods and the RIA were strong (competitive ELISA, r=0.82; sandwich ELISA, r=0.92; both p<0.001). However, Deming regression and Bland-Altman analysis indicated lack of agreement of the 3 methods preventing direct comparison of results. The equations of the regression lines are: Competitive ELISA=1.48 x RIA-0.88; High sensitivity sandwich ELISA=0.77 x RIA+1.01. CONCLUSIONS: Fully automated measurement of adiponectin by ELISA is feasible and substantially more rapid than RIA. The investigated ELISA test systems seem to exhibit analytical characteristics allowing for clinical application. In addition, there is a strong correlation between the ELISA methods and RIA. These findings might promote a more widespread use of adiponectin measurements in clinical research.

BACKGROUND ; AIMS: Pancreatic and bile duct carcinomas represent highly aggressive malignancies that evolve from secretin receptor-rich ductular cells. With premessenger RNA splicing abnormalities common in cancer, we evaluated whether an abnormal secretin receptor spliceoform were present, characterized it, and developed a serum assay for it. METHODS: Cancer cell lines and healthy and neoplastic tissue were studied by nested reverse-transcription polymerase chain reaction and sequencing. A promising spliceoform was isolated and characterized, and monoclonal antibodies were raised to 2 distinct regions. A dual antibody enzyme-linked immunosorbent assay was developed and applied to blinded serum samples from 26 patients with pancreatic carcinoma, 10 patients with chronic pancreatitis, and 14 controls. RESULTS: Each of 9 pancreatic cancer specimens and no normal tissue expressed a secretin receptor variant with exons 3 and 4 deleted. This encoded a 111-residue peptide with its first 43 residues identical to wild-type receptor, but, subsequent to a shift in coding frame and early truncation, the next 68 residues were unique in the transcriptome/proteome. This nonfunctional soluble protein did not bind or signal in response to secretin and was secreted from transfected MiaPaCa-2 cells. Elevated serum levels of this variant were present in 69% of pancreatic cancer patients, 60% of chronic pancreatitis patients, and 1 of 14 controls. CONCLUSIONS: We identified a novel abnormal spliceoform of the secretin receptor in pancreatic and bile duct cancers and developed a dual antibody sandwich enzyme-linked immunosorbent assay to measure it in the circulation. Initial application of this assay in patients with pancreatic cancer and chronic pancreatitis was promising, but additional validation will be required to evaluate its clinical utility.

The GH-IGF axis has profound effects on the local and systemic regulation of bone metabolism and may be important for quality of fracture healing. To test the hypothesis that deficiency of the GH/IGF axis may play a role in the pathogenesis of fracture non-union we investigated whether alterations of serum concentrations of the GH-IGF axis could be related to failed fracture healing compared to timely fracture healing in trauma patients. Serum probes were prospectively collected from 186 patients with surgical treatment of long bone fractures up to 6 months after surgery. Samples from 14 patients with atrophic type of non-union have been compared to 14 matched patients with normal bone healing. Postoperative time courses of serum concentrations have been analyzed using commercially available chemiluminescence sandwich assays (GH), fully automated assay systems (IGF-I, IGFBP-3) or sandwich immunometric assays (ALS). Comparison between both collectives revealed significantly lower serum concentrations of GH dependent ALS during early (1st week after surgery) and of both IGFBP-3 and ALS during late stages of fracture healing (6 and 8 weeks after surgery) in non-union patients, coinciding clinically with failed fracture healing. Tendentially lower serum levels of IGF-I in the non-union group over the entire investigation period were statistically not significant. We have been able to show time courses of serum concentrations of the GH/IGF-I axis during normal and failed fracture healing in humans. An impairment of the GH/IGF-I axis might be involved in the biochemical mechanisms determining delayed or failed fracture healing.

BACKGROUND: Cerebral revascularization may be indicated either for blood flow preservation or flow augmentation, often in clinical situations where neither endovascular nor standard surgical intervention can be performed. Cerebral revascularization can be performed by using a temporary occlusive or a non-occlusive technique. Both of these possibilities have their specific range of feasibility. Therefore non-occlusive revascularization techniques have been developed. To further reduce the risks for patients, less time consuming, sutureless techniques such as laser tissue soldering are currently being investigated. METHOD: In the present study, a new technique for side-to-side anastomosis was developed. Using a "sandwich technique", two vessels are kept in close contact during the laser soldering. Thoraco-abdominal aortas from 24 different rabbits were analyzed for laser irradiation induced tensile strength. Two different irradiation modes (continuous and pulsed) were used. The results were compared to conventional, noncontact laser soldering. Histology was performed using HE, Mason's Trichrome staining. FINDINGS: The achieved tensile strengths were significantly higher using the close contact "sandwich technique" as compared to the conventional adaptation technique. Furthermore, tensile strength was higher in the continuously irradiated specimen as compared to the specimen undergoing pulsed laser irradiation. The histology showed similar denaturation areas in both groups. The addition of a collagen membrane between vessel components reduced the tensile strength. CONCLUSION: These first results proved the importance of close and tight contact during the laser soldering procedure thus enabling the development of a "sandwich laser irradiation device" for in vivo application in the rabbit.

Molecules are the smallest possible elements for electronic devices, with active elements for such devices typically a few Angstroms in footprint area. Owing to the possibility of producing ultrahigh density devices, tremendous effort has been invested in producing electronic junctions by using various types of molecules. The major issues for molecular electronics include (1) developing an effective scheme to connect molecules with the present micro- and nano-technology, (2) increasing the lifetime and stabilities of the devices, and (3) increasing their performance in comparison to the state-of-the-art devices. In this work, we attempt to use carbon nanotubes (CNTs) as the interconnecting nanoelectrodes between molecules and microelectrodes. The ultimate goal is to use two individual CNTs to sandwich molecules in a cross-bar configuration while having these CNTs connected with microelectrodes such that the junction displays the electronic character of the molecule chosen. We have successfully developed an effective scheme to connect molecules with CNTs, which is scalable to arrays of molecular electronic devices. To realize this far reaching goal, the following technical topics have been investigated. 1. Synthesis of multi-walled carbon nanotubes (MWCNTs) by thermal chemical vapor deposition (T-CVD) and plasma-enhanced chemical vapor deposition (PECVD) techniques (Chapter 3). We have evaluated the potential use of tubular and bamboo-like MWCNTs grown by T-CVD and PE-CVD in terms of their structural properties. 2. Horizontal dispersion of MWCNTs with and without surfactants, and the integration of MWCNTs to microelectrodes using deposition by dielectrophoresis (DEP) (Chapter 4). We have systematically studied the use of surfactant molecules to disperse and horizontally align MWCNTs on substrates. In addition, DEP is shown to produce impurityfree placement of MWCNTs, forming connections between microelectrodes. We demonstrate the deposition density is tunable by both AC field strength and AC field frequency. 3. Etching of MWCNTs for the impurity-free nanoelectrodes (Chapter 5). We show that the residual Ni catalyst on MWCNTs can be removed by acid etching; the tip removal and collapsing of tubes into pyramids enhances the stability of field emission from the tube arrays. The acid-etching process can be used to functionalize the MWCNTs, which was used to make our initial CNT-nanoelectrode glucose sensors. Finally, lessons learned trying to perform spectroscopic analysis of the functionalized MWCNTs were vital for designing our final devices. 4. Molecular junction design and electrochemical synthesis of biphenyl molecules on carbon microelectrodes for all-carbon molecular devices (Chapter 6). Utilizing the experience gained on the work done so far, our final device design is described. We demonstrate the capability of preparing patterned glassy carbon films to serve as the bottom electrode in the new geometry. However, the molecular switching behavior of biphenyl was not observed by scanning tunneling microscopy (STM), mercury drop or fabricated glassy carbon/biphenyl/MWCNT junctions. Either the density of these molecules is not optimum for effective integration of devices using MWCNTs as the nanoelectrodes, or an electroactive contaminant was reduced instead of the ionic biphenyl species. 5. Self-assembly of octadecanethiol (ODT) molecules on gold microelectrodes for functional molecular devices (Chapter 7). We have realized an effective scheme to produce Au/ODT/MWCNT junctions by spanning MWCNTs across ODT-functionalized microelectrodes. A percentage of the resulting junctions retain the expected character of an ODT monolayer. While the process is not yet optimized, our successful junctions show that molecular electronic devices can be fabricated using simple processes such as photolithography, self-assembled monolayers and dielectrophoresis.

The study is based on experimental work conducted in alpine snow. We made microwave radiometric and near-infrared reflectance measurements of snow slabs under different experimental conditions. We used an empirical relation to link near-infrared reflectance of snow to the specific surface area (SSA), and converted the SSA into the correlation length. From the measurements of snow radiances at 21 and 35 GHz , we derived the microwave scattering coefficient by inverting two coupled radiative transfer models (the sandwich and six-flux model). The correlation lengths found are in the same range as those determined in the literature using cold laboratory work. The technique shows great potential in the determination of the snow correlation length under field conditions.
