Este estudo objetivou analisar a prática docente relacionada à orientação sexual enquanto tema transversal em escolas públicas no munícipio de Curralinhos-PI, e neste encalço, identificar as formas de práticas docentes relacionadas à orientação sexual enquanto tema transversal; caracterizar os fatores condicionantes para a realização da abordagem da orientação sexual em sala de aula; verificar a contribuição das oficinas do Programa Saúde e Prevenção na Escola (SPE) na formação continuada da prática docente e, a percepção de professores e alunos, quanto à relevância da abordagem do tema em questão de forma sistemática e transversal nas referidas escolas. Trata-se de um estudo com abordagem qualitativa e quantitativa com função de complementaridade, realizada em duas escolas públicas (X e Y) do município de Curralinhos-PI, de ensino fundamental e ensino médio, respectivamente, envolvendo uma amostra de 8 (oito) professores, que participaram das oficinas do SPE e 116 alunos, sendo 54 da escola X e 62 da escola Y. Os instrumentos da coleta de dados foram a entrevista e o questionário semiestruturados, originários de um roteiro de questões-guia. A coleta de dados foi realizada no segundo semestre de 2011, nos meses de agosto a setembro. Os resultados apontaram que é grande a necessidade de se discutir e implementar a orientação sexual no espaço escolar, pois embora a maioria dos professores entrevistados reconheçam a importância de se trabalhar a orientação sexual como tema transversal e demonstrarem conhecer esta proposta dos Parâmetros Curriculares Nacionais (PCNs), ainda são difusas as práticas incorporadas em suas atividades pedagógicas. Quanto aos alunos, evidenciaram que ainda se fazem presentes dúvidas e angústias, assim como dificuldades para a lida com o tema da sexualidade.

Active queue management (AQM) policies are those policies of router queue management that allow for the detection of network congestion, the notification of such occurrences to the hosts on the network borders, and the adoption of a suitable control policy. This paper proposes the adoption of a fuzzy proportional integral (FPI) controller as an active queue manager for Internet routers. The analytical design of the proposed FPI controller is carried out in analogy with a proportional integral (PI) controller, which recently has been proposed for AQM. A genetic algorithm is proposed for tuning of the FPI controller parameters with respect to optimal disturbance rejection. In the paper the FPI controller design metodology is described and the results of the comparison with random early detection (RED), tail drop, and PI controller are presented.

The levels of health-related phytochemicals were determined in lettuce leaf and in strawberry, raspberry and blueberry fruits grown in near-commercial conditions under plastic films of three different UV transparencies. In the red lettuce Lollo Rosso, total phenolics, anthocyanin, luteolin and quercetin levels were all raised by changing from a UV blocking film to a film of low UV transparency, and to a film of high UV transparency. The related green lettuce, Lollo Biondo, cultivated under the same conditions, showed virtually no phytochemical responses to the same variation in UV levels. Overall, the phenolic levels of strawberries, raspberries, and blueberries were unresponsive to the UV transparency of the plastic film under which the crops were grown. The significance of these findings is discussed in relation to the nutritional quality of soft fruit and salad crops which are increasingly being grown commercially under plastic tunnels.

The phenolic compositions of fecal water samples from ten free-living human subjects without marked dietary restrictions were monitored before and after intake of raspberry puree (200 g/day, 4 days) using gas chromatography-mass spectrometry. No single phenolic component was increased in all subjects after intake, but a majority of subjects had significant elevations in phenylacetic acid (7/10), 4-hydroxyphenylacetic acid (6/10), 3-hydroxyphenylacetic acid (5/10), 3-phenylpropionic acid and 3-(4-hydroxyphenyl)propionic acid. The levels of 3,4-dihydroxbenzoic acid were elevated in 8/10 subjects, significantly for 6 subjects (p < 0.05), and not significantly reduced in the other 2 subjects. In addition, unlike most other fecal metabolites, the increase was always >2-fold. This metabolite may be representative of the increased colonic dose of cyanidin anthocyanins. The colonic microbiota varied greatly between individuals, and supplementation with raspberries did not produce any statistically significant alterations in the profile of colonic bacteria, nor was a common pattern revealed to account for the interindividual variations observed in the fecal water phenolic profiles.

The regulation of phosphoinositide (PI) 3-kinase activities has been linked to many normal and disease-related processes, including cell survival, cell growth and proliferation, cell differentiation, cell motility, and intracellular vesicle trafficking. However, as the family of enzymes has now grown to include eight true members, in three functional classes, plus several related protein kinases that are also inhibited by the widely used PI 3-kinase selective inhibitors, wortmannin and LY294002, extended methodologies are required to identify which type of kinase is involved in a particular cellular process, or protein complex, under study. A robust in vitro PI 3-kinase assay, suitable for use with immunoprecipitates, or purified proteins, is described here together with a series of modifications of substrate and assay conditions that will aid researchers in the identification of the particular class and isoform of PI 3-kinase that is involved in a signaling process under investigation.

Oxidised low density lipoprotein (LDL) may be involved in the pathogenesis of atherosclerosis. We have therefore investigated the mechanisms underlying the antioxidant/pro-oxidant behavior of dehydroascorbate, the oxidation product of ascorbic acid, toward LDL incubated With Cu2+ ions. By monitoring lipid peroxidation through the formation of conjugated dienes and lipid hydroperoxides, we show that the pro-oxidant activity of dehydroascorbate is critically dependent on the presence of lipid hydroperoxides, which accumulate during the early stages of oxidation. Using electron paramagnetic resonance spectroscopy, we show that dehydroascorbate amplifies the generation of alkoxyl radicals during the interaction of copper ions with the model alkyl hydroperoxide, tert-butylhydroperoxide. Under continuous-flow conditions, a prominent doublet signal was detected, which we attribute to both the erythroascorbate and ascorbate free radicals. On this basis, we propose that the pro-oxidant activity of dehydroascorbate toward LDL is due to its known spontaneous interconversion to erythroascorbate and ascorbate, which reduce Cu2+ to Cu+ and thereby promote the decomposition of lipid hydroperoxides. Various mechanisms, including copper chelation and Cu+ oxidation, are suggested to underlie the antioxidant behavior of dehydroascorbate in LDL that is essentially free of lipid hydroperoxides. Mature nonstructural protein-15 (nsp15) from the severe acute respiratory syndrome coronavirus (SARS-CoV) contains a novel uridylate-specific Mn2+-dependent endoribonuclease (NendoU). Structure studies of the full-length form of the obligate hexameric enzyme from two CoVs, SARS-CoV and murine hepatitis virus, and its monomeric homologue, XendoU from Xenopus laevis, combined with mutagenesis studies have implicated several residues in enzymatic activity and the N-terminal domain as the major determinant of hexamerization. However, the tight link between hexamerization and enzyme activity in NendoUs has remained an enigma. Here, we report the structure of a trimmed, monomeric form of SARS-CoV nsp15 (residues 28 to 335) determined to a resolution of 2.9 A. The catalytic loop (residues 234 to 249) with its two reactive histidines (His 234 and His 249) is dramatically flipped by approximately 120 degrees into the active site cleft. Furthermore, the catalytic nucleophile Lys 289 points in a diametrically opposite direction, a consequence of an outward displacement of the supporting loop (residues 276 to 295). In the full-length hexameric forms, these two loops are packed against each other and are stabilized by intimate intersubunit interactions. Our results support the hypothesis that absence of an adjacent monomer due to deletion of the hexamerization domain is the most likely cause for disruption of the active site, offering a structural basis for why only the hexameric form of this enzyme is active.

The effects of temperature, photosynthetic photon flux density (PPFD) and photoperiod on vegetative growth and flowering of the raspberry (Rubus idaeus L.) 'Autumn Bliss' were investigated. Increased temperature resulted in an increased rate of vegetative growth and a greater rate of progress to flowering. Optimum temperatures lay in the low to mid 20degreesC range. Above this the rate of plant development declined. Increased PPFD also advanced flowering. While photoperiod did not significantly affect the rate of vegetative growth, flowering occurred earliest at intermediate photoperiods and was delayed by extreme photoperiods. These responses suggest that there is potential for adjusting cropping times of raspberry grown under protection by manipulating the environment, especially temperature.
